Physiological Functionality and Nitrite Scavenging Ability of Fermentation Extracts from Pine Needles (솔잎발효추출물의 효소적 저해활성 및 아질산염 소거작용)

  • Effects on the physiological functionality, such as tyrosinase, xanthine oxidase, angiotensin converting enzyme and Nitrite scavenging ability were also observed by pine needle fermentation extract(PFE) and the difference in the consistency of pine needle ethanol extracts(PE 80, PE 50) was found. In the inhibition effect on tyrosinase, PFE showed 5-38% higher than that of PE 80 and PE 50. In the inhibition on XOase, PFE, PE 80 and PE 50 showed 62.77%, 64.90%, 55.9% respectively. In the inhibition effect on ACE, PFE, PE SO and PE 50 showed 78.02%, 69.82% and 21.75% respectively. Among these, PFE showed the highest ACE inhibition effect. In the inhibition effect on nitrite scavenging ability, the pine needle extracts showed a high effect in pH 3.0. As the result of the research using HPLC for the organic acid, all the samples(PFE, PE 80, PE 50) showed higher contents of the ascorbic acid concerned with the effect of the antioxidative. PFE showed the highest contents of the ascorbic acid.

Studies on the Physiological Functionality of Pine Needle and Mugwort Extracts (솔잎과 쑥 추출물의 기능성 점토)

  • This study was conducted to investigate the physiological functionality of the hot water and 70% acetone extracts obtained from the pine needle and mugwort. Flavanol tannin content was above 60% of the total polyphenol in pine needle extracts. 70% Acetone extract from pine needle exhibited inhibition percentage of about 82.2% on the mutagenicity of Trp-P-1. Angiotensin I converting enzyme(ACE) inhibition activity was 61% and 50% in the hot water extract and in the 70% acetone extract respectively. Electron donating ability(EDA) of the hot water and 70% acetone extracts obtained from the pine needle was significantly good above 80%. 70% Acetone extract from pine needle showed inhibitary effect against the polyphenol oxidase(PPO) of water dropwort. The nitrite scavenging ability was appeared in all the extracts examined and it showed $80{\sim}90%$ at pH 3.0. The oxidative stability was determined by POV. Results showed that ethyl acetate fraction is better antioxidants than chloroform and butanol fractions in the soybean oil. Then, in the mugwort extracts, antimutagenicity of 70% acetone extract was similar to that of pine needle. Ratio of ACE inhibition activity was higher than that of pine needle extracts. EDA of hot water and 70% acetone extracts was 45%. Extracts from the mugwort were lower in nitrite scavenging ability about $20{\sim}30%$ than extracts from pine needle.

Hypoglycemic and Angiotension Converting Enzyme Inhibitory Effect of Water and Ethanol Extracts from Haesongi Mushroom (Hypsizigus marmoreus)

  • Water and ethanol extracts were prepared from the haesongi mushroom (Hypsizigus marmoreus) to measure functional components. The ability of the extracts to inhibit angiotensin-converting enzyme (ACE) and their hypoglycemic effects were also determined; the latter was measured by $\alpha$-amylase and glucosidase inhibition. Extraction yield, protein content, total phenol, and $\beta$-glucan in the water extracts were 55.86, 17.71, 1.89, and 21.93%, respectively. The respective values for the ethanol extracts were lower than those for water extracts. Both water and ethanol extracts showed dosedependent ACE inhibition, the effect of the former being greater. The water extract inhibited ACE activity by 95.34% at 40 mg/mL. The $IC_{50}$ values of the water extracts were 63.32 and 0.41 mg/mL for $\alpha$-amylase and glucosidase, respectively. Thus, the water extracts had a greater hypoglycemic effect than the ethanol extracts. From these results, water is a better solvent than ethanol to extract from the haesongi mushroom functional components that show ACE inhibition and have hypoglycemic effects.

3D-QSAR of Angiotensin-Converting Enzyme Inhibitors: Functional Group Interaction Energy Descriptors for Quantitative Structure-Activity Relationships Study of ACE Inhibitors

  • A new set of functional group interaction energy descriptors relevant to the ACE (Angiotensin-Converting Enzyme) inhibitory peptide, QSAR (Quantitative Structure Activity Relationships), is presented. The functional group interaction energies approximate the charged interactions and distances between functional groups in molecules. The effective energies of the computationally derived geometries are useful parameters for deriving 3D-QSAR models, especially in the absence of experimentally known active site conformation. ACE is a regulatory zinc protease in the renin-angiotensin system. Therapeutic inhibition of this enzyme has proven to be a very effective treatment for the management of hypertension. The non bond interaction energy values among functional groups of six-feature of ACE inhibitory peptides were used as descriptor terms and analyzed for multivariate correlation with ACE inhibition activity. The functional group interaction energy descriptors used in the regression analysis were obtained by a series of inhibitor structures derived from molecular mechanics and semi-empirical calculations. The descriptors calculated using electrostatic and steric fields from the precisely defined functional group were sufficient to explain the biological activity of inhibitor. Application of the descriptors to the inhibition of ACE indicates that the derived QSAR has good predicting ability and provides insight into the mechanism of enzyme inhibition. The method, functional group interaction energy analysis, is expected to be applicable to predict enzyme inhibitory activity of the rationally designed inhibitors.

Studies on the Radical Scavenging Effects and the Inhibitory Effects on ACE Activity of Several Flavonoids (각종 Flavonoids의 라디칼 봉쇄능과 ACE 활성 억제능에 관한 연구)

  • This study was carried out to identify the biophysical utility of bioflavonoids by the determination of their antioxidative activities, radical scavenging activity and inhibitory effect on the ACE activity. The results obtained were as follows; All flavonoids experimented greatly inhibited the linoleic acid oxidation from the early period of oxidation, and the radical scavenging ability was also greater in genistein and daidzein than other flavonoids, generally showing donating ability. Rutin has the metal-chelating ability with C $u^{2+}$ and $Mg^{2+}$, which means to have the inhibitory effect on the promotive oxidation of lipid by metal ion. All flavonoids experimented inhibited the angiotensin converting enzyme (ACE) activity, which was greater in genistein and daidzein than other flavonoids.s.

Antioxidant activity and inhibition activity against α-amylase and α-glucosidase of Smilax China L. (청미래덩굴(Smilax China L.) 추출물의 항산화 및 α-amylase와 α-glucosidase 저해활성)

  • This study was carried out to analyze the effects of water and 70% ethanol extract on the antioxidative and antidiabetic activities of Smilax china L., a vine shrub belonging to the lily family. The activities of the extracts were measured based on the total phenolic and flavonoid contents and through on the results of the antioxidant tests, such as the electron-donating ability, ABTs radical scavenging activities, SOD-like activity, xanthine oxidase inhibition effect, antioxidant protection factor (PF), TBARs content and ACE inhibition activity, and ${\alpha}$-glucosidase, and ${\alpha}$-amylase inhibition activity. The resulting total phenolic and flavonoid contents of the 70% ethanol extract from S. china L. were greater than those of the water extract from S. china L. With regard to the results of the antioxidant tests, such as the electron-donating ability, ABTs radical scavenging activity, SOD-like activity, xanthine oxidase inhibition effect, antioxidant protection factor (PF), and TBARs content, those from the 70% ethanol extract from S. china L. were greater than those from the water extract from S. china L. Also, with regard to the ACE inhibition effect and ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibition, those from the 70% ethanol extract from S. china L. were greater than those from the water extract from S. china L. All these findings show that the 70% ethanol extract from S. china L. has greater antioxidative and antidiabetic effects and can be used as a preventive agent for oxidation and diabetes.

The Biological Activity of Deer Antler Extract in vitro (In vitro에 의한 녹용 추출물의 생리 활성 효과)

  • Our research objective was to examine the in vitro biological activity of deer antler(Nogyong in Korean) extract, including the antioxidative, nitrite scavenging, and tyrosinase inhibitory effects, as well as the antithrombotic, and angiotensin I converting enzyme(ACE) inhibitory activities. The carbohydrate, protein, fat, and mineral contents of the deer antler were 7.6%, 65.3%, 3.2% and 23.9%, respectively. The electron donating ability(EDA) by the reduction of 2,2'-diphenyl-1-picrylhydrazyl(DPPH) was 67.1%, and the inhibition rate of lipid peroxidation by the thiocyanate method using linoleic acid was 92.1% in 100 mg/ml of extract. The nitrite scavenging effects were pH dependent, and were highest at pH 1.2 and lowest at pH 6.0. The sample inhibition rate against tyrosinase was above 64.0%. The platelet aggregation induced by ADP(adenosine-5'diphosphate) was inhibited up to 51.7%, and the inhibitory effect was dependent on the sample concentration. Lastly, the inhibition rate of ACE was 47.5% in 100 mg/ml of deer antler extract.

Physiological Activities of Extracts from Phellinus linteus on Brown Rice added Rice Bran, Pine Needle and Tumeric Powder (미강과 솔잎, 강황 분말을 첨가한 현미에 배양한 상황버섯 균사체 추출물의 생리활성에 관한 연구)

  • This study was carried out to examine Electron donating ability (EDA), nitrite scavenging, tyrosinase inhibition, ACE inhibition activity and fibrinolytic activity of culture extracts from Phellinus linteus which was grown added rice bran, pine needles and turmeric in brown rice. Electron donating ability of Phellinus linteus extract (PLE) was lower in the water extract than the ethanol extract. Nitrite scavenging activity was the highest in PLE from ethanol extract than water extract. Especially, when the pine needles was addition treatment, the nitrite scavenging activity was about 70% at pH 1.2 by ethanol extract. Tyrosinase inhibition activity of PLE was highest in the water extract than ethanol extract, and inhibition rate was the most higher in the extract by hot water added pine needles. ACE inhibition activity were very low effective at water and ethanol extract. Fibrinolytic activities were similarly strong in rice bran, pine needles and turmeric powder. Especially, when rice bran was added, showed the activity was increased about 5% than plasmin. Therefore, It may be used for the food industry as natural source of bioactive compound after further investigation, such as in vivo experiment.

  • This study was carried out to examine the electron donating ability (EDA), nitrite scavenging, tyrosinase inhibition, ACE inhibition and fibrinolytic activity of culture extracts from Fomitella fraxinea which was grown added rice bran, pine needles and turmeric in brown rice. Electron donating ability was higher in the extract by hot water than ethanol. Nitrite scavenging activity was the highest from ethanol extract of culture products added to the pine needles, showed the scavenging activity was approximately 50%. Tyrosinase inhibition activity was highest in the extract by hot water, showed inhibition rate of rice bran was 72.49%, pine needles 87.34% and turmeric powder 82.75%, respectively. ACE inhibition activity were very low effective at all water and ethanol extracts. Fibrinolytic activities were similarly strong in the rice bran, pine needles and turmeric powder. Especially, when the rice bran and turmeric powder were added, showed the activities were increased about 10% than plasmin. Therefore, it may be used for the food and cosmetic industry as natural source of bioactive compound after further investigation such as in vivo experiment.

Studies on Biological Activity from Antler extract added Medical plants (한약재를 가미한 녹용추출물의 생리활성에 관한 연구)

  • This study was designed to investigate biological activity of antler extract added medical plants. The scavenging activity of DPPH radical was low scavenging activity at 0.01% concentration. But in the 0.05% and higher concentration, electron donating ability(EDA) is above 50% except Kongindangagam(48.5%) and significantly good above 70% in the 4 extracts. Superoxide dismutase(SOD)-like activity was 44.3% and 45.1% extracts of Ohjayenjongwhangami and M(market sample), Inhibition of xanthine oxidase were above 50% at 0.5% concentration except Boshingiwhangwhangami and from 62.4% to 84.9% in the 4 extracts. Inhibition rate of boshingiwhangwhangami was hasty increased from 33.5% to 77.5% at 1.0% concentration and others the higher concentration, the more increasing inhibition. Angiotension I-converting enzyme(ACE) inhibitory activities were high activity all of extracts. In the 0.5% concentration, ACE inhibition was above 80%. Especially 0.01% concentration of M was presented 81.8%. The study which conducted to investigate the effect of feeding antler extract group for 50 days on sperm concentration, Ca contents of serum, kidney and femur in rats was higher than that saline group.
