전임상 혈관분석을 위한 초분광 이미징 시스템의 활용 (Application of Hyperspectral Imaging System to Analyze Vascular Alteration for Preclinical Models)

  • 본 논문에서는 초분광 이미징시스템을 이용하여 획득된 헤모글로빈 포화이미지와 다양한 이미지프로세싱을 통해 얻어진 실시간 혈관 변화과정을 마이크로미터/밀리초 부터 밀리미터/시간에 이르는 시공간 해상도로 제공하여, 다양한 질병에 기인한 혈관의 생성 및 변화 등과 같은 고유한 생리적 특성뿐만 아니라 혈관간의 산소이동, 혈관질환의 치료효과의 검증 등 다목적 영상장비로의 개발이 가능하다. 이는 질병으로 인한 혈관의 변이과정을 관찰하기 위해 최근 다양한 임상 및 전임상 영상장비들이 개발되고 있으나 높은 개발비용과 환자들이 감수해야하는 위험부담에 비해 상대적으로 낮은 해상도와 제한된 만족도를 제공한다 한계점을 극복할 수 있다. 새로운 혈관의 생성 및 기존의 모세혈관 변화는 암 전이 및 발전과정 뿐만 아니라 다양한 질병의 종류에 따라 다른 특성을 보이며 이를 통한 생리학적 분석이 가능하므로 혈관의 연구를 통한 질병종류 및 유무의 판단은 진단 과정의 핵심 요소이며 새로운 치료법의 효과를 평가할 수 있는 중요한 근거가 될 것으로 기대한다.

후성 유전학적 리프로그래밍과 클로닝 (Epigenetic Reprogramming and Cloning)

  • 포유동물의 초기 발생과정 중 접합체가 전능성이나 다능성을 가지기 위해서는 전반적인 DNA 메틸화를 포함하는 후성 유전학적 리프로그래밍의 복잡한 과정을 거쳐야만 한다. 본 연구팀에서는 공여핵의 후성 유전학적 리프로그래밍 과정을 조사하기 위하여 소 복제수정란에서 메틸화 양상을 분석하였다. 복제수정란의 비정상적인 메틸화 양상이 다양한 반복염기서열에서 관찰되었지만 single-copy유전자들의 염기서열은 정상적인 메틸화 양상을 보여주었다. 전반적으로 복제수정란의 전반적인 메틸화 상태는 정상수정란과 완전히 다른 양상을 보여주었다. 또한 복제 배반포의 영양외배엽세포에서 특이적으로 높은 메틸화 수준은 현 복제동물에서 빈번히 나타나는 불완전한 태반형성에 작용할 수 있을 것이다. 결론적으로 복제수정란의 비정상적 발생은 공여핵의 불완전한 후성 유전학적 리프로그래밍에 기인할 수 있다는 사실을 제시하게 되었다. 이러한 공여핵의 후성 유전학적 과정의 이해는 복제수정란의 비정상적 발생을 보다 분명히 밝힐 수 있을 것이다.

Frey씨 증후군의 진단에 있어서 Thermography의 유용성 (The Effectiveness of Thermography in Diagnosis of Frey's Syndrome Following Parotidectomy)

  • Objectives and Backgrounds : After parotidectomy, some of patients complain gustatory sweating, facial flushing and discomfort in the same area. A series of these symptoms are supposed to be caused by the aberrant regeneration of the secretory parasympathetic fibers to sweat glands and blood vessels of the skin following parotidectomy. In this study, we want to compare the efficacy of thermography to the Minor's starch-iodine test for determining the presence of Frey's syndrome. Materials and Methods : 48 patients who underwent total or superficial parotidectomy from March 2002 to December 2004 were selected for this study. A subjective clinical questionnaire and the objective Minor's starchiodine test were performed to evaluate the incidence of this syndrome. Total 21 patients were confirmed as positive Frey's syndrome and infrared thermography was performed for them. Result : Frey's syndrome occurred in 21 patients(43.8%). The average temperature of parotidectomy site and normal opposite area were $27.65^{\circ}C\;and\;26.41^{\circ}C$ respectively. Thermography showed temperature difference in 20 patient(95.2%) and the difference of temperature was statistically significant above $1.0^{\circ}C$(p<0.001). The severity of symptoms were related with the difference of temperature(p<0.05). Conclusion : Thermography is useful, non-invasive, simple and quantifying method to diagnose Frey's syndrome. Additionally, this geographic diagnosis is available to show the accurate area for botulinum toxin injection.

Caenorhabditis elegans: A Model System for Anti-Cancer Drug Discovery and Therapeutic Target Identification

  • The nematode Caenorhabditis elegans (C. elegans) offers a unique opportunity for biological and basic medical researches due to its genetic tractability and well-defined developmental lineage. It also provides an exceptional model for genetic, molecular, and cellular analysis of human disease-related genes. Recently, C. elegans has been used as an ideal model for the identification and functional analysis of drugs (or small-molecules) in vivo. In this review, we describe conserved oncogenic signaling pathways (Wnt, Notch, and Ras) and their potential roles in the development of cancer stem cells. During C. elegans germline development, these signaling pathways regulate multiple cellular processes such as germline stem cell niche specification, germline stem cell maintenance, and germ cell fate specification. Therefore, the aberrant regulations of these signaling pathways can cause either loss of germline stem cells or overproliferation of a specific cell type, resulting in sterility. This sterility phenotype allows us to identify drugs that can modulate the oncogenic signaling pathways directly or indirectly through a high-throughput screening. Current in vivo or in vitro screening methods are largely focused on the specific core signaling components. However, this phenotype-based screening will identify drugs that possibly target upstream or downstream of core signaling pathways as well as exclude toxic effects. Although phenotype-based drug screening is ideal, the identification of drug targets is a major challenge. We here introduce a new technique, called Drug Affinity Responsive Target Stability (DARTS). This innovative method is able to identify the target of the identified drug. Importantly, signaling pathways and their regulators in C. elegans are highly conserved in most vertebrates, including humans. Therefore, C. elegans will provide a great opportunity to identify therapeutic drugs and their targets, as well as to understand mechanisms underlying the formation of cancer.

쌀겨발효추출물의 항혈전효과 (Anti-thrombotic activity of fermented rice bran extract with several oriental plants in vitro and in vivo)

  • Although the effects of the rice bran have recently been investigated, there is no information regarding platelet physiology available. However, it is well known that fermented natural plants have a beneficial effect on cardiovascular diseases. Therefore, this study was conducted to investigate whether fermented rice bran extract (FRBE) with several plants (Artemisia princeps, Angelica Gigantis Radix, Cnidium officinale, and Camellia sinensis) affected agonist-induced platelet aggregation, and if so, what the underlying mechanism of its activity was. We performed several experiments, including in vitro platelet aggregation, intracellular calcium concentration and adenosine triphosphate release. In addition, the activation of integrin ${\alpha}_{II}b{\beta}3$ was determined using fibrinogen binding. Thrombus formation was also evaluated in vivo using an arterio-venous shunt model. The FRBE inhibited collagen-induced platelet aggregation in a concentration-dependent manner. FRBE significantly and dose dependently attenuated thrombus formation using rat arterio-venous shunt. FRBE suppressed the intracellular calcium mobilization in collagen-stimulated platelets. We also found that FRBE inhibited extracellular stimuli-responsive kinase 1/2, p38-mitogen-activated protein kinases and c-Jun N-terminal kinase phosphorylation. These results suggested that FRBE inhibited collagen-induced platelet aggregation, which was mediated by modulation of downstream signaling molecules. In conclusion, FRBE could be developed as a functional food against aberrant platelet activation-related cardiovascular diseases.

폐격리증을 동반한 기관지 내 평활근종 -1예 보고- (Endobronchial Leiomyoma Combined with Pulmonary Sequestration -A case report -)

  • 호흡곤란을 주소로 내원한 42세 여자환자가 단순 흉부 촬영상 좌측 폐의 무기폐 소견을 보였다. 기관지 내시경과 흉부 컴퓨터 단층촬영상 좌측 주기관지에 종괴가 발견되었다. 조직검사상 종괴는 기관지 평활근종으로 진단되었다. 개흉술을 실시하여 좌측 주기관지절개를 하고 종괴를 제거하자 중엽은 정상적으로 환기가 되었다. 또한 하행대동맥에서 좌폐하엽으로 공급되는 이상동맥이 발견되어 좌폐하엽 절제수술을 동시에 실시하였다. 병리검사상 좌폐하엽은 내엽형 폐격리증으로 진단되었다. 저자들은 기관지 평활근종과 내엽형 폐격리증이 공존하는 드문 질환을 치험하였기에 보고하는 바이다.

Anti-proliferative and Apoptotic Effects of Basella rubra (L.) Against 1, 2-Dimethyl Hydrazine-induced Colon Carcinogenesis in Rats

  • Colorectal cancer is a very prevalent diagnosed cancer. The current study was performed in order to examine the role of BRAE (Basella rubra aqueous extract) in regulating aberrant crypt foci (ACF) formation, cell proliferation and inhibition of apoptosis in a colon carcinogenesis model in male Wistar rats. Rats were randomly allocated into six groups. Group I served as control, and group II acted as a drug control administered BRAE (250mg/kg b.w.) orally for 30 weeks. Rats in group III-VI were given subcutaneous injections of DMH (25mg/kg b.w. weekly) for 15 weeks to initiate colon carcinogenesis. Those in group IV and VI were administered BRAE along with DMH injections. Rats in group V were administered with BRAE after cessation of DMH injection. After 30 weeks of experimental period colons were obtained from experimental groups and analyzed for ACF incidence, argyrophilic nucleolar organizing region-associated proteins (AgNOR) count, histopathological and immunohistochemical changes. Only in DMH exposed groups were ACF and AgNOR numbers increased. Administration of BRAE appreciably decreased the numbers of ACF and AgNOR in BRAE treated groups. Histopathological findings revealed a high level of dysplastic changes with decreased number of goblet cells found only in only DMH injected rats. Administration of BRAE in treated group rats reversed these changes. Expression markers for cell proliferation (PCNA and Ki67) were elevated in DMH treated rats, but reduced with BRAE treatement. This expression was reversed with apoptosis markers (p53 and Caspase-3). Thus the results results of the present study were found to be significant and confirmed the potential efficacy of BRAE against colon carcinogenesis.

Parameters Involved in Autophosphorylation in Chronic Myeloid Leukemia: a Systems Biology Approach

  • Background: Chronic myeloid leukemia (CML) is a stem cell disorder characterized by the fusion of two oncogenes namely BCR and ABL with their aberrant expression. Autophosphorylation of BCR-ABL oncogenes results in proliferation of CML. The study deals with estimation of rate constant involved in each step of the cellular autophosphorylation process, which are consequently playing important roles in the proliferation of cancerous cells. Materials and Methods: A mathematical model was proposed for autophosphorylation of BCR-ABL oncogenes utilizing ordinary differential equations to enumerate the rate of change of each responsible system component. The major difficulty to model this process is the lack of experimental data, which are needed to estimate unknown model parameters. Initial concentration data of each substrate and product for BCR-ABL systems were collected from the reported literature. All parameters were optimized through time interval simulation using the fminsearch algorithm. Results: The rate of change versus time was estimated to indicate the role of each state variable that are crucial for the systems. The time wise change in concentration of substrate shows the convergence of each parameter in autophosphorylation process. Conclusions: The role of each constituent parameter and their relative time dependent variations in autophosphorylation process could be inferred.

Association between RASSF1A Promoter Hypermethylation and Oncogenic HPV Infection Status in Invasive Cervical Cancer: a Meta-analysis

  • Cervical carcinoma is the main cause of cancer-related mortality in women and is correlated with more than 15 risk cofactors, including infection of cervical cells with high-risk types of HPV (hrHPV). Indeed, both aberrant methylation of the RASSF1A promoter and hrHPV infection are often observed in cervical carcinomas. The purpose of our meta-analysis was to evaluate the role of RASSF1A promoter methylation and hrHPV infection in cervical cancer. Our meta-analysis involved 895 cervical cancer patients and 454 control patients from 15 studies. Our results suggested that RASSF1A promoter hypermethylation increased the risk of cervical cancer (OR=9.77, 95%CI=[3.06, 31.26], P=0.0001, $I^2=78%$). By grouping cases according to cancer subtypes, we found that HPV infection was higher in cervical squamous cell carcinomas (SCCs) than in cervical adenocarcinomas/adenosquamous cancers (ACs/ASCs) (OR=4.00, 95%CI=[1.41, 11.30], P=0.009, $I^2=55%$). Interestingly, HPV infection tended to occur in cervical cancers with relatively low levels of RASSF1A promoter methylation (OR=0.59, 95%CI=[0.36, 0.99], P=0.05, I2=0%). Our study provides evidence of a possible interaction between HPV infection and RASSF1A promoter methylation in the development of cervical cancers.

Aberrant Expression of CCAT1 Regulated by c-Myc Predicts the Prognosis of Hepatocellular Carcinoma

  • Background: CCAT1 has been reported to be linked with pathogenesis of malignancies including colon cancer and gastric cancer. However, the regulatory effect of CCAT1 in hepatocellular carcinoma (HCC) remains unclear. The purpose of this research was to identify any role of CCAT1 in the progression of HCC. Materials and Methods: Real time-PCR was performed to test the relative expression of CCAT1 in HCC tissues. A computation screen of CCAT1 promoter was conducted to search for transcription-factor-binding sites. The association of c-Myc with CCAT1 promoter in vivo was tested by Pearson correlation analysis and chromatin immunoprecipitation assay. Additionally, Kaplan-Meier analysis and Cox proportional hazards analyses were performed. Results: c-Myc directly binds to the E-box element in the promoter region of CCAT, and when ectopically expressed increases promoter activity and expression of CCAT1. Moreover, Kaplan-Meier analysis demonstrated that the patients with low expression of CCAT1 demonstrated better overall and relapse-free survival compared with the high expression group. Cox proportional hazards analyses showed that CCAT1 expression was an independent prognostic factor for HCC patients. Conclusions: The findings demonstrated CCAT1, acting as a potential biomarker in predicting the prognosis of HCC, is regulated by c-Myc.
