Effect of Immune Activity on Berberis koreana Palibin by Ultra High Pressure Low Temperature Process (초고압 저온처리에 의한 매자나무의 면역 활성)

  • This study was performed to investigate a methodology of improving immune activities of Berberis koreana Palibin by ultra high pressure on low temperature extraction process. Extraction yield was enhanced up to more than 35% by above process, compare to the control and extraction average. The cytotoxicity on human kidney cell (HEK293) was showed below 20.4%, 21.6% in adding 1.0 mg/ml of the highest concentration. Generally, the extracts by ultra high pressure extraction process showed less toxicity about 5% than the other processes. It could tell that toxic materials that existing in the plant could be reduced or broken by ultra high pressure process due to can be broken bond such the hydrogen bond, the electrostatic bond, the van der Waals bond, and the hydrophobic bond, can be broken by high pressure. The result could be employed to develop a new type of functional food from B. koreana Palibin by low temperature high pressure process.

Methanol Extracts of Stewartia koreana Inhibit Cyclooxygenase-2 (COX-2) and Inducible Nitric Oxide Synthase (iNOS) Gene Expression by Blocking NF-κB Transactivation in LPS-activated RAW 264.7 Cells

  • Cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) are involved in various pathophysiological processes such as inflammation and carcinogenesis. In a search for inhibitors of COX-2 and iNOS production we found that extracts of Stewartia koreana strongly inhibited NO and $PGE_2$ production in LPS-treated macrophage RAW 264.7 cells. We have now shown that the mRNA and protein levels of iNOS and COX-2 are reduced by the Stewartia koreana extract (SKE). SKE inhibited expression of an NF-${\kappa}B$ reporter gene in response to LPS, and gel mobility shift assays revealed that SKE reduced NF-${\kappa}B$ DNA-binding activity. The extract also inhibited LPS-induced phosphorylation of $I{\kappa}B-{\alpha}$ and nuclear translocation of p65. Administration of the extract reduced the symptoms of arthritis in a collagen-induced arthritic mouse model. These results indicate that Stewartia extracts contain potentially useful agents for preventing and treating inflammatory diseases.

Variation in Needle Morphology of Natural Populations of Abies nephrolepis Maxim. and A. Koreana Wilson in Korea (분비·구상나무 천연집단(天然集團)의 침엽특성(針葉特性) 변이(變異))

  • Characteristics of needle morphology and anatomy were examined in 14 populations of Abies nephrolepis (Trautv.) Maxim. and A. koreana Wilson. Additionally we studied the classification index to distinguish between the species by the method of discriminant analysis. Characteristics of needle for A. nephrolepis could be distinguished from those for A. koreana by flatten arrangement, thin and long length for needle form, many stomata row, and marginal position of resin duct Nested ANOVA showed that there were statistically significant differences among populations as well as among individuals within populations in all 9 needle traits. For the needle indices such as needle thickness, number of stomata row, and the distance between resin duct and vascular for both species, variance components among populations were larger than those among individuals within populations. The characteristics that contributed most to the separation of A. nephrolepis and A. koreana according to the discriminant analysis using stepdisc procedures were needle index and thickness of needle, needle arrangement index, distance between resin duct and vascular, and number of stomata row.

Cytotoxic Compounds from the Roots of Pulsatilla koreana

  • Seven compounds including hederagenin 3-[(O-${\alpha}$-L-rhamnopyranosyl-($1{\rightarrow}2$)-${\alpha}$-L-arabinopyranosyl) (1), $3{\beta}$-[(O-${\alpha}$-L-rhamnopyranosyl-($1{\rightarrow}2$)-${\alpha}$-L-arabinopyranosyl)oxy]olean-12-en-28-oic acid (2), caffeic acid methyl ester (3), ferulic acid (4), orebiusin A (5), latifonicinin C (6) and 5-(hydroxymethyl)-2-furfuraldehyde (7) were isolated from the ethyl acetate fraction of the roots of Pulsatilla koreana. Their chemical structures were established based on physicochemical and spectroscopic data analyses. All isolates were investigated for their cytotoxic activity against cancer cell lines. Among them, compound 1 showed inhibitory activity against A549, COLO 205, and L1210 cancer cell lines with $IC_{50}$ values of 15.8, 36.5, and 22.8 ${\mu}g$/mL, respectively.

Newly Recorded Naturalized Species in Korea, Silene antirrhina L. (Caryophyllaceae) (한국 미기록 귀화식물: 가는끈끈이장구채(석죽과))

  • A naturalized species, Silene antirrhina L is newly reported in Korea. S. antirrhina L. was found along the Geumho riverside in Daegu. S. antirrhina L. is similar to S. koreana Kom. and S. armeria L. in that it has dark glutinous areas. S. antirrhina L. can be distinguished from congeneric species by the stamens and styles of its sepal and seed surface which are finely papillate. The Korean name of S. antirrhina L. "Ganeun-Kkunkkunijangguchea" was newly formulated based on the overall characteristics of this thin and small plant and on the Korean name of S. koreana Kom. photos of the habitat and a description and key of characteristics are provided.

Anti-oxidative effects of fractionated Pulsatilla koreana $N_{AKAI}$ extracts (백두옹 분획층의 항산화 효과)

  • Objectives : This study was designed to investigate the effects of anti-oxidation of fractionated Pulsatilla koreana $N_{AKAI}$ (PK) extracts. And we examined to determine that a certain fractionated extract has the best anti-oxidative effects between the fractionated PK extracts. Methods : Anti-oxidative effects of fractionated PK extracts was measured by scavenging activities of DPPH, superoxide, nitric oxide (NO) and peroxynitrite radicals. And also scavenging activities of anti-oxidation in lipopolysaccharide (LPS)-treated RAW 264.7 cell was measured. After these examination, we determined a fraction that has best anti-oxidative effects. Results : Fractionated PK extracts inhibited radicals effectively. Also in RAW 264.7 cell, intracellular oxidation has inhibited by PK extracts. In these tests, ethyl acetate (EA) fraction has the best anti-oxidative effects among PK extracts. Conclusions : This results demonstrate that PK extracts exhibit anti-oxidative effects. And EA fraction has the best inhibition effects among the six fractions of PK.

A Cytotoxic Secocycloartenoid from Abies koreana

  • Two triterpenoids, 24-methylene-3,4-seco-cycloart-4(28)-en-3-oic acid (1) and 3-oxo-$9{\beta}$-lanosta-7,22Z,24-trien-26,23-olive (6) were isolated from Abies koreana, together with $\beta$-sitosterol (2), maltol (3), ${\beta}-sitosterol-O-{$\beta}-D-glucoside$ (4), and hexacosylferulate (5). The structures of the compounds were established based on the spectroscopic data. The cytotoxic activities of triterpenoids have been evaluated using the sulforhodamine B (SRB) method. Compound 1 showed moderate cytotoxicities against human lung carcinoma (A549), ovarian carcinoma (SK-OV-3), malignant melanoma (SK-MEL-2), and colon carcinoma (HCT-15) cell lines.

Recently Augmented Natural Habitats of Forsythia koreana (Rehder) Nakai and Abeliophyllum distichum Nakai in Korea (개나리와 미선나무의 새로운 자생지 보고)

  • This study was performed to report the augmented wild populations of Forsythia koreana (Rehder) Nakai and Abeliophyllum distichum Nakai in Korea. Forsythia koreana was found in Budong-myeon, Cheongsong-gun, Gyeongsangbuk-do. The overall size of the natural habitat of Forsythia koreana is $5m{\times}5m$. A total of 12 individuals were recorded in the wild, and 3 of them were in the seedling stage. The wild habitat is in a secondary forest, which is co-dominated by Pinus densiflora Siebold & Zucc and Robinia pseudoacacia L, with 15% coverage. The coverage of the shrub layer is 40% and is dominated by Spiraea prunifolia for. simpliciflora Nakai. The natural habitat of Abeliophyllum disitichum, which is recorded in Uisong-up, Gyeongsangbuk-do, is distributed in sites characterized by boulders along streams, with a patchwork of 15 scattered areas over a habitat size of $50m{\times}15m$. Quercus variabilis Blume is a dominant species in this habitat, with this species dominating at the sub-tree layer. Celtis aurantiaca Nakai also dominates at the shrub layer. The natural ranges of these two species in Korea were mapped based on the terms of the extent of occupancy (EoO).

A Study on the Application of Cosmetic Materials and the Physiological Activities of Forsythia koreana Nakai (개나리꽃의 항산화효과 및 화장품소재에 관한 연구)

  • Objectives : This is the study of the application as the ingredients of cosmetics through the examination of the function for physiological activity of Forsythia koreana Nakai. Methods : Forsythia koreana Nakai, which had been extracted, concentrated, and freeze drying with water and ethanol, have been used for the experiment. The effects on electronic donating ability, SOD-like activity, xanthine oxidase inhibition, whitening effect, nitric oxide inhibition have been investigated in the physiological activity measurement of function experiment. Results : According to the physiological activity measurement of function experiment, it had been found that the electron donating ability, at over 100ppm of water extract and ethanol extract showed relatively high donating ability by more than 80%, and as a result of measuring. In the xanthine oxidase inhibition test, 500ppm of water extract showed an effect of 18% and ethanol extract showed an effect of 31.5%. In the tyrosinase inhibition test, 5000ppm of water extract showed an effect of 60% and ethanol extract showed an effect of 85%. In the anti-inflammatory test, the water extract and ethanol extract inhibited the generation of nitric oxide. Conclusions : The results indicated that extract of Forsythia koreana Nakai can be used as a natural ingredients with biological function in cosmetics ingredients.

Inhibitory Effect of Extracts from Rhododendron Brachycarpum and Abies Koreana E.H. Wilson on Degranulation and Cytokine Expression in RBL-2H3 Cells (만병초와 구상나무 추출물의 RBL-2H3 세포 탈과립, 싸이토카인 유전자 발현에 미치는 영향)

  • Ethanol extracts from Rhododendrom brachycarpum and Abies koreana were investigated for their anti-allergic activities using RBL-2H3 cell line. After treatment with ethanol extracts of various concentrations on the immune response induced mast cell by concanavalin A (Con A), the expressions of cytokine interleukin-4 (IL-4), interleukin-13 (IL-13) were determined by using RT-PCR and the degranulation of mast cells was determined by measuring ${\beta}$-hexosaminidase release. Expression level of IL-4 was decreased by the extract from Rhododendrom brachycarpum in $10^{-7}$, $10^{-5}$ and $10^{-3}%$ concentrations. Expression level of IL-13 was also decreased by both extracts. ${\beta}$-Hexosaminidase release by RBL-2H3 cells was inhibited at the $10^{-5}$ and $10^{-3}%$ concentration of extracts from Rhododendrom brachycarpum and Abies koreana, respectively. These results demonstrate that ethanol extracts of Rhododendron brachycarpum and Abies koreana exert anti-allergic effects by regulating the reduction of IL-4 and IL-13 genes expression and also the secretion of ${\beta}$-hexosaminidase.
