The Effect of Hongdeung-tang on Endoxotin-induced Disseminated Intravascular Coagulation (홍등탕(紅藤湯)이 내독소로 유발된 파종성혈관내응고증에 미치는 영향)

  • Objective : This experimental study was carried out to investigate anti-coagulation effects and mechanism of Hongdeung-tang on the dissentimenated intravascular coagulation Methods : Each 10 rats were classified as normal gorup, control group and treated group. The control group was given a 0.5mg/ml endotoxin intra-vein injection and was occurred Disseminated Intravascular Coagulation(DIC). The treated group were orally administrated 1ml Hongdeung-tang concentration once a day for 6 days and then it was treated the same as the control group. We took the blood from heart and were investigated number of Platelet and white blood cell(WBC), fibrinogen concentration, prothrombin time(PT), activated partial thromboplastin time(APTT), interleukin-6(IL-6), Tumor Necrosis $Factor-{\alpha}(TNF-{\alpha})$, Prostaglandin $E2(PGE_2)$ from them. Results : There were a statistically significant increase of platelet and fibrinogen concentration in the treated group compared to the treated group. The PT was a significantly reduced in treated group compared to the control group. There was no significant change in numbers of WBC and APTT between control and treated group. On the contrary, the number of PT, IL-6, $TNF-{\alpha}$, $PGE_2$ were significantly decreased in treated group compared to the control group. Conclusion : DIC which was occurred by endotoxin intravenous injection seems to related to the increase of IL-6, $TNF-{\alpha}$, $PGE_2$ and Hongdeung-tang has an effect that supress those factors. we can assume Hongdeung-tang has an anti-coagulation effect by those supressing effect.

Screening of Anticoagulant Activities in Extracts from Edible Herbs (허브 추출물의 항응고 활성 검색)

  • In order to search for blood anticoagulant substance from herbal extracts, we extracted with cold water (Fr. I), methanol (Fr. II) and hot water (Fr. III) from 93 commercially available herbs. The anticoagulant activity of herbal extracts was examined through the intrinsic pathway by activated partial thromboplastin time (APTT), the extriansic pathway by prothrombin time (PT) and the common pathway by thrombin time (TT). Onion, garlic, clove, fenugreek, mugwort, thyme and sage had on anticoagulant activity with intrinsic pathway and mint, italian seasoning, rosemary, turmeric, tarragon and wasabie had on anticoagulant activity with common pathway. Nobody had on anticogulant activity with extrinsic pathway. Among commerical herbs tested, clove cultivated in holland was selected because it showed the most potent anticoagulant activity among the samples investigated.

Antithrombosis of Chungpesagantang and Its Ingredients

  • The possibility of Chungpesagantang, which has been recommended for stroke patients in Korean Sasang Constitutional Clinics, and its ingredients as a novel antithrombotic agent was evaluated. Chungpesagantang potently inhibited ADP and collagen-induced rat platelet aggregation in vitro as well as ex vivo in a dose-dependent manner. Puerariae Radix and Rhei Rhizoma potently inhibited ADP-induced rat platelet aggregation ex vivo. However, Puerariae Radix did not inhibit both in vitro ADP and collagen-induced rat platelet aggregations. Chungpesagnatang and its ingredients except Rhei Rhizoma did not affect certain plasma clotting times, such as APTT, PT, and TT. Chungpesagantang and its ingredients Raphani semen and Scutellariae Radix showed significant protection against death due to pulmonary thrombosis in mice.

A Study of Anticoagulation Activity from Perillae Folium Extract (자소엽(紫蘇葉) 추출물의 항응혈(抗凝血) 활성에 관한 연구(硏究))

  • Objectives: This research was investigated to find out the effect of the anticoagulant Perillae folium extract. Methods: To examine an active effect of anticoagulation in Perillae folium extract, the study measured Prothrombin time(PT) and activated partial thromboplastin time(APTT) of human plasma in vitro and measured bleeding time and arterio-venous shunt model in rats in vivo. Results: Bleeding time of Perillae folium extract in vivo had a significant increase 1.6 times and thrombus weight of Perillae folium extract had a significant reduction of thrombus weight as 68%. Perillae folium extract had an effect of anticoagulation by operating on extrinsic pathway factor II, V, VII, X and intrinsic pathway factor VIII, IX, X, VI, VII in the coagulation system. Conclusions: Considering the above mentioned results, it is judged that a Perillae folium extract has a control effect of thrombus creation.

  • Curdlan is a natural $\beta$-1,3-glucan produced by Agrobacterium biovar 1. In this study, the anticoagulant activity of sulfoalkyl derivatives of curdlan was investigated by carrying out activated partial thromboplastin time (APTT) assay and compared with that of o-sulfonated curdlan. Approximately 100-fold higher concentration of o-sulfonated curdlan than heparin was required to obtain the same level of the clotting time. Anticoagulant activity of curdlan derivatives was dependent on the degree of sulfation in prolonging the clotting time. However, the chain length of the substituent did not play a role in prolonging the clotting time. The curdlan derivatives enhanced thrombin inhibition by mediating through antithrombin III. The inhibition of thrombin by o-sulfonated curdlan was found to be approximately 10-fold weaker than that by heparin.

  • The possibility of Chungpesagantang, Sunghyangjunggisan and Yangkyuksanwhatang, which have been recommended for stroke patients with constipation in traditional Korean clinics, was evaluated as a novel antithrombotic agent. Chungpesagantang potently inhibited ADP- and collagen-induced rat platelet aggregation in vitro, but Sunghyangjunggisan and Yangkyuksanwhatang did not inhibit it. However, all tested herbal formulae inhibited in vivo ADP- and collagen-induced rat platelet aggregation and extended the bleeding time of mice tails. All these herbal formulae did not affect certain plasma clotting times such as APTT, PT and TT. Among these herbal formulae, Chungpesagantang showed significant protection against death due to pulmonary thrombosis in mice. Based on these findings, these herbal formulae should prevent thrombosis and cardiovascular diseases such as myocardial infraction stroke, and arteriosclerosis.

  • Introduction: We previously reported that a new glycosaminoglycan, acharan sulfate (AS) from the African giant snail Achatina fulica showed anticoagulation activity in vitro, but it was much less than that of heparin. In the present study, the anticoagulant activity of AS was investigated in vivo. Methods: AS and heparin were administered to rats in various concentrations and anticoagulant activities were measured. Both were also compared in thrombin-induced Results: Intravenous administration of acharan sulfate prolonged the coltting time (APTT) in mice and rats in a dose-dependent manner. (omitted)

  • Acquired hemophilia A (AHA) is an uncommon autoimmune bleeding disorder in which autoantibodies that affect the functions of factor VIII (FVIII) are present in the blood. The initial diagnosis of AHA is difficult as the presentations of AHA differ from those of congenital hemophilia A. Moreover, the treatment of AHA is more complex due to the presence of autoantibodies against FVIII. Here, we present a case report of postpartum AHA, to increase the perception and knowledge regarding the recognition and management of such cases. We present a young female with the chief complaint of vaginal bleeding and upper arm ecchymosis. Laboratory results exhibited isolated prolonged activated partial thromboplastin time (APTT) and FVIII inhibitors. The patient was treated with corticosteroids, FVIII concentrates, and a bypassing agent. In conclusion, unexplained postpartum bleeding, unmanageable with basic hemostatic measures, should lead to clinical suspicion of an acquired bleeding disease.

Active Effect of Antivoagulant Effects in chaenomelis Fructus Water Extract (모과 추출물의 항응혈 활성)

  • Objectives : This research was investigated anticoagulant effect of the Chaenomelis Fructus extract. Methods : To examine an active effect of anticoagulation in Chaenomelis Fructus extract, the study measured Prothrombin time(PT) and activated partial thromboplastin time (APTT) of human plasma in vitro and measured bleeding time and arterio-venous shunt model in rats in vivo. Results : Bleeding time of Chaenomelis Fructus extract in vivo had a significant increase as about 1.6 times and thrombus weight of Chaenomelis Fructus extract had a significant reduction of thrombus weight as 50%. A Study on the Antioxidative, Anti-inflammatory and Anti-thrombogenic Effects of Zanthoxylum piperitum DC. Extract (초피나무 추출물의 항산화, 항염증 및 항혈전 효능에 관한 연구)

  • Effects of root, stem and leaf extracts of Zanthoxylum piperitum on the inhibition of lipid peroxidation in the hepatic microsome of rat, DPPH radical scavenging activity, soybean lipoxygenase activity and activated partial thromboplastin times (APTT) were examined in vitro. The highest inhibition of hepatic microsomal lipid peroxidation was observed by ethyl acetate fraction of the root and stem extracts. The high inhibition of lipid peroxidation was observed in the leaf, the root and the stem in order. The DPPH radical scavenging activity of ethyl acetate fraction was higher than that of n-butanol fraction and it was similar to the root and the steam extract. It was similar to the inhibition of hepatic microsomal lipid peroxidation. The DPPH radical scavenging activity was the highest in 0.50 mg/mL of ethyl acetate fraction, and it was 4.4-fold higher than that of a-tocopherol, as an antioxidant standard. The DPPH radical scavenging activity was dependent on the extract concentration in the range of $0.12\~5.00$ mg/mL. The soybean lipoxygenase activity of ethyl acetate fraction was higher than that of n-butanol fraction and it was similar to the root and the stem extracts. The soybean lipoxygenase activity was the highest in 0.50 mg/mL of ethyl acetate fraction. The soybean lipoxygenase activity was dependent on the extract concentration in the range of $0.12\~5.00$ mg/mL. The leaf extract showed the highest antithrombogenic effect followed by the stem and then the root extract. The activated partial thromboplastin times were dependent on the extract concentration in the range of $0.10\~2.00$ mg/mL.
