인진쑥 배지에서 배양한 노루궁뎅이버섯 균사체 추출물(HEAC)의 알코올 대사촉진 활성 (Enhanced Activities of Alcohol Methabolism by Extracts from Hericium erinaceum Hypha Cultivated with Artermisia capillaris(HEAC))

  • 인진쑥 배지에서 배양한 노루궁뎅이버섯(HEAC), 노루궁뎅이 버섯과 인진쑥 추출물들의 음주 후 체내 알코올 농도의 감소 효과를 확인하였으며, 또한 혈중 알코올 탈수소효소 및 아세트알데히드 탈수소효소의 활성을 조사하여 알코올 분해 활성을 조사하였다. 체내 알코올 농도 측정결과 HEAC의 경우 170분, 노루궁뎅이 버섯 추출물은 210분후에 혈중 알코올이 검출되지 않았다. 또한, HEAC는 알코올 탈수소효소 활성에서 대조군 보다 시간에 따라 154%, 노루궁뎅이버섯 추출물은 148%로 비교적 높은 활성을 나타내었다. 아세트알데히드 탈수소활성에서는 HEAC와 노루궁뎅이버섯 추출물이 대조구에 비해 104-111%이상의 효소활성이 유지되는 것을 확인하였으며 이들 추출물들은 알코올 분해 촉진작용이 매우 우수함을 알았다.

국산 홍합과 뉴질랜드 초록입 홍합 열수 추출물의 알코올분해효소 활성에 미치는 영향 및 DPPH 라디칼 소거능과 Angiotensin Converting Enzyme 저해 활성 (Effects of Hot Water Extracts of Domestic Blue Mussel and New Zealand Green Lipped Mussel on Alcohol Metabolizing Enzymatic, DPPH Radical Scavenging, and Angiotensin Converting Enzyme Inhibitory Activities)

  • 우리나라 남해안 연안에서 주로 양식되는 진주담치(국산 홍합)와 세계적으로 기능성이 잘 알려진 뉴질랜드 초록입 홍합의 열수 추출물을 제조하여 생리활성을 비교하였다. 모든 홍합 추출물들은 알코올대사와 관련한 alcohol dehydrogenase(ADH)와 acetaldehyde dehydrogenase(ALDH)의 활성을 상승시켰으며, 특히 ADH의 활성을 크게 향상시켰다. 국산 홍합의 육질 추출물은 초록입 홍합의 육질 추출물에 비하여 ADH 상승 효과는 비슷하였으며, ALDH 상승 효과는 약간 낮았다. DPPH 라디칼 소거능으로 조사한 항산화능은 국산 홍합의 육질 추출물이 초록입 홍합의 육질 추출물 보다 높았다. 항고혈압과 관련한 ACE 저해능의 경우에는 국산 홍합 육질 추출물이 초록입 홍합의 육질 추출물에 비해 10 mg/mL 농도에서는 낮았으나 20 mg/mL 이상의 농도에서는 유의차(P<0.05)를 보이지 않았다. 이상의 결과는 국내에서 양식되고 있는 홍합도 세계적으로 초록입 홍합에 못지않은 생리활성을 가지고 있음을 시사한다.

Effects of Dried Persimmon Snacks on Alcohol Metabolism in Men

  • Persimmon has been known to help alcohol intoxication in Korea for a long time and is prepared as a processed food. The effect of dried persimmon snacks on alcohol metabolism was determined in vivo. Eight Korean men (25~27 years old) were administered 3.5 mL/kg of alcohol (22.5%, w/v) with or without a dried persimmon snack (2g/kg). The levels of alcohol in the blood and of acetaldehyde in the blood and the urine were determined by alcoholmeter and assay using alcohol dehydrogenase and HPLC, respectively. All subjects showed decreased levels of alcohol in blood, and six subjects showed a decrease of alcohol in urine after consumption of ethanol with dried persimmon snacks. Concentration of acetaldehyde in the blood an urine decreased significantly in three and five of the eight subjects, respectively. Reduction was more significant for alcohol than for alcohol than for acetaldehyde with administration of ethanol and dried persimmon snacks. This in vivo result suggests that dried persimmon snacks are effective in decreasing th concentration of alcohol after alcohol intake.

  • Alcohol consumption causes numerous consequences on the health of the human body. Heavy drinking on a daily base has caused liver diseases. Furthermore, some products such as acetaldehyde produced from alcohol metabolism are more toxic than alcohol itself. This study was carried out to evaluate the role of Lactobacillus casei on alcohol metabolism, especially, the removal of the toxic effect of alcohol. The maximum alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activities from L. casei were observed at 4 hr of culture. L. casei was confirmed to produce the ADH and ALDH by the SDS-PAGE. From in vivo test using SD rats with 22% alcoholic drink, blood alcohol concentration (BAC), glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) of the rats feeding the medium containing L. casei were lower than those of the rats feeding the medium containing an alcoholic drink only This demonstrates that the ADH and ALDH produced by L. casei have virtual functions to detoxicate the alcohol in vivo and the fermentation broth of L. casei can be used as an alcohol detoxification drink.

주상(酒傷)에 응용되는 성주청간탕(醒酒淸肝湯)이 알코올대사 및 간장애에 미치는 영향 (Effects of the Sungjuchunggan-tang(Xingjiuqinggan-tang) on alcoholic metabolism and alcoholic liver damages)

  • Objectives : The aim of this study was to investigate effects of the Sungjuchunggan-tang(Xingjiuqinggan-tang), which has been used for alcoholic diseases in Oriental medicine, on alcoholic metabolism and alcoholic liver damage. Methods : We evaluated the activities of alcohol dehydrogenase(ADH)and aldehyde dehydrogenase(ALDH), and measured the levels of AST, ALT, glucose, triglyceride, HDL-cholesterol, LDL-cholesterol and phospholipid in serum of guinea pigs. Results and Conclusions : In this experiment, Sungjuchunggan-tang, Pharbitidis Semen and Puerariae Radix significantly suppressed the activity of ADH which is a precursor enzyme of acetaldehyde. Sungjuchunggan-tang and Pharbitidis Semen significantly suppressed the activity of ALDH, which is a catabolic enzyme, and increased its level. Due to alcohol consumption, level of ALT and AST were significantly reduced. These experimental results suggest that Sungjuchunggan-tang and Pharbitidis Semen inhibit the formation of acetaldehyde in the metabolism of alcohol, and affect the recovery of weakened liver functions caused by alcohol.

복합생약 HO-Series의 숙취개선 임상적 유용성 평가 (Evaluation of Clinical Usefulness of Herbal Mixture HO-Series for Improving Hangover)

  • The purpose of this study is to investigate the hangover relieving effect of HO-series. HO-S1 is an herbal mixture, which consists of extracts from Flower of Pueraria lobata Ohwi, Glycyrrhiza glabra Linné, Fruit of Lycium chinense Miller, Poria cocos Wolf, Acanthopanax sessiliflorum Seeman, Scutellaria baicalensis Georgi, Atractylodes lancea De Candlle and Zingiber officinale Roscoe. HO-S2 is a candidate that has been performed to ultra filtration based on HO-S1. HO-S3 is a mixture of amino acids and vitamins based on HO-S2. HO-01 is the final beverage base produced based on HO-S3. The antioxidant activity of HO-series was similar to that of vitamin C or trolox. The production of t-BHP induced reactive oxygen species(ROS) was significantly blocked in the presence of HO-series. In vivo study, AUC of alcohol and acetaldehyde concentrations in HO-S2 and HO-S3 treated groups significantly decreased. Hepatic alcohol dehydrogenase(ADH) and acetaldehyde dehydrogenase(ALDH) activity were significantly higher in HO-S2 and HO-S3 treated groups. And 2E1 activity and glutathione were significantly elevated, while the malondialdehyde level was not significantly in liver tissue. After alcohol exposure, the sensitivity scores of blood alcohol and acetaldehyde concentration and hangover symptoms were significantly decreased in the HO-01 intake group compared with the non-intake group. ALDH activity was significantly increased in the HO-01 intake group. HO-series have antioxidant activity and a protective effect from ROS. HO-S2, HO-S3 and HO-01 are potentially highly beneficial in relieving hangover, as it scavenges reactive free radicals and boosts the endogenous antioxidant system.

천연소재복합물의 알코올 및 아세트알데히드 분해활성 및 호기중 알코올 농도 감소 (Natural herbal extract complex induces the degradation of alcohol and acetaldehyde and reduces the breath alcohol concentration)

  • 본 연구는 천연소재 12종의 페놀성 화합물 함량, 라디칼 소거능, 알코올탈수소효소(ADH), 아세트알데히드탈수소효소(ALDH) 활성도를 측정하고 상승효과를 가지도록 배합한 복합물의 숙취해소에 미치는 효과를 규명하는데 목적이 있다. 우수한 개별 소재를 선정하고 배합비를 달리하여 효능 평가를 통해, 배합비가 효능에 미치는 상관관계를 분석하였다. 8종의 추출물 중 보이차, 소나무, 갈근 순으로 ADH 활성이 높았고, 갈근은 배합량과 효능 간의 양의 상관관계, 보이차, 소나무 등은 음의 상관관계를 가지는 것으로 나타났다. 8종의 소재로 배합한 F1~F7 을 대상으로 ADH, ALDH 활성평가를 통해 F.7에서 상승효과가 가장 높은 것을 확인하여 최적 배합비로 도출하였다. F.7를 섭취한 시험군에서 알코올 섭취 30분 후부터 호기 알코올 농도가 섭취 직후 호기중 알코올 농도 대비 58%로 유의적으로 감소되기 시작하였고, 120분 후 27%로 호기중 알코올 농도가 감소되었다. 음주 후 발생하는 숙취증상 설문 결과 대조군에 비해 시험군에서 음주 후 발생하는 피곤함과 갈증에서 유의적인 개선효과 (p<0.05)가 있었다.

Acetobater sp.KM Alcohol Dehydrogenase의 분리 및 특성 (Purification and Characterization of Alcohol Dehydrogenase from Acetobacter sp. KM)

  • Membrane-bound alcohol dehydrogenase(ADH) was purified to homogeneity from the acetic acid producing bacteria, Acetobacter sp. KM. The enzyme was solubilized and extracted with Triton X-100 and purified using the Mono-Q ion exchange chromatography and Superose 12 gel filtration chromatography. The enzyme was purified to 12-fold with a yield of 30%. The molecular weight of the purified enzyme was to be 335 KDa. SDS-PAGE of the enzyme showed two subunits with molecular weights of 79 KDa and 49 KDa. It indicated that the enzyme consisted of three subunits of the 79 KDa and two subunits of the 49 KDa. The purified .ADH preferentially oxidized straight chain aliphatic alcohol except methanol. Formaldehyde, acetaldehyde and glutaraldehyde were also oxidized. The apparent Km for ethanol was 1.04 mM and the optimum pH and temperature were 5.0∼6.0 and 32$^{\circ}C$, respectively. V2O5 and divalent cation such as ZnCl2 and NiCl2 inhibited enzymatic activity.

Lactobacillus brevis HY7401 섭취가 쥐의 혈중 알코올 수준에 미치는 영향 (Effect of Lactobacillus brevis HY7401 Intake on the Serum Ethanol Concentration in Rats)

  • Lactobacillus 균주들의 에탄올과 acetaldehyde의 대사 가능성을 시험관내 및 생체내에서 시험하였다. ADH와 ALDH 활성은 L. brevis 균주들이 다른 Lactobacillus 균주들과 비교하여 높게 나타났으며, 특히 L. brevis HY7401은 가장 높은 ADH와 ALDH 활성을 나타냈다. L. brevis HY7401$(10^9\;cfu/mL)$을 2주간 하루에 1 mL씩 쥐에게 급여한 후 시험 14일째에 에탄올(4g/kg BW)을 경구투여하고 혈중 에탄올 양을 측정한 결과, L. brevis HY7401 급여에 의해 혈중 에탄올 수준이 대조구에 비해 유의적으로 감소하는 것으로 나타났다. 에탄올 투여 6시간후의 혈중 에탄올 양이 각각 $1,195{\pm}92.9$(대조군) 그리고 $454{\pm}150mg/L$(HY7401군)으로 나타났다. 또한 L. brevis HY7401 급여한 군에서 소장내 에탄올 농도가 대조구와 비교하여 약 1/2수준이었으며, 초산 농도는 2배정도 높은 것으로 확인되었다. 따라서 이 연구결과들은 사람으로부터 분리된 L. brevis HY7401이 높은 ADH와 ALDH 활성을 갖고 있어 에탄올과 acetaldehyde를 대사한다는 것을 의미하고 있다. 또한 L. brevis HY7401을 쥐에게 급여함으로써 혈중 에탄올 수준을 낮출 수 있는 것으로 확인되었다.

염건 조피볼락(Sebastes schlegeli) 제조 중 ADH 및 ALDH의 활성변화 (Changes in Alcohol Dehydrogenase (ADH) and Acetaldehyde Dehydrogenase (ALDH) Activity during the Processing of Salt-Dried Rockfish Sebastes schlegeli)

  • The objective of this study was to determine the processing conditions for salt dried rockfish Sebastes schlegeli by sun drying and cold-air drying, as measured by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activity. We processed salt dried rockfish samples. The salinity of rockfish samples was within 1% following salting with 25% salt brine for 3 h. The moisture content of salt dried rockfish was found to reduce linearly from 70.12 to 39.5 g/100 g over the same time interval. The water activities of salt dried rockfish by sun and cold-air drying were 0.94 and 0.87, respectively, after three days of drying. Acid values (AV) were 10.71 and 5.96 mg KOH/g, respectively, after the three day drying period. The ADH activity in a water extract from salt dried rockfish following sun and cold-air drying for 24 h was 228.5% and 226.1% at 13.3 mg/mL, respectively, and was higher than that when drying lasted for 48 and 72 h. The ALDH activity was not affected but both ADH and ALDH activity tended to decrease as the drying time increased from 24 to 72 h. The conditions of processing for the best quality of salt dried rockfish were determined to be drying with a cold-air system for 24 h. These results indicated that water extracts from salt dried rockfish have valuable biological attributes owing to the metabolizing of alcohol and can provide useful information for the design of drying systems for salt dried rockfish.
