Proteomic and Phenotypic Analyses of a Putative YggS Family Pyridoxal Phosphate-Dependent Enzyme in Acidovorax citrulli

  • Acidovorax citrulli (Ac) is a phytopathogenic bacterium that causes bacterial fruit blotch (BFB) in cucurbit crops, including watermelon. However, there are no effective methods to control this disease. YggS family pyridoxal phosphate-dependent enzyme acts as a coenzyme in all transamination reactions, but its function in Ac is poorly understood. Therefore, this study uses proteomic and phenotypic analyses to characterize the functions. The Ac strain lacking the YggS family pyridoxal phosphate-dependent enzyme, AcΔyppAc(EV), virulence was wholly eradicated in geminated seed inoculation and leaf infiltration. AcΔyppAc(EV) propagation was inhibited when exposed to L-homoserine but not pyridoxine. Wild-type and mutant growth were comparable in the liquid media but not in the solid media in the minimal condition. The comparative proteomic analysis revealed that YppAc is primarily involved in cell motility and wall/membrane/envelop biogenesis. In addition, AcΔyppAc(EV) reduced biofilm formation and twitching halo production, indicating that YppAc is involved in various cellular mechanisms and possesses pleiotropic effects. Therefore, this identified protein is a potential target for developing an efficient anti-virulence reagent to control BFB.

Expression of a Recombinant Cry1Ac Crystal Protein Fused with a Green Fluorescent Protein in Bacillus thuringiensis subsp. kurstaki $Cry^-B$

  • To investigate the co-expression and crystallization of a fusion gene between the Bacillus thuringiensis crystal protein and a foreign protein in B. thuringiensis, the expression of the Cry1Ac fused with green fluorescent protein (GFP) genes in a B. thuringiensis $Cry^-B$ strain was examined. The cry1Ac gene was cloned in the B. thuringiensis-E. coli shuttle vector, pHT3101, under the control of the native cry1Ac gene promoter, while the GFP gene was inserted into the XhoI site upstream of the proteolytic cleavage site, in the middle region of the crylAc gene (pProAc-GFP). The B. thuringiensis $Cry^-B$ strain carrying pProAc-GFP (ProAc-GFP/CB) did not produce any inclusion bodies. However, the transformed strain expressed fusion protein forms although the expression level was relatively low. Furthermore, an immu­noblot analysis using GFP and Cry1Ac antibodies showed that the fusion protein was not a single spe­cies, but rather multiple forms. In addition, the N-terminal fragment of Cry1Ac and a non-fused GFP were also found in the B. thuringiensis $Cry^-B$ strain after autolysis. The sporulated cells before autolysis and the spore-crystal mixture after autolysis of ProAc-GFP/CB exhibited insecticidal activities against Plutella xylostella larvae. Accordingly, the current results suggest that a fusion crystal protein produced by the transfomant, ProAc-GFP/CB, can be functionally expressed but easily degraded in B. thuring­iensis.

  • 본 논문에서는 Custom Power의 전력품질의 향상을 위하여 PWM Buck-Boost AC-AC 컨버터를 사용, 항상 일정한 출력전압을 얻는 시스템을 보이고 제어를 위한 동적 모델링 및 제어기 설계의 일례를 보였다. 그 결과, 시스템 상태방정식을 유도하였고 전달함수를 구였으며 일정한 동작점 하에서 출력전압을 기준전압으로 regulation 제어하기 위한 제어기에 대하여 다루었다. 끝으로, 실험을 통하여 제안된 모델링과 시스템 설계가 타당함을 확인하였다.

Expression of a Fusion Protein with Cry1Ac Protein and a Scorpion Insect Toxin in Acrystalliferous Bacillus thuringiensis Strain

  • Expression of a fusion protein between B. thuringiensis crystal protein, Cry1Ac1 and a scorpion insect toxin (AaIT, Androctonus australis Hector insect toxin) in acrystalliferous B. thuringiensis strain (Cry-B strain) was examined. The cry 1Ac1 gene was cloned in B. thuringiensis-E coli shuttle vector, pHT3101, under the control of the native cry 1Ac1 gene promoter (pProAc) and a gene encoding AaIT was inserted in XhoI site in the middle of the cry 1Ac1 gene (pProAc-ScoR). B. thuringiensis Cry-B strain carrying pProAc-ScoR (PyoAc-ScoR/CB) produced an inclusion body of irregular shape and the expressed fusion protein is approximately 65 kDa in size. Sporulated cells and spore-crystal mixtures of ProAc-ScoR/CB had insecticidal activity against Plutella xylostella larvae, showing $LT_50$ of ProAc-ScoR/CB (22.59 hrs) lower than that of ProAc/CB (30.06 hrs) at $1{\times}{10^7} {CEU/cm^2}$. These results suggest that the fusion protein including a B. thuringiensis crystal protein and an AaIT may be functionally expressed in B. thupingiensis. Moreover, we verified the additive toxicity of AaIT, which is a new feasible candidate for insect control.

A New Waveshaper for Harmonic Mitigation in Vector Controlled Induction Motor Drives

  • This paper deals with a new wave shaping technique for cost effective harmonic mitigation in ac-dc converter feeding Vector Controlled Induction Motor Drives(VCIMD's) for improving power quality at the point of common coupling(PCC). The proposed harmonic mitigator consists of a polygon connected autotransformer based twelve-pulse ac-dc converter and a small rating passive shunt filter tuned for $11^{th}$ harmonic frequency. This ac-dc converter eliminates the most dominant $5^{th},\;7^{th},\;and\;11^{th}$ harmonics and imposes the reduction of other higher order harmonics from the ac main current, thereby improving the power quality at ac mains. The design of autotransformer is carried out for the proposed ac-dc converter to make it suitable for retrofit applications, where presently a 6-pulse ac-dc converter is used. The effect of load variation on VCIMD is also studied to demonstrate the effectiveness of the proposed ac-dc converter in a wide operating range of the drive. Experimental results obtained on the developed laboratory prototype of the proposed harmonic mitigator are used to validate the model and design of the ac-dc converter.

EFFECTS OF ACTIVATED CARBON ON GROWTH, RUMINAL CHARACTERISTICS, BLOOD PROFILES AND FEED DIGESTIBILITY IN SHEEP

  • This study was carried out to investigate the effects of activated carbon (AC) on growth, ruminal charateristics, blood profiles and feed digestibility in sheep, using roughage-based or concentrate-based diets. Twelve Suffolk breed of sheep of similar age and weight were distributed into 4 groups in a $2{\times}2$ factorial design. Two groups were fed a roughage-based diet with (R + AC) and without AC (R - AC), while the other two were fed a concentrate-based diet with (C + AC) and without AC (C - AC), respectively. The addition of 0.3% AC was based on dry matter of feed offered to animals. The incorporation of AC in roughage and concentrate based diets had no marked effects on feed intake, daily gain and feed conversion of the animals within experimental diets. The results obtained might be due to the low level of AC added in the diet. The animal on both concentrate-based diets were higher than the roughage-based diets in terms of daily gain and feed conversion ratio. However, it was observed that the animals provided with AC in the concentrate-based diet did not suffer from diarrhea and easily adjusted to high concentrate feeding. Further, the pH value for all diets before feeding was noted to be similar. After feeding, however, pH was shown to be higher in R + AC (p < 0.05) than in C + AC diet. Rumen protozoa number was decreased after feeding for both + AC diets, but in C - AC diet it was higher than in the roughage-based diets. For ammonia-nitrogen, C - AC was found to be higher than C + AC diet and the roughage-based diets before feeding. Total volatile fatty acid concentration, propionate and valerate molar ratios for both diets and time of collection were not affected. However, acetate, butyrate and valerate molar ratios were observed to be affected by diets and time of collections. The diets with AC increased (p < 0.05) before feeding for acetate molar ratio, but not different within diet, however, the roughage diets were found to be higher (p < 0.05) in acetate than the concentrate diet. In the blood parameters, the glutamic pyruvic transaminase (GPT), red and white blood cell (RBC, WBC) counts and packed cell volume (PCV) did not differ within and among the diets. Likewise, the WBC differential count in both diets with either - AC or + AC were similar in trend. However, lymphocyte count was noted to be increased in R + AC than the R - AC diet. The addition of AC in both diets did not affect nutrient digestibilities within diets.

저전압 스마트 배전시스템을 위한 Quasi Z-소스 AC-AC 컨버터 (A Quasi Z-Source AC-AC Converter for Low Voltage Smart Distribution System)

  • 본 논문에서는 저전압 스마트 배전시스템을 위한 Quasi Z-소스 AC-AC 컨버터를 제안하였다. 제안된 시스템은 Quasi Z-소스 AC-AC 컨버터의 교류 입력 전압에 대하여, 동상의 출력 전압을 부스트 하고, 역상의 출력 전압을 벅-부스트하는 고정 주파수 가변 전압 VVCF(Variable Voltage Constant Frequency)의 출력특성을 가지고 있다. 또한 Quasi Z-소스 AC-AC 컨버터의 shoot-through 스위치 단에 벅-컨버터를 연결하여 Quasi Z-소스 AC-AC 컨버터의 동작과 함께 저압의 직류 전압을 출력 할 수 있다. PSIM 시뮬레이션과 실험에 의하여, 제안된 시스템이 동상 부스트 모드로 동작하는 경우, PI 제어된 벅 컨버터에 의하여 직류 전압을 출력할 수 있음을 확인하였다.

  • 본 논문에서는 3상 Quasi Z-소스 AC/AC 전력변환기를 제안한다. 제안된 전력변환기는 3대의 단상 Quasi Z-소스 AC/AC 컨버터로 구성되어 있으며, 간단한 듀티 비 제어만으로도 동상 및 역상의 벅-부스트 출력 전압의 발생이 가능하다. 제안된 방식은 PSIM 시뮬레이션을 통하여 타당성을 확인하였다.

  • In this paper, a PWM Buck-Boost AC-AC converter for improvement of power quality of custom power is presented. The PWM Buck-Boost AC-AC converter is modelled by using circuit DQ transformation whereby the design guideline is obtained. Based on the analysis, the converter system is implemented with the design criteria and the experimental results show the validity of modelling and analysis.

  • This paper presents controller design of a static var compensator using PWM Cuk AC-AC converter. The PWM Cuk AC-AC converter is modelled by using complex circuit DQ transformation and perturbed around the operating point whereby the small signal system characteristics is analytically obtained. Finally, the PSIM simulations show the validity of the modelling and analysis.
