대두(Glycine max)의 ${\beta}$-1,3-glucanase를 분리동정하고 benzyladenine(BA)이 이 효소의 세포내 함량과 활동도에 미치는 영향을 조사하였다. 또 세포벽의 callose함량과 protoplast의 세포벽재생에 미치는 BA의 영향을 조사하여, cytokinin이 식물의 세포벽재생을 촉진하는 기능이 있음을 확인하고 세포벽재생에 있어서 cytokinin의 작용기구를 검토하였다. 대두 ${\beta}-1,3-glucanase$는 21 kD의 polypeptide로 동정되었는데 이 polypeptide의 세포내함량과 효소활성은 BA처리에 의하여 저하되었다. 그리고 callus세포벽의 callose함량과 protoplast의 세포벽재생율이 BA처리에 의하여 증가되었다. 이 결과들은 cytokinin이 세포의 ${\beta}-1,3-glucanase$수준을 저하시켜 callose분해를 억제함으로써 세포벽 재생을 촉진할 수 있음을 보여주었다.
본 연구는 옥수수 뿌리에서 cytokinin ($N^6$-benzyladenine; BA)과 IAA를 각각 혹은 같이 처리하여 ethylene 생성에 미치는 영향을 알아보았다. 두 호르몬을 외부에서 동시에 처리하면 ethylene 생성은 각각 처리하였을 때 보다 함께 처리하면 상승적으로 증가하였다. 예를 들면, $10^{-4}\;M$ BA와 $10^{-4}\;M$ IAA를 처리하면 ethylene 생성량은 각각 대조구의 87%와 170% 증가되지만 함께 처리하면 대조구의 480%가 증가된다. 이러한 상승적 효과는 에틸렌생성에 관여하는 효소인 ACC synthase의 활성과 유전자 발현 수준에서도 볼 수가 있었다. 그러나 에틸렌 생성에 관여하는 다른 효소인 ACC oxidase의 활성에서는 관찰되지 않았다. 이러한 결과는 cytokinin인 BA와 IAA가 옥수수 뿌리에서 ACC synthase에 영향을 미쳐 ethylene 생성을 상승적으로 촉진시킨다는 사실을 제시한다.
대두(Glycine max var. Acme)의 자엽조직에서 유래된 callus의 부유배양세포로부터 protoplast를 분리하고, 이들 protoplast의 viability, 세포벽재생 및 세포분열에 대한 benzyladenine(BA)의 영향과 protoplast에 의한 BA의 흡수특성을 조사하였다. Protoplast의 viability는 BA처리에 의하여 증가되었고, 세포벽재생과 세포분열 그리고 callue의 생장도 BA처리에 의하여 촉진되었다. Protoplast에 의하여 흡수된 BA의 양은 BA 처리후 약 20시간에 최대에 이르렀고, 이중에서 약 2/3가 6시간 이내에 흡수되었다.
담배에서 기내 생장, 엽록소 및 광합성 효소인 rubisco와 rubisco activase에 미치는 벤질 아데닌(BA)의 영향을 연구하였다. 여러 농도의 BA 처리 후 11주에 2 ${\mu}M$의 농도에서 기내 생장과 엽록소의 함량이 가장 현저하였다. Rubisco에 대한 이러한 효과가 rubisco activase와 관련되어 있을 것이라는 가정하에 이 효소의 함량과 활성을 측정한 결과, 2 ${\mu}M$ BA에서 가장 증가하였다. 이러한 결과들을 종합하면 BA에 의한 rubisco의 활성화 효과는 rubisco activase와 관련되어 있음을 보여준다.
감자의 생장점배양시 기본배지의 종류 및 기본배지에 첨가되는 2,4-D, NAA 및 Benzyladenine이 감자의 생장점으로부터의 기관의 분화 및 생장과 Callus의 유기에 미치는 영향을 구명하고져 본실험을 준행하였던 바 그 결과를 요약하면 다음과 같다. 1. 기본배지중에 Benzyladenine (BA)의 농도를 증가시킬수록 감자 생장점에서 유기된 경의 신장은 현저히 촉진되었으나 BA를 첨가함 배지상에서는 근 및 Callus는 전혀 유기되지 않았다. 2. NAA를 0.5ppm 이상 함유하고 있는 배지상에서는 경의 분화가 완전히 억제되었고 모두 Callus가 유기되었으며 NAA의 농도를 증가시킬수록 Callus의 생장은 촉진되었다. 3. 2.4-D 1.0ppm 이하 함유하는 배지상에서 경의 분화가 완전히 억제되었고 Callus가 유기되었으며 Callus 생장율은 2.4-D 농도가 높을수록 많았는데 2,4-D보다는 NAA 첨가배지사에서 Callus의 생장율은 현저한 증가를 보였다. 4. NAA만을 첨가한 배지상에서는 경의 분화 및 생장이 억제되었으나 BA와 NAA를 조합처리하였을 때는 NAA 0.01ppm에서보다 0.1ppm에서 경의 분화 및 생장이 현저히 증가되었으며 2,4-D와 BA의 조합처리에서도 같은 경향이었던 바 BA와 NAA 혹은 2,4-D의 조합처리시에는 경의 분화 및 신장이 촉진되는 방향으로의 교호작용이 현저하였다. 5. 엽편보다는 경편에서 Callus의 유기 및 생장이 양호하였으며 이들 경엽조직편으로부터의 Callus 유기 및 생장에는 NAA보다는 2,4-D가 더욱 효과적이 었다. 6. MS 표준배지보다는 MS배지의 농도를 $\frac{1}{2}$ 로 희석하여 배지중의 무기성분 농도를 저하시킨 배지상에서 거의 경의 분화 및 신장이 현저히 촉진되었다. 7. 감자 생장점배양시 경의 유기 및 생장에는 MS \frac{1}{2} 배지에 BA 1.0ppm 및 NAA 혹은 2,4-D 0.1ppm을 첨가한 배지가 가장 효과적이었다.
A protocol is described for rapid multiplication of Zanthoxylum piperitum DC. (Rutaceae), an important aromatic and medicinal plant, through shoot-tip explant cultures. Murashige and Skoog (MS) medium supplemented with various concentrations of N-6-benzyladenine (BA), N-6-benzylaminopurine (BAP) and thidiazuron (TDZ), in single or in combination with ${\alpha}-naphthaleneacetic$ acid (NAA), was used to determine the rate of shoot proliferation. N-6-benzyladenine (BA) used at 0.5mg/l, was the most effective in initiating multiple shoot proliferation at the rate of 23 microshoots per shoot-tip explants after 40 days of culture. Shoot multiplication increased 1.2-fold in each successive subculture. Induction of rooting (98%) was achieved by transferring the shoots to the same basal medium containing 2 mg/l indole-3-butyric acid (IBA). Plantlets went through a hardening phase in a controlled growth chamber, prior to in vivo transfer. These results represented that possible application for the mass production of plantlets through in vitro culture system of Zanthoxylum piperitum DC.
To establish direct multipropagation through organogenesis from nodal explants of Anoectochilus formosanus Hayata, the nodes were cultured on LS medium containing various concentrations of 6-benzyladenine(BA). High plant regeneration and adventitious bud formation were obtained from supplemented with 4.0mg/l of BA. Plant height was promoted by adding 0.3% activated charcoal. Plantlet regeneration capacity from nodes was depended on nodal parts on the stem, upper position was the best comparing with intermediate and lower.
The genetic status of somatic embryo-derived plantlets of Cymbopogon flexuosus was examined by randomly amplified polymorphic DNA (RAPD) analysis. Auxins such as 2, 4-dichlorophenoxyacetic acid (2, 4-D) (1-4 mg/l) were used in Murashige and Skoog (MS) medium for induction of calli from rhizomatous explants of Cymbopogon flexuosus. Optimum calli were induced on MS medium supplemented with 2, 4-dichlorophenoxyacetic acid (2, 4-D) (3.5 mg/l) alone or in combination with $N^6-benzyladenine$ (2 mg/l). Somatic embryogenesis was achieved from long term calli when cultured on MS medium containing 2, 4-dichlorophenoxyacetic acid (2, 4-D) (2 mg/l) along with $N^6-benzyladenine$ (BA) (1-2 mg/l). Regeneration was achieved when freshly induced embryogenic calli were sub-cultured on MS medium supplemented with $N^6-benzyladenine$ (3 mg/l) alone. Long-term cultured embryos showed profuse minute rooting on regeneration medium supplemented with N6 -benzyladenine (3 mg/l). Microshoots were rooted in the presence of indole-butyric acid (IBA) (2 mg/l). DNA samples from the mother plant and 18 randomly selected regenerated plants from a single callus were subjected to RAPD analysis with 6 arbitrary decamer primers for the selection of putative somaclones. A total of 64 band positions were scored, out of which 19 RAPD bands were polymorphic. From genetic similarity coefficient based on RAPD band data sharing, it was found that the majority of the clones were almost identical or more than 92% similar to the mother plant, except CL2 and CL9 (66%) which showed highest degree of genetic change with CL2 and CL9 showing presence of two non-parental bands each.
The effect of NAA and Benzyladenine(BA) for determination times on the formation of adventitious shoots, roots trichmoes and calli in MS basal medium was investigated in leaf segments from ecotype 'Nosses' of Arabidopsis thalliana. Adventitious shoots, roots, trichomes and calli were formed fromed from leaf segments in a wede range of NAA and BA. The optimal combination of hormones for adventitious shoots formation, 20mg/L NAA for trichome formation, 100mg/L for callus formation. Inductive times for formation of adventitious shoots, roots, trichomes and calli were determined at 14, 4, 6 and 18 days respectively by periodical transfer of leaf segments from hormines containing media to hormone free medium.
This study was carried out to reveal optimum conditions for in vitro propagation of 3 Korean native azalea species, Rhododendron mucronulatum, R. yedonese var. poukhanense, and R. shlippenbachii, which are useful for landscape proposes. Seeds and meristems from three azalea species were cultured on 1/2MS, Hyponex, and Anderson media containing growth of regulators benzyladenine(BA) and 2-isopentenyadenosine(2ip). The results were as follows. 1. In the culture of R. schlippenbachii and R. mucronulatum seeds, in vitro seedlings germinated and grew well on he 1/2MS and Anderson media, while R. yedoense var. poukhanense on Hyponex media containing 6.0mg/$\ell$ 2ip. 2. When the meristems of R. mucronulatum were cultured on Andeson media containing 9.0mg/$\ell$ 2ip, the survival rate of meristems was 23.0% in 6 weeks after culture, and the survival rate of R. schlippenbachii was 46.0% o nthe same media containing 12.0mg/$\ell$ 2ip. The survial rate of R. yedoense var. poukhanense was 92.0% onHyponex media containing 0.5mg/$\ell$ BA and 9.0mg/$\ell$ 2ip. When the meristems of R. mucronulatum and R. yedoense var. poukhanense were cultured on Hyponex media containing 12.0mg/$\ell$ 2ip, they showed the most excellent growth. R. schlippenbachii grew well on Anderson media containing 9.0mg/$\ell$ 2ip. When in vitro shoots of R. yedoense var. poukhanense were subcultured to solid medium, they grew well in shoot growth on Hyponex media containing 6.0mg/$\ell$ 2ip.
