Blood Pressure Reduction of SHR Treated with EA-contaning Lentinus edodes Mycelial Solid Culture Extract (Eritadenine 함유 표고버섯균사체 고체배양 효소추출물(EA-LEMSCEE)의 spontaneous hypertension rat 혈압감소)

  • Eritadenine (EA), derived from Lentinus edodes (LE), reduced low-density lipoprotein (LDL), triglyceride (TG), and phospholipids in bloods, and fatty acid depositions in animals and humans. Previously, we reported that EA inhibited angiotensin-converting enzyme (ACE) activity in vitro. Now, we report that EA reduced blood pressures in spontaneous hypertension rats (SHR). EA-containing LE mycelial culture enzyme extract (EA-LEMSCEE) was prepared from LE mycelial solid cultures and the hot-water extract of LE fruit bodies. Both EA and EA-LEMSCEE inhibited ACE activity in immortalized human umbilical endothelial cells (EA.hy926). EA-LEMSCEE treatments (7.5 mg/kg, 22.5 mg/kg) significantly reduced systolic and diastolic blood pressure in SHR. At five weeks of treatment, EA-LEMSCEE treatment significantly reduced systolic and diastolic blood pressure, similar to the positive control (captopril, CP; 4 mg/kg) treatment. In addition, the LEMSCEE without EA decreased systolic and diastolic blood pressures compared to the control, but not significant. EA-LEMSCEE decreased renin and ACE activities, and angiotensin II (Ang II) contents in SHR compared to the control. After five weeks of treatment, the effect of EA-LEMCEE was similar to that of CP. These results indicate that EA and EA-LEMSCEE reduce blood pressure by inhibiting the renin and ACE activity of SHR. Furthermore, these results imply that EA or EA-LEMSCEE could be used as an antihypertension agent in humans.

  • The present study was conducted to elucidate the body modulating function of fermented seafood products. Angiotensin converting enzyme (ACE) acts in blood pressure regulation, converting angiotensin I to the potent vasoconstrictor angiotensin II and inactivating the vasodilator bradykinin to raise blood pressure. Salted and fermented anchovy which is one of the traditional fermented seafood in Korea was tested for inhibitory activity against ACE. ACE inhibitory activity of salted anchovy during the period of fermentation was increased with the elapse of fermentation days until fermentation of 60 days, but thereafter decreased inversely. When the fermented product was extracted with $50\%$ ethanol, the ACE inhibitory activity was the highest. And the ACE inhibitory activity was proportion to the content of $50\%$ ethanol soluble peptide-nitrogen of the fermented product. From the profiles of gel permeation chromatography on a Bio-Gel P-2 of $50\%$ ethanol soluble fraction obtained from salted and fermented anchovy fermented for 60 days at an ambient temperature, the higher activity fractions were C'($IC_{50}=97{\mu}g\;protein/ml$) and D'($IC_{50}=65{\mu}g\;protein/ml$). Amino acid analysis showed that the large quantify of threonine, glutamic acid, lysine for C' and serine, proline for D', respectively.

Antihypertensive Effect and Safety of Imidapril on the Patient with Essential Hypertension (본태성 고혈압에 대한 Imidapril의 강압효과 및 마른기침 발현율에 대한 고찰)

  • Imidapril(Tanatril$^{(R)}$), a newly developed ACE inhibitor, has been used to treat hypertension and congestive heart failure. This study was designed to assess the antihypertensive effect and safety of Imidapril(Tanatril$^{(R)}$) in patients with essential hypertension. 5-10mg of imidapril(Tanatril$^{(R)}$) was administered once a day in 30 patients with essential hypertension and followed up for 8 weeks. We tested the drug's effectiveness, safety, and the incidence of imidapril induced dry coughs. After 8 weeks of treatment with imidapril, 76.2%(16/21) of patients showed lowered blood pressure and 47.6% showed normal blood pressure. The overall incidence of adverse effects was 33.3%(7/21), and among these adverse effects, dry cough was shown in only 9.5%. Thus, we concluded that imidapril(Tanatril$^{(R)}$) is as safe and effective as other ACE inhibitors, especially with imidapril showing very little incidence of dry cough compared to other ACE inhibitors.

  • This study was carried out to understand hyrolytic characteristics of $\beta-casein$ by enzyme in goat milk and to measure the inhibition effect of the ACE of the hydrolysate. In order to conduct the experiment, $\beta-casein$ of goat milk was separated using Mono S HR 5/5, a cation exchange column. The separated $\beta-casein$ was treated with trypsin of animal hydrolysis enzymes, in an effort to verify the characteristics of hydrolysis. The inhibition activity of ACE was measured and the results are as follows. By analyzing the hydrolysate separated from the trypsin-processed $\beta-casein$ of goat milk, the inhibition effect of the ACE was measured trypsin-hydrolyzed $\beta-casein$ demonstrated a $25.36\pm0.79\%$ of inhibition effect and the $IC_{50}$ of the hydrolysate from the trypsin-processed $\beta-casein$ reached $308.7\pm2.77({\mu}g/mL)$.

Akebiae Caulis Inhibits Oxidative Stress through AM PK Activation (AMPK 활성화를 통한 목통의 항산화 효과)

  • Akebiae Caulis is a galenical originated from Akebia quinata Decaisne species. It is commonly used in the treatment of oposiuria, inflammation, nociceptive and fever. Here, we investigated the effect of Akebiae Caulis extract (ACE) to protect hepatocyte against the malfunction of mitochondria and apoptosis. Arachidonic acid (AA)+iron promoted excessive reactive oxygen species (ROS) production and exerted a deleterious effect on mitochondria. Treatment with ACE protected hepatocytes from AA+iron-induced cytotoxicity, as shown by alterations in the protein levels related with apoptosis such as poly(ADP-ribose) polymerase, pro-caspase 3, Bcl-XL and Bcl-2. Moreover, AA+iron-induced $H_2O_2$ production, GSH depletion and mitochondrial dysfunction were alleviated by ACE pretreatment. As a potential molecular mechanism for the ACE-mediated cytoprotection, phosphorylation of AMP-activated protein kinase (AMPK), a key regulator in determining cell survival or death, was increased by ACE. Moreover, ACE treatment enhanced inactive phosphorylation of glycogen synthase kinase-$3{\beta}$ ($GSK3{\beta}$), downstream substrate kinase of AMPK. More importantly, ACE prevented a decrease in the $GSK3{\beta}$ phosphorylation derived by AA+iron, which might contribute to mitohondiral protection and cell survival. To further identify essential compounds in Akebiae Caulis for the protection of AA+iron-mediated cytotoxicity, we found that betulin in combination with hederagenin protected from AA+iron-induced mitochondrial dysfunction. Betulin+hederagenin treatment also increased inactive phosphorylation of $GSK3{\beta}$ in common with ACE. These results suggest that ACE protected hepatocytes against oxidative stress and mitochondrial dysfunction, which is mediated with inactive $GSK3{\beta}$ phosphorylation downstream of AMPK.

Studies on antioxidant, anti-inflammatory and tyrosinase inhibitory activity of Ganoderma lucidum fermented Artemisia capillaris extract (인진쑥 영지버섯 균사 발효 추출물의 항산화, 항염 및 티로시나제억제 활성 연구)

  • This study investigated whether Ganoderma lucidum (Y2)-mediated fermentation of Artemisia capillaris extract (ACE) could synergistically enhance its antioxidant, anti-inflammatory, and tyrosinase-inhibiting activities. Both G. lucidum extract and fermented ACE exhibited 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging ability, but with poorer efficacy than ACE (even at a low ACE concentration). Viability of RAW264.7 macrophages was significantly reduced in the presence of ACE (150 mg/mL and above). However, this effect was greatly mitigated upon G. lucidum-mediated ACE fermentation. Additionally, relative to the same concentration ($25{\mu}g/mL$) of G. lucidum mycelial extract, ACE exhibited an improved ability to significantly inhibit RAW264.7 macrophage nitric oxide (NO) production. Finally, relative to the same concentration ($200{\mu}g/mL$) of a positive control (arbutin), fermented ACE exhibited an approximately 3.66 times higher capacity for tyrosinase inhibition. These results suggest that G. lucidum-fermented ACE possesses enhanced tyrosinase-inhibiting activity and may be of utility as a skin-lightening agent.

A Study on the analysis of constitutional genes (체질유전자 분석에 관한 연구)

  • There have been several the reports that the mechanism of Sasang consititution might be understood in the level of genes. Previous study of the authors showed that HLA types and constitutional information had significant relationships. One hundred subjects who showed Taeum characteristics were selected in the present study. HLA-A, HLA-B, HLA-DRB1, ACE, ${\beta}-IIAR$, ${\beta}-IIIAR$, UCP-1, and ALDH2 polymorphisms were analyzed. Also, ACE, ${\beta}-IIAR$, ${\beta}-IIIAR$, UCP-1, and ALDH2 analyses were performed on the 100 samples of previous study who showed Taeyang characteristics. Despite of several significant differences of HLA allele frequencies between Taeum-inclined group and normal control group, this significance level was not sufficient ro support the association between constitution and HLA genes, because of the raised alpha error rate. The polymorphisrns of ACE, ${\beta}-IIAR$, ${\beta}-IIIAR$, UCP-1, and ALDH2 genes did not show relationship between Taeyang-inclined and Taeum-inclined groups, whereas BMI showed difference between Taeyang-inclined and Taeum-inclined groups. ALDH2 in Taeyang-inclined group confirmed the protective role of ALDH2*2 allele against alcoholism.

Inhibitory Effect against Angiotensin Converting Enzyme and Antioxidant Activity of Panax ginseng C. A. Meyer Extracts (인삼 추출물의 Angiotensin Converting Enzyme 저해 효과와 항산화 활성)

  • The study was performed for elucidating angiotensin converting enzyme (ACE) inhibitory activity and comparing antioxidative activity of Panax ginseng extracts prepared at different conditions. Total phenolic content, inhibitory activity on ACE and antioxidative effects were tested on 10 ethanolic extracts and correlation coefficient between total phenolic content and physiological activity was calculated. Yield and total phenolic content of 50% ethanolic extract prepared at $85^{\circ}C$ exhibited the highest value as 42.52% and 0.82%, respectively. Among the fractions obtained from 50% ethanolic extract prepared at room temperature, water fraction showed the highest value in yield as 72.08% and ethyl acetate fraction did in total phenolic content as 6.59%. In the test on ACE inhibitory activity, 50% ethanolic extract obtained at room temperature indicated the strongest effect of 93.8% which was higher than 85.2% of commercialized ACE inhibitor and solvent fractions showed potent inhibitory activity in order of hexane fraction, diethyl ether fraction, ethyl acetate fraction, butanol fraction and water fraction at concentration of $4000{\mu}g/ml$. 50% Ethanolic extract prepared at $85^{\circ}C$ had the most potent inhibition effect on human LDL oxidation as 78.2% at $200{\mu}g/ml$ and the other extracts also did above 60%. Diethyl ether fraction and ethyl acetate fraction showed strong inhibition activity $(34.38%{\sim}78.13%)$ on LDL oxidation at concentration of $10{\sim}200\;{\mu}g/ml$. From the statistical analysis via SAS program, correlation coefficient between total phenolic content and ACE inhibitory effect was 0.6353 at P<0.05. Conclusively, this report showed that the most efficient extraction condition for elevating inhibitory activity on ACE and LDL oxidation, phenolic content and yield from Panax ginseng was 50% ethanol extraction at room temperature or high temperature condition. And Panax ginseng would be used for preventing hypertension or atheroscrelosis for man via inhibitory action on ACE and LDL oxidation.

The Relationshin between ACE Inhibitory Activity and Degradations of Sulfur Containing Materials in Dolsan Leaf Mustard Juice

  • This Study was tarried out to investigate the relationship ACE inhibitory activity and degradations of sulfur containing materials in Dolsan leaf mustard juice (DLMJ). The changes of sulfur containing materials which were treated with autolysis, myrosinase, ascorbate and papain were studied, as well as the changes of ACE inhibitory activity in DLMJ. At $37^{\circ}C$, sulfur contain-ing materials by autolysis decreased most rapidly from $0.43\%$ to $0.13\%$ in the second day. Conversely. ACE inhibitory activity increased most from $66\%$ to $87\%$. in the second day at $37^{\circ}C$. As myrosinase concentrations increased more, sulfur containing materials in DLMJ decreased more. The ACE inhibitory activities at 0, 0.5, 1, 2, and 4 Units of myrosinase for 240 min later were 70, 74, 75, 82, and $85\%$, respectively. At 1 mM ascorbate. concentrations of Sulfur containing materials in DLMJ decreased more significantly on the second day than on the other days. At 1 mM ascorbate for 6 days, ACE Inhibitory activity reached a maximum of about $92\%$. And, an increase of papain concentration was noted in accordance with a decreased sulfur containing materials. The maximum rate of AEC inhibitory activity at control, 3, 6, and 12 Units of papains treatments was shown as 70, 70, 75, and $78\%$ at 60 min, respectively. These results suggested that the degradation of sulfur containing materials led to the increase of ACE inhibitory activity. Consequently, it was suggested that ACE inhibiting was significantly related to the degradatives of sulfur containing materials.

Association between the Angiotensin-converting Enzyme Gene Insertion/Deletion Polymorphism and Essential Hypertension in Young Pakistani Patients

  • Several studies have demonstrated the importance of angiotensin-converting enzyme (ACE) insertion (I)/deletion (D) polymorphisms in the pathogenesis of hypertension. This study sought to determine the association between the ACE I/D polymorphism and essential hypertension in young Pakistanis. The frequency of the ACE I/D polymorphism was established by a comparative cross-sectional survey of Pakistani patients suffering from essential hypertension and ethnically matched normotensive controls. Samples were collected from tertiary care hospitals in northern Pakistan. Hypertensive individuals were defined as those with a systolic blood pressure > 140 mmHg and/or diastolic blood pressure > 90 mmHg on three separate occasions, or those currently receiving one, or more, anti-hypertensive agents. DNA samples obtained from hypertensive (n=211) and normotensive (n=108) individuals were typed by PCR. The frequency of the ACE I/I genotype was significantly higher in hypertensive patients, aged 20-40 years, than in normotensive controls of the same age group ($\chi^2$ = 4.0, P = 0.041). Whereas no overall significant differences were observed between the I/I, I/D and D/D ACE genotypes (One way ANOVA, F=0.672; P=0.413). The association between the ACE I/I genotype and essential hypertension in individuals aged $\leq$ 40 years suggests that ACE has a role in early onset essential hypertension in Pakistan.
