Analysis and Stability Test of the Extracts from Epimedii Herba, Atractylodis Rhizoma Alba and Polygalae Radix for Toxicity Study (음양곽, 백출 및 원지의 독성평가를 위한 성분분석 및 안정성 시험)

  • A simple and reliable reverse phase HPLC method was developed to determine pharmacologically active marker compounds of Epimedii Herba, Atractylodis Rhizoma Alba, and Polygalae Radix. The stability test of water-extracts of the three natural medicines has been evaluated for six months. However, no significant changes in the content of the marker compounds of each extract were observed during the time of investigation.

  • As digital contents circulation environment goes on-line by fast development of the latest Internet, contents demand is growing rapidly. However, XrML hard to change dynamically and has controversial point that hard to reflect user's request maximum after create license and issues to user, in this paper study of active digital license administration model who can reflect user request dynamically and support reusability of license XrML eXrML model who design extension design and suggested. Expect that this presents efficient administration plan in administration study of license base as well as study for digital contents administration.

The Family Policy of the Lee administration from the Social Investment Perspective : contents analysis and policy implication (사회투자론적 관점에서 본 새 정부의 가정정책 : 분석과 과제)

  • The purpose of this study is to analyze and assess the family policy of the Lee administration and to suggest agendas for sustainable future family policy. Traditionally, families are a unique unit of production and reproduction of family members and social laborers. In addition, families are the social safety net help people survive in a society. Therefore, family policies play an important role in contemporary society. In this context, we review the literature related to family and social policy, and the contents of Ministry for Health, Welfare and Family Affairs, which is the main authority in Korea. The results are as follow: Firstly, the Lee administration ignored the data showing that family policy should be a unique policy, and not a sub-division of welfare policy. Secondly, there is no difference between the active welfare policy of the Lee administration and policies of past administrations. Finally, this study suggests that subjects of future oriented family policy should focus on co-developing programs for the individual, family, and society, to develop problem-prevention and family needs- ordered policy, and to make integrated family policy through laws and delivery systems such as Healthy Family Centers.

  • In terms of coastal and marine tourism, Busan's Haeundae had the nation's representativeness, and is pushing for many kinds of related policies to revitalize the special tourism zone. Due to a drop in the number of beach users, it is inevitable for the Haeundae Special Tourist Zone to face active responses to new trends emerging in the global tourism market. Therefore, the purpose of this study is to present a tourism strategy for making the Suyeong River as a more competitive ourist resource along the zone. First, a language network analysis is conducted through interviews to understand the ideas of interest groups for the river cruise activation project. Second, the frame structure of stakeholders is used to analyze solutions by comparing the similarities and differences in recognition frames of interest groups. Third, we intend to analyze the detailed frame types of stakeholders and present new alternatives based on the structure of relationships between types.

Screening and Characterization of an Enzyme with ${\beta}-Glucosidase$ Activity from Environmental DNA

  • A novel ${\beta}-glucosidase$ gene, bglA, was isolated from uncultured soil bacteria and characterized. Using genomic libraries constructed from soil DNA, a gene encoding a protein that hydrolyzes a fluorogenic analog of cellulose, 4-methylumbelliferyl ${\beta}-D-cellobioside$ (MUC), was isolated using a microtiter plate assay. The gene, bglA, was sequenced using a shotgun approach, and expressed in E. coli. The deduced 55-kDa amino acid sequence for bglA showed a 56% identity with the family 1 glycosyl hydrolase Chloroflexus aurantiacus. BglA included two conserved family 1 glycosyl hydrolase regions. When using $p-nitrophenyl-{\beta}-D-glucoside$ (pNPG) as the substrate, the maximum activity of the purified ${\beta}-glucosidase$ exhibited at pH 6.5 and $55^{\circ}C$, and was enhanced in the presence of $Mn^{2+}$. The $K_m\;and\;V_{max}$ values for the purified enzyme with pNPG were 0.16 mM and $19.10{\mu}mol/min$, respectively. The purified BglA enzyme hydrolyzed both pNPG and $p-nitrophenyl-{\beta}-D-fucoside$. The enzyme also exhibited substantial glycosyl hydrolase activities with natural glycosyl substrates, such as sophorose, cellobiose, cellotriose, cellotetraose, and cellopentaose, yet low hydrolytic activities with gentiobiose, salicin, and arbutin. Moreover, BglA was able to convert the major ginsenoside $Rb_1$ into the pharmaceutically active minor ginsenoside Rd within 24 h.

Determination of Dimethyl Disulfide, Diallyl Disulfide, and Diallyl Trisulfide in Biopesticides Containing Allium Sativum Extract by Gas Chromatography

  • BACKGROUND: Garlic (Allium sativum) extract has been allowed as commercial biopesticide material for pesticidal activity in the Environmentally-friendly Agriculture Promotion Act. Nine commercial biopesticides containing A. sativum extract have been marketed in Korea. However, the analytical method of the active substances in these materials has not been studied. METHODS AND RESULTS: Cartridge clean-up method for the determination of dimethyl disulfide(DMDS), diallyl disulfide(DADS), and diallyl trisulfide(DATS) in biopesticides containing A. sativum extract was developed and validated by gas chromatography(GC). The clean-up method was optimized using hydrophilic lipophilic balance (HLB) solid phase extraction(SPE) cartridges for the bioactive sulfides in biopesticides containing A. sativum extract, and the eluate was analyzed to quantify the DMDS, DADS, and DATS using the GC. The developed method was validated, and the LOQ and recovery rates of DMDS, DADS, and DATS were 0.226, 0.063, and $0.051mg\;L^{-1}$ and 80.6, 84.8, and 73.1%, respectively. From the nine commercial biopesticide samples, contents of DMDS, DADS, and DATS were analyzed using the developed method and results showed $2.3mg\;L^{-1}$, respectively. CONCLUSION: The developed method could be used in determining the quality of biopesticides for the manufacture of commercial biopesticides containing A. sativum extract.

Antigenicity of a Water Soluble Dimethyl Dimethoxy Biphenylate Derivative(DDB-S), a New Antihepatitis Agent (새로운 간염치료제인 수용성 DDB 유도체 (DDB-S)의 항원성 평가)

  • Dimethyl dimethoxy biphenylate (DDB) is an agent used to treat hepatits. DDB-S (DDB-soluble), a new DDB derivative, was synthsized to increase water solubility of the original DDB. In the present study, the antigenic potential of DDB-S was examined by active systemic anaphylaxis (ASA), passive cutaneous anaphylaxis (PCA) and passive hemagglutination (PHA) tests. The experimental groups consist of a low dosage group, a high dosage group, he group emulsified with Freund's complete adjuvant (FCA, ASA test) or an alum (PCA and PHA tests) and the macromolecule conjugate group emulsified with FCA or an alum. In the ASA test, all experimental groups showed negative responses whereas the positive control group given ovalbumin plus FCA showed severe anaphylactic responses. In the heterologous PCA test using mice and rats, positive responses were not detected in any of the experimental groups. In the PHA test, all experimental groups showed negative responses whereas the positive control group given ovalbumin plus an alum showed 512~2048 PHA titers. These results demonstrated that DDB-S does not have any antigenic potential. These can be utilized as a part of preclinical data for the development of DDB-S as an intravenous injection.

Migration patterns of old population in Korea (우리나라 노인인구의 지역간 이동 특성 - 16개 특.광역시도별 $2000{\sim}2006$년 비교를 중심으로 -)

  • Although the majority of elderly are residentially stable, many western studies have recently reported that there has been a gradual growth of retirement migration. However, little is known about that in Korea. Therefore, this study intended to examine the trend and nature of elderly migration. For this purpose, inter- and intra-province migration among the aged 65 and over were analyzed, using the migration data of the Korea National Statistical Office, compiled from the 2000 and 2006 resident registration records. The results were as follows. First, there was an increase of elderly migrants to 531 thousand persons in 2006 from 383 thousand persons in 2000. Secondly, the migration rate of elderly has steadily grown with a 11.7% in 2000 and 12% in 2006. Also, the move of female among old population was active but this trend tended to decrease in 2006. Thirdly, the number of provinces which showed a positive net inter-province migration of old population increased from 7 provinces in 2000 to 8 provinces in 2006. Fourthly, we found that the destinations of elderly migrants were mostly neighboring province except the Chungnam, Jeonbuk, and Jeju. These results suggested that elderly migration would be expected to increase gradually and be important in the future of Korea.

Chemical Characteristics and Efficacy of Combined Pesticide Granules Formulated by Different Ways (제제방법별(製劑方法別) 혼합입제(混合粒劑) 농약(農藥)의 특성(特性)과 약효(藥效))

  • A comparative study of chemical characteristics, efficacy and relative merits of extruded and coated granules, containing fungicide and insecticide, was conducted under laboratory and field conditions. Probenzole (3-allyloxy-1,2-benzisothiazole 1,1-dioxide) and carbofuran (2,3-dihydro-2,2-dimethyl-7-benzofuranyl methyl carbamate) were chosen as toxicants for rice blast(Pyricularia oryzae) and brown planthopper (Nilaparvata lugens) control, respectively. Stability of active ingredients in coated granule was superior to the extruded one under accelerated temperature. Active ingredient dissolution of coated one into distilled water showed slow release pattern. Pesticide residues in rice (Oryzae sativa, Chucheongbyeo) shoot applied with extruded one at the dosage of 3kg/10a retained higher levels than those with coated one on two days after application, while the residue levels were a reversed tendency on eight days after application. Efficacy on rice blast exhibited minute differences between the granules, on the other hand efficacy on brown planthopper by coated one was of higher rank than that by extruded one. Production cost of the combined pesticide granule by coating method could be cut down by 6% as compared to extrusion method.

Identification and characterization of the phytocystatin family from Brassica rapa

  • Phytocystatins, which are inhibitors of plant cysteine peptidases, are involved in the regulation of protein turnover and in the defense against insect pests and pathogens. Extensive searches in the Brassica rapa genome allowed the prediction of at least eight different phytocystatin genes on seven chromosomes in the B. rapa genome. Structure comparisons based on alignments of the all BrCYS ($\underline{B}$. $\underline{r}apa$ $phyto{\underline{cys}}tatin$) proteins using the CLUSTALW program revealed conservation of the three consensus motifs known to interact with the active site of cysteine peptidases. According to the phylogenetic analysis based on the deduced amino acid sequences, the eight BrCYS proteins were divided into several clusters related to the orthologous phytocystatin. The predicted three-dimensional structure models of the eight BrCYS proteins demonstrate that all of these proteins are similar to the reported crystal structure of oryzacystatin-I (OC-I). Digital northern and RT-PCR analyses indicated that the eight BrCYS genes exhibit different expression patterns in B. rapa tissues and respond differently to abiotic stimuli. The differences in gene structure and expression between the eight BrCYS genes suggest that these proteins may play diverse physiological roles in B. rapa and may interact with cysteine peptidases through different mechanisms.
