Composite Membrane Preparation for Low Pressure Using Salting-Out Method and Its Application to Nanofiltration Process (염석법에 의한 저압용 역삼투막 제조 및 NF로의 적용)

  • Nanofiltration composite membranes were prepared through the ion exchange polymers coating onto the porous microfiltration polyethylene (PE) membrane surfaces the salting-out and phase separated and pressurization (PSP) methods. The existence of coating on the surfaces was confirmed by the scanning electronic microscopy. The resulting membranes were characterized under the various conditions, such as the coating material, coating time, ionic strength etc., in terms of flux and rejection for NaCl 100 ppm solution. Under the same coating conditions of 10,000 ppm coating solution concentration and 3 atm coating pressure for both the coating materials of PEI and PSSA_MA, the flux 91.2 LMH and rejection 64.6% were obtained for PEI whereas 122.7 LMH and 38.1% were observed for PSSA_MA. From this study, it may be concluded that the composite membrane preparation is possible.

Activation of Small GTPases RhoA and Rac1 Is Required for Avian Reovirus p10-induced Syncytium Formation

  • The first ORF of the ARV S1133 S1 segment encodes the nonstructural protein p10, which is responsible for the induction of cell syncytium formation. However, p10-dependent signaling during syncytium formation is fully unknown. Here, we show that dominant negative RhoA, Rho inhibitor C3 exoenzyme, ROCK/Rho-kinase inhibitor Y-27632 and Rac1 inhibitor NSC23766 inhibit p10-mediated cell fusion. p10 over-expression is concomitant with activation and membrane translocation of RhoA and Rac1, but not cdc42. RhoA and Rac1 downstream events, including JNK phosphorylation and transcription factor AP-1 and $NF-{\kappa}B$ activation, as well as MLC expression and phosphorylation are simultaneously activated by p10. p10 point mutant T13M possessed 20% fusion-inducing ability and four p10 fusion-deficient mutants V15M, V19M, C21S and L32A reduced or lost their ability to activate RhoA and Rac1 signaling. We conclude that p10-mediated syncytium formation proceeds by utilizing RhoA and Rac1-dependent signaling.

The Effect of Sesame Oil, Perilla Oil and Beef Tallow on Body Lipid Metabolism and Immune Response (한국에서 상용되는 식용유지로 사육된 흰쥐의 체내 지방대사 및 면역능력에 대한 연구)

  • The research was designed to study the effect of different fat sources and levels on Body lipid metabolizm and immune responses in Sprague-Dawely strain male rats. These effect of different fat sources compared with sesame Oil, Perilla oil and Beet tallow. Fat sources were divided into 3 groups respectively 7%, 15%, 30% fat level on diet weight basis. The experimental period was 54days. 1) The body weight gain was significantly low in NF group. In Sesame oil group and perilla oil group, low fat level groups were higher than medium, high fat level groups. But in Beef tallow group, high fat level groups were higher than low and Medium groups. 2) The weight of liver, kidney and epididymal fat ped tend to increse with increasing body weight. 3) The contents of triglyceride and total lipid in serum were significantly different with dietary fat sources and Perilla oil group was the lowest. 4) The contents of triglyceride and total lipid in liver were significantly different with dietary fat levels and high fat level group was higher than low fat level group. 5) Perilla oil group, compared with Beef tallow group, showed the higher excretion of cholesterol through feces and the higher deposit of cholesterol in liver. Therefore serum cholesterol level of Perilla oil group was lower than that of Beef tallow group. But eventhough Sesame oil is vegetable oil, Sesame oil did not showed an effect like Perllia oil on serum cholesterol level. 6) Weight of thymus decreased with fat levels particularly in vegetale oil. And it had on effect on mitogen response, mitogen response decreased with fat level in vegetable oil. But in Beef tallow, there was no difference in fat level.

Effects of Angiopoietin-2 on the Proliferation and Activity of Ostoeblasts and Osteoclasts (Angiopoietin-2가 조골세포와 파골세포의 성장과 활성에 미치는 영향)

  • The present study was undertaken to determine the possible cellular mechanism of action of angiopoietin-2 in bone metabolism. The effects on the osteoblasts were determined by measuring 1) cell viability, 2) alkaline phosphatase (ALP) activity, 3) gelatinase activity, and 4) nitric oxide production. The effects on the osteoclasts were investigated by measuring 1) tartrate-resistant acid phosphatase (TRAP)(+) multinucleated cells (MNCs) formation, and 2) resorption areas after culturing osteoclast precursors. Angiopoietin-2 treatment showed a significant increase in both the viability and ALP activity of osteoblasts. Angiopoietin-2 increased the activity of gelatinase and nitric oxide production. In addition, angiopoietin-2 decreased the osteoclast generation induced by macrophage-colony stimulating factor (M-CSF) and receptor activator of NF-kB ligand (RANKL), and inhibited osteoclastic activity in (M-CSF)-dependent bone marrow macrophage (MDBM) cell cultures. Taken these results, angiopoietin-2 may be a regulatory protein within the bone marrow microenvironment.

Technology of selective absorber coatings on solar collectors using black chromium+3 sulfate acid on substrates (흑색 황산3가크롬을 이용한 태양열 흡열판 선택흡수막 도금기술)

  • One of the most important factors that have a large influence on performance of the solar water heater system is performance of the solar collector, more detailedly, coating technology on the surface of the solar collector, which can provide high solar absorptance and low emittance. The core of the coating technology is to coat solar selective surfaces. In this study, various performance experiments are carried out using $Cr_2(SO_4)_3{\cdot}15H_2O$ coating technology. Here, IGBT(Insulated Gate Bipolar Transistor) of 5000A-15V was used as the surface processing rectifier which can stably output power and also can control voltage and current. The plating solution mainly contains black chrome$^{+3}$ concentration, H-y Conductivity, N-u Complex, NF Additive and NC-2 Wetter. Before applying the black chrome coating on the copper plate, optimal conditions are provided by using various preprocessing methods such as removal of fat, activation, electrolytic polishing, nickel strike, copper sulfate plating and bright neckel plating, and then the automatic continuous coating experiment are performed according to plating time and cathode current density. In the experiment, after the removal of fat, chemical polishing, nickel strike and activation processes as the preprocessing methods, the black chrome coating was performed in a plate solution temperature of $28^{\circ}C$ and a cathode current density of $18A/cm^2$ for 90 seconds. The thickness of chrome and nickel on the coated plate is $0.389{\mu}m$, $159{\mu}m$ respectively. As a result of the coating experiment, it showed the most excellent performance having a high solar absorptance of 98% and a low emittance of $5{\pm}1%$ when the black chrome surface had a thickness of $0.398{\mu}m$.

Anti-Inflammatory Principles from the Fruits of Evodia rutaecarpa and Their Cellular Action Mechanisms

  • The fruits of Evodia rutaecarpa Benth (Rutaceae) has long been used for inflammatory disorders and some anti-inflammatory actions of its constituents such as dehydroevodiamine, evodiamine and rutaecarpine were previously reported. Since the pharmacological data is not sufficient to clearly establish the scientific rationale of anti-inflammatory medicinal use of this plant material and the search for its active principles is limited so far, three major constituents (evodiamine, rutaecarpine, goshuyuamide II) were evaluated for their anti-inflammatory cellular action mechanisms in the present study. From the results, evodiamine and rutaecarpine were found to strongly inhibit prostaglandin $E_2$ synthesis from lipopolysaccharide-treated RAW 264.7 cells at $1-10{\mu}M$. Evodiamine inhibited cyclooxygenase-2 induction and NF-kB activation, while rutaecarpine did not. On the other hand, goshuyuamide II inhibited 5-lipoxygenase from RBL-1 cells $(IC_{50}=6.6{\mu}M)$, resulting in the reduced synthesis of leukotrienes. However, these three compounds were not inhibitory against inducible nitric oxide synthase-mediated nitric oxide production from RAW cells up to $50{\mu}M$. These pharmacological properties may provide the additional scientific rationale for anti-inflammatory use of the fruits of E. rutaecarpa.

Innate Immunity Activation and Anti-Inflammation Effects of Evodia Rutaecarpine Water Extract (오수유 물 추출물의 선천 면역 활성과 염증 억제 효과)

  • Objectives: This study was designed to examine immuno-modulatory effects of Evodia Rutaecarpine by activating innate immune system and inhibiting inflammation. Methods: First, Cell cytotoxicity was examined with 4T1 breast carcinoma and TG-induced macrophage. To investigate activating innate immune system of Evodiamine Rutacarpine Extract (ERE) on macrophage, we tested tumor necrosis factor-alpha (TNF-α), interleukin-12 (IL-12), and interleukin-6 (IL-6). In addition, TNF-α and nitric oxide (NO) induced by lipopolysaccharide (LPS) were measured after treating with ERE to observe innate immune modulating effect of ERE on RAW 264.7 cell. Also, mitogen-activated protein kinase (MAPK) and nuclear factor κB (NF-κB) were examined by western blot analysis. Results: In cytotoxicity analysis, ERE significantly affected tumor cell growth above specific concentration. Also, ERE significantly affected macrophage growth above specific concetration. As compared with the control group, the production of TNF-α, IL-12 and IL-6 were increased in TG-induced macrophage. As compared with the control group, TNF-α and IL-6 were significantly up-regulated in RAW 264.7 cell. The expression of TNF-α and NO induced by LPS after treating ERE was significantly decreased compared with control group. In addition, We observed ERE inhibited the phosphorylation levels of p-extracellular signal-regulated kinase (p-ERK), p-Jun N-terminal kinase (p-JNK), and p-p38 in western blotting by treating ERE on RAW 264.7 cell. Conclusions: ERE seems to have considerable impact on the anti-cancer effect by activation of innate immune system and inflammation control.

Morin alleviates fructose-induced metabolic syndrome in rats via ameliorating oxidative stress, inflammatory and fibrotic markers

  • Metabolic syndrome (MBS) is a widespread disease that has strongly related to unhealthy diet and low physical activity, which initiate more serious conditions such as obesity, cardiovascular diseases and type 2 diabetes mellitus. This study aimed to examine the therapeutic effects of morin, as one of the flavonoids constituents, which widely exists in many herbs and fruits, against some metabolic and hepatic manifestations observed in MBS rats and the feasible related mechanisms. MBS was induced in rats by high fructose diet feeding for 12 weeks. Morin (30 mg/kg) was administered orally to both normal and MBS rats for 4 weeks. Liver tissues were used for determination of liver index, hepatic expression of glucose transporter 2 (GLUT2) as well as both inflammatory and fibrotic markers. The fat/muscle ratio, metabolic parameters, systolic blood pressure, and oxidative stress markers were also determined. Our data confirmed that the administration of morin in fructose diet rats significantly reduced the elevated systolic blood pressure. The altered levels of metabolic parameters such as blood glucose, serum insulin, serum lipid profile, and oxidative stress markers were also reversed approximately to the normal values. In addition, morin treatment decreased liver index, serum liver enzyme activities, and fat/muscle ratio. Furthermore, morin relatively up-regulated GLUT2 expression, however, down-regulated NF-κB, TNF-α, and TGF-β expressions in the hepatic tissues. Here, we revealed that morin has an exquisite effect against metabolic disorders in the experimental model through, at least in part, antioxidant, anti-inflammatory, and anti-fibrotic mechanisms.

Hataedock Treatments for Dermatophagoides Farinae-induced Atopic Dermatitis in NC/Nga Mice Treated with High-fat Diet

  • Hataedock (HTD) treatment is a traditional preventive therapy for the fetal toxicosis- the acute allergic disease after childbirth, mainly manifested by a variety of skin allergies such as scab, phlegm. The aim of this study was to investigate the efficacy of HTD treatments for the alleviation of inflammation in Dermatophagoides farinae-induced obese NC/Nga mice. 20 mg/kg of Coptidis Rhizoma, Glycyrrhizae Radix (CRGR) extracts as a remedy of HTD treatments were orally administered to NC/Nga mice. We induced obesity in the mice by high-fat diet. To induce skin allergies, the extracts of Dermatophagoides farinae were topically applied on the NC/Nga mice at 4th-6th and 8th-10th weeks. Structural and molecular changes in the skin tissues were measured by immunohistochemical staining. HTD treatment decreased the atopic dermatitis (AD)-like symptoms including hemorrhage, erythema, erosion, edema, and dryness. HTD treatment suppressed the mast cell activation confirmed by reduction of $Fc{\varepsilon}RI$, substance P, and serotonin. The expression of several inflammatory mediators including nuclear factor-kappa B ($NF-{\kappa}B$) p65, inducible nitric oxide synthase (iNOS), vascular endothelial growth factors (VEGFs) was also decreased by HTD treatment. HTD treatment suppressed the allergic, inflammatory responses in the skin tissues of the NC/Nga mice by reducing mast cells and down-regulating several inflammatory mediators.

Soybean Fermented with Bacillus amyloliquefaciens (Cheonggukjang) Ameliorates Atopic Dermatitis-Like Skin Lesion in Mice by Suppressing Infiltration of Mast Cells and Production of IL-31 Cytokine

  • The present study was conducted with the aim to investigate the ameliorative effects of a new soybean product (cheonggukjang) fermented with Bacillus amyloliquefaciens SCGB1 (SFBA) in atopic dermatitis (AD) mouse model. Visual evaluation of AD induction in the mice indicated the remarkable control of SFBA in reducing the pathological severity of AD-like skin lesions reported as the SCORAD score of AD clinical symptoms. The results revealed that SFBA reduced dorsal skin and epidermal thickness to a similar extent with prednisolone. Further analysis revealed the dominance of SFBA in restraining mast cell infiltration in the dermis; immunoglobulin-E expression in serum; and TH2 IL-4 cytokine and itch-related IL-31 cytokine in the mice skin and serum. SFBA also suppressed scratching behaviours in mice induced by compound 48/80. Further histological findings also revealed the alleviation of collagen fiber deposition in dermal skin of the AD mice model. These actions of SFBA were examined to be mediated by its suppression of the phosphorylation activation of key signalling molecules such as $NF-{\kappa}B$ and MAPK responsible for the induction of cytokine production. Thus, SFBA can be considered as a promising functional food for managing clinical, histological and immunological spectra associated with AD.
