Effect of Liver Damage by Combined Administration of Ethanol and Xylene in Rats (흰쥐에게 Ethanol과 Xylene 병행 투여시 간손상에 미치는 영향)

  • To evaluate an effect of liver damage on ethanol and xylene exposure, experiments on normal male rats of the S-D strain were performed in 4 groups. The biochemical results suggest that the ethanol group had significantly higher levels of AST, ALT, LDH and also, the xylene group had notably higher levels of AST, ALT, LDH along with MMHPA than those of the control groups. The levels of AST, ALT and LDH in the ethanol+xylene group were drastically higher than those in the control, ethanol and xylene groups. But, there were significantly lower ALP levels in the xylene and ethanol+xylene groups than both the control and ethanol groups. The histological features of rat livers treated with alcohol, or xylene proved to be normal. But the rat livers treated with ethanol+xylene showed mild to moderate necrosis and inflammation as well as minimal fatty changes. The results in this experiment suggest that liver functions decreased when medicated together with xylene and ethanol rather than solely with xylene.

The Effect of He-Ne IR Laser Therapy on The Healing of Acute Septic Knee Arthritis in Rat (흰쥐의 화농성 슬관절염 치유에 He-Ne IR레이저 처치의 효과)

  • The purpose of this study was to find out the effect of He-Ne, IR laser therapy for 4 and 10 minutes on healing of septic arthritis. Sprague-Bawley rats (212.1$\pm$10 g) were randomly assigned to one control and three arthritis groups. Septic arthritis groups were divided into arthritis control, 4 or 10 minute laser therapy groups. Arthritis was experimentally induced by intraperitoneally administration of staphylococcus aureus in citrate buffer(pH 4.3). These result were obtained as follows: AST, LDH, ALP, CK activities and IgG concentration in 10 minute laser group were signifantly increased compare with those in the arthritis group. AST, LDH and CK activities in 4 minute laser group were significantly decreased compare with those in the arthritis group. We think that laser therapy in 4 minute may effective on the healing of septic arthritis. And we will have to study of the effect of laser therapy on healing mechanism, appropriate time of septic arthritis.

Effect of Kojinyumia(固眞飮子) on Galactosamie Induced hepatoxicity on Rats (고진음자(固眞飮子)가 galactosamine으로 유발한 흰쥐의 간중독에 미치는 영향)

  • The purpose of this study is to observe the protective effect of Kojinyumja on serum reaction and hepatic tissue in galactosamine treated rats. In this study, the experimental rats divided four group(Normal group, Control group, Sample A group, and Sample B group) : Under the same condition, normal and control group were administered water, sample A, B group were administered Kojinyumja for 8days. And then, both control group and Sample B group were injected to abdomen with galactosamine for 1day. The rates of of glutathione, lipid peroxide, GOT, GPT, ${\gamma}$-GTP, ALP, LDH, and contents of bile acid level were measured. The results are as follows : The glutathione rate significantly increased in sample group, the others (lipid peroxide, GOT, GPT, ${\gamma}$-GTP, ALP, LDH, bile acid) significantly decreased in sample group.

Effects of Chungganhaeju-tang(Qingganjiejiu-tang) on Alcohol Metabolism and Alcoholic Liver Damages (청간해주탕이 알코올대사 및 손상간에 미치는 영향)

  • Objectives: This experiment was conducted to verify the effects of Chungganhaeju-tang(Qingganjiejiu-tang) on the alcohol metabolism and liver functions, by measuring the activity levels of ADH and ALDH, as well as glucose, triglyceride, and BUN. Damage of the liver cells caused by alcohol was determined through the examination of serum levels of AST, ALT, ALP, LDH, and uric acid. Methods: Sprague-Dawley rats were used in this experiment and the rats were divided into control and experiment groups. Chungganhaeju-tang(Qingganjiejiu-tang) extract was orally administered in the experiment group for three weeks. Each group was further classified into two sub-groups, and control group's blood was taken without oral ingestion of alcohol, while the experiment group' s blood was withdrawn after ingestion of alcohol. Evaluation of damage level was done considering the presence of extract and alcohol. Results: In this experiment, Chungganhaeju-tang(Qingganjiejiu-tang) significantly suppressed the activity of ADH which is a precursor enzyme of acetaldehyde, but didn't cause significant changes in the activity of ALDH which is a catabolic enzyme. Decreased glucose level due to alcohol consumption was recovered back to the normal level and increased levels of triglyceride, BUN, AST, ALT, ALP, LDH, and uric acid were significantly reduced. Conclusions: These experiment results suggest that Chungganhaeju-tang(Qingganjiejiu-tang) inhibits the formation of acetaldehyde in the metabolism of alcohol, and affects the recovery of weakened liver functions due to alcohol.

  • Background and Objective : Recently it is true that increasing interest in Oriental medicine and usage of herbal medicine. On the other hand, there are many concerns about the stability of herbal medicine. So, this study is carried out to see whether or not the treatment with Sanghan-Bang(傷寒方) is effective on the liver function besides its effectiveness in treating facial palsy. Methods : We examined serum TP, Albumin, TB, AST, ALT, ALP, LDH, ${\gamma}$-GGT levels, and facial palsy states in 37 patients with facial palsy who had admission treatment in an oriental medical hospital. Results and Conclusion : 1. We found the levels of TP, albumin, TB, and ${\gamma}$-GGT had decreased statistically significant. Also, AST, ALT, LDH and ALP levels had decreased however it was not statistically significant. 2. We analyzed the herbal group that may be efficient to the liver function; Daehwang-Jae(16 case), Injin-Jae(12 case), and Chija-Jae(6 case) in this order. 3. From the eight cases using oriental medicinal concoctions using one of Daehwang, Injin-Jae or Chija as their main component, the liver function test results displayed decreasing values, re-entering into normal LFT ranges. 4. As we compared the admission House-Brackmann Scale with discharge, the treatment was efficient and statistically significant.

Effects of Acupuncture at Taekjung and Ungyo$(GV_{28})$ Bloodletting on Liver Function (택중침자(澤中鍼刺) 및 간교$(GV_{28})$사혈(瀉血)이 간기능(肝機能)에 미치는 영향(影響))

  • Effects of acupuncture at Taekjung and Ungyo($GV_{28}$) bloodletting on serum glutamic oxaloacetic transaminase (GOT) activity, glutamic pyruvic transaminase activity(GPT), total Protein, Albumin, Alkalin Phosphatase(ALP), ${\gamma}$-GT, lactic dehydrogenase (LDH), total cholesterol and triglyceride were determined. 1) In the Ungyo bloodletting group, GOT, GPT, ALP, ${\gamma}$-GT and LDH activity showed a tendency to decrease compared to those of $CCl_4$ only group and Taekjung acupuncture group. 2) Serum albumin content showed a tendency decrease by treatment of $CCl_4$ and to increase by treatment of Ungyo bloodletting. 3) The values of serum total protein, total cholesterol and triglyceride showed no significant difference among all the treatment groups. 4) All the biological values showed no significant difference between the $CCl_4$ only group and Taekjung acupuncture group except the values of albumin. Results from this study indicate that the Ungyo bloodletting showed the possibility of liver function recovery in liver-harmed rats by $CCl_4$.

Anti-stress Effect of Cholic acid Derivatives in Restraint Stress Induced Rats (구속스트레스를 가한 흰쥐에서 Cholate류의 항스트레스 작용에 관한 연구)

  • This study was done to investigate whether cholic acid derivatives have anti-stress activity and what is a cause of this anti-stress effect. Seven cholic acid derivatives (cholic acid, taurocholic acid, ursodeoxycholic acid, tauroursodeoxychoic acid, chenodeoxy cholic acid, dehydrocholic acid, hyodeoxycholic acid) were used, silymarin and valproic acid were used as positive controls. Stress was induced by restraint immobilization technique plus water immersion (24hrs) and adrenal weight, spleen weight, adrenal ascorbic acid, serum cholesterol, lactate dehydrogenase (LDH), alkaline phosphatase (ALP), adrenal cholesterol, glucose and corticosterone levels were measured as stress indicators. Most cholic acid derivatives markedly decreased the adrenal weight, and TUDCA and DHCA increased the spleen weight. The restraint stress induced increments in serum LDH, ALP and cholesterol were attenuated by most cholic acid derivatives. Cholic acid, taurocholic acid and tauroursodeoxycholic acid only increased the content of adrenal ascorbate. While valproic acid showed an inhibitory effect against stress, silymarin did not. Our findings suggest that most cholic acid derivatives have anti-stress effect and that their anti-stress effect is, in part, related to choleretic activity.

Effects of Turmeric (Curcuma longa L.) Supplementation on Blood Urea Nitrogen and Enzyme Activities in Dyslipidemic Rats (울금(Curcuma longa L.)이 이상지질혈증 흰쥐의 혈액요소질소(BUN) 및 효소활성에 미치는 영향)

  • The purpose this study was to investigate the influences of 5% turmeric (Curcuma longa L.) supplementation on enzyme activities such as aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), amylase, lipase and catalase in serum of dyslipidemic rats. Sprague-Dawley (SD) rats (24 male) were divided into four groups, namely the ND group (normal-nondyslipidemic diet), NT group (normal-nondyslipidemic diet+5% turmeric), DD group (control-dyslipidemic diet), and DT groups (dyslipidemic diet+5% turmeric). Serum concentrations of blood urea nitrogen (BUN), creatinine and uric acid were significantly decreased (p<0.05) by turmeric supplementation diet. The activities of AST, ALT, ALP, LDH, amylase and lipase in sera of turmeric diet group were significantly decreased (p<0.05). The catalase activity in serum of turmeric supplementation group was significantly increased than dyslipidemic diet (p<0.05). In vivo experiment with dyslipidemic rats showed that ingestion of turmeric were effective in kidney and hepatic functional enzyme activities. Which suggests that turmeric material could be used for further studies as a potential source for nutraceutical foods.

Effects of Antarctic Ocean Krill Meal with NaF Oral Administration on Serum and Tissues in Rats (남극해 크릴 분말과 NaF 처리가 흰쥐의 혈청 및 조직에 미치는 영향)

  • The purpose of this study was to probe the influences of krill (Euphausia superba) meal with NaF oral administration on a dose-effect relationship between fluoride levels of krill meal and serum enzyme activity such as alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) in rats fed experimental diets for 5 weeks. There were no significant decreases in the activities of ALP, AST, ALT, and LDH in sera among krill meal diet groups (KF10, KF20, KF30). However, these groups were significantly (p<0.05) lower enzyme activities than normal diet (ND) plus NaF 10 mg group (NF). The fluoride levels of serum and organ tissues (liver, brain, heart, lung, kidney) in NaF 10 mg groups (NF, KF10, KF20, KF30) were significantly increased by adding krill meal in comparison with normal diet group. The results indicate that a difficult to found toxicity to the tissues from krill meal diet groups.

The Beneficial Effect of Melatonin for Toluene Hepatotoxicity in Rats

  • Toluene is mainly metabolized in liver by oxidative pathway. Oxigen free radicals occur through the process of toluene metabolism Therefore it causes tissue and cell min by the oxygen free radicals from the metabolism of toluene. Melatonin acts as a highly efficient free radical scavenger that protects cells from damage by oxygen free radicals. To test this hypothesis, toluene hepatotoxicity was induced by an abdominal injection of toluene. To see if the melatonin protects the rat's liver, melatonin was administrated orally, at the time of each toluene injection. Aspartate aminotransferase(AST), alanin aminotransferase(ALT), latic dehydrogenase(LDH) and alkaline phosphatase(ALP) levels in serum were measured to estimate hepatic function. Malondialdehyde(MDA), which gives an indirect index of oxidative injury was also measured. Hippuric acid is the last metabolic Production of toluene was measured by HPLC. There were significantly higher in AST, ALT, LDH, MDA and hippuric acid in toluene group, but there were no significant difference in melatonin group except ALT and hippuric acid. There was significantly lower in ALP level in toluene group, but there was no significant difference melatonin group, suggesting a significant hepatotoxicity due to oxygen free radicals through the process of toluene metabolism Melatonin treatment significantly protected hepatic function and free radical-mediated injury in the liver against toluene-induced changes. Accordingly, this study shows that melatonin is helpful in protecting liver injury by acute toluene intoxication.
