LC/MS에 의한 퉁퉁마디의 Flavonoids 정량 분석 (Quantitative Analysis of Flavonoids from Salicomia herbacea L. Extracst by LC-MS)

  • 본 연구는 염전에서 자라고 있는 퉁퉁마디를 채집하여 플라보노이드 성분을 LC/MS로 정량하였다. 퉁퉁마디 분말에서 flavonol계 물질인 quercetin (124.43 ppm), rutin (2.57 ppm), quercetin-3-${\beta}$-glucoside (3992.49 ppm), quercetin-3',4'-glucoside(0.08 ppm) 그리고 isorhamnetin (27.81 ppm)이 검출되었다. 특히 quercetin-3-${\beta}$-glucoside가 차지하는 비중은 건초에서 99%, 분말에서 96% 이상으로 높게 나타났다. 이러한 결과들은 염생식물인 퉁퉁마디가 항산화제로서의 높은 기능성 물질 임을 시사해 주고 있다.

  • 유채유를 이용해서 만든 새로운 정련비누와 현재 시중에서 사용되고 있는 정련 비누로, 정련염색 및 물성시험을 통하여 얻어진 결과를 비교 분석하여 요약하면 다음과 같다. 1. 정련율에서는 OC(올레인산으로 만든 비누)를 사용한 것이 가장 좋고 RS(유채유로 만든 비누)를 사용한 것이 다음으로 모두 MS(시중에서 사용되고 있는 비누)를 사용한 것보다 우수하였다. 2. 완충력을 측정한 결과 RS와 OS는 비슷하였지만 MS는 크게 떨어졌다. 3. 오염성에 미치는 영향으로는 OS와 RS로 정련한 것이 반점이 없었으나, MS로 정련한 것은 염반이 발생하였다. 4. 강·신도에서는 큰 차이가 없었으나, MS로 정련한 것이 OS, RS로 정련한 것보다 다소 낮았다. 5. 정련 후의 방추도 및 백도는 OS, RS, MS순으로 높게 나타났다.

Development of a Matrix-prespotted Plate for Enhancing the Reproducibility of Serum Glycan Analysis by MALDI-TOF-MS

  • Matrix Assisted Laser Desorption/Ionization-Time-of-Flight mass spectrometry (MALDI-TOF-MS) is the most widely used MS technique for glycan analysis. However, the poor point-to-point and sample-to-sample reproducibility becomes a limit in glycan biomarker research. A prespotted MALDI plate which overcomes the large crystal formation of 2,5-dihydroxybenzoic acid (DHB) has been developed and applied for glycan analysis. A homogeneous matrix coated surface without a crystal structure was formed on a hydrophilic/ hydrophobic patterned surface using a piezoelectric device. The reproducible MALDI-TOF-MS data have been presented using MALDI imaging of beer glycan as well as serum glycan eluted from 10% and 20% ACN elution fractions. The glycan profile from the serum glycan by MALDI-TOF-MS with a DHB prespotted plate was highly conserved for 10 different spectra and the coefficient of variations of significant ion peaks of MALDI data varies from 3.59 to 19.95.

차량 통신 기술을 위한 WME 모듈과 MLME 모듈 간의 상호 메시지 처리과정 설계 및 구현 (The Design and Implementation of the Mutual Message Processing between WME Module and MLME Module for Vehicle Communication Technology)

  • WAVE(Wireless Access for Vehicular Environment) System is a communication technology to monitor system failure and vehicle functions and management services to prevent possible incidents of driving a vehicle. In this paper, we have designed and implemented the mutual message processing through parameter between WME management module that manages WAVE system and MLME that manages the upper layer MAC(Media Access Control) module. Also, in order to verify the validity, we have carried out experiments to compare the speed of data processing by dividing data of 1Mbyte, 2Mbyte, 3Mbyte into the packets of 2KByte and 4KByte. Experiments data processing speed of 2KByte packet were shown about 173.62ms in 1MByte, 2MByte about 352.61ms, 3MByte about 550.13ms and, data processing speed of 4KByte packet, 1MByte approximately 87.56ms, 2MByte about 177.94ms, 3MByte about 277.18ms. Therefore, in WAVE system, the mutual messages processing through the parameters between WME and MLME module can be utilized in the various service of ITS(Intelligent Transportation Systems) depending on the speed of data processing.

Simultaneous Determination of Anthraquinone, Flavonoids, and Phenolic Antidiabetic Compounds from Cassia auriculata Seeds by Validated UHPLC Based MS/MS Method

  • A systematic isolation and characterization study for Cassia auriculata (CA) seeds resulted in identifying antidiabetic compounds 1,3,8-trihydroxyanthraquinone and quercetin, quercetin-3-O-rutinoside, gallic acid, caffeic acid, ferulic acid, and ellagic acid. The ultra-high-performance liquid chromatography based triple quadrupole mass spectrometry methodology was developed and validated for simultaneous identification and confirmation of these compounds from CA seeds. Multiple reaction monitoring (MRM) based quantification method was developed with MRM optimizer software for MS1 and MS2 mass analysis. The method was optimized on precursor ions and product ions with the ion ratio of each compound. The calibration curves of seven bioactive analytes showed excellent linearity (r2 ≥ 0.99). The quantitation results found precise (RSD, < 10 %) with good recoveries (84.58 to 101.42%). The matrix effect and extraction recoveries were found within the range (91.66 to 102.11%) for the CA seeds. This is the first MS/MS-based methodology applied to quantifying seven antidiabetic compounds in CA seeds and its extract for quality control purposes.

세종지역 유통 식육의 플루오로퀴놀론계 항생제 잔류 연구 (Monitoring for fluoroquinolones residues in raw meat in Sejong)

  • This study describes an analytical method based on LC-MS/MS for the quantitation of 5 fluoroquinolones (Enrofloxacin, Ciprofloxacin, Marbofloxacin, Norfloxacin and Danofloxacin) in meat, and was applied to 230 meat samples for validation. Quantitation was performed based on a matrix-matched calibration to compensate for the matrix effect on the electrospray ionization. Good linear calibrations (R2≥0.998) were obtained for all fluoroquinolones at 6 concentrations of 1~50 ㎍/kg. Satisfied recoveries of all fluoroquinolones were demonstrated in spiked meat at three levels from 10 to 50 ㎍/kg. The recoveries ranged between 75.8~99.2% in beef, 80.1~99.6% in pork and 72.2~99.8% in chicken, respectively. The limits of quantitation (LOQs) for fluoroquinolones ranged from 0.7 to 3.2 ㎍/kg. We also monitored fluoroquinolones residue in the sample (beef 107, pork 71, chicken 52) using LC-MS/MS. Residues of fluoroquinolones which exceeded maximum residue limits (MRL) were not exceed in any of the 230 samples.

MS-5, a Naphthalene Derivative, Induces Apoptosis in Human Pancreatic Cancer BxPC-3 Cells by Modulating Reactive Oxygen Species

  • Pancreatic cancer is one of the most fatal cancers with a poor prognosis. Standard chemotherapies have proven largely ineffective because of their toxicity and the development of resistance. Therefore, there is an urgent need to develop novel therapies. In this study, we investigated the antitumor activity of MS-5, a naphthalene derivative, on BxPC-3, a human pancreatic cancer cell line. We observed that MS-5 was cytotoxic to BxPC-3 cells, as well as inhibited the growth of cells in a concentration- and time- dependent manner. Flow cytometry analysis revealed that the percentage of annexin V-positive cells increased after MS-5 treatment. We also observed cleavage of caspases and poly (ADP-ribose) polymerase, and downregulation of Bcl-xL protein. Flow cytometry analysis of intracellular levels of reactive oxygen species (ROS) and mitochondrial superoxide suggested that MS-5 induced the generation of mitochondrial superoxide while lowering the overall intracellular ROS levels. Thus, MS-5 may be potential candidate for pancreatic cancer treatment.

역행 차폐를 통해 본 시각작업기억의 공고화 및 비교처리 과정 (The Consolidation and Comparison Processes in Visual Working Memory Tested under Pattern-Backward Masking)

  • 변화탐지 과제를 사용한 최근의 시각작업기억 연구는 기억된 표상을 감각적 표상과 대조하는 비교처리 과정이 상대적으로 신속하게 수행될 가능성을 보고하였다[1]. 이러한 가설을 검증하기 위해, 본 연구에서는 기억항목에 대한 공고한 표상 형성이 요구되는 시점 또는 기억항목과 검사항목에 대한 비교 처리가 요구되는 시점에 역행패턴차폐 자극을 제시하여 두 처리 과정에서의 차폐 간섭 효과의 발현 유무를 비교하였다. 실험 1에서는 네 개의 기억항목 또는 검사항목의 제시에 뒤이어 64ms 또는 150ms의 차폐출현간격을 두고 차폐자극이 제시되었으며 피험자는 기억과 검사항목 간 차이 유무를 보고하는 변화탐지 과제를 수행하였다. 실험 결과, 기억항목에 뒤이어 차폐가 제시된 경우(기억차폐 시행)에는 차폐출현간격에 관계없이 변화탐지 정확도가 저조했으나 검사항목에 뒤이어 차폐가 제시된 경우(검사차폐시행)에는 차폐출현간격 64ms 조건에 비해 150ms 조건에서 변화탐지 정확도가 상대적으로 높았다. 실험 2에서는 항목의 개수를 변화시키고(1, 2, 3, 4개) 차폐출현간격을 세분화(117ms, 234ms, 350ms, 584ms)시켜 항목 개수의 증가와 차폐출현간격의 감소에 따른 간섭 효과의 증감패턴을 조사하였다. 기억차폐시행에서는 항목의 개수가 늘어나고 차폐출현간격이 짧아 질수록 점차 증가하는 간섭패턴이 관찰되었으나, 검사차폐시행에서는 이러한 패턴이 상대적으로 미미하였다. 이러한 결과는 시각작업기억의 비교처리과정이 공고화 과정에 비해 상대적으로 신속하고 정확하게 수행된다는 기존 연구의 제안을 지지한다.

LC-MS/MS를 이용한 비글견 혈장 중 파록세틴의 고감도 분석 (Sensitive determination of paroxetine in canine plasma by liquid chromatography-tandem mass spectrometry (LC-MS/MS))

  • 본 연구에서는 비글견 혈장 중의 파록세틴을 액체상추출법(LLE)으로 전처리하고 액체크로마토그래피-탠덤질량분석기(LC-MS/MS)로 신속하게 분석하는 방법을 개발하였다. 파록세틴과 내부표준물질로 사용한 플루오세틴을 TBME(tert-butyl methyl ether)로 추출하고 상층액을 취하여 건조시킨 후, 이동상 $100{\mu}L$로 재분산하여 LC-MS/MS에 주입하였다. HPLC 분석조건으로 Capcell Pak UG120($2.0{\times}150mm$, $5{\mu}m$) 컬럼을 사용하였으며, 이동상은 50% 아세토니트릴(pH 3, formic acid로 조정) 용액을 사용하였고, 유량은 0.2 mL/min으로 하였다. MS/MS의 SRM(selective reaction monitoring) 방법으로 파록세틴과 플루오세틴의 선구 이온, 생성 이온을 각각 m/z $330{\rightarrow}192$, m/z $310{\rightarrow}148$로 분석한 결과 0.02~5 ng/mL의 농도범위에서 상관계수($R^2$) 0.9993으로 좋은 직선성을 나타내었다. 또한 정량한계는 0.02 ng/mL이며, 정밀성은 일내 및 일간 변동계수가 7.67% 이하이고, 정확도는 92.96~102.99%로 비글견 혈장 중의 파록세틴의 약물동력학 연구에 이용될 수 있는 충분한 감도와 특이성, 직선성, 정밀성 및 정확성을 갖고 있음을 확인하였다.

MS-2 system을 이용한 황색포도구균에 대한 moxalactam과 fosfomycin의 병용효과 측정 (Efficiency of Combined Action of Moxalactam and Fosfomycin Determined by MS-2 System Against Penicillin-resistant Staphylococcus aureus Isolates)

  • Twenty strains of penicillin(PC)-resistant Staphylococcus aureus ($MIC{\geqq}32U/ml$) were chosen randomly from recent isolates and submitted to the present experiment to see what effect the combined antibiotic action of moxalactam(MX) and fosfomycin(FM) would bring about on the cells, using MS-2 system. 1. The conventional agar dilution method and the rapid automatic MS-2 system were used in measuring the MICs of MX and FM against each strain and the comparison of the data obtained revealed no significant difference between the two methods in the titer and distribution of the MICs. 2. The automatic MS-2 system was, therefore, used alone in determining the combined growth inhibitory effect of MX and FM because of its more rapidness, and the obtained results were that most of the PC-resistant strains(16 out of 20, 80%) were synergistically inhibited by the two antibiotic combination while additive effect was observed in the remaining 4 strains(20%). 3. Thus, it is suggested that the growth of PC-resistant staphylococcal cells may be synergistically inhibited by MX and FM combination and the efficiency of two antibiotic action as well as MIC of single antibiotic may be more rapidly determined by the MS-2 system than by the conventional method.
