토우슬과 회우슬의 열수 추출물의 성분 및 생리활성 비교 (A Comparison of Components and Biological Activities Between the Hot Water Extracts of Achyranthes japonica Nakai and Achyranthes bidentata Blume)

  • 식용 및 약용으로 사용 가능한 우슬은 국가마다 다른 기원식물을 가지고 있으며, 국내에서는 토우슬 및 회우슬을 사용하도록 규정하고 있다. 현재까지 다양한 우슬에 대한 많은 연구가 이루어져 왔으나, 식용으로 이용되고 있는 토우슬과 회우슬의 열수 추출물에 대한 비교연구는 전무한 상태이다. 본 연구에서는 기능성 우슬차 및 우슬식혜 제조를 목표로, 토우슬과 회우슬의 열수 추출물과 이들의 순차적 분획물을 조제하여 각각의 유용성분 및 in-vitro 항균활성, 항산화, 및 항당뇨 활성을 비교 평가하였다. 그 결과, 회우슬의 열수 추출물은 토우슬보다 수용성 당류 및 다양한 폴리페놀 성분을 다량 포함하고 있으며, 토우슬보다 높은 명도와 낮은 황색도를 나타내었다. 항세균 활성 평가에서는 토우슬의 ethylacetate 분획과 회우슬의 n-hexane 분획에서 우수한 활성이 나타났으며, 활성 음이온, 활성 양이온 및 nitrite 소거능 및 환원력 측정의 항산화 활성 평가결과, 토우슬 추출물이 회우슬보다 우수하며, 특히 토우슬의 ethylacetate 분획에서 강력한 항산화 활성을 확인하였다. 또한 in-vitro 항당뇨 활성평가 결과, 토우슬 및 회우슬의 ethylacetate 분획에서 우수한 ${\alpha}$-amylase 저해활성을, 각각의 butanol 분획에서 ${\alpha}$-glucosidase 저해활성을 확인하였다. 본 연구결과는 토우슬과 회우슬의 열수 추출물들은 추출효율과 성분에서 많은 차이가 있으며, 특히 이들의 분획물의 경우 성분 및 활성이 더욱 차이가 나타남을 제시하고 있다. 따라서, 우슬의 항균, 항산화 및 항당뇨 활성을 이용한 천연물 신약 개발시에는 토우슬의 ethylacetate 분획을 이용하는 것이 적합하며, 우슬차 및 우슬식혜 제조시에는 토우슬의 열수 추출물이 적합함을 확인하였다.

식물의 Ecdysteroids 함량에 미치는 광과 식물생장조절제의 영향 (Effects of Light and Some Plant Growth Regulators on Ecdysteroids Contents in Polypodium vulgare L. and Achyranthes japonica Nakai)

  • 미역고사리(Polypodium vutgare L.)와 쇠무릎(Achyranthes japonica Nakai)에 있어서 광과 methyl jasmonate(MJ), 6-benzylaminopurine(BA), thidiazuron(TDZ), 2,4-dichlorophenoxyacetic acid(2,4-D)가 식물체 중의 ecdysteroids (${\beta}$-ecdysone+polypodine B) 함량에 미치는 영향을 조사하였다. 수광량을 조절하여 재배한 미역고사리의 잎과 근경 모두 상대광도가 높을수록 ecdysteroids 함량이 감소하였고 쇠무릎 유묘의 배양시 광 조건일 때가 암 조건일 때 보다 ecdysteroids함량이 낮아 식물의 ecdysteroids 함량은 광에 의해 저하되는 것으로 나타났다. 미역고사리 전엽체와 쇠무릎 유묘의 배양계에서 식물체 중의 ecdysteroids 함량은 배지 중의 MJ와 BA 그리고 TDZ의 농도가 높아짐에 따라 증가된 반면 2,4-D의 농도와는 무관하게 일정한 수준으로 유지되어,식물의 ecdysteroids 함량은 MJ와 cytokinin처리에 의해 증가되지만 auxin처리에 의해서는 영향을 받지 않는 것으로 나타났다.

꾸지뽕나무 잎과 우슬 복합물의 골 질환 개선 소재 개발가능성에 대한 연구 (Study on the Potential of Development of Materials for Bone Disease Improvement of Cudrania tricuspidata Leaf and Achyranthes japonica Nakai Complex)

  • This study was conducted to suggest the Cudrania tricuspidata leaf and Achyranthes japonica Nakai Complex (CAC) possibility of use as a functional natural material for improving bone disease. Cudrania tricuspidata leaf and Achyranthes japonica Nakai were mixed in the same amount, extracted with hot water, and then powdered and used in the study. After, the cytotoxicity of CAC for osteoblasts (MG63 cell), osteoclasts (differentiated RAW264.7 cell), and macrophages (RAW264.7 cell) were evaluated by MTT assay, and ALP assay and TRAP assay were performed to confirm the differentiation capacity of osteoblasts and osteoclasts, respectively. In addition, the anti-inflammatory effect in macrophages was evaluated by ELISA, qRT-PCR, and western blot assay. CAC did not proliferated osteoblasts and osteoclasts, but increased ALP activity against osteoblasts differentiation and decreased TRAP activity against osteoclasts differentiation. CAC did not proliferated macrophages but decreased nitric oxide production. Also, decreased NOS2, IL1B, IL6, PTGS2, and TNFA gene expression, and JNK and p38 protein phosphorylation in a concentration-dependent manner, but ERK protein phosphorylation was not changed. As a result, CAC increased the differentiation and activation of osteoblasts, inhibited the differentiation and activation of osteoclasts, and regulated the expression of inflammatory cytokines in macrophages. Therefore, it is thought that CAC can be used as a functional natural material that prevents bone disease and has an anti-inflammatory effect.

Synergistic Antimicrobial Effect of Achyranthes japonica Nakai Extracts and Bifidobacterium Supernatants Against Clostridium difficile

  • The synergistic antimicrobial effect of Achyranthes japonica Nakai (AJN) and Bifidobacterium extracellular factors against Clostridium difficile were measured using a turbidity method. Each broth supernatant of Bifidobacterium infantis ($68.8{\pm}0.02%$) and Bifidobacterium adolescentis ($33.2{\pm}0.2%$) obtained by adding ethyl acetate soluble fractionate from A. japonica Nakai ethanolic extracts (AJNEA, 100 ppm, no inhibition) showed high synergistic antimicrobial activity against C. difficile. In addition, the antimicrobial activity in a laboratory medium and yogurt products against C. difficile were evaluated. In yogurt prepared with a starter 5 (Lactobacillus acidophilus: Streptococcus thermophilus: B. adolescentis =1 : 1 : 1) and a starter 4 (L. acidophilus: S. thermophilus: B. infantis=1 : 1 : 1) and 0.5% AJNEA powder, high antimicrobial effects were recorded that measured 79.0 and 65.2%, respectively. The results indicated the potential of AJN extract for use as an antimicrobial agent. In addition, the efficiency of the antimicrobial activity of the extracts was further improved in combination with lactic acid bacteria, which suggests that they have the potential to be used as a highly effective antibiotic-tolerant microorganism prevention system. Such a strategy can be used for alternative drugs or functional food additives for treatment of antibiotic-associated diarrhea.

Effect of fermented Achyranthes japonica (Miq.) Nakai extract on osteoarthritis

  • The present study was conducted to evaluate the anti-inflammatory and anti-arthritic effects of fermented Achyranthes japonica (Miq.) Nakai extract (FAJE). The FAJE was effective in nitrogen oxide (NO) scavenging in RAW264.7 cells. In the case of experimental Sprague Dawley (SD) rats injected with monosodium iodoacetate (MIA), the levels of $TNF-{\alpha}$ and $IL-1{\beta}$ in blood increased in the osteoarthritis-induced group while decreasing in the group administered with FAJE. In addition, MMP-2 and MMP-9 in cartilage tissues increased in the osteoarthritis-induced group, but decreased in the group treated with FAJE. Cartilage examination indicated that the osteoarthritis-induced group exhibited cartilage erosion and cell degeneration, but in the FAJE administered group the tissue, conditions were recovered and cartilage proteoglycan was increased. Therefore, FAJE clearly showed anti-inflammatory effects and this suggests it is effective for recovery from osteoarthritis induced by MIA.

Pharmacological studies on roots of Achyranthes aspera Linn

  • The crude methanol extract of the roots of Achyranthes aspera Linn. was investigated for its possible antinociceptive, diuretic and neuropharmacological activities in animal models. At the dose of 250 and 500 mg/kg body weight, the extract showed a significant antinociceptive effect in acetic acid induced-writhing in mice comparable to that produced by diclofenac sodium, used as standard drug. The crude extract produced significant diuretic effect at the dose of 500 mg/kg of body weight comparable to that produced by furosemide, used as standard drug. The extract also potentiated significantly the pentobarbital induced sleeping time in mice; decreased the open field score in open field test, decreased the number of hole crossed from one chamber in the hole cross test and decreased the head dip responses. The obtained results provide a support for the use of this plant in traditional medicine and its further investigation.

Achyranthes japonica Nakai Water Extract Suppresses Binding of IgE Antibody to Cell Surface FcεRI.

  • Achyranthes japonica Nakai (AJN) water extract has a variety of physiological properties, including anti-diabetic, anti-cancer, anti-inflammatory, anti-microbial, and anti-oxidative activities. In the present study, the inhibitory effects of AJN extract were investigated in high affinity immunoglobulin E receptor ($Fc{\varepsilon}RI$)-mediated KU812F cells activation. AJN extract showed suppressive effects on histamine release and intracellular calcium [$Ca^{2+}$]i elevation from anti$Fc{\varepsilon}RI$ antibody (CRA-1)-stimulated cells in a dose-dependent manner. Flow cytometric analysis showed that AJN extract treatment caused a dose-dependent decrease in the cell surface $Fc{\varepsilon}RI$ expression and the binding between the cell surface $Fc{\varepsilon}RI$ and the IgE antibody. Moreover, reverse transcription-polymerase chain reaction analysis showed that levels of the mRNA for the $Fc{\varepsilon}RI$ ${\alpha}$ chain was decreased by treatment with AJN extract. These results indicate that AJN extract may exert anti-allergic effects via the inhibition of calcium influx and histamine release, which occurs as a result from the downregulation of the binding of IgE antibody to cell surface $Fc{\varepsilon}RI$. This mechanism may occur through $Fc{\varepsilon}RI$ expression inhibition.

Prevention of Arthritic Inflammation Using an Oriental Herbal Combination BDX-1 Isolated from Achyranthes bidentata and Atractylodes japonica

  • An oriental herbal combination (BDX-1) was isolated from Achyranthes bidentata and Atractylodes japonica. We previously tested the clinical effectiveness of BDX-1 in rheumatoid arthritis (RA) patients and found that it has a beneficial therapeutic effect. Here, we provide experimental evidence for the effectiveness of BDX-1 on RA in murine models. The oral administration of BDX-1 was found to markedly inhibit collagen-induced arthritis, adjuvant-induced arthritis, and zymosan-induced inflammation. It also inhibited carrageenan-induced acute edema and acetic acid-induced writhing response. In addition, the biological activity of BDX-1 was found to be strongly increased by fermentation. Our results suggest that BDX-1 could be useful for the treatment of rheumatoid arthritis.

우슬뿌리 추출물의 Cathepsin B에 대한 저해효과 (Inhibition Effect of Achyranthes japonica N. Root Extract on Cathepsin B)

  • 우슬이 민간요법으로 관절염 치료에 우수한 한약재로 알려진 바 우슬 추출물이 cathepsin B 에 대한 저해력을 검토하기 위하여 각종 용매로 우슬 추출물을 순차 분획하고 분획 추출물에 대하여 cathepsin B에 대한 저해활성을 검토하였으며 우슬의 지표물질인 20-hydroxy ecdysone이 추출분획에 검출되는지 TLC 및 HPLC를 이용하여 분석하였으며 우슬의 지표물질이 cathepsin B에 대한 저해활성여부를 확인한바 다음과 같은 결과를 얻었다. 우슬 뿌리에 대해 methanol/water(4:1, v/v) 추출물을 ethyl acetate, chloroform, chloroform/methanol(3:1, v/v), methanol의 각 용매로 분획한 결과, 분획 F4(methanol 분획)에서 수율이 가장 높아 $8.27\%$를 나타내었다. 각 분획별에 따라 cathepsin B에 대한 저해활성을 측정한 결과 F4분획물에서 가장 저해활성이 높았으며 F1분획에서도 높은 저해활성을 나타내었다. F4분획물 중 우슬의 지표물질인 20-hydroxy ecdysone이 검출되었으며 우슬에 함유되어있는 함량은 $0.33\%$이었다. 20-hydroxy ecdysone에 대하여 cathepsin B저해활성을 보기 위하여 cathepsin B저해활성제로 알려진 leupeptin과 활성을 비교해본 결과 기질을 BANA로 사용하였을 경우 leupeptin은 저해율이 $92\%$인데 비하여 20- hydroxy ecdysone은 $88\%$이었으며 F4분획물은 $97\%$를 나타내었고 기질을 CLN으로 사용하였을 경우 leupeptin은 저해율이 $62\%$인데 비하여 20-hydroxy ecdysone은 $36\%$이었으며 F4추출물은 $67\%$를 나타내었다.
