Polymorphisms of tumor necrosis factor-alpha promotor gene in Kawasaki disease and relation to the risk of coronary artery lesion (가와사키병에서의 TNF-alpha 유전자의 다형성 및 관상동맥 합병증과의 연관성)

  • Purpose : The purpose of this study was to investigate the polymorphisms of the TNF-alpha promotor gene, its susceptibility to Kawasaki disease (KD) and to assess whether the TNF-alpha promotor gene polymorphism was related the risk of coronary artery lesions (CALs). Methods : From January 2003 to January 2007, 51 children (30 boys and 21 girls) with KD and 48 children forming an age-matched control group were studied. DNA from the peripheral blood of all the children was sampled, and the DNA polymorphisms of the 5' flanking regions of the TNF-alpha promoter gene at position -308 [guanine (G) to adenine (A)] were determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Then, the relationship between KD and the TNF-alpha promotor gene polymorphisms was evaluated. Results : The A allele frequency of the -308 site of the TNF-alpha promotor gene was 17.6% (9/51) for children with KD and 6.8% (3/48) for the control group children, but this result was not statistically significant. Twenty-four patients experienced CALs within 60 days after the onset of symptoms. KD children with TNF-alpha -308 A allele had lower frequencies of CALs (12.5% versus 22.2%, P>0.05). Conclusion : The DNA polymorphism of the -308 site TNF-alpha gene was not associated with susceptibility to KD and a risk of CALs. Multicenter, large-scale randomized controlled trials are needed for further study.

Ethanol Tolerance of Campylobacter jejuni by Ethanol Shock (Ethanol 충격에 의한 Campylobacter jejuni 의 Ethanol 내성)

  • The responses of C. jejuni to ethanol shock were studied for their survival. synthesis of ethanol shock proteins, and increased survival at higher concentration of ethanol upon prior treatments of ethanol. When C. jejuni were shocked with ethanol at 1. 3. and 5% for 60. 30 and 10 minutes, respectively. those cells synthesized the ethanol shock proteins of 90, 66, 60, 45, and 24 kd in molecular weight. When the C. ,jejuni shocked with 1 and 3% ethanol were exposed to 3 and 5% ethanol for 30 minutes. their survival rates were increased by $10^1$~$10^2$ as compared with those of the cells without ethanol-shock. In the same way. C. ,jejuni shocked with 5% ethanol for 10 minutes :.bowed about 102 times higher survival rates than the cells without ethanol-shock. This result suggests that C jejuni shocked with I-5% ethanol for 10-30 minutes synthesized five kinds of ethanol shock proteins. and that the shock proteins contributed to increase ethanol tolerance for their survival at the higher concentrations of ethanol.

Antifungal activity of a chitinase purified from bean leaves (강낭콩 잎에서 정제한 키틴분해효소의 항균활성)

  • In order to elucidate the plant-microorganism relationship, we purified an ethylene-inducible, basic 30 KD endochitinase from bean leaves and studied its antifungal activity by a hyphal extension-inhibition assay. The purified chitinase was effective in the inhibition of hyphal growth of Aspergillus fumigatus, Botrytis cinerea, Fusarium oxysporum, Rhizoctonia solani, while microbial chitinases of Serratia marcescens and Streptomyces griceus, egg white lysozyme and papya protease didn't affect hyphal growth of the fungi. The chitinase degraded the cell walls of Micrococcus lysodeikticus, suggesting the lysozyme activity of the chitinase. We discussed the implication of the bifunctional chitinase/lysozyme activities of the protein with hydrolysis of chitin in the rapidly extending hyphae of the fungi.

  • Bombyx mori nuclear polyhedrosis virus (BmNPV) was successfully multiplied in the nuclear of BmN4 cells cultured with insect Grace's medium. By electron microscopic observation, the virons had a single nucleocapsid in an envelope. Polyhedral protein synthesis of BmNPV in BmN4 cells was detected at 18 hr p.i. and polyhedral protein was a singlepolypeptide with a M.W of 30 kd. At 48 hr p.i. polyhedra formation was observed by inverted mociroscope and electron microscope. Genome analysis of BmNPV by restriction endonucleases was not revealed the difference between virus produced in vivo and that in vitro.

  • The surface patterns and the structures of transverse section of the egg-shell of the sikmoth, Bombyx mandarina, have been described by scanning electron microscope. Three spatially differentiated cross section, called lamellar, conic pillar and cover layers, are found on the mature eg-shell. Silkmoth chorion proteins were detected more than 80 components from a single chorion by two-dimensional electrophoresis. Major protein components of the egg-shell have bee identified on the basis of their isoelectric points and molecular weights, pH 4-6 and 6-30 kd. Several protein components are found entirely or predominantly in th cover layers.

  • Purple non-sulfur bacteria, Rhodobacter sphaeroides KD131 grew to reach the maximum cell concentration in 45 hrs of incubation in the synthetic media containing (NH4)2SO4, L-aspartic acid and succinic acid as the carbon and nitrogen sources, respectively, at 30oC under 8 klux irradiance using halogen lamp. The strain produced hydrogen from the middle of the logarithmic growth phase and continued until the cell growth leveled out. The strain grew and produced hydrogen under the irradiance of 3-30 klux, but cell growth was inhibited over 100 klux. In addition, anaerobic/light culture condition was better than the aerobic/dark on the hydrogen production. Among various photo-bioreactors examined, the flat-vertical reactor manufactured using clear acrylic plastic material showed the best hydrogen production rate at the given culture condition.

  • This experiment was conducted to obtain basic information of biochemical changes in the process of senescence by measuring the total RNA, protein, protease activity and electrophoretic pattern of protein in tobacco plant. The content of soluble protein increased by 15 days after leaf emergence and its level was not changed from 15 to 35 days after leaf emergence. The content of total RNA showed a maximum value at 15 days after leaf emergence and then decreased rapidly until 30 days after leaf emergence. The activity of protease of neutral fraction was higher than that of acidic fraction and rapidly increased up to the end of senescence after 50 days after leaf emergence. According to the analysis of electrophoresis, polypeptide band of 61kd was developed after 35 days after leaf emergence and increased by the end of senescence.

A Study on the Mechanism of Analgesic Action of Piperine (Piperine의 진통작용 기전에 관한 연구)

  • It was carried out to detect the analgesic action of piperine by hot-plate method and to elucidate its mechanism in rats. Piperine (30mg/kg i.p.) produced profound analgesia, which was blocked by naloxone (10mg/kg). Chronic intraperitoneal administration of piperine significantly increased the contents of $\beta$-endorphin in rat midbrain. In the chronic piperine-treated groups, significant decreases of maximum opiate binding were observed. However, Kd value in these groups were not changed.

Association between Kawasaki disease and acute respiratory viral infections (가와사끼병과 급성 호흡기 바이러스 감염증의 연관성에 관한 연구)

  • Purpose:The etiology of Kawasaki disease (KD) is still unknown. Recently, an association between human coronavirus NL63 (HCoV-NL63) and KD was implicated. Hence, we attempted to determine the association between KD and acute respiratory viral infections. Methods:Nasopharyngeal aspirate samples were obtained from 54 patients diagnosed with KD at the Seoul National University (SNU) Children's Hospital and SNU-Bundang Hospital between October 2003 and September 2006. Viral diagnoses of 11 respiratory viruses were made using multiplex reverse transcriptase-polymerase chain reaction (RT-PCR): respiratory syncytial virus (RSV), adenovirus, rhinovirus (RV), parainfluenza viruses (PIVs) 1 and 3, influenza viruses (IFVs) A and B, human metapneumovirus (HMPV), human bocavirus (HBoV), HCoV OC43/229E, and HCoV-NL63. Clinical data were reviewed retrospectively. Results:The median age was 32 months (6 months-10.4 years). Respiratory symptoms were observed in 37 patients (69%). The following respiratory viruses were identified in 12 patients (22%): RV (n=4), PIV-3 (n=2), HBoV (n=2), and adenovirus, RSV, PIV-1, IFV-A, and HCoV-NL63 (n=1). Co-infection with PIV-3 and RV was observed in one patient. Respiratory symptoms were observed in 7 (58.3%) and 30 (71.4%) patients of the virus-positive and virus-negative groups (P>0.05). Response rate to intravenous immunoglobulin administration was 67% (n=8) and 86% (n=36) in the virus- positive and virus-negative groups (P>0.05). Conclusion:Respiratory symptoms were commonly observed in KD patients but the association between respiratory viruses and KD were not found. Large multicenter-based investigations are required to confirm the association between acute respiratory viral infections and KD.

Usefulness of echocardiographic findings in the early diagnosis of Kawasaki disease (가와사끼병의 조기 진단에서 심장초음파검사의 유용성)

  • Purpose : The early diagnosis of Kawasaki disease (KD) is difficult sometimes, especially in atypical or incomplete cases presenting 4 or less principal clinical features without coronary arterial lesions (coronary arterial ectasia or aneurysm). The authors investigated the incidence of echocardiographic abnormalities in patients with KD to discover whether abnormal echocardiographic findings might be helpful in the early diagnosis of KD. Methods : Echocardiography was done in the acute stage of 103 patients with KD (Kawasaki group) and 40 patients with other acute febrile illnesses (control group). Abnormal echocardiographic findings were classified into 4 categories and defined as follows; 1) significant pericardial effusion, 2) significant valvular dysfunctions, 3) left ventricular systolic dysfunction, 4) coronary arterial ectasia or aneurysm. Results : In the Kawasaki group, significant pericardial effusion was present in 24 patients (23.3 percent), significant valvular dysfunctions in 30 patients (29.1 percent), left ventricular systolic dysfunction in 10 patients (9.7 percent), and coronary arterial lesions in 27 patients (26.2 percent). In the control group, significant pericardial effusion was present in only one patient (2.5 percent). The number of patients with any one of echocardiographic abnormalities was 57 (55.3 percent) in Kawasaki group and one (2.5 percent) in control group. The sensitivity of echocardiography in the diagnosis of KD was 55.3 percent, the specificity 97.5 percent, the positive predictive value 98.3 percent, and the negative predictive value 45.9 percent. Conclusion : If abnormal echocardiographic findings, even if other than coronary arterial lesions, are confirmed in patients in whom KD is suspected, it seems desirable to inifiate specific treatment for KD.
