APON, the subscriber network using bus or tree topology, requires MAC protocol, because it shares the uplink channel for many subscribers to transmit data to OLT. Although various MAC schemes, which consider each QoS characteristics of traffic are proposed considering transmission characteristics of ATM traffic class, existing schemes reduce the network efficiency by increasing transmission overhead in frame. In this paper, we propose new MAC scheme, which shows similar performance as those of existing schemes but does not increase transmission overhead. To prevent increasing transmission overhead, the proposed scheme encodes the information about each cell information of traffic of the local queue in RAU transmitted periodically from ONU to OLT Thus, the proposed MAC scheme shows similar performance as those of existing ones and increases efficiency of entire network.

In this paper, we proposed the MAC protocol of APON supporting multi-class traffic such as CBBUVBR, ABR, UBR, to guarantee the required QoS of each service. For this, we analyze the performance of variousrequest mechanisms and employee the different request mechanism for each traffic classes. Upstream anddownstream frame structures to minimize transmission overhead are proposed based on our request mechanism.The proposed MAC protocol applies the different priority to permit distribution process. CBBWBR traffic, withthe stringent requirements on CDV or delay, is allocated prior to any other class. ABR traffic, which hasnon-strict CDV or delay criteria, uses flexibly the available bandwidth but ensures a minimum cell rate (MCR).UBR traffic is allocated with lowest priority for the remaining capacity. The performance of proposed protocol isevaluated in terms of transfer delay and 1-point CDV with various offered load. The result of simulation showsthat the proposed protocol guarantees the required QoS of the corresponding category, while making use of theavailable resources in both an efficient and dynamical way.

In the APON(ATM Passive Optical Network), the transmission of the upstream traffic is based on a TDMA(Time Division Multiple Access) method that an OLT(Optical Line Termination) permits ONUs(Optical Network Units) sending cells by allocating time slots. Because the upstream is not a streaming mode, the cell synchronizer has to be operated in the burst mode. Also, the cell phase monitor is required to prevent collisions between cells which are transmitted by multiple ONUs through a single optical fiber. In this paper, a TDMA burst cell synchroniser is implemented with the FPGA(Field Programmable Gate Array) being used in the APON based on G.983.1 for transmitting upstream cells. It has two main functions which are the upstream data recovery and the phase monitoring. The former is to recover the upstream data and clock in the OLT by seeking the preamble which is the overhead of the upstream time slot and by aligning the phase of the bit and cell with the system clock. The latter is to provide the information to the ONU to compensate for the equalization delay by monitoring continuously the phase difference between adjacent cells to avoid the cell collision on the upstream.

Fuzzy extractor is a biometric encryption that generates keys from biometric data where input values are not always the same due to the noisy data, and performs authentication securely without exposing biometric information. However, if a user registers biometric data on multiple servers, various attacks on helper data which is a public information used to extract keys during the authentication process of the fuzzy extractor can expose the keys. Therefore many studies have been conducted on reusable fuzzy extractors that are secure to register biometric data of the same person on multiple servers. But as the key length increases, the studies presented so far have gradually increased the number of key recovery processes, making it inefficient and difficult to utilize in security systems. In this paper, we design an efficient and reusable fuzzy extractor based on LWE with the same or similar number of times of the authentication process even if the key length is increased, and show that the proposed algorithm is reusably-secure defined by Apon et al.[5].

This paper presents the window-based permit distribution scheme for efficient medium access control to support multiclass traffic in APON(ATM over passive optical network). The proposed MAC protocol considers the characteristics of QoS(Quality of Service) for various traffic classes. A periodic RAU(request access unit) in upstream direction, includes dedicative request fields for each traffic category within the request slot. The transmission of upstream cell is permitted by the proposed window-based spacing scheme which distributes the requested traffic into several segments in the unit of one spacing window. The delay sensitive traffic source such as CBR or VBR with the stringent requirements on CDV and delay, is allocated prior to any other class. In order to reduce the CDV, so that the permit arrival rate close to the cell arrival rate, Running-Window algorithm is applied to permit distribution processing for these classes. The ABR traffic, which has not-strict CDV or delay criteria, is allocated flexibly to the residual bandwidth in FIFO manner. UBR traffic is allocated with the lowest priority for the remaining capacity. The performance of proposed protocol is evaluated in terms of transfer delay and 1-point CDV according to various offered load. The simulation results show that our protocol has the prominent improvement on CDV and delay performance with compared to the previous protocol.

MAC (Medium Access Control) protocol is necessary for a OLT (Optical Line Termination) to allocate bandwidth to ONUs (Optical Network Units) dynamically in ATM PON (Passive Optical Network) operated in a kind of optical subscriber network having tree topology. The OLT collect information about ONUs and provide all permission with each ONU effectively by means of MAC protocol. Major functions of MAC protocol are composed of the algorism for distributing permission demanded by a ONU dynamically and allocation all permission used in APON properly. Sometimes MAC get to be a element of limiting the whole operation speed and occupy a most frequent operation part of the TC (Transmission Convergence) function module so it have to be designed to guarantee the best quality for each traffic. This paper introduce the way of implementation of a algorism which satisfy all of the upper renditions. This MAC algorism allocate bandwidth according to a number of working ONU and the information of the queue length dynamically and distribute permission for same interval to minimize delay variation of each ONU cell. MAC scheduler for the dynamic bandwidth allocation which is introduced in this paper has look-up table structure that makes programming possible. This structure is very suitable for implementation and operated in high speed because it require very simple and small chip size.
Hepatitis B treatments using immune therapy are gaining interest because of the improvements in dendritic cell performance for antigen presentation, which induces an appropriate immune response and raises patient survival rates. This research aims to produce a significant amount of the HBcAg antigen, which can induce an immune response and have a curative effect on HBV infection. In this study, the HBV subgenotype B3 of the HBcAg gene was used, which is dominant in Indonesia. Further, Lactococcus lactis bacteria was used as the host because of its safety and tightly regulated protein expression. The codon usage for the HBcAg gene was optimized to improve protein expression in L. lactis, which is important because a codon is not random between species. The HBcAg gene is attached to a pNZ8148 plasmid and transformed into the L. lactis NZ3900 expression host. The results confirm that a positive protein band (21 kDa) in two fractions of purified HBcAg was recognized by both western blotting and dot blot hybridization, even if the HBcAg optimized codon has higher GC contents than that suggested for L. lactis expression. Overall, this research strengthens the broad use of L. lactis bacteria for any protein expression, including higher protein expression of codon optimized HBcAg gene compared to non-optimized genes. Furthermore, the improvement in the codon optimization of the HBcAg gene significantly increases the total protein expression by 10-20%, and the expression level of the codon optimized HBcAg increases 1.5 to 3.2-times that of the native HBcAg.
Aptamers are short, chemically synthesized, single-stranded DNA or RNA oligonucleotides that fold into unique three-dimensional structures. In this study, we aim to determine the antibiofilm activity and binding specificity of the six polyclonal DNA aptamers (S15K3, S15K4, S15K6, S15K13, S15K15, and S15K20) on Staphylococcus aureus BPA-12 and Escherichia coli EPEC 4. Aptamer S15K6 showed the highest percentage of antibiofilm activity against S. aureus BPA-12 (37.4%) as shown by the lowest OD570 value of 0.313. Aptamer S15K20 showed the highest percentage of antibiofilm activity against E. coli EPEC 4 (15.4%) as shown by the lowest OD570 value of 0.515. Aptamers S15K13 and S15K20 showed antibiofilm activities against both S. aureus BPA-12 and E. coli EPEC4, and thus potentially have broad reactivity. Furthermore, based on the binding capacity and Kd values from our previous study, the binding specificity assay of selected polyclonal DNA aptamers (S15K3 and S15K15) against S. aureus BPA-12, E. coli EPEC 4, S. aureus BPA-6, S. agalactiae, E. coli MHA-6, and Listeria monocytogenes were performed using qPCR. Aptamers S15K3 and S15K15 showed specific binding to S. aureus BPA-12, E. coli EPEC 4, S. aureus BPA-6, and S. agalactiae, but could not bind to E. coli MHA-6 and L. monocytogenes. Therefore, this study showed that the polyclonal DNA aptamers have antibiofilm activity and were able to bind to S. aureus BPA-12 and E. coli EPEC 4 bacteria.

Type I Interferons (IFNα) are known for their role as biological anticancer agents owing to their cell-apoptosis inducing properties. Development of an appropriate, cost-effective host expression system is crucial for meeting the increasing demand for proteins. Therefore, this study aims to develop codon-optimized IFNα-2b in L. lactis NZ3900. These cells express extracellular protein using the NICE system and Usp45 signal peptide. To validate the mature form of the expressed protein, the recombinant IFNα-2b was screened in a human colorectal cancer cell line using the cytotoxicity assay. The IFNα-2b was successfully cloned into the pNZ8148 vector, thereby generating recombinant L. lactis pNZ8148-SPUsp45-IFNα-2b. The computational analysis of codon-optimized IFNα-2b revealed no mutation and amino acid changes; additionally, the codon-optimized IFNα-2b showed 100% similarity with native human IFNα-2b, in the BLAST analysis. The partial size exclusion chromatography (SEC) of extracellular protein yielded a 19 kDa protein, which was further confirmed by its positive binding to anti-IFNα-2b in the western blot analysis. The crude protein and SEC-purified partial fraction showed IC50 values of 33.22 ㎍/ml and 127.2 ㎍/ml, respectively, which indicated better activity than the metabolites of L. lactis NZ3900 (231.8 ㎍/ml). These values were also comparable with those of the regular anticancer drug tamoxifen (105.5 ㎍/ml). These results demonstrated L. lactis as a promising host system that functions by utilizing the pNZ8148 NICE system. Meanwhile, codon-optimized usage of the inserted gene increased the optimal protein expression levels, which could be beneficial for its large-scale production. Taken together, the recombinant L. lactis IFNα-2b is a potential alternative treatment for colorectal cancer. Furthermore, its activity was analyzed in the WiDr cell line, to assess its colorectal anticancer activities in vivo.
