Pseudolaric Acid B Induces Apoptosis Through p53 and Bax/Bcl-2 Pathways in Human Melanoma A375-S2 Cells

  • Pseudolaric acid B is a major compound found in the bark of Pseudolarix kaempferi Gordon. In our study, pseudolaric acid B inhibited growth of human melanoma cells, A375-S2 in a time and dose-dependent manner. A375-S2 cells treated with pseudolaric acid B showed typical characteristics of apoptosis including morphologic changes, DNA fragmentation, sub-diploid peak in flow cytometry, cleavage of poly-ADP ribose polymerase (PARP) and degradation of inhibitor of caspase-activated DNase (ICAD). P53 protein expression was upregulated while cells were arrested at the $G_2/M$ phase of the cell cycle. There was a decrease in the expression of anti-apoptotic Bcl-2 and Bcl-xL proteins, whereas pro-apoptotic Bax was increased. The two classical caspase substrates, PARP and ICAD, were both decreased in a time-dependent manner, indicating the activation of downstream caspases.

A Thermostable Protease Produced from Bacillus sp. JE 375 Isolated from Korean Soil (한국의 토양으로부터 내열성 단백질 분해효소를 생산하는 Bacillus sp. JE 375의 선별)

  • A thermophilic microorganism, strain JE 375, which produces a thermostable protease, was isolated from soil and compost in Korea. This gram-positive, rod-shaped, catalase positive, motility positive, and hemolysis ${\beta}$ containing organism was implicated in glucose fermentation, mannitol fermentation, xylose oxidation, aerobic activity and spore formation. The color of the colony was yellowish white. The temperature range for growth at pH 6.5 was between 55 and $70^{\circ}C$, with an optimum growth temperature of $65^{\circ}C$. This result confirmed the strain JE 375 as a thermophilic microorganism. The enzyme was produced aerobically at $65^{\circ}C$ during 20 hr in a medium (pH 6.5) containing 1% trypton. 1% maltose, 0.5% yeast extract and 1% NaCl. The 16S rDNA of strain JE 375 had 97.6% sequence similarity with the 16S rDNA of Bacillus caldoxyloyticus. On the basis of biochemical and physiological properties and phylogenetic analysis, we named the isolated strain as Bacillus sp. JE 375. The thermostable protease from Bacillus sp. JE 375 had been partially purified and characterized. The molecular weight of the enzyme was deduced from SDS-PAGE and gel chromatography as 55 kDa and its optimal temperature was $60^{\circ}C$. The enzyme showed its highest activity at pH 7.5 and was stable from pH 7.0 to 8.0.

Antiproliferative and Apoptotic Effects of Sasa quelpaertensis Nakai in Human Cancer Cells (제주조릿대의 인간 암세포 증식 저해와 자연사멸 효과)

  • Plants are an invaluable source of potential new anti-cancer drugs. Sasa quelpaertensis Nakai (Korean name, Jeju-Joritdae) is one of these plants with medical value, which is a bamboo grass widely distributed in Mt. Halla on Jeju Island, Korea. Here, we investigated the apoptotic effects of S. quelpaertensis leaf extracts in six human cancer cell lines (A549, MCF-7, HepG-2, Hela, HCT116 and A375). MTT assay signified the antiproliferative nature of S. quelpaertensis extracts against all tested cancer cells: S. quelpaertensis displayed slight cytotoxicity against A549, MCF-7 and HepG-2 cells, whereas it was exclusively cytotoxic to Hela, HCT116 and A375 cells. Apoptotic cells were evaluated using PI staining of DNA fragmentation by flow cytometry (sub-G1 peak). PI staining indicated increasing accumulation of Hela, HCT116 and A375 cells at sub-G1 phase. Further events like generation of nitric oxide ($NO^{\bullet}$) were accompanied in the S. quelpaertensis Nakai-induced apoptosis. Augmented $NO^{\bullet}$ generation resulted in the DNA fragmentation of Hela, HCT116 and A375 cells by treatment with S. quelpaertensis leaf extracts. These results suggest that S. quelpaertensis may be a potential natural resource for treating cancer cell. To identify the exact mechanisms of molecular mechanism of S. quelpaertensis induced apoptosis awaits further investigation.

  • A rapid, simple and sensitive spectrophotometric method was developed for the determination of cobalt(II) using 2-hydroxy-3-methoxy benzaldehyde thiosemicarbazone (HMBATSC) as a analytical reagent. The metal ion in aqueous medium forms a brown coloured complex with HMBATSC at pH 6.0. The complex has two absorption maxima at 375 nm and 390 nm. At 375 nm, the reagent shows considerable absorbance, while at 390 nm the reagent does not shows appreciable absorbance. Hence, analytical studies were carried out at 390 nm. Beer's law is obeyed in the range of 0.059-2.357 μg ml-1 of Co(II). The molar absorptivity and Sandall's sensitivity of the method are 2.74×104 l mol-1 cm-1 and 0.0024 μg cm-2 respectively. The interference of various diverse ions has been studied. The complex has 1:2 [Co(II)- HMBATSC] stoichiometry. A method for the determination of cobalt(II) by second order derivative spectrophotometry has also been proposed. The proposed methods were applied for the determination of cobalt(II) in alloy steels, vitamin B12 and in some biological samples.

Optical Properties of $Bi_2S_3$ Thin films ($Bi_2S_3$ 薄膜의 光學的 特性)

  • $Bi_2S_3$ polycrystalline and $Bi_2S_3$ amorphous thin tilm were grown by the evaporation method. The measured lattic parameters were $a=1.708{\AA},\;b=0.351{\AA},\;and\;C=3.943{\AA}$ at substrate temperature $210^{cric}C$ were shown to have the orthorhombic structure. The energy gape of $Bi_2S_3$ polycrystalline that was made from thin film were measured to be 1.375eV at $289^{cric}K.$ The optical band gap of $Bi_2S_3$ amorphous thin film was measured to be 1.71eV at $289^{cric}K.$ It was supposed to mechanism that a photon absorption was changed at the center of 674nm (1.84eV)

  • Layered semiconductor materials can be a promising candidate for large-area thin film transistors (TFTs) due to their relatively high mobility, low-power switching, mechanically flexibility, optically transparency, and amenability to a low-cost, large-area growth technique like thermal chemical vapor deposition (CVD). Unlike 2D graphene, series of transition metal dichalcogenides (TMDCs), $MX_2$ (M=Ta, Mo, W, X=S, Se, Te), have a finite bandgap (1~2 eV), which makes them highly attractive for electronics switching devices. Recently, 2D $MoS_2$ materials can be expected as next generation high-mobility thin-film transistors for OLED and LCD backplane. In this paper, we investigate in detail the electrical characteristics of 2D layered $MoS_2$ local bottom-gated transistor with the same device structure of the conventional thin film transistor, and expect the feasibility of display application.

Triptolide Inhibits Proliferation and Induces Apoptosis of Human Melanoma A375 Cells

  • Triptolide, a diterpenoid obtained from Tripteryglum wilfordii Hook.f, has attracted interest for its antitumor activities against human tumor cell lines in recent years. This report focuses on anti-proliferative and pro-apoptotic activities in human melanoma A375 cells assessed by CCK8 assay, Hoechst 33258 staining and flow cytometry. In addition, triptolide-induced arrest in the S phase was also observed. Caspase assays showed the apoptosis induced by triptolide was caspase-dependent and probably through intrinsic apoptotic pathways. Furthermore, expression of NF-${\kappa}B$ (p65) and its downstream factors such as Bcl-2, Bcl-$X_L$ was down-regulated. Taken together, the data indicate that triptolide inhibits A375 cells proliferation and induces apoptosis by a caspase-dependent pathway and through a NF-${\kappa}B$-mediated mechanism.

Practical and Verifiable C++ Dynamic Cast for Hard Real-Time Systems

  • The dynamic cast operation allows flexibility in the design and use of data management facilities in object-oriented programs. Dynamic cast has an important role in the implementation of the Data Management Services (DMS) of the Mission Data System Project (MDS), the Jet Propulsion Laboratory's experimental work for providing a state-based and goal-oriented unified architecture for testing and development of mission software. DMS is responsible for the storage and transport of control and scientific data in a remote autonomous spacecraft. Like similar operators in other languages, the C++ dynamic cast operator does not provide the timing guarantees needed for hard real-time embedded systems. In a recent study, Gibbs and Stroustrup (G&S) devised a dynamic cast implementation strategy that guarantees fast constant-time performance. This paper presents the definition and application of a cosimulation framework to formally verify and evaluate the G&S fast dynamic casting scheme and its applicability in the Mission Data System DMS application. We describe the systematic process of model-based simulation and analysis that has led to performance improvement of the G&S algorithm's heuristics by about a factor of 2. In this work we introduce and apply a library for extracting semantic information from C++ source code that helps us deliver a practical and verifiable implementation of the fast dynamic casting algorithm.

  • This study was to determine the incidence of pulp stone in each tooth by means of analyzing 375 orthopantomograph taken from 375 patients (252 male), 125 female). The pictures were grouped by age, 20s, 30s, 40s, 50s, and sex. The results were as follows 1. In the group of 20s, male showed the highest incidence in the first molar (40~50%) and the second highest was in the 2nd molar about 35%~50%. In the female group, the first molar and the second molar showed 25%~40%. 2. In the group of 30s, the highest value was found on the first molar and the next was the 2nd molar which appeared 30%~40% in male. In the female, the first molar was 55%~70% and the 2nd molar 30%~50% 3. In the group of 40s, the first molar represented highest value about 45%~60% and the 2nd molar was 40~60% in the male. Female group also showed in the first molar 30%~50% and the second molar 25~30%. 4. In the group of 50s, the highest incidence was 40%~60% on the first molar and next was 35%~50% on the second molar in male, in femalegroup showed 30%~50% on the first molar and 25%~50% on the second molar. 5. The incidence in upper was higher than that of lower. 6. Comparison between right and left showed right was slightly higher than left side.

  • S.deltoides (Compositae) distributed widely in Korea. China. It is edible as a food additive. but there has been no study on chemical constituents. Therefore. we isolated nine compounds from S.deltoldes. On the basis of spectroscopic evidence. the structure of these compounds were characterized as lupeol( 1), $\alpha$-amyrin(2).$\beta$-amyrin (3), ursolic acid(4), nonacosanol(5), nonacosanoic acid(6). mixture of $\beta$-sitosterol. stigmasterol and campesterol (7), $\beta$-sitosteryl-3-O-$\beta$-D-glucopyranoside(8), stigmasteryI-3-O-$\beta$-D-glucopyranoside(9).
