Purification and Characterization of Complement-activating Acidic Polysaccharides from the Fruits of Capsicum annuum

  • Hot water-soluble crude polysaccharide (HCAP-0) that was obtained from the fruits of Capsicum annuum showed potent anti-complementary activity. The activity was unchanged by pronase digestion, but decreased by periodate oxidation. The HCAP-0 was fractionated by DEAE ion-exchange chromatography to give two major fractions, HCAP-II and III. These two fractions were finally purified by gel filtration to give HCAP-IIa, HCAP-IIIa1, and IIIa2 fractions that had high anti-complementary activities. The HCAP-IIIa1 and IIIa2 consisted of homogeneous polysaccharides. The anti-complementary activities were unaffected by treatment with polymyxin B, indicating that the modes of complement activation were not due to preexisting lipopolysaccharide. The molecular weight and sugar content of HCAP-IIIa2 had potent anti-complementary activity. The highest yields were 55 kDa and 75.9%, and the molar ratio of galactose (Ara:Gal, 1.0:4.6) was higher than other sugars. The crossed immuno-electrophoresis showed that both classical and alternative pathways were activated by HCAP-IIIa2.

  • 본 논문은 홍삼 산성 다당류 (RGAP)의 생체외 쥣과 NK 세포 활성 향상에 대한 내용으로 주요 내용은 다음과 같다. RGAP는 Yac-1 종양 세포에서 NK 세포 활성을 향상시켰다. 비장 세포에 16시간 동안 RGAP 10 및 $100{\mu}g$/ml을 처리한 결과, NK 활성이 239와 250% 증가 하였다. 인터페론 (IFN)-$\gamma$ 생성은 17-125% 증가하였고, 종양 괴사 인자 (TNF)-${\alpha}$ 생성도 15-100% 증가 하였다. NK 세포의 세포 독성 증가는 NK 세포가 생성하는 이 두 시토카인 증가가 원인이다. 따라서 RGAP는 NK 세포에 미치는 면역증진 효과가 있다는 내용이다.

Antitumor Activities of Red Ginseng Acidic Polysaccharide(RGAP) as an Immunomodulator

  • A red ginseng acidic polysaccharide(RGAP) with immunomodulating antitumor activities was isolated from Korean red ginseng, The molecular weight of RGAP was estimated to be 12-450 kDa by gel filtration chromatography, RGAP was found to increase survival rate and to inhibit of tumor growth significantly in a dose dependent manner in mice transplanted with tumor cells. RGAP significantly promoted nitric oxide(NO) production from peritoneal macrophages bothin vivo and in vitro. Western blot analysis exhibited a newly synthesized inducible nitric oxide synthase(iNOS) protein band in the RGAP treated group. It seems likely that immunomodulating antitumor activities of RGAP are mainly mediated by NO production of macrophage. RGAP was further purified by ultrafiltration and anion exchange chromatography on DEAE-sepharose, followed by gel filtration on Sephacryl S-300 to give an active fraction(GFP) with stronger NO production in murine macrophages. GFP increased survival rate ten times compared to RGAP in male ICR mice transplanted with sarcoma 180 and also showed more potent tumoricidal activities of natural killer cells than RGAP. Sugar $composition(mol\%)$ of GFP was found to be arabinose:rhamnose:xylose:galacturonic acid:mannose:galactose:glucose(10:9:1:25:8:20:27) by GC/MS. The results suggest that clinical trials of RGAP in immunotherapy against cancer are highly feasible.

Novel analysis procedure for red ginseng polysaccharides by matrix-assisted laser desorption/ionization time-of-flight/time-offlight mass spectrometry

  • Background: Red ginseng polysaccharides (RGPs) have been acknowledged for their outstanding immunomodulation and anti-tumor activities. However, their studies are still limited by the complexity of their structural features, the absence of purification and enrichment methods, and the rarity of the analytical instruments that apply to the analysis of such macromolecules. Thus, this study is an attempt to establish a new mass spectrometry (MS)-based analysis procedure for RGPs. Methods: Saponin pre-excluded powder of RG (RG-SPEP, 10 mg) was treated with 200 µL of distilled water and centrifuged for 5 h at 1000 rpm and 85 ℃. Ethanol-based precipitation and centrifugation were applied to obtain RGPs from the heated extracts. Further, endo-carbohydrase treatments were performed to produce specific saccharide fragments. Solid-phase extraction (SPE) processes were implemented to purify and enrich the enzyme-treated RGPs, while matrix-assisted laser desorption/ionization time-of-flight/time-of-flight (MALDI-TOF/TOF) MS was employed for the partial structural analysis of the obtained RGPs. Results: Utilizing cellulase, porous graphitized carbon (PGC), hydrophilic interaction chromatography (HILIC), and MALDI-TOF/TOF MS, the neutral and acidic RGPs were qualitatively analyzed. Hexn and Hexn-18 (cellulose analogs) were determined to be novel neutral RGPs. Additionally, the [Unknown + Hexn] species were also determined as new acidic RGPs. Furthermore, HexAn (H) was determined as another form of the acidic RGPs. Conclusion: Compared to the previous methods of analysis, these unprecedented applications of HILIC-SPE and MALDI-TOF/TOF MS to analyze RGPs proved to be fairly effective for fractionating and detecting neutral and acidic components. This new procedure exhibits great potential as a specific tool for searching and determining various polysaccharides in many herbal medicines.

20년 이상 장기저장된 홍삼의 이화학적 특성변화 및 품질안정성 (The Changes of Physicochemical Characteristics and Quality Stability of Korean Red Ginseng (Panax ginseng C.A. Meyer) Stored over 20 Years)

  • This study was investigated the changes of quality stability and physicochemical characteristics of the Korean red ginsengs stored for a long times over 20 years. The Korean red ginsengs were stored for 4 to 22 years in canned packaging with polypropylene film and wooden box at room temperatures. The unusal phenomena such as discoloration and pin hole in packaging were not observed. General bacteria showed the vlaues of below 100 CFU/g, coliform groups and molds were not found in any samples stored for 22 year. Any samples also were not detected in mycotoxins. The contents of moisture, ash and crude saponin were the levels of 10.6~11.1%, 3.8~4.2% and 4.1~4.7% during the whole storage periods, respectively. The contents of maltol, which has been known as characteristic flavour and antioxidant of Korean red ginseng, showed remarkably increasing tendency from 0.10 mg/g for 4 years to 2.53 mg/g for 22 years during the storage. The contents of AFG (arginyl-fructosyl-glucose), arginine and free sugar were slightly decreased. Acidic polysaccharide and ginsenoside were not changed significantly during the storage periods. The contents of acidic polysaccharide and total ginsenosides were the 75.1~76.3 mg/g and 15.1~16.6 mg/g, respectively. The sums of ginsenoside-Rg1,-Rb1 and -Rg3s were the ranges of 9.3~9.9 mg/g and PD (ginsenoside-Rb1, -Rb2,-Rc,-Rd,-Rg3s,-Rg3r)/PT (ginsenoside-Rg1,-Rg2,-Re,-Rf,-Rh1) saponin ratios were the levels of 1.4~1.5. These results suggest that Korean red ginsengs stored for long periods show relatively stable quaility stabilities and not significantly changed the contents of ginsenoside and polysaccharide during the storage up to 22 years.

Rheological Properties of a Novel High Viscosity Polysaccharide, A49-Pol, Produced by Bacillus polymyxa

  • An exopolysaccharide, designated as A49-Pol, was produced from Bacillus polymyxa KCTC 8648P in nitrogen sufficient conditions. The viscosity of the culture broth increased up to $2\times10^4$cP in 38 hours of culture and then decreased to $1.5\times10^4$CP at 48 hours. The $1.0\%$ (w/v) solution of purified A49-Pol represented pseudoplasticity with a viscosity of $2.7\times10^4$cP which was two times higher than xanthan at the same concentration. The viscosity of the A49-Pol solution was also greatly affected by its concentration in comparison with the xanthan solution. The viscosity of $1.0\%$ A49-Pol solution was 930-fold higher than its $0.2\%$ solution, whereas the corresponding viscosities of xanthan solution experienced only l7-fold difference. The viscosity was observed to be maximum at pH 7.0 in both A49-Pol and xanthan solutions, and gradually decreased as the pHs of the polysaccharide solutions went to acidic or alkaline regions. The viscosity of A49-Pol solution was very sensitive to temperature compared to xanthan and decreased with increasing temperature. The viscosity of $0.6\%$ solution of A49-Pol was 8,100 cP at $10^{\circ}C$ and 55 cP at $50^{\circ}C$. The viscosity was also affected by the presence of surfactants such as Span 20 and Triton X-l00 ; with $0.5\%$ Triton X-l00 (v/v), the viscosity of A49-Pol solution increased by $50\%$.

압출성형 산삼배양근의 이화학적 성질 및 침출특성의 비교 (Comparison of Physicochemical Properties and Release Characteristics of Extruded Tissue Cultured Mountain Ginseng)

  • 본 연구에서는 조직배양에 의해 생산된 산삼배양근을 홍삼화하여 부가가치를 향상시키고 새로운 식품소재의 개발을 목적으로 산삼배양근을 압출성형 하였다. 압출성형 산삼 배양근의 일반성분, 조사포닌, 말톨, 산성다당체, 페놀성화합물 등의 화학적 성분과 갈색도, 적색도, 침출특성의 물리적 변화를 살펴보았다. 실험결과 주근의 함량이 높은 홍삼이 총당과 환원당 함량은 산삼배양근과 비교하여 높게 나타났으며, 압출성형 산삼배양근의 총당과 환원당은 증가하였다. 반면 미세근으로 이루어진 산삼배양근의 총아미노산 함량이 주근으로 이루어진 홍삼과 비교하여 높게 측정되었으며, 압출성형 산삼배양근의 아미노산 함량은 다소 감소하는 경향을 나타내었다. 유효성분인 조사포닌 함량은 산삼배양근을 압출성형을 통하여 증가시킬 수 있음을 확인하였으며, 배럴온도 $120^{\circ}C$에서 압출성형한 산삼배양근이 9.60% 가장 높게 측정되었다. 말톨은 홍삼에서만 확인되었으며, 산삼배양근의 경우 압출성형을 통해서도 말톨의 생성은 나타나지 않았다. 산성다당체는 산삼배양근($0.24\;{\mu}g/g$)이 가장 낮았지만 압출성형을 하였을 때 배럴온도 $120^{\circ}C$에서 $0.75\;{\mu}g/g$으로 홍삼의 $0.83\;{\mu}g/g$과 비슷한 수준까지 증가시킬 수 있었다. 페놀성화합물 함량은 압출성형을 통해 감소하였다. 산삼배양근의 갈색도와 적색도는 압출성형으로 증가하였으며, 배럴온도가 증가함에 따라 증가하였다. 갈색도를 기준으로 한 침출특성과 침출속도상수 역시 배럴온도가 증가함에 따라 증가를 확인할 수 있었다. 압출성형은 유효성분인 조사포닌 및 산성다당체 등을 향상시킬 뿐만 아니라 침출력을 향상시켜주므로 향후 고가의 산삼배양근을 더욱 효율적으로 가공할 수 있는 공정으로 확인되었다.

발효 조건이 막걸리 술덧 유래 다당의 면역자극 활성에 미치는 효과 (Effect of Crude Polysaccharide Isolated from Mageoli Mash under Different Fermentation Conditions on Immune-Stimulating Activity)

  • 본 연구에서는 가장 대중적인 한국의 전통주 막걸리의 제조과정이 기능성 분획에 미치는 영향을 확인하고자 하였다. Saccharomyces cerevisae 균주별, 발효온도별, 발효기간별로 다르게 만든 막걸리로부터 ethanol 침전법을 이용해 crude polysaccharides(CP-M)를 분리하여 peritoneal macrophage 활성화 효과와 세포 배양액 중의 interleukin(IL)-6와 -12 농도에 미치는 영향을 조사하였다. S. cerevisae 89-1-1, 98-2, 268-3, 113-4의 4가지 효모 중 S. cerevisae 113-4로 발효한 CP-M을 peritoneal macrophage에 $10{\mu}g/mL$ 처리했을 때 세포 배양액 중 nitric oxide(NO) 농도 $33.3{\mu}M$로 가장 높았으며, IL-6와 -12의 농도 역시 116.3 pg/mL와 59.8 pg/mL로 가장 높게 나타났다. S. cerevisae 113-4를 접종하여 온도별로 발효한 발효물에서 분리한 CP-M 중 $25^{\circ}C$에서 발효 후 얻은 CP-M을 peritoneal macrophage 세포에 처리했을 때, NO 농도가 Control 대비 2.7~3.3배, IL-6 농도는 5.7배까지 증가하였다. 그러나 15, 20, $30^{\circ}C$에서 발효하여 얻은 CP-M의 경우 peritoneal macrophage의 NO와 IL-6 생성량에는 Control(무처리구)과 비교하여 영향이 없었다. 발효기간별 영향을 관찰한 결과 5일째 술덧에서 분리한 CP-M에 의한 NO 생성량이 가장 높아 0일째 MCP에 비해 2.2배 증가하였다. 그러나 급격하게 IL-6와 -12의 농도가 감소한 10일째 MCP를 제외하고는 발효기간에 따른 IL-6와 -12의 유의적인 농도 차이는 없었다. 이상의 결과에서 가장 활성이 좋은 발효 조건은 S. cerevisae 113-4를 접종하여 $25^{\circ}C$에서 5일간 발효시킨 술덧의 CP-M이었다. 이때 CP-M의 조성은 neutral sugar 함량 78.6%, acidic polysaccharide 11.6%, 단백질 9.8%였다. 특히 neutral sugar의 구성당 비율은 mannose 47.8 mole%, glucose 29.6 mole%, galactose 12.7 mole%였다. 이상의 결과에서 면역증강 활성이 있는 기능성 조다당 CP-M은 mannose 함량이 높은 yeast 유래의 extracellular polysaccharide로 추정하였다.

아가리쿠스 버섯에서 생리활성물질의 추출 및 정제 (Extraction and Purification of Bioactive Materials from Agaricus blazei Fruiting Bodies)

  • ${\beta}$-Glucan a kind of polysaccharide which is particularly abundant in Agaricus blazei is known as the bioactive materials especially anticancer agents. The process development of the isolation and the purification process of water soluble ${\beta}$-glucans from A. blazei was achieved. and the process operation variables were optimized. Crude polysaccharides (CR.PS) were obtained from A. blazei by hot water extraction filtration solvent precipitation dialysis and freeze drying. Neutral and acidic fraction of polysaccharides were separated from crude polysaccharides by ion chromatography and then high molecular weight and low molecular weight fraction were separated from neutral fraction by gel chromatography. Quantitative and qualitative analysis of each compounds were performed with FT-IR NMR spectroscopy. Based on these analysis the optimal conditions of temperatures operating time organic solvent volume for precipitation and dialysis time were determined.

다당류를 생산하는 Bacillus polymyxa KS-1의 분리 및 생산 다당류의 특성 (Isolation of Exopolysaccharide-Producing Bacillus polymyxa KS-1 and Some Properties of Exopolysaccharide)

  • 미생물에 의해 체외로 분비하는 다당류생산 균주를 토양으로부터 분리하여 이 균주에 대한 형태학, 생리학적 성질과 화학적 동정방법을 이용하여 동정하였으며 이 균주에 의해 생성된 다당류의 물리화학적 성질을 조사하였다. 분리된 균주는 Bacillus sp.중에 Bacillus polymyxa와 아주 유사한 형태와 특성을 나타내어 Bacillus polymyxa KS-1으로 동정하였으며 생산된 다당류의 용매에 대한 용해도, 정색반응 및 적외선 흡수 spectrum을 조사한 결과 물, formic acid, formaldehyde에는 잘 용해되었으며, 다당류내 pentose, ketose 그리고 전분을 함유되지 않았으며 uronic acid가 함유되지 않음을 알 수 있었다.
