Effect of trans-10, cis-12 Conjugated Linoleic Acid on Calcium-Dependent Reactive Oxygen Species and Nitric Oxide Production and Nuclear Factor-${\kappa}B$ Activation in Lipopolysaccharide-Stimulated RAW 264.7 Cells (LPS 자극 RAW 264.7 세포에 있어서 칼슘의존성 ROS와 NO 생산 및 NF-${\kappa}B$ 활성에 대한 CLA의 억제효과)

  • Trans-10, cis-12-conjugated linoleic acid (t10c12-CLA) has been shown to participate in the regulation of anti-inflammatory effects. The objectives of this study were to examine the effects of t10c12-CLA on reactive oxygen species (ROS) and nitric oxide (NO) production and nuclear factor-kappaB (NF-${\kappa}B$) activation in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells and to determine whether these effects were associated with change of intracellular calcium ion ($Ca^{2+}$). ROS production was increased in LPS-stimulated RAW 264.7 cells, and this effect was suppressed by 1,2-bis-(o-aminophenoxy) ethane-N,N,N',N'-tetraacetic acid-acetoxymethyl ester (BAPTA/AM), a calcium chelator. t10c12-CLA suppressed ROS production in LPS-stimulated RAW 264.7 cells, which was further more decreased by treatment with BAPTA/AM. These indicated that t10c12-CLA decreases $Ca^{2+}$-dependent ROS production in LPS-stimulated RAW 264.7 cells. Similarly, NF-${\kappa}B$ p65 DNA binding activity and NO production were decreased by treatment with either t10c12-CLA, BAPTA/AM, or t10c12-CLA and BAPTA/AM combination. However, there were no differences between t10c12-CLA and BAPTA/AM treatment in NO production of LPS-stimulated RAW 264.7 cells. These data indicate that t10c12-CLA inhibits the increases in ROS and NO production and the NF-${\kappa}B$ activation in LPS-stimulated condition. These results suggested that CLA exerts potent anti-inflammatory effects by suppression of LPS-induced ROS and NO production, and NF-${\kappa}B$ activationn via $Ca^{2+}$-dependent pathway.

Anti-cholesterol Effects and Molecular Mechanism Study of Mixture of Atractylodes Macrocephala and Amomum Villosum Extracts (백출과 양춘사 추출 혼합물의 항콜레스테롤 효과 및 기전 연구)

  • Atractylodes macrocephala (AM) and Amomum villosum (AV) are the most common herbs in Korean Medicine to treat digestive diseases. In this study, we investigated the cholesterol lowering effects of mixtures of AM and AV extracts on high cholesterol diet (HCD) induced dyslipidemia mouse model. We classified animals into six different groups; Group 1: Normal diet, Group 2: HCD, Group 3: AV extracts : AM extracts (1:1) (200 mg/kg) + HCD, Group 4: AV extracts : AM extracts (1:2) (200 mg/kg) + HCD, Group 5: AV extracts : AM extracts (1:3) (200 mg/kg) + HCD, Group 6: Simvastatin 40 mg/kg + HCD. After 4 weeks of oral administration of respective drugs, we checked body, liver and epididymal fatweights along with liver and serum triacylglyceride (TG) concentration, total and low density lipoprotein (LDL) cholesterol in serum. Moreover, 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) reductase (HMGCR), LDL receptor (LDLR), and sterol regulatory element-binding protein 2 (SREBP2) were detected by RT PCR or western blot analysis. The overall results showed that mixtures of AM and AV extracts inhibited HCD-induced increases of total cholesterol and LDL cholesterol in serum. Those effects seem to be caused by AM and AV extracts through inhibition of HMGCR expression. And thus blood cholesterol is induced into the liver by increasing LDLR expression, which is regulated by SREBP2 transcrption factor. The cholesterol lowering effects and mechanism of mixtures of AM and AV extracts was similar to the statin. We have identified the potential mixtures of AM and AV extracts as a new treatment for dyslipidemia.

Developing an Embedded Method to Recognize Human Pilot Intentions In an Intelligent Cockpit Aids for the Pilot Decision Support System

  • Several recent aircraft accidents occurred due to goal conflicts between human and machine actors. To facilitate the management of the cockpit activities considering these observations. a computational aid. the Agenda Manager (AM) has been developed for use in simulated cockpit environments. It is important to know pilot intentions performing cockpit operations accurately to improve AM performance. Without accurate knowledge of pilot goals or intentions, the information from AM may lead to the wrong direction to the pilot who is using the information. To provide a reliable flight simulation environment regarding goal conflicts. a pilot goal communication method (GCM) was developed to facilitate accurate recognition of pilot goals. Embedded within AM, the GCM was used to recognize pilot goals and to declare them to the AM. Two approaches to the recognition of pilots goals were considered: (1) The use of an Automatic Speech Recognition (ASR) system to recognize overtly or explicitly declared pilot goals. and (2) inference of covertly or implicitly declared pilot goals via the use of an intent inferencing mechanism. The integrated mode of these two methods could overcome the covert goal mis-understanding by use of overt GCM. And also could it overcome workload concern with overt mode by the use of covert GCM. Through simulated flight environment experimentation with real pilot subjects, the proposed GCM has demonstrated its capability to recognize pilot intentions with a certain degree of accuracy and to handle incorrectly declared goals. and was validated in terms of subjective workload and pilot flight control performance. The GCM communicating pilot goals were implemented within the AM to provide a rich environment for the study of human-machine interactions in the supervisory control of complex dynamic systems.

  • As the digital system with high sampling rates is required to process numerous data, it is difficult to realize the real time processing for this system. By using the nonlinear energy tracking signal operator, in this paper four kinds of AM/FM demodulators are proposed and designed by the undersampling scheme of decreasing the sampling rate. Particularly, the undersampling frequency and 3dB bandwidth are controllable of these systems and their conditions are respectively. Through the analysis of the designed detectors, useful results are obtained in respect to characteristics and errors.

  • Some of modern communication systems make use of broadband digital modulation with 5 MHz and 10 MHz. In the broadband communication systems, the nonlinear properties of power amplifier have a great effect on all systems. This paper analyzed the ACPR characteristics of WLL reverse link power amplifier with the varying AM and PM nonlinearities and suggests the required backoff from 1 dB compression point to satisfy the ACPR specifications. Also the comparions between the measured and the simulated characteristics are presrnted.

Sequence Analyses of PCR Amplified Partial SSU of Ribosomal DNA for Identifying Arbuscular Mycorrhizal Fungi in Plant Roots

  • The genomic DNAs were extracted from roots of Glycine max and Sorghum bicolor, and compared with those from spores of two arbuscular mycorrhizal(AM) fungi, Glomus mosseae and Scutellospora heterogama. The partial small subunit(SSU) of ribosomal RNA genes were synthesized and amplified by polymerase chain reaction with the fungal specific primers, AM1 and NS31. By the recent molecular techniques, the presence of another AM fungal DNA were confirmed in the roots of two plants, and three sequences of rDNA fragments amplified were identified to be close to those of G. caledonium, G. fasiculatum and G. proliferum. The two AM fungi, both, were found to colonize at the cortical layers of plant roots collected in the fields, together.

Determination of methamphetamine in the hair of Korean abuser by CG/MS (GC/MS에 의한 한국인의 모발중 Methamphetamine검출)

  • A sensitive method for the determination of methamphetamine(MA) and amphetamine(AM) in hair was developed by gas chromatography/mass spectrometry using stable isotope-labeled internal standards, amphetamine-d$_{5}$ and methamphetamine-d$_{5}$. Hair sample was washed with MeOH, incubated with MeOH(I% HCI) overnight at $37^{\circ}C$ while stirring and extracted using solid phase extraction column on a vacuum manifold. The extract obtained was pentafluoropropionated, and applied to GC/MS. The calibration curves of MA and AM were linear from 2.5 to 250 ng (r>0.99 for both). The limit of detection was 0.1 ng/mg in hair and cut-off level was set at 0.25 ng/mg for both. Hair samples of 27 MA abusers showed positive results in the range 0.7 to 106.8 ng/mg. AM, its metabolite, was detected in 20 out of 27 samples. The ratio of MA versus AM was 4.6~38.3 in specimens. Hair analysis for methamphetamine by GC/MS is an effective method for identifying long-term drug abusers.

  • The streptococcal plasmid pAM$\beta_{1}$ (erythromycin resistance)was transferred via conjugation from Streptococcus faecalis DS 5 to Lactobacillus casei YIT 9018 by a filter mating method. The transfer frequency depended greatly on the type, pore size and side(front or back) of membrane filter. Water passing through a membrane under reduced pressure induced very tight contact between the cells, increased the transconjugation frequency about 1.3 to 37-fold when Millipore membrane filter (0.45.$\mu$m, front side up) was used.

A Study on Bi-metal 3D printing Technology Development based on Laser Technology

  • Additive manufacturing(AM) can create complex shapes directly in 3D CAD models with internal geometry compared to conventional subtraction manufacturing. AM technology has the advantage of adopting various materials as well as the reduction of material. However, the high cost of AM is still a significant barrier preventing the wider adoption of AM in industries. This paper analyzes the technical application cases for solving these entry barriers and proposes a bi-metal 3D printing technology as an anticipated application to overcome the difficulty. The paper investigates the complications for current 3D metal printing technology to conduct bi-metal 3D printing and addresses ongoing solution research based on laser technology.
