Anti-hypertensive Effects of DHP1501, Ethanolic Extracts from Eleutherococcus sessiliflorus Fruits, via Inhibition of Angiotensin Converting Enzyme and Activation of Endothelial Nitric Oxide Synthase (오가피열매 주정추출물, DHP1501의 ACE 억제 및 eNOS 활성화를 통한 항고혈압 효능)

  • The fruits of Eleutherococcus sessiliflorus (Rupr. & Maxim.) S. Y. Hu (Araliaceae), as edible fruits, were traditionally used for ingredients of wine or tea in Eastern Asia. In addition to, the fruits of E. sessiliflorus were known for having antioxidant and anti-inflammatory effects. Recently, we investigated that the ethanolic extracts of E. sessiliflorus fruits (DHP1501) have effects on hypertension via vasorelaxation and decrease of blood pressure. In the present study, we investigated that the gene and protein expression levels of endothelial nitric oxide synthase (eNOS) was increased by treatment of DHP1501 in HUVECs. Moreover, we confirmed the angiotensin converting enzyme inhibitory activity of DHP1501 through in vitro tasks. Therefore, DHP1501 could be a candidate of functional food for alleviating hypertension.

  • Milk-derived bioactive peptides have been found to exhibit various physiological activities such as angiotensin-converting enzyme (ACE) inhibitory, antibacterial, and antioxidative effects. Bioactive peptides can be used in the formulation of functional foods, nutraceuticals, and natural drugs because of their beneficial effects. However, the degree of variability in the composition, functionality, and sensory properties of such peptides has greatly limited their use in the food industry. In this review, we discuss the main peptides obtained from milk proteins and summarize findings from previous studies on the production and biological activities of these peptides. In addition, we compare the methods used to separate and identify the structure of the bioactive peptides and highlight current investigations into engineering and implementation of technologies that would allow more efficient isolation of bioactive peptides for functional food production. To improve human health, further molecular biology studies will also be required to elucidate the complex network of interactions between food microorganisms and the digestive system.

  • In this study, we synthesized the aminoclay (AC) with magnesium ions and 3-aminopropyl triethoxysilane (APTES). At the same time, propolis intercalated aminoclay (PIAC) and coptis extract intercalated aminoclay (CIAC) were synthesized by intercalating natural chemicals between clay sheets. Clay synthesis and natural chemical intercalation were confirmed through SEM, particle size analyze, FT-IR, TGA and XRD. In particular, the characterization of intercalation of natural chemicals was determined by analyzing the interlayer distance from XRD data. The antimicrobial property of PIAC and CIAC was checked by minimum inhibitory concentration (MIC) test and increased compared with that of the pristine aminoclay (AC).

  • To enhance application of FPC in food industry, salmon FPC hydrolysates were produced and their functional properties were investigated. Hydrolysates of salmon FPC showed high solubilities ranged from approximately $80\~95\%$. Emulsifying activity index (EAI) values of hydrolysates were higher than that of egg white, however sugar ester showed the highest EAI value of 32.5. Though sugar ester had the best emulsifying stability value $({\Delta}EAI)$ of 9.65, hydrolysates showed better ${\Delta}EAI$ value than that of egg white. Foam Activity of hydrolysates was predominantly better than those of controls and also foam stability value showed similiar trend. Osmolality of hydrolysates was increased with the increase of degree of hydrolysis (DH) and concentration of protein, but it showed very lower osmolality than that of NaCl. Flow property of hydrolysates showed different upward-downward curves, and hysteresis loop increased with the increase of DH. ACE inhibitory activity showed similiar value ranged from $21.1\~24\%$ at all DH values.

  • The biological activities of macrophage migration inhibitory factor (MIF) might be mediated through a classical receptor-mediated or non-classical endocytic pathway. JAB1 (C-Jun activation domain-binding protein-1) promotes the degradation of the tumor suppressor, p53, and the cyclin-dependent kinase inhibitor, p27. When MIF and JAB1 are bound to each other in various intracellular sites, MIF inhibits the positive regulatory effects of JAB1 on the activity of AP-1. The intestinal parasite, Anisakis simplex, has an immunomodulatory effect. The molecular mechanism of action of As-MIF and human JAB1 are poorly understood. In this study, As-MIF and hJAB1 were expressed and purified with high solubility. The structure of As-MIF and hJAB1 interaction was modeled by homology modeling based on the structure of Ace-MIF. This study provides evidence indicating that the MIF domain of As-MIF interacts directly with the MPN domain of hJAB1, and four structure-based mutants of As-MIF and hJAB1 disrupt the As-MIF-hJAB1 interaction.

Biological Treatment of Nutrients and Heavy Metals in Synthetic Wastewater Using a Carrier Attached to Rhodobacter blasticus

  • The removal efficiencies of nutrients (N and P) and heavy metals (Cu and Ni) by Rhodobacter blasticus and R. blasticus attached to polysulfone carriers, alginate carriers, PVA carriers, and PVA + zeolite carriers in synthetic wastewater were compared. In the comparison of the nutrient removal efficiency based on varying concentrations (100, 200, 500, and 1000 mg/L), R. blasticus + polysulfone carrier treatment showed removal efficiencies of 98.9~99.84% for N and 96.92~99.21% for P. The R. blasticus + alginate carrier treatment showed removal efficiencies of 88.04~97.1% for N and 90.33~97.13% for P. The R. blasticus + PVA carrier treatment showed removal efficiencies of 18.53~44.25% for N and 14.93~43.63% for P. The R. blasticus + PVA + zeolite carrier treatment showed removal efficiencies of 26.65~64.33% for N and 23.44~64.05% for P. In addition, at the minimum inhibitory concentration of heavy metals, R. blasticus (dead cells) + polysulfone carrier treatment showed removal efficiencies of 7.77% for Cu and 12.19% for Ni. Rhodobacter blasticus (dead cells) + alginate carrier treatment showed removal efficiencies of 25.83% for Cu and 31.12% for Ni.

Biological activity of functional foods and anti-microbial activity of phenolics from Sambucus sieboldiana var. pendula leaves

  • This study aimed to investigate the biological and antibacterial activities of phenolics from Sambucus sieboldiana var. pendula leaf extract. The phenolic contents of the S. sieboldiana var. pendula extracted with water and ethanol was 11.60 and 12.39 mg/g, respectively. The inhibitory effects of the water and ethanol extracts on the angiotensin converting enzyme (ACE) were 71.94-92.08 and 48.42-78.33%, respectively, and on xanthine oxidase (XOase) were 42.11-100 and 35.61-100%, respectively, at a phenolic concentration of 50-200 ㎍/mL. All these effects were found to be concentration-dependent. Additionally, the leaf extracts exhibited excellent antibacterial activities against Streptococcus mutans, Staphylococcus aureus, and Propionibacterium acnes. Hence, S. sieboldiana var. pendula was confirmed to have excellent antihypertensive, antigout, and antimicrobial properties.

Functional properties of hydrolysate from silkworms fed Cudrania tricuspidata (효소처리 누에 분말의 생리 활성에 관한 연구)

  • In this study, silkworms fed Cudrania tricuspidata leaves were powdered, and hydrolytic enzymes, including viscozyme, papain, and flavourzyme, were added to verify the functionality of the different mixtures. In the general component analysis, the C. tricuspidata silkworm (CS) group exhibited higher crude protein and ash content than did the other groups, and the enzyme-treated groups exhibited higher carbohydrate content than the CS group. The DPPH and ABTS radical scavenging activities were significantly higher in the CS treated with viscozyme (CSV) and the CS treated with viscozyme/flavourzyme (CSVF) than in the other groups, with the CSV group showing the highest reducing power. ACE inhibitory activity was significantly higher in the CS treated with visocozyme/papain (CSVP) than in the CS group. In conclusion, rather than using powdered silkworms fed C. tricuspidata leaves, it would be more effective to use hydrolysates from C. tricuspidata silkworms as raw materials for functional foods.

Physicochemical Characteristics and Biological Activities of Rice and Amaranth Fermented by Bacillus subtilis KMKW4 (Bacillus subtilis KMKW4를 이용한 백미 및 아마란스 발효물의 이화학적 특성 및 생리활성)

  • This study examined the cultural characteristics and biological activities of fermented rice and amaranth with Bacillus subtilis KMKW4. These samples were made with various amounts of rice and amaranth [100:0 (R100), 90:10 (R90), 80:20 (R80), and 70:30 (R70)]. B. subtilis KMKW4 was used as starter for the fermentation, and its cultures at the lated logarithmic growth were inoculated for final concentration of 2% (v/v). Number of viable cells of fermented R80 (7.67 log CFU/mL) was greater than those of R100, R90, and R70 (7.48 log CFU/mL, 7.38 log CFU/mL, and 7.09 CFU/mL, respectively) during the fermentation period (120 h). Amylase activities of fermented R80 and R100 were 57.77 U/mL and 19.91 U/mL, respectively. Furthermore, amylase activities of fermented freeze-dried powders of R100 and R80 were 24.31 U/g and 9.12 U/g, respectively. Free sugar contents of R100 and R80 increased after fermentation, and that of R80 (5,454.15 mg/100 g) significantly increased compared to that of R100 (4,274.85 mg/100 g). The free amino acid content of R80 was higher than that of R100. DPPH and superoxide radical scavenging activities of 5 mg/mL of fermented freeze-dried powder (R80) were 44.21% and 89.76%, respectively. ACE inhibition rates and ${\alpha}$-glucosidase inhibitory activities were significantly higher in R80 than R100. This study suggested that fermentation of R80 might be a new potential source of antioxidant, anti-diabetic, and anti-hypertensive agents applicable to grain enzyme-containing foods.

Biological Activity of Extracts from Acanthopanax sessiliflorum Fruit (오가피(Acanthopanax sessiliflorum) 열매 추출물의 생리활성)

  • This study was carried out to determine the biological activity of Acanthopanax sessiliflorum fruit extracts. The phenolic compound contents of the extracts were 21.4 and 15.8 mg/g in hot water and 60% ethanol extracts. The total anti-oxidant activities of the water and the 60% ethanol extracts at a 200 ${\mu}g/mL$ phenolic concent ration were at $92.4{\pm}0.8$ and $89.2{\pm}1.1%$ in terms of the DPPH radical scavenging activity, $98.3{\pm}1.1$ and $96.5{\pm}3.5%$ in terms of the ABTS radical decolorization, $2.0{\pm}0.6$ and $1.2{\pm}2.8$ PF in terms of the anti-oxidant protection factor, and $66.3{\pm}0.8$ and $61.4{\pm}2.3%$ in terms of the TBARs inhibitory activity. The activities that inhibited the angiotensin-converting enzyme and xanthin oxidase were at $85.1{\pm}3.2$ and 0% in the water extracts and $59.3{\pm}1.5$ and $9.5{\pm}0.8%$ in the 60% ethanol extracts at the 200 ${\mu}g/mL$ phenolic concentration. The tyrosinase and elastase inhibitory activities were at $56.6{\pm}1.8$ and $53.1{\pm}1.1%$ in the water extracts and $33.7{\pm}2.2$ and $22.4{\pm}3.1%$ in the 60% ethanol extracts. The astringent effect of the water and the 60% ethanol extracts were at $50.5{\pm}0.9$ and $11.5{\pm}4.1%$.
