Hsp90 is a good drug target molecule that is involved in regulating various signaling pathway in normal cell and the role of Hsp90 is highly emphasized especially in cancer cells. Thus, much efforts for discovery and development of Hsp90 inhibitor have been continued and a few Hsp90 inhibitors targeting the N-terminal ATP binding site are being tested in the clinical trials. There are no metabolic signature molecules that can be used to evaluate the effect of Hsp90 inhibition. We previously found a potential C-domain binder named PPC1 that is a synthetic small molecule. Here we report the metabolomics study to find signature metabolites upon treatment of PPC1 compound in lung cancer cell line, A549 and discuss the potentiality of metabolomic approach for evaluation of hit compounds.

Ferroelectric PZT(10/90)/(90/10)heterolayered thin films were fabricated by the alkoxide-based Sol-Gel method Electric and dielectric properties of PZT(10/90)/(90/10) heterolayered thin films have been investigated, focusing on the effect of PZT/PZT and PZT/electrode interface on the heterolayered films. Dielectric constant increased with increasing the number of coatings. Increasing the number of coatings, remanent polarization and coercive field were decreased and the values of the PZT-6 heterolayered thin films deposited on Pt/Ti/$SiO_2$/Si were $7.18{\mu}C/cm$, $68.5kV/cm^2$, respectively. Leakage current, densities were increased with increasing the number of coatings, and the value of the PZT-4 film deposited on the Pt/Ti/$SiO_2$/Si substrate was about $7{\times}10^{-8}A/cm^2$ at 0.05MV/cm.

We have fabricated a superconducting YBCO system according to the pyrolysis method and low pressure apparatus. In our experiment, the X-ray diffraction pattern of the non doped YBaCuO layer indicated that the superconductor contained only 90K phase crystal. The critical temperature and critical current density for a thick layer at $650^{\circ}C$ were Tc=90 K and $Jc=6{\times}10^{4}A/cm^2$ at 90K. In low pressure apparatus, the 90 K phase YBaCuO was grown at a lower temperature compared with the normal system. Tc and Jc at $650^{\circ}C$ were Tc = 90 K and $Jc=6{\times}10^{4}A/cm^2$ at 90K.

하이어 풀러렌의 구성요소인 나머지 단위에 대하여 연구하였다. 나머지 단위에 2개의 $C_1$이 존재할 때마다 $C_{90}$의 에너지는 약 11.0 kcal/mol 정도 증가하였으며 나머지 단위의 평면성이 $1.0{\beta}$만큼 증가할 때마다 5.6 kcal/mol 정도의 에너지가 증가하였다. 따라서 46개의 $C_{90}$ 이성질체 중에서 나머지 단위에 $C_1$도 존재하지 않고 나머지 단위의 평면성이 적은 것이 안정하다는 사실을 알 수 있었다. 제 존재가 확인되는 $C_{90}$이성질체들은 이러한 조건을 만족하고 있다.

녹병은 Puccinales 목의 병원성 곰팡이가 일으키는 식물병으로서 다양한 식물에 발병한다. 최근 많은 연구자들은 녹병을 효과적이고 친환경적으로 방제하기 위하여 연구를 활발히 진행하고 있다. 본 연구에서는 당후박(Magjia90) 추출물로부터 제조된 유제(EC10)와 수화제(WP20)의 들깨 녹병과 잔디 녹병에 대한 방제효과를 포장에서 조사하였다. Magjia90-EC10과 Magjia90-WP20은 들깨 녹병에 대하여 47.9% 내지 69.6%의 방제효과를 보였고, 수화제는 유제보다 우수한 효과를 보였다. 또한 Magjia90-WP20은 전디 녹병에 대하여 65.7% 내지 80.5%의 높은 방제효과를 보였다. 다른 한편으로, 두 제제를 처리한 식물체 잎에서 어떠한 약해도 발견되지 않았다. 이상의 결과는 Magjia90이 녹병 방제를 위한 천연물 살균제로서 사용이 가능하다는 것을 강력하게 나타낸다.

본 연구에서는 담수사육 감성돔을 대상으로 수온을 상승시켰을 때, 세포적 스트레스 측면에서 HSP90 및 HSP70 mRNA의 발현 정도를, 신경-내분비적 스트레스 측면에서 혈장 cortisol 및 glucose 농도를 조사하였다. RT-PCR법을 이용하여 생식소로부터 HSP90 (891 bp) 및 HSP70 (465 bp) cDNA 단편을 클로닝 하여, 타 종과 그 상동성을 비교해 본 결과, 감성돔 HSP90은 참돔 HSP90과 99%, 무지개송어 HSP90과 95%, 대서양 연어HSP90과 94%, zebrafish HSP90과 94%로 나타났으며, 감성돔 HSP70은 무지개송어 HSP70과 96%, silver seabream HSP70과 95%, zebrafish HSP70과 95%의 상동성을 나타내었다. 감성돔의 사육수온을 $30\;^{\circ}C$로 상승시켰을 때, HSP90 mRNA는 모든 조직에서 그 발현 정도가 $20\;^{\circ}C$ 실험구에 비하여 $7{\sim}9$배 정도 높았으나, HSP70 mRNA는 생식소에서만 발현하는 것으로 나타났다. 혈장 cortisol 및 glucose 농도는 $20\;^{\circ}C$ 실험구에 비하여 $30\;^{\circ}C$ 실험구에서 유의하게 증가한 것으로 나타났다.

Ribosomal S6 kinase is a family of serine/threonine protein kinases involved in the regulation of cell viability. There are two subfamilies of ribosomal s6 kinase, (p90rsk, p70rsk). Especially, p90rsk is known to be an important downstream kinase of p44/42 MAPK. We investigated the role of p90rsk on ethanol-induced cell proliferation of HepG2 cells. HepG2 cells were treated with 10~50 mM of ethanol with or without ERK and p90rsk inhibitors. Cell viability was measured by MTT assay. The expression of pERK1, NHE1 was measured by Western blots. The phosphorylation of p90rsk was measured by ELISA kits. The expression of Bcl-2 was measured by qRT-PCR. When the cells were treated with 10~30 mM of ethanol for 24 hour, it showed significant increase in cell viability versus control group. Besides, 10~30 mM of ethanol induced increased expression of pERK1, p-p90rsk, NHE1 and Bcl-2. Moreover treatment of p90rsk inhibitor attenuated the ethanol-induced increase in cell viability and NHE1 and Bcl-2 expression. In summary, these results suggest that p90rsk, a downstream kinase of ERK, plays a stimulatory role on ethanol-induced hepatocellular carcinoma progression by activating anti-apoptotic factor Bcl-2 and NHE1 known to regulate cell survival.

제주도산 돼지의 품질 특성을 알아보기 위하여 생체중 90∼100 kg(JJ90) 및 110∼120 kg(JJ110)의 일반돼지와 생체중 90∼100 kg(JB90)의 개량 흑돼지 등심에 대한 구성 아미노산과 유리 아미노산 함량, pH, 보수력, 경도, 씹힘성, 색도 및 기호도를 평가하였다. 구성 아미노산 총량은 18.15∼20.22%로 시료들 사이에 유의한 차이가 없었으며, 모두 glutamic acid, aspartic acid, lysine 및 leucine이 많이 함유되어 있었다. 유리아미노산 총량은 JB90이 0.891%로 JJ90과 JJ110보다 많았으며, taurine, serine, glycine, alanine, valine과 leucine이 유의적으로 높게 나타났다. 적색도 및 황색도는 JB90이 JJ90과 JJ110보다 유의적으로 높았다. 보수력, 경도, 씹힘성, 다즙성 및 종합적인 기호도는 JJ90보다 JJ110과 JB90이 우수하였으나 유의적 차이가 나타나지 않았다.

본 논문에서는 90$^{\circ}$ 하이브리드 결합기에 저역통과 여파기 특성을 결합하여 전력분배와 하모닉 저지특성효과를 보여주고 있다. 일반적인 90$^{\circ}$ 하이브리드 결합기는 위상차를 갖고 전력을 분배, 합성하는 데에만 쓰이고 있기 때문에 비선형 소자와 결합함으로써 생기는 하모닉 성분을 억제하려면 여파기를 따로 설계해야 하는 단점이 있다. 그래서 본 논문에서는 90$^{\circ}$ 하이브리드 결합기에 저역통과 여파기 특성을 결합함으로써 위상차와 전력분배는 물론 여파기 특성인 감쇠효과를 이용하여 하모닉 성분을 크게 감소시킬 수 있는 구조를 제시하였다.

Hsp90, an evolutionarily conserved molecular chaperone, is involved in the folding, stabilization, activation, and assembly of a wide range of 'client' proteins, thus playing a central role in many biological processes. Especially, several oncoproteins act as Hsp90 client proteins and tumor cells require higher Hsp90 activity than normal cells to maintain their malignancy. For this reason, Hsp90 has emerged as a promising target for anti-cancer drug development. It is still largely unknown how Hsp90 can recognize structurally unrelated client proteins. However, recent progress in structural studies on Hsp90 and its interaction with various co-chaperones has broadened our knowledge of how the Hsp90 ATPase activity, which is essential for its chaperone function, is regulated and coupled with the conformational changes of Hsp90 dimer. This review focuses on the roles of various Hsp90 co-chaperones in the regulation of the Hsp90 ATPase cycle, as well as in the selection of client proteins. In addition, the current development of Hsp90 inhibitors based on the structural information will be discussed.
