Effect of Administering Recombinant Bovine Somatotropin to Breeding Cows on Weight Gain and Prevention of Diarrhea in Suckling Calves (번식우에 대한 rBST투여가 포유기 송아지의 비육과 설사 예방에 미치는 영향)

  • This study was carried out to investigate weight gain and prevention of diarrhea in suckling calves which were injected with recombinant bovine somatopropin (rBST). A total of 101 breeding cows were assigned to the six groups according to the administered dosage and injected time, respectively. Groups T-1, T-3, T-4 and T-6 were injected starting 1 week before calving and groups T-2 and T-5 were injected on the calving day. The six groups were injected five times at two week intervals. Groups T-1 and T-2 were injected with 250mg composite rBST, Group T-3 was injected with 375 mg composite rBST. Groups T-4 and T-5 were injected with 500mg composite rBST, And group T-6 was injected with 500mg rBST-S. The control group was not injected with BST. The groups injected with 500 mg BrST had a lower rate of morbidity from diarrhea than the control group or the groups injected with 250 mg rBST (T-1 and T-2). Weight gain was higher in group T-4 than in the control group or groups T-1 and T-2. In Korean Native Cattle, the total weight gain was greater in group T-4 than in the control group (p<0.05). In crossbred cows, total weight gain was the highest in group T-4, and the total weight gain rate was greater than in group T-4 and the control group (p<0.05). The results of the hematological values showed that injections of rBST did not affect the level of the RBC, TP and BUN in the breeding cows at 9 weeks after postpartum or the neonatal calves. The results of this study indicate that injecting breeding cows with 500 mg rBST before calving would be effective in the preventing of diarrhea and in increasing weight gain of calves from birth to weaning.

Peri-Operative Liver Fibrosis and Native Liver Survival in Pediatric Patients with Biliary Atresia: A Systematic Review and Meta-Analysis

  • No systematic review to date has examined histopathological parameters in relation to native liver survival in children who undergo the Kasai operation for biliary atresia (BA). A systematic review and meta-analysis is presented, comparing the frequency of native liver survival in peri-operative severe vs. non-severe liver fibrosis cases, in addition to other reported histopathology parameters. Records were sourced from MEDLINE, Embase, and CENTRAL databases. Studies followed the PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analyses) guidelines and compared native liver survival frequencies in pediatric patients with evidence of severe vs. non-severe liver fibrosis, bile duct proliferation, cholestasis, lobular inflammation, portal inflammation, and giant cell transformation on peri-operative biopsies. The primary outcome was the frequency of native liver survival. A random effects meta-analysis was used. Twenty-eight observational studies were included, 1,171 pediatric patients with BA of whom 631 survived with their native liver. Lower odds of native liver survival in the severe liver fibrosis vs. non-severe liver fibrosis groups were reported (odds ratio [OR], 0.16; 95% confidence interval [CI], 0.08-0.33; I2=46%). No difference in the odds of native liver survival in the severe bile duct destruction vs. non-severe bile duct destruction groups were reported (OR, 0.17; 95% CI, 0.00-63.63; I2=96%). Lower odds of native liver survival were documented in the severe cholestasis vs. non-severe cholestasis (OR, 0.10; 95% CI, 0.01-0.73; I2=80%) and severe lobular inflammation vs. non-severe lobular inflammation groups (OR, 0.02; 95% CI, 0.00-0.62; I2=69%). There was no difference in the odds of native liver survival in the severe portal inflammation vs. non-severe portal inflammation groups (OR, 0.03; 95% CI, 0.00-3.22; I2=86%) or between the severe giant cell transformation vs. non-severe giant cell transformation groups (OR, 0.15; 95% CI, 0.00-175.21; I2=94%). The meta-analysis loosely suggests that the presence of severe liver fibrosis, cholestasis, and lobular inflammation are associated with lower odds of native liver survival in pediatric patients after Kasai.

Detection of Fusarium verticillioides Contaminated in Corn Using a New Species-specific Primer (종 특이 primer를 이용한 옥수수 오염 Fusarium verticillioides의 PCR 검출)

  • Fusarium verticillioides (teleomorph: Gibberella moniliformis), a member of the Gibberellea fujikuroi species complex, causes rots of corn stalks and ears, and produces a group of mycotoxins known as fumonisins that are harmful to animals and humans. Here, we focus on the development of a species-specific PCR primer set for differentiating F. verticillioides from other fumonisin-producing Fusarium species belonging to the species complex, such as F. proliferatum, F. fujikuroi, and F. subglutinans that are frequently associated with corn. The specific primers (RVERT1 and RVERT2) derived from the nucleotide sequences of RNA polymerase II beta subunit (RPB2) gene amplified a 208 bp-DNA fragment from only F. verticillioides isolates among the potential fumonisin-producing species examined; all of these isolates were shown to carry FUM1 required for fumonisin biosynthesis. The PCR detection limit using this specific primer set was approximately 0.125 pg/${\mu}l$ genomic DNA of F. verticillioides. In addition, the F. verticillioides-specfic fragment was successfully amplified from genomic DNAs of corn samples contaminated with Fusarium spp. This primer set would provide a useful tool for the detection and differentiation of potential fumonisin-producing F. verticillioides strains in cereal samples.

  • This study was designed to ultimately develop a highly efficient mass production technology of bacterial cellulose isolated from the citrus gel fermented by G. hansenii TL-2C. Pilot equipment made with FRP vessel length (665 mm) ${\times}$ width (375 mm) ${\times}$ height (210 mm) was developed for mass production of the citrus gel. To develop the optimal conditions for mass production of citrus gel, comprised of citrus juice (6,000 mL) diluted 100 times, containing 5% seed bacteria, 10% sucrose, and 1% ethanol, citrus juice was fermented at $30^{\circ}C$ for 14 days, and gel productivity in pilot system was examined. BC was isolated and purified from the citrus gel, and their chemical composition and physicochemical properties were investigated.

Antiviral Effects of the Culture Filtrate from Serratia marcescens Gsm01, against Cucumber mosaic virus (CMV)

  • The potential antiviral effects of the culture filtrates (CF) from Serratia marcescens strain Gsm01 against yellow strain of Cucumber mosaic virus (CMV-Y) were investigated. The culture filtrate of S. marcescens strain Gsm01 applied on Chenopodium amaranticolor showed high inhibitory activity, likewise no necrosis appeared when applied on the tobacco plants 2 days before CMV-Y inoculation. When plants were challenge inoculated with CMV-Y for eighteen days, the disease incidence in plants with culture filtrate of S. marcescens Gsm01 did not exceed 59%, whereas 100% of control plants were severely infected. The results of double antibody sandwich-enzyme linked immunosorbent assay (DAS-ELISA), reverse transcriptase polymerase chain reaction (RT-PCR), dot blotting, and western blotting showed that culture filtrate treatment highly affected the accumulation of CMV-Y or its CP protein gene in the treated plant leaves. It was also observed that the culture filtrate had no RNase activity on genomic RNAs of CMV-Y, suggesting that culture filtrate may not contain ribosome inactivating proteins (RIPs) or proteins with RNase activity. These data shows that culture filtrate of S. marcescens strain Gsm01 seems to be a promising source of antiviral substance for the practical use.

Purification and Properties of Ribosome-inactivating Proteins from the Leaves of $Cucurbita\;moschata\;D_{UCHESNE}$ (호박$(Cucurbita\;moschata\;D_{UCHESNE})$잎에서 리보즘불활성화 단백질의 분리 및 특성)

  • Two ribosome-inactivating proteins, PRIP 1 and PRIP 2 have been isolated from the leaves of $Cucurbita\;moschata\;D_{UCHESNE}$. Crude extracts were purified through ammonium sulfate precipitation and column chromatography using DE-52 cellulose, S-Sepharose, FPLC Suprose 12 HR and FPLC Mono-S. The molecular weights of PRIP 1 and PRIP 2 were 31,000 and 30,500, respectively. PRIP 2 was thermostabe and maintained its activity even after the incubation of the protein at $50^{\circ}C$ for 30 min. In a cell free in vitro translation system using rabbit reticulocyte lysate, protein synthesis was inhibited by the addition of PRIP 1 and PRIP 2. The $IC_{50}$ of PRIP 1 and PRIP 2 were 0.82 nM and 0.79 nM, respectively. The comparison of N-terminal amino acid sequences of the PRIP 1 and PRIP 2 with known RIPs revealed that PRIP 1 shows sequence similarity with Luffin B from Luffa cylindrica and Trichokirin from Trichosanthes kirilowii Maximowicz and PRH) 2 has sequence similarity with Momordin II and MAP 30 from Momordica charantia.

Chordoma - A Clinicopathologic Review of 4 Cases - (척삭종 - 4례의 임상 및 병리학적 검색 -)

  • Chordoma is a slow-growing malignant neoplasm arising from the remnants of the primitive notochord. It accounts for 1 to 4% of all malignant bone tumors. It occurs exclusively along the spinal axis. Authors experienced four cases of chordoma occurred in the sacrococcygeal region. There were two male and two female patients, with a mean age of 63.5 years(range, 57~75 years). Tissue was obtained by wide excision in two patients, by incisional biopsy in one patient and by needle biopsy in the other. Adjuvant radiation therapy was performed on all the patients after their biopsy. The mean diameter of the tumors was 7.6cm(range, 5.5 to 13.0cm). Grossly, tumor was multiobulated, soft and myxoid gelatinous mass. Microscopically, the tumor showed lobulated feature divided by fibrous septa within it. There were physaliphorous cells with vacuolated bubbly cytoplasm. And small uniform, round, and non-vacuolated tumor cells were also present. On immunohistochemical stain, all the cases were immunoreactive for cytokeratin, epithelial membrane antigen(EMA) and vimentin, respectively. One of the 4 cases was positive for S-100 protein. All the cases were negative for CEA.

A Study on the Effect of Social Worker's Job Characteristics on Their Job Satisfaction (사회복지사의 직무특성이 직무만족에 미치는 영향에 관한 연구)

  • The objective of this study examined the effect of job characteristics on the job satisfaction of social worker employed by 1st social agency, hospitals. Data were collected by 240 social workers. Based on Hackman and Oldham's(1975) theory of core job characteristics, this study was to defined other element of job characteristics, in social worker's duties, focused on moderating effect of self-confidence for job satisfaction, and compared to social worker's job characteristics between 1st social agency and hospitals. The results of this study were summarized as follows; first, job autonomy, feedback among Hackman and Oldham's 5 core job characteristics showed positive influence on job satisfaction. The other job characteristics found out significantly effected social worker's job satisfaction than Hackman and Oldham's 5 core job characteristics. Second, identities of social worker's job, cognition of professional level, necessity, and self-confidence variables were found to have a statistically significant strong relationship with job satisfaction of 1st setting social worker. The other side, necessity, self-confidence, sex, social worker's academic career variables positive influenced medical social worker's job satisfaction. Third, the moderating variables which had significant difference among social worker's working conditions. 1st setting social worker's self-confidence partial moderated the effect of job characteristics on social worker's job satisfaction. But, medical social worker's self-confidence not moderated the effect of job characteristics on job satisfaction. As a result, Social worker's job satisfaction depended on job characteristics and self-confidence. Therefore, to assure the job satisfaction, redesign for social worker's job characteristics, reinforced their self-confidence. Staff-development education program, supportive environment, supervision program is strongly recommended.

A study on Middle School Students' Perception and Knowledge for Kimchi in Masan and Changwon City (마산, 창원지역 중학생의 김치에 대한 지식 및 인식조사)

  • The purpose of this study was to investigate the knowledge and perception for Kimchi in middle school students. A questionnaire was used as the instrument of investigation. The subjects were 375 male students and 278 female students in Masan and Changwon City. The main results of this study are as follows. Ninety-three point three percent of the subjects were nuclear family type and 61.7% of respondent's mother had job. The average knowledge score for Kimchi of the subjects was 5.27 out of possible 10 points and the average value score on Kimchi was 41.25 out of 50 points. Eighty-six point one percent of students think they should eat Kimchi because Kimchi is good for the health(38.6%) and Kimchi is our traditional food(31.4%), and it was significantly difference between gender(p<0.01), male students more realized 'Kimchi is good for the health(41.6%)', whereas female students more realized 'Kimchi is our traditional food(38.9%)'. The subjects perceived that the additive material making for Kimchi were powdered red pepper(80.2%), garlic(62.5%), salt(62.0%), salted, fermented shrimp(50.2%). ginger(49.5%), green onion(39.8%), salted, fermented anchovy(34.9%), sesame(25.0%), carrot(22.4%). More than 70% of the subjects estimated for Kimchi 'Kimchi is our traditional food(84.1%)', 'Kimchi must be developed the international food'(73.7%), 'Kimchi must be in succession(75.8%)', 'Kimchi is very nutritious food(70.3%)'. However, they thought Kimchi have to be improved more hygienically(38.8%) and less stimulative taste(25.7%). The knowledge for Kimchi showed a positive correlation with the value on Kimchi and the preferences for Kimchi, and the value on Kimchi was a positively related to the preferences for Kimchi(p<0.01). Therefore it is need to the development of program for the knowledge for Kimchi and the value on Kimchi in order to improvement the preferences for Kimchi.

A Study on the Development of Diagnosing System of Defects on Surface of Inner Overlay Welding of Long Pipes using Liquid Penetrant Test (PT를 이용한 파이프내면 육성용접부 표면결함 진단시스템 개발에 관한 연구)

  • A system for diagnosing surface defects of long and large pipe inner overlay welds, 1m in diameter and 6m in length, was developed using a Liquid Penetrant Test (PT). First, CATIA was used to model all major units and PT machines in 3-dimensions. They were used for structural strength analysis and strain analysis, and to check the motion interference phenomenon of each unit to produce two-dimensional production drawings. Structural strength analysis and deformation analysis using the ANSYS results in a maximum equivalent stress of 44.901 MPa, which is less than the yield tensile strength of SS400 (200 MPa), a material of the PT Machine. An examination of the performance of the developed equipment revealed a maximum travel speed of 7.2 m/min., maximum rotational speed of 9 rpm, repeatable position accuracy of 1.2 mm, and inspection speed of $1.65m^2/min$. The results of the automatic PT-inspection system developed to check for surface defects, such as cracks, porosity, and undercut, were in accordance with the method of ASME SEC. V&VIII. In addition, the results of corrosion testing of the overlay weld layer in accordance with the ferric chloride fitting test by the method of ASME G48-11 indicated that the weight loss was $0.3g/m^2$, and met the specifications. Furthermore, the chemical composition of the overlay welds was analyzed according to the method described in ASTM A375-14, and all components met the specifications.
