Biological Activities of Methanol Extracts from Green Tea Seed (녹차종자 메탄올추출물의 생리활성)

  • This study was to investigate the biological activities of green tea seed methanol extract (GTSME) and compared those of green tea methanol extract (GTME) for using green tea seed as the functional food material. The hydrogen-donating activity of GTSME was over 50% at the $100 {\mu}g/mL$ concentration the activity of GTME was 21.86% at the $1000{\mu}g/mL$ concentration compared with that of control. The MDA (malondialdehyde) production was 60 Mol/g and 50 Mol/g in the mouse liver homogenate teated with GTME and GTSME of $1000{\mu}g/mL$ concentrations, respectively, and the values were lower than 86 Mol/g of control. GTME and GTSME of $1000{\mu}g/mL$ concentration inhibited the proliferation of over 50% and over 20% in A549 and SW480 human cancer cells, respectively. The morphology transformation was shown in the cancer cells treated with GTSME of $500{\mu}g/mL$ with the decrease of cell numbers lower than that of control cells numbers. The NO production was increased in a dose dependent manner in the RAW264.7 macrophage cells treated with GTME and GTSME of 1, 10, 100 and $1000{\mu}g/mL$ concentrations, and the NO production by GTSME was $2.04{\mu}M$ at $100{\mu}g/mL$ concentration, and the value was higher than $0.77{\mu}M$ by GTME.

  • Sphagnum palustre is a semi aquatic moss. S. palustre has been used as Korean traditional medicine to treat cardiac pain and stroke. This study was carried out to analyze phytochemical constituents of S. palustre and investigate the biological activity for the promotion of human health. At first, we isolated seven compounds from the ethanolic extract of this plant, and their structures were characterized by spectroscopic methods. Their structures were characterized to be Coumarin(1), Caffeic acid(2), Quercetin(3), Astragalin(4), Luteolin(5), Chlorogenic acid(6), Rutin(7) were for the first time reported from this source. The ethanol extract from S. palustre which was tested for its anticancer activity against three human tumor cell line by in vitro assay.

Anti-Cancer Activity of the Leave Extracts of Rodgersia podophylla through β-catenin Proteasomal Degradation in Human Cancer Cells (도깨비부채(Rodgersia podophylla) 잎 추출물의 인간 암세포의 β-catenin 분해 유도 활성)

  • In this study, we evaluated the effect of Rodgersia podophylla leave extracts (RPL) on ${\beta}-catenin$ level in human cancer cells. RPL dose-dependently inhibited cell proliferation in SW480, A549, MDA-MB-231, PC-3 and AsPC-1 cells. RPL dramatically decreased ${\beta}-catenin$ protein level in all cancer cells. However, decreased level of ${\beta}-catenin$ mRNA expression was observed in A549 and AsPC-1 cells. In addition, RPL dramatically attenuated cyclin D1 mRNA expression in all cancer cells. MG132 decreased the downregulation of ${\beta}-catenin$ protein level induced by RPL in all cancer cells, while RPL-induced downregulation of ${\beta}-catenin$ was inhibited by the inhibition of $GSK-3{\beta}$ by LiCl in MDA-MB-231 cells. RPL phosphorylated ${\beta}-catenin$ and $GSK-3{\beta}$. In addition, the inhibition of $GSK-3{\beta}$ by LiCl attenuated RPL-induced ${\beta}-catenin$ phosphorylation. Based on these findings, RPL may be a potential candidate for the development of chemopreventive or therapeutic agents for human cancer.

Biological activities and physicochemical properties of polysaccharides from Gloiopeltis furcata prepared by using various enzymes (효소종류에 따른 불등풀가사리 유래 다당류의 이화학적 특성 및 생리활성)

  • In this study, the biological activities and physicochemical properties of polysaccharides from Gloiopeltis furcata were investigated. Polysaccharides were isolated by enzymes treatment (celluclast, flavourzyme, papain, termamyl, viscozyme) followed by ethanol precipitation and lyophilization. The yield of polysaccharides by enzymes treatment group were 52.8-66.4%. The major constituents in viscozyme treatment group were total sugar (71.04%), protein (7.22%), uronic acid (23.18 g/100 g), and sulfate (28.27%), respectively. The DPPH radical scavenging activity and ferric reducing antioxidant potential of the viscozyme treatment group at 5 mg/mL were 23.10% and $218.50{\mu}M$, respectively. The protective effects against $H_2O_2$-induced cytotoxicity in L132 cell of viscozyme treatment group at $1{\mu}g/mL$ was 85.64%. The viscozyme treatment group increased the production of nitric oxide (NO) in a dose-dependent manner. The antitumor activity of viscozyme treatment group (at $25{\mu}g/mL$) in A549, HeLa, SNU719 and MCF7 was 69.57%, 52.74%, 61.06% and 68.64%, respectively. All of data showed that the biological activities and chemical characteristics of enzymes treatment group are higher than that of the control group. The polysaccharides isolated from Gloiopeltis furcata investigated herein are useful as functional materials agents.

Effects of Loquat (Eriobotrya japonica Lindl.) Ethanol Extracts of Different Aerial Parts on Antioxidant Activity and Antiproliferation of Human Cancer Cells (비파 부위별 에탄올 추출물의 항산화 활성 및 암세포 증식 억제효과)

  • The current study was carried out to determine the effects of the seed, flesh (seedless fruit), and leaf of loquat (Eriobotrya japonica Lindle.) on antioxidative activity and anti-proliferation in human cancer cells. Total polyphenol contents of loquat seed, flesh, and leaf ethanol extracts were found to be 17.77, 32.32, and 28.08 mg/g, respectively. Also, total flavonoid contents of loquat seed, flesh, and leaf ethanol extracts were found to be 18.77, 28.73, and 21.35 mg/g, respectively. The $IC_{50}$ values of DPPH hydroxyl scavenging of loquat seed, flesh, and leaf ethanol extracts were 0.049, 0.063, and 0.042 mg/mL, respectively. Antioxidative indexes of loquat leaf and seed ethanol extracts was highly and it was similar to the BHA and BHT. The antioxidative activities in loquat seed and leaf ethanol extracts were higher in loquat flesh. The antiproliferation effect of loquat seed and leaf ethanol extracts on liver cancer cell line (H460), stomach cancer cell line (AGS) and lung cancer cell line (A549) showed higher values compared with the flesh ethanol extracts. These results indicate that loquat ethanol extracts may play a positive role in antioxidative properties and cancer prevention.

Lack of Cytotoxicity of the Colorant in Conjugated Linoleic Acid against Human Cancer and Normal Cells (Conjugated linoleic acid 황갈색의 인체암세포와 인체정상세포에 대한 세포독성)

  • The cytotoxicity of the colorant in conjugated linoleic acid (CLA) was investigated in human cancer cell lines and a normal human cell line. Commercially-available CLA with a brown color (designate crude CLA; c-CLA) was distilled in a vacuum (10 mmHg-$220^{\circ}C$, 10 mmHg-$235^{\circ}C$, 10 mmHg-$240^{\circ}C$, and 20 mmHg-$260^{\circ}C$) for 30 min to obtain pure CLA (distilled CLA; d-CLA) and dark brown-colored CLA (residual CLA; r-CLA) samples. No color intensity was shown in the d-CLA sample obtained under 10 mmHg-$220^{\circ}C$ conditions of distillation when the L (brightness), a (red/blue), and b (yellow/green) parameters were analyzed, whereas the r-CLA sample showed a dark brown color. The composition of CLA isomers in both the d- and r-CLA samples, as compared to that of the c-CLA sample, was not significantly different when analyzed by gas chromatography. When the cytotoxicity of the r-CLA and d-CLA samples obtained under 10 mmHg-$220^{\circ}C$ conditions were compared against human breast cancer cells (MCF-7), human lung cancer cells (A-549), human colon cancer cells (HT-29), human prostate cancer cells (PC-3), and human neuroblastoma cells (SK-N-SH), no significant cytotoxicity was seen in the cell lines. These results suggest that the color or colorant in the CLA samples did not have any effects on the proliferation of human cancer and normal cells and imply that the colorant in commercially available CLA samples is safe for human consumption.

Effect of Particle Size on Antioxidant Activity and Cytotoxicity in Purple Corn Seed Powder (검정찰옥수수 종실 분쇄 정도에 따른 항산화 및 Cytotoxicity 활성 효과)

  • This study was carried out to evaluate antioxidant and anticancer activity of crudes extracts from colored corn (Zea mays. L.) among particle size with different pulverizing methods (pin mill and ultra fine pulverizer). In cytotoxicity test using extracts from the flours grounded by pin mill and ultra fine pulverizer respectively, it showed that A-549 was the highest anticancer activity among in vitro using 4 cancer cell line types (Hep-G2, A-549, HCT-116 and MCF-7). Assessing the antioxidant activities of crude extracted from different pulverizing methods was measured by trolox equivalent antioxidant capacity (TEAC) and ferric reducing antioxidant power (FRAP). Higher scanvening activity against free radical was observed in the crude extracted from small particle size flours of colored corn than in those of the big particle size.

Antioxidative and Antitumor Activities of Crude Polysaccharide Fraction from P leurotus eryngii (큰느타리버섯(Pieurotus eryngii) 조다당체 분획의 항산화 및 항종양활성)

  • The aims of this study were to investigate the antioxidative and antitumor effects of crude polysaccharide fraction from Pleurtus eryngii (CPPE). CPPE inhibited autoxidation of linoleic acid at 1,000 $\mu$g/mL concentration, and the inhibitory rates of lipid peroxidation in rat liver microsome were 5.28, 9.40, and 32.5% at 10, 100, and 1,000 $\mu$g/mL concentrations, respectively. After treatment with CPPE for 72 hours, the inhibitory rates against MCF-7, A549 and AGS cell lines showed 42.3, 33.4 and 26.7% at concentration of 1,000 $\mu$g/mL, respectively. Results of CPPE treatment at 100 and 300 mg/kg/day for 7 days in sarcoma-180 bearing-mice showed survival rates of 70 and 90%, respectively. Body weights of mice treated with CPPE were significantly decreased when compared with the control.

Biological Activities of the Vaporized Liquid of Water-boiled Pine Needle (솔잎 열수 증류액의 생리활성 효과)

  • This study was performed to determine the antioxidative, antimutagenic, and anticancer effects of vaporized liquid of water-boiled pine needle(VLP) using DPPH free radical donating method, Ames test, and cytotoxicity. VLP showed the highest electron donating activities $(18.4\;{\mu}L)$. The inhibition rate of VLP $(200\;{\mu}L/plate)$ in the Salmonella. typhimurium TA100 strain showed $45.9\%$ inhibition against the mutagenesis induced by MNNG. In addition, the suppression of with same concentration of VLP in the S. typhimurium TA100 strains showed $85.5\%$ inhibition against 4NQO, respectively. The suppressions under the same condition against Trp-P-1 in the TA98 and TA100 strains were $91.0\%$ and $62.1\%$, respectively. The cytotoxic effects of VLP against the cell lines with human lung carcinoma (A549), human hepatocellular carcinoma (HepG2) , human gastric carcinoma (AGS), human breast adenocarcinoma (MCF-7) and human cervical adenocarcinoma (HeLa) were inhibited with increase of the VLP concentration. The treatment of $50\;{\mu}L/well$ VLP showed strong cytotoxicities of $78.7\%,\;90.3\%,\;90.8\%,\;62.3\%$ and $93.7\%$ against A549, HepG2, AGS, MCF-7 and HeLa, respectively.

Antioxidant and anticancer properties of hot water and ethanol extracts from the roots of Smilax china L. (발계(Smilax china L.) 열수 및 에탄올 추출물의 항산화 및 항암활성)

  • The biological activities of Smilax china L. rhizome (SCR), hot water (SCRW) and 70% ethanol extract (SCRE) were analyzed. The total phenolic contents of SCRW and SCRE were 51.7 and 100.5 mg/g, respectively. The measured flavonoid content of SCRW ($67.7{\mu}g/g$) was almost double that of SCRE ($31.7{\mu}g/g$). SCRE ($IC_{50}=42.4{\mu}g/mL$) exhibited stronger antioxidant activity in the DPPH system than the positive control ${\alpha}$-tocopherol ($71.3{\mu}g/mL$) or butylated hydroxy anisole ($53.8{\mu}g/mL$) did. SCRE ($IC_{50}=50.3{\mu}g/mL$) also showed stronger ABTS radical scavenging activity, as did ${\alpha}$-tocopherol ($67.1{\mu}g/mL$). The SOD-like activity and Tyrosinase inhibition activity of SCRW and SCRE showed almost the same pattern. The best SOD-like activity and tyrosinase inhibition activity were measured as 24.9% and 20.3% in SCRW at $1,000{\mu}g/mL$, respectively. The cytotoxic effects of the SCR extracts were analyzed via MTT assay on human cancer and normal cells. SCRW and SCRE did not show cytotoxicity up to the concentration of $1,000{\mu}g/mL$ against the normal human cell line HEK293. Against human breast cancer cells (MCF-7), SCRW inhibited MCF-7 growth (by 27.6%) better than the anticancer drug cyclophosphamide (15.5%) at $1,000{\mu}g/mL$. SCRE ($1,000{\mu}g/mL$) inhibited the growth of human lung cancer cells A549 (37.6%) and human stomach cancer cells AGS (53.6%) more effective than did SCRW (21.0% and 35.4%) or CPA (22.2% and 31.7%). These results suggest the potential use of SCRE and SCRW as an excellent antioxidant and antiproliferative substance, respectively.
