We investigated the effect of cytosolic and extracellular $Ca^{2+}$ on $Ca^{2+}$ signals in pancreatic acinar cells by measuring $Ca^{2+}$ concentration in the cytosol($[Ca^{2+}]_c$) and in the lumen of the ER($[Ca^{2+}]_{Lu}$). To control buffers and dye in the cytosol, a patch-clamp microelectrode was employed. Acetylcholine released $Ca^{2+}$ mainly from the basolateral ER-rich part of the cell. The rate of $Ca^{2+}$ release from the ER was highly sensitive to the buffering of $[Ca^{2+}]_c$ whereas ER $Ca^{2+}$ refilling was enhanced by supplying free $Ca^{2+}$ to the cytosol with $[Ca^{2+}]_c$ clamped at resting levels with a patch pipette containing 10 mM BAPTA and 2 mM $Ca^{2+}$. Elevation of extracellular $Ca^{2+}$ to 10 mM from 1 mM raised resting $[Ca^{2+}]_c$ slightly and often generated $[Ca^{2+}]_c$ oscillations in single or clustered cells. Although pancreatic acinar cells are reported to have extracellular $Ca^{2+}$-sensing receptors linked to phospholipase C that mobilize $Ca^{2+}$ from the ER, exposure of cells to 10 mM $Ca^{2+}$ did not decrease $[Ca^{2+}]_{Lu}$ but rather raised it. From these findings we conclude that 1) ER $Ca^{2+}$ release is strictly regulated by feedback inhibition of $[Ca^{2+}]_c$, 2) ER $Ca^{2+}$ refilling is determined by the rate of $Ca^{2+}$ influx and occurs mainly in the tiny subplasmalemmal spaces, 3) extracellular $Ca^{2+}$-induced $[Ca^{2+}]_c$ oscillations appear to be triggered not by activation of extracellular $Ca^{2+}$-sensing receptors but by the ER sensitised by elevated $[Ca^{2+}]_c$ and $[Ca^{2+}]_{Lu}$.
The effects of the hormonal factor (CCK) a and neural factor(carbachol) on the exocrine function of the pancreas were studied in th is experiment. A superfusion technique was used for in vitro study of stimulus-secreti- o on coupling in isolated pancreatic acinarce 11s frQm the rats fed heated or raw soybean diet. Chymotrypsin secretion was higher in cells from the raw soybean group than in those from the heated soybean group with both kinds of stimulants(CCK and carbachol), whereas, amylase secretion was higher inthe h heated soybean group than in the raw soyb bean group. This indicated that chymotrvpsin a and amylase secretion from the acinar cells are not parallel with CCK and carbachol sti imulation.

The ultrastructural investigations of the parotid gland of Korean native goat were carried out by transmission electron microscopy. The results were as follows; 1. The acini of parotid gland were composed of light and dark acinar cells. 2. In the light acinar cells, the secretory granules were classified into three types according to their electron densities and dense bodies. One type of granules was low electron density and had no dense bodies. Another type was low electron density and had dense bodies, and the other type was low electron density and had granular dense bodies. 3. The secretory granules of dark acinar cells showed high electron density and were also calssified into three types by dense bodies as the same way as in the light acinar cells. 4. The intercalated ducts consisted of simple cuboidal epithelium. The nuclei of epithelial cells were oval or round form, located at the central part, and had infolding nuclear membranes and one or two nucleoli. 5. The cells of both of the striated and excretory ducts were composed of light cells, dark cells, specific light cells and basal cells. 6. The nerve terminals were distinguished into two types. One had large granular synaptic vesicles, and another had small agranular synaptic vesicles.
The role of $Cl^-$ channels in regulatory volume decrease (RVD) in human salivary gland acinar cells was examined using a whole-cell patch clamp technique. Human tissues were obtained from healthy volunteers or from patients with oromaxillofacial tumors. During the measurements, $K^+$-free solutions were employed to eliminate contamination of whole-cell conductance by $K^+$ currents. When the cells were exposed to a 70% hypotonic solution, outward-rectifying currents, which were not observed in the resting state, were found to have significantly increased both in human labial and parotid gland acinar cells. The amplitudes of the currents were reduced in a low $Cl^-$ bath solution. Furthermore, the addition of $100{\mu}M$ 5-Nitro-2- (3-phenyl propylamino) benzoic acid (NPPB) or $100{\mu}M$ 4,4'-diisothio cyanatostilbene-2,2'-disulphonic acid (DIDS), known to partially block $Cl^-$ channels, significantly inhibited these currents. Its outward-rectifying current profile, shift in reversal potential in a low $Cl^-$ bath solution and pharmacological properties suggest that this is a $Ca^{2+}$-independent, volume activated $Cl^-$ current. We conclude therefore that volume activated $Cl^-$ channels play a putative role in RVD in human salivary gland acinar cells.

The ultrastructure of the secretory granules of the submandibular gland was examined in the Korean spider shrew, Sorex caecutiens, lesser white-toothed shrew, Crocidura suaveolens and big white-toothed shrew, C. lasiura. The mucous and serous acinar granules of S. caecutiens with a border of the lucid corona differed from those of C. suaveolens and C. lasiurar with a dense lateral border. The mucous acinar granules of C. lasiura with several bands producing a variety of patterns in the matrix were similar to those of C. suaveolens. The serous acinar granules of C. lasiura had a homogenous pale center surrounded by minute dense specks or had an unusual substructure showing a geometric pattern according to the inclusions in the pale matrix of the granules. This is the first report of an unusual substructure showing a geometric pattern of the serous acinar granules of C. lasiura. The myelin-like body was observed in the granular duct cell of the three species of shrew. The myelin-like body of S. caecutiens with layers of unit membranes was different from that of C. suaveolens and C. lasiura with paired membranes. Therefore, the layers composing of the paired membranes of the myelin-like body might be one of the characteristics of Crocidura.

Background : The different features of high-resolution CT(HRCT) findings of active pulmonary tuberculosis(TB) were studied between acid fast bacilli(AFB) smear or culture positive and negative group. Methods : We prospectively evaluated 36 patients who had been confirmed for active pulmonary tuberculosis by the smear or culture of AFB in sputum(n=25), and changes on serial chest radiographs(n=11). The patients were divided into 3 groups by the results of sputum AFB stain and culture. Group 1(n= 11) is negative in both AFB stain and culture; group 2(n=13) is negative in AFB stain but positive in culture ; and group 3(n=12) is positive in both AFB stain and culture. We evaluated the findings of HRCT in each group randomly. Result : On the HRCT scans, acinar nodule(100%), macronodule(75%), and cavity(75%) in group 3 were more frequently found than group 1(63%. 18%, 9%) and group 2(46%, 15%, 23%)(p<0.05). The centrilobular nodule and branching structure were more frequently observed in group 3(92%) than in group 1(54%)(p<0.05), but were similarly observed in group 2(77%)(p>0.05). AFB positive group was statistically different than the negative group in the HRCT findings with to acinar nodule(100% vs 54%), macronodule(75% vs 17%), and cavity(75% vs 17%)(p<0.05). TB culture positive group was statistically different than the negative group in the HRCT findings with respect to acinar nodule(72% vs 45%) and cavity(48% vs 9%)(p<0.05). Conclusions : HRCT scans are helpful in determining disease acitivity in sputum AFB stain-negative pulmonary tuberculosis. When HRCT shows centrilobular nodule and branching structure, acinar nodule, macronodule, cavity, further studies as sputum induction and bronchoscopy can be performed to determine the presence of bacilli in patients of AFB stain-negative tuberculosis.
The effects of chromium picolinate supplementation in pig diet were evaluated by measuring the in vitro lipogenic and lipolytic activities in adipose tissue and the protein synthetic activity in liver acinar cell in culture. Thirty-two male and thirty-two female pigs were randomly assigned to one of four dietary groups: Control, 100 ppb, 200 ppb, and 400 ppb of Cr in the form of picolinate. The chromium picolinate supplementation (p < 0.01) increased the in vitro lipolytic activity in adipose tissue of pig, but had no effects on lipogenesis. The chromium picolinate effect was greater in female pigs than in male pigs on lipolytic activity. The results from the studies with the liver acinar cells in culture indicated that chromium picolinate supplementation increased protein synthetic activity (p < 0.05). It was observed through this experiment that chromium picolinate functions not only on fat degradation but also on retained protein synthesis.
In order to know the histological changes of rat preputial gland during normal sexual cycle and sex hormone treatment, matured Wistar rats (B.W.about 200g) were used for the experiment. Rats were subcutaneously given $2{\mu}g$ 17-${\beta}$-estradiol (Sigma) and 2.5g progesterone (Nakarai Chem., Japan) daily in 0.5ml propylen glycol for ten days respectively. The results obtained are as follow: At the stage of estrus and metestrus, the eosinophilic crystalloid granules and large vacuoles in the acinar cells appeared numerously, and the excretory ducts were severely extended. The developed connective tissue between the acinus were also found. At the stage of proestrus and diestrus, however, the small vacuoles and a few eosinophilic crystalloid granules appeared in the acinar cell. In the estradiol treatment, on the other hand, severely extended excretory ducts and a small number of the eosinophilic crystalloid granules compared with the progesterone treatment were found. The cyst-like structure was found, and in that the acinus disappeared completely. In the progesterone treatment, remarkably extended excretory ducts and nomerous appearance of oval formed eosinophilic granules in the acinar cells were found. As these findings, it could be suggested that the secretion of rat preputial gland was active at the estrus and metestrus stage, and estrogen may concerned in the secretion, land progesterone in the formation of secretory products.

The purpose of this study was to investigate the irradiation effects on the secretory granules of the rat submandibular gland acinar cell. For this study, Sprague-Dawley strain rats were irradiated to their neck region with the dose of 5Gy and l0Gy by 6MV X-radiation, and sacrificed on the experimental periods after irradiation. The authors observed the ultrastructural changes of the secretory granules of the rat submandibular gland acinar cell under a trasmission electron microscope. The results were as follows: In the transmission electron microscopic examination, secretory granules were blurred in limiting membrane on the 3hours groups after irradiation. And they showed decrease in number, irregularities in shape and distributional pattern, and inhomogeneous internal electron density on the Iday and 3days groups. After then, these changes were recovered in shape and distributional pattern on the 14days groups, and changes of internal electron density and limiting membrane were recovered on the 28days groups after irradiation. Among the intracellular organelles, rough endoplasmic reticulum was scattered, but golgi complex was not changed. And such pathologic changes were earlier and more prominent in 10Gy irradiated groups than in 5Gy irradiated groups.
