Effect of Arsenic Types in Soil on Growth and Arsenic Accumulation of Pteris multifida (토양 비소 오염원의 종류가 봉의꼬리의 생육 및 비소 축적에 미치는 영향)

  • This study was carried out to analyze the effect of arsenic types on growth and arsenic accumulation ability of Pteris multifida. Among arsenic pollution sources, Sodium arsenate, Calcium arsenate, Sodium arsenite and Potassium arsenite were treated in horticultural compost contaminated with $500mg{\cdot}kg^{-1}$. P. multifida was cultivated for 12 weeks. The results of study, Calcium arsenate treatment showed slightly decreased growth of P. multifida. But, growth of P. multifida cultivated in the remaining arsenic treatment was similar to untreated control plot. With only short-term cultivation of 4 weeks, aerial part of P. multifida in Sodium arsenate treatment showed high arsenic accumulation of $2,289.5mg{\cdot}kg^{-1}DW$. The arsenic accumulation ($2,956.0mg{\cdot}kg^{-1}DW$) was the highest at 12 week. On the other hand, underground part showed the highest arsenic accumulation in Potassium arsenite treatment ($2,470.2mg{\cdot}kg^{-1}DW$) and Calcium arsenate treatment accumulated $1,060.7mg{\cdot}kg^{-1}DW$ of arsenic. Regardless of arsenic types, aerial part of P. multifida was absorbed more than $1000mg{\cdot}kg^{-1}DW$ of arsenic. And removal of arsenic in soil was also higher. Therefore, Pteris multida is considered to be suitable phytoremediation meterial of various arsenic contaminated areas.

Effects of Dietary Vitamin E Supplementation on Color Stability, Lipid Oxidation and Reducing Ability of Hanwoo (Korean Cattle) Beef during Retail Display

  • The effects of dietary vitamin E supplementation (control; 200 IU/head/day, 500 IU/head/day and 1,000 IU/head/day) on color stability, lipid oxidation and total reducing ability in M. longissimus and M. semimembranosus from Hanwoo (Korean cattle) steer during retail display ($3{\pm}1^{\circ}C$, 1,200 lux) were investigated. The $L^{*}$, $a^{*}$, $b^{*}$, $C^{*}$, R630-R580 values and TRA (total reducing ability) of 1,000 IU group were significantly (p<0.05) higher than those of the other groups. The $a^{*}$, $C^{*}$, R630-R580 and TBARS values were significantly (p<0.05) higher in M. longissimus than in M. semimembranosus. The $a^{*}$, $C^{*}$, R630-R580 values and TRA for two beef muscles declined gradually during storage and the decline was more rapid in control and 500 IU groups. Hue angle and metmyoglobin (%) for two beef muscles increased (p<0.05) as display time increased, and 1,000 IU group had a lower rate of metmyoglobin accumulation during retail display. The TBARS values for two beef muscle were significantly (p<0.05) lower in 1,000 IU group than in the other groups over time. Consequently, the meat from 1,000 IU vitamin E-supplemented Hanwoo steer extended retail display life compared to the control and 500 IU/head/day.

Reduction of Ammonia Accumulation and Improvement of Cell Viability by Expression of Urea Cycle Enzymes in Chinese Hamster Ovary Cells

  • Previously, we developed a CHO cell line (CHO-OTC1-Al9) that expresses the first two enzymes in the urea cycle and exhibits a higher ammonia-removing ability and faster growth rate than a vector-controlled CHO cell line (CHO-neo-5). The current study was undertaken to develop a cell line with an ammonia-removing ability higher than the cell line developed previously. To accomplish this, CHO cell lines expressing the first three, first four, or all five enzymes of the urea cycle were constructed using a stable transfection method. Finally, the CHO-AS-16, CHO-AL-19, and CHO-Arg-11 cell lines expressing the first three, first four, and all five enzymes of the urea cycle, respectively, were selected and found to exhibit higher ammonia-removing ability than the CHO-OTC1-Al9 cell line. Among the three selected cell lines, CHO-AL-19 showed the highest ammonia-removing ability and highest cell viability at a higher cell density, with 40% and 15% lower ammonia concentration in the, culture media than that of CHO-neo-5 and CHO-OTC1-A19 cell lines, respectively. CHO-AL-19 also showed 44% and 10% higher cell viability than the CHO-neo-5 and CHO-OTC-Al9 cell lines, at a higher cell density, respectively. The ammonia concentrations in the culture media were expressed as the ammonia concentration/cell, and the CHO-AL-19 cells revealed 45-60% and 20% lower ammonia concentration/cell than the CHO-neo-5 and CHO-OTC1-Al9 cells, respectively.

Effect of Fish Oils on Brain Fatty Acid Composition and Learning Performance in Rats

  • The effects of sardine oil(high in eicosapentaenoic acid : EPA) and tuna oil(high in docosahexaenoic acid : DHA, also high in EPA) on fatty acid composition of brain and learning ability were evaluated in male weanling Sprague-Dawley rats and compared with the effects of corn oil and beef tallow. Animals assigned by randomized block design to one of the four experimental diet groups containing dietary lipid at 15%(w/w) level were given ad libitum for 7 weeks. Food intake and body weight gain of the fish oil groups were significantly lower than those of the corn oil and beef tallow groups. However, brain weights of the groups were not significantly different. In the brain fatty acid composition, the corn oil group showed high concentrations of n-6 fatty acids, the fish oil groups of n-3 fatty acids, and the beef tallow group of saturated fatty acids. Brain EPA and DHA contents of the fish oil groups showed significantly higher than the other groups while the brain ratio of saturated/monounsaturated/polyunsaturated fatty acid was controlled in a narrow range. In a maze test, the fish oil groups appeared to arrive at the goal faster than the corn oil and beef tallow groups. It explained that EPA in diets might efficiently convert to DHA resulting in DHA accumulation in brain tissue and might increase the learning performance as DHA did.

Two Threonine Residues Required for Role of AfsKav in Controlling Morphogenesis and Avermectin Production in Streptomyces avermitilis

  • AfsKav is a eukaryotic-type serine/threonine protein kinase, required for sporulation and avermectin production in Streptomyces avermitilis. In terms of their ability to complement SJW4001 (${\Delta}afsK$-av), afsK-av mutants T165A and T168A were not functional, whereas mutants T165D and T168D retained their ability, indicating that Thr-165 and Thr-168 are the phosphorylation sites required for the role of AfsKav. Expression of the S-adenosylmethione synthetase gene promoted avermectin production in the wild-type S. avermitilis, yet not in the mutant harboring T168D or T165D, demonstrating that tandem phosphorylation on Thr-165 and Thr-168 in AfsKav is the mechanism modulating avermectin production in response to S-adenosylmethione accumulation in S. avermitilis.

Isolation of Bacillus sp. SHL-3 from the Dry Soil and Evaluation of Plant Growth Promoting Ability

  • Excess use of chemical fertilizer causes soil acidification and accumulation of salt, and thus might bring to desertification of soil. To overcome this problem, it needs limited usage of chemical fertilizer and increased usage of natural fertilizer as an alternative. In this study, dry soil-borne Bacillus sp. SHL-3, which was isolated from arid and barren soil, with plant growth promoting activity was isolated for identification and to determine optimal culture condition. A bacterial strain SHL-3 had the IAA productivity ($5.16{\pm}0.10mg\;L^{-1}$), ACC deaminase activity ($0.36{\pm}0.09$ at 51 hours) and siderophore synthesis. It was identified as genus Bacillus sp.. Also, optimal culture condition of SHL-3 were $20^{\circ}C$ and pH 7 in LB medium. Bacillus sp. SHL-3 had up to 4% salt tolerance in the medium. We evaluated the plant growth promotion ability of SHL-3 using yam (Dioscorea japonica Thunb.). As a result, Bacillus sp. SHL-3 was effective on the increase of the shoot length (202.4% increase for 91 days). These results indicate that Bacillus sp. SHL-3 can serve as a promising microbial resource for the biofertilizers of soil.

Cadmium Accumulation and Tolerance of Iris pseudacorus and Acorus calamus as Aquatic Plants Native to Korea (자생 수생식물 노랑꽃창포와 창포의 카드뮴 축적 및 내성)

  • This study was conducted to find out the cadmium (Cd) accumulation and tolerance of Iris pseudacorus and Acorus calamus as aquatic plants native to Korea for Cd removal in water. In the range of Cd concentration from $10{\mu}M$ to $130{\mu}M$, the Cd lethal dose 50 ($LD_{50}$) was $78.5{\mu}M$ in I. pseudacorus and $47.6{\mu}M$ in A. calamus. In I. pseudacorus, superoxide dismutase and peroxidase as antioxidants were relatively effective against oxidative stress caused by Cd, while catalase, superoxide dismutase, and polyphenolics were effective in A. calamus. The polyphenolics known as typical antioxidants were not detected in I. pseudacorus. In both species, the Cd accumulation in plants increased with the higher Cd concentration and the longer processing period. Also, the absorbed Cd was accumulated mainly in the roots. The amount of Cd accumulated in the shoot part was maximally $548.1mg{\cdot}kg^{-1}$ (82.1% to Cd accumulated in the root part) in I. pseudacorus and $121.4mg{\cdot}kg^{-1}$ (13.7%) in A. calamus, which implied that both species all were enough evaluated as Cd hyper-accumulators based on 0.01% or more Cd accumulation in the shoot. Especially I. pseudacorus showed outstanding ability to move well Cd into the shoots from the roots and high tolerance to Cd stress.

A Study on Ability of Cognitive Impairment According for Decreased Balance Function of Pneumoconiosis Patients

  • Purpose: Pneumoconiosis is a chronic lung disease in which the lungs become hardened due to the accumulation of fine mineral dust that has been breathed in at industrial sites. The lungs are unable to function properly, resulting in complications of several respiratory diseases. Methods: The subjects were 36 elderly adults (pneumoconiosis patients 18, healthy elderly 18) over the age of 65 years. The respiratory function was assessed using a spirometer to measure forced vital capacity (FVC), first second of forced expiration (FEV1), FEV1/FVC, and peak expiratory flow n(PEF). The static balance function test was determined by the center of mass (COM), ankle angle range, hip angle range, sway parameters, and reciprocal compensatory index (RCI). Cognitive function was measured by applying MoCA-k. Results: Comparison of respiration functions between the two groups showed statistically significant differences in FVC, FEV1, FEV1/FVC, and PEFs (p<0.05). Cognitive abilities showed statistically significant differences due to higher cognitive skills in the control group as compared to the patient group (p<0.05). Most results of the static balance capability measurements showed statistically significant differences between the groups (p<0.05). However, no statistically significant differences were obtained for COM AP Range, Angle AP Range, RCI ML Range, and AP Range (p>0.05). Conclusion: The results of the current study indicate that patients suffering from pneumoconiosis have less cognitive ability and balance function as compared to healthy elderly people. Therefore, we expect an improvement in the balance ability and cognitive function through systematic management and strengthening lung function training to improve the quality of life of pneumoconiosis patients.

The Study on Proper Way for Engineering University Education: Based on the Perception of Current Competencies and Expected Competencies of Engineering Freshmen (공과대학 신입생의 핵심역량 인식수준을 통한 공학교육방향 연구)

  • This paper is a study on the perception of current competency and expected competency of engineering freshmen by extracting core competencies acquired from university education. It also aims to suggest proper way for engineering university education. This study extracts competencies in the following five areas as core competencies: 'knowledge on major area', 'cultural ability', 'foreign language ability', 'basic learning ability', 'intercommunication ability'. To achieve this purpose, this study surveyed 'C' university engineering department freshmen (584 students) with questionnaires about their perception of core competencies. The results are as follows. First, engineering freshmen perceived current competencies were weak in every area, especially their capacities in 'foreign language ability' area were perceived to be weakest. Their demand for education is the highest in 'foreign language ability' area, and the second higher in 'knowledge on major area'. Secondly, there exists meaningful difference between perception of current competency and expected competency depending on the gender, high school department (science/liberal arts), high school location, types of college admissions, and types of mathematics in NAST. According to these results, this study suggests enhancement of foreign language (English) education in engineering department, design and implementation of various educational program to overcome individual difference, promoting importance of competencies in the 'cultural abilty' and 'intercommunication abilty', necessity of continuous adjustment and complementation for engineering educational program through accumulation of feedback processes, activation of career education through engineering education and special programs.

Factors Influencing Problem Solving Abilities of Freshmen Nursing Students (간호대학 신입생들의 문제해결력에 미치는 영향 요인)

  • Purpose: The purpose of this study was to investigate factors which influence freshmen nursing students' problem solving abilities. Method: The data were collected from 119 students by means of self-reported questionnaires measuring problem solving abilities, critical thinking disposition, and self-directed learning, on March 7th and 11th, 2010. Result: For problem solving ability categories, the level of problem discovery was the highest followed by solution plan, problem definition, solution evaluation and solution implementation. The influencing factors on problem solving abilities of nursing students were critical thinking disposition, self-directed learning and gender. Conclusion: Using the results of this study as a foundation, nursing education curriculum should be comprised of critical thinking skills and self-directed skills for the improvement of nursing students' problem solving abilities. It is also recommended that further studies be conducted in order to investigate comprehensive curriculum designs that allow for maximum accumulation of these proficiencies.
