Studies on the Physiological Chemistry of Flower Organ and Seed in Ginseng Plant. IV. Variation of Free Amino Acids in the Flower and Seeds of the $F_1$ Plants of the Combinations Panax ginseng ${\times}$ Panax quinquefolium and Panax ginseng ${\times}$ Panax japonicus.   • The sterile phenomenon is frequently found in the inter-species hybrids of ginseng as in other plants. It is known that among the hybrids between Panax Ginseng (PG) and Panax Quinquefolium (PQ), and between Panax Ginseng and Paxax Japonicus (PI), PG${\times}$PI is fertile only very rarely, while PG ${\times}$ PQ is always sterile. Therefore, in order to clarify the relationship between this sterility phenomenon and the metabolism of free amino acids, the changes of free amino acids through the formation of the flower organs and seeds of two hybrids, PG ${\times}$ PQ and PG ${\times}$ PI were investigated by thin layer chromatography. The results are summarized as follows: 1. Distinct differences in the quantity and number of free amino acids were recognized between PG ${\times}$ PQ, PG ${\times}$ PI and their parent plants. From the hybrid PG ${\times}$ PQ, 19 kinds of ninhyrin sensitive substances were detected in all. They were (1) 17 amino acids: alanine, valine, leucine, phenylalanine, proline, hydroxy-proline, serine, threonine, tyrosine, aspartic acid, glutamic acid, lysine, arginine, ${\gamma}$-amino butyric acid, ${\beta}$-alanine, cysteic acid and tryptophan, and (2) two amides: asparagine and glutamine. From the hybrid PG ${\times}$ PI, in addition to the above 19 substances, methionine and one unknown substance were detected. 2. Generally, alanine, as partie acid, glutamic acid, cysteic acid and asparagine were detected in large amounts in the two hybrids as in PG, PG and PJ but it was a noticeable fact concerning these two hybrids that the largest quantity of asparagine was found at microspore satge and pollen mature stage. 3. The decrease of cysteic acid in the two hybrids at the red ripened stage was the same as in PQ and PJ but opposite to the change in PG. The detection of methionine in PG ${\times}$ PJ was worthy of notice. 4. The change of proline was conspicuously different from that in their parent plants. It was detected as a trace of color at the micros pore stage while asparagine was detected in the greatest amount at that time. It is well known that the quantity of proline is closely related to the sterility of plant. This fact was also found true in the formation of ginseng seeds. It was reported as well that asparagine accumulated when proline decreased. 5. The deficiency of proline seemed to be closely related with the sterility of hybrids and with the degradation of pollen in anther. 6. The difference in the changes of free amino acids between the selfed lines of PG, PQ and PJ, and their hybrids seemed to be caused by the transformation of gene-action system by hybridization. On these phenomena along with proline metabolim and its physiological role in seed formation further studies are required.

The Test of Combining Ability and Heterosis on the Silkworm(Bombyx mori) Breeding (누에 견.사질에 관한 잡종강세 및 조합능력검정)

  • The study was conducted to obtain the genetic information on heterosis and combining ability of the quantitative characters for F1 hybrid breeding in silkworms. Six parental varieties and each set of 30 diallel crosses in F1's were used as materials, and bred on the randomized complete block design with three replications. Fourteen characters were observed with the twenty samples in each tray. The data were analyzed for (1) heterosis and combining ability in F1 hybrid. The heterosis in the weight and the length of cocoon showed positively high at 24.51%, and 23.4%, respectively and the weight of the whole cocoon as well as the weight of the whole cocoon layer showed a siginificant heterosis ranging from 15.56% to 15.71% and from 17.14% to 19.01%, but the fifth and the total instar period showed negative heterosis. It was found that the combination between, C70XRomogua and N9 X Romogua showed highly a negative heterosis on the fifth instar period and for the cocoon weight. The female of N9+Sansuian and the male of Romogua X Sansurian have a high heterosis effect, on the cocoon shell weight, and Sansurian X Romogua(reciprocal) on the length and the weight of cocoon filament with no regard to sexuality. The significant maternal and cytoplasmic effect on heterosis of the cocoon weight and the cocoon shell weight were observed with the combinations, N9 X C5, N63 X C70 and on the length of the cocoon filament with the combinations, Sansurian X N63, Sansurian X C5, Sansurian X C70 and N9 X C70, N63 X C70 on the weight of cocoon filament. As mean squared of GCA, SCA and RCA were significant with these combining ability for all characters resulted from additive and non-additive altogether and there is a significant difference between reciprocals. Sansurian showed a negative GCA effect on the fifth and total larval duration, but the higher positive GCA effects took places with varieties N9 and C5 on the length, width, weight of cocoon, cocoon shell weight, percentage of cocoon shell weight, length and weight of cocoon filament, percentage of raw-silk with no regard to both generations and silkworm sexuality. The values of SCA between the cross combinations varied generation-wise and sex-wise. It was shown that SCA value for the fifth instar period was highly negative for Sansurian X C70, Romogua X C70, Sansurian X C5, Romogua X C5, but it was positive effect on the cocoon weight, cocoon shell weight with N9 X C5, and C70 X Sansurian, on the length of cocoon filament with N9 X C5, Romogua X Sansurian on the weight of cocoon filament between Romogua and N63 and on the percentage of raw-silk between the combination of Sansurian X Romoga.

Application of Oceanic Camp Program for the Enhancement of Inquisitiveness and Affection to Ocean: from 2004 to 2012 (해양에 대한 호기심과 친근감 향상을 위한 해양캠프 프로그램의 적용: 2004~2012년)

  • In order to enhance scientific interest and a sense of affinity about ocean, the programs of the oceanic camp 'oceanic summer school' were developed and applied to $4^{th}$ and $9^{th}$-grade elementary and middle school students for 9 years from 2004 to 2012. It was composed of oceanic training for snorkeling, a tour to oceanic institutes and museums near the camp academy place, experimental learning in oceanic-related field, field trips for ocean and earth sciences, and lectures on various subjects of ocean. We developed and implemented 9-kinds of inquiry surveys to evaluate changes in cognitive and affective characteristics, and ocean literacy of students participated at the present oceanic summer camp. Based on the statistical analysis, affective characteristics such as interest, inquisitiveness, passion, and so on, were enhanced. Analysis of ocean literacy revealed that cognitive characteristics of the students were increased by 40%. We presented parents' responses on the programs of oceanic summer school. Some students with less initial interest of ocean have positively changed to make up their minds to be a oceanographer in several years later. In light of this, the summer school can be evaluated to be successfully functioned as a long-term support system for potentially young-talented students in the field of ocean science. This study addresses that long-term implementation of the summer oceanic camp may trigger students with potential talent toward in-depth science in the near future even though it could not bring positive effect immediately. This addresses the necessity of policy supports in order that various programs like the scientific camp should be more constructively developed and executed for next-generation manpower in oceanographic fields.

To support policy-making and evaluation on optimal farm production, disaster management, effective agro-environmental resource management etc., providing tools such as GIS. 4. To improve farm management and farming technologies by efficient farm management, risk management, effective information or knowledge transfer etc., realizing competitive and sustainable farming with safe products. 5. To provide systems and tools to secure food traceability and reliability that has been an emerging issue concerning farm products since serious contamination such as BSE and chicken flu was detected. 6. To take an important and key role for industrialization of farming or lam business enterprise, combining the above roles.

Modulation of Immune Parameters by Aging Process (노화에 따른 면역지표의 변화에 관한 연구)

  • The purpose of this study was to investigate the effects of aging process on the immunity in human subjects. In this investigation, nineteen families of three generations (daughters on college age, their mothers, and grandmothers) participated to avoid genetic variation among individuals. Dietary food records, anthropometric measurements and biochemical assessments of serum nutrients were used to evaluate the nutritional status of subjects. The immune parameters of subjects were assessed by the total and differential WBC count. Total B and T lymphocytes, and T cell subsets were quantified by flowcytometer. Serum immunoglobulin G, A, M concentrations were also measured as an index of humoral immunity. The result of this study can be summarized as follows: 1. Along with the aging process, body fat was found to be increased whereas lean body mass and total body water were diminished. Since there were no significant difference in serum vitamin E levels in all age groups, serum retinal concentrations tended to decrease as one gets old. 2. Although total number of T lymphocytes seemed to be unchanged, B lymphocytes and NK cell numbers were increased by aging. The Percentage of CD8 + lymphocytes was lower in the elderly subjects compared with the younger, resulting in higher ratio of CD4 +/CD8 + lymphocytes in the elderly. Serum Ig G and Ig A levels remained unchanged, but IgM levels were significantly decreased as the age processes continue. Taking all together, it could be suggested that the alteration of immune cell population by aging is selective and possibly nonage factors such as nutrition may be attributable to the change of immunity in the elderly. The nutritional status and aging process may selectively affect both the cell-mediated (CD8 +, CD4 + CD8 + ratio, NK cell) and humoral (B lymphocyte, Immunoglobulin M, G) immune parameters in human subjects.

A Pursuit of Innovation in the Korean Genetics-Genomics Research System through a Culturalist Strategy (문화적 전략을 통한 한국 유전학-유전체학 연구체계의 혁신 모색)

  • The Korean genetics and genomics reveal a firm willingness to participate in and contribute to the production of creative scientific knowledge at a world level at present, though they have short past histories of introduction from the Western counterparts and those of education for the next generations. But the Korean genetics and genomics have been developed in a fragmented and biased manner. By reconfiguring the various research projects of genomics into the Genome Project of Korea, which reflect a worldly trend in life science, but have been established in a scattered fashion in Korea, and incorporating some neglected areas of genetics, such as human genetics and theoretical and population genetics which can be reconstructed in a new way, a genetics-genomics research system can be formulated on the multi-tiered perspective of concept, knowledge, and institution, while the system being a subsystem of the national research system of life science in Korea. Innovation can be pursued in the systematic practice through a culturalist strategy. The culturalist strategy with the practice based on the research system consists of 1) intensification of fundamentalness of genetics and genomics, 2) advancement of communitarianism in geneticist-genomicist community, 3) research on the cultural bio-species along with the promotion of scientific arts and culture, and 4)formation of the Korean science studies of genetics-genomics and the diffusion of the knowledge produced. The first two strategy components are the ones that intends to bring out changes in the structural aspect of the scientist community in Korea. The third is the one that attempts to magnify the interface between the scientist community and the Korean society at large and increase its connectivity between both, while the fourth is the one that has an intentionality toward the Korean society outside of the scientist community. This culturalist strategy is intended to increase the cultural constructivity of the genetics-genomics research system in Korea.

Effect of Temperature on Development and Life Table Parameters of Tetranychus urticae Koch (Acari: Tetranychide) Reared on Eggplants (가지에서 온도별 점박이응애 발육특성 및 생명표 통계량)

  • Temperature dependent development of Tetranychus. urticae Koch was studied on the leaf of eggplant at 17, 22, 27, 32 and $37^{\circ}C$. T. urticae showed a minimum mortality at $27^{\circ}C$ and it increased at higher or lower temperatures than $27^{\circ}C$. The hatchability was low at 17 and $37^{\circ}C$. The duration of development decreased with increasing temperatures i.e., 5.3d at $37^{\circ}C$ and 25.8d at $17^{\circ}C$. Linear regression analysis of temperature vs. rate of development yielded the higher $r^2{\geq}0.88$ resulting in a good fit of the estimated line in the range of $17{\sim}37^{\circ}C$. Developmental zero temperature was $12.5^{\circ}C$ for the entire immature stage of female and $12.8^{\circ}C$ for that of male. Thermal constants were 80.5 and 74.7 degree days for those of female and male, respectively. Adult life span and oviposition period decreased with increasing temperatures. The number of eggs laid per female peaked at 141.0 eggs at $27^{\circ}C$, while that was a minimum 78.0 eggs at $37^{\circ}C$. Rate of hatchability, ratio of female, and $R_o$ were increased up to $27^{\circ}C$, and than declined thereafter. Intrinsic rate of natural increase (Rm) increased with rising temperatures and showed a maximum 0.5652 at $37^{\circ}C$. Also, ${\lambda}$ increased with increasing temperature. Doubling time (Dt) and generation time (T) decreased with increasing temperature.

Studies on the Mechanism of Varietal Resistance of Rice to the Brown Planthopper(Nilaparvata lugens $ST{\AA}L$ (벼멸구에 대한 수도품종(水稻品種)의 저항성기작(抵抗性機作)에 관(關)한 연구(硏究))

  • This paper was performed to study the nature of varietal resistance of some Korean-new rice cultivars to the brown planthopper (BPH), Nilaparvata lugens ($ST{\AA}L$). The rice cultivars tested were Cheongcheong, Gaya, Hangangchal, Samgang, Nampoong and Yeongpoong which have been reported as having resistant genes for the BPH. The check varieties were Jinheung, Sangpoong and Chucheong for susceptible and IR-36 for resistant. The factors studied were referred to the seedling responses, preference in feeding and oviposition of BPH, antibiosis (nymphal development, adult emergence and sex ratio, adult body weight, population build-up, feeding amount and amylase activity), and chemical composition (inorganic components, chlorophyll contents, cell wall components, amino acids and esterase isozymes) of leaf- sheath and/or roots of rice plants. In conclusion, the natures of varietal resistance of rice cultivars to the BPH were not only correlated with the resistant reaction of rice plant, but also they were related with the non preference in feeding and oviposition and those resistant cultivars had the antibiotic effects to the insects. Their antibiotic effects of rice cultivars to the BPH would be related with some of the chemical components of rice plants, such as the contents of magnesium oxide (MgO), and chlorophyll and the different esterase isozymes.

Health Behavior and Health Condition of the Rural Young-Old and the Rural Old-Old in an Agricultural District (농촌 전기노인과 후기노인의 건강행태와 건강상태)

  • Objectives: The purpose of this study is to garner useful information through a comparative analysis of health behaviors and health states between the young-old and old-old elderly in a rural Korean area. Methods: We define the young-old elderly as those 65 to 74 years of age, and the old-old as those over 70. The survey was administered in October and November of 2009 at senior citizen centers in Sangju City, Kyongsangbuk-do, South Korea. The number of subjects surveyed approximated the demographics of the aged population of the administrative district of centers of 24 eup, myeon, and dong. Results: Compared with the young-old elderly, the old-old were vulnerable to population sociological characteristics. While there were many cases of contraction of diseases, only a small percentage of old-old elderly were engaged in regular exercise. In addition, the old-old elderly lagged behind the young-old in terms of physical activity, mental and oral health, hearing, and vision. Conclusions: The vulnerability of the old-old elderly in terms of physical and mental health needs to be acknowledged as various characteristics of the elderly that appears according an age group. A variety of disease prevention and health promotion programs that focus on the health behavior and status of the young-old and old-old elderly need to be developed and put into practice.

Development and Application of Training Program for RI-Biomics Manpower through Analysis of Educational Demands (교육수요 분석을 통한 RI-Biomics 전문인력 양성 프로그램 개발 및 적용)

  • RI-Biomics is a promising radiation convergence technology that combines radiation with bio science as new growth power technology. Many developed countries are focusing active support and constant exertion to dominate the RI-Biomics market in advance. In order to achieve global leadership in the RI-Biomics field, we need more highly advanced technologies and professional manpower. In fact, we have less manpower compared to technology we currently hold. In this study, we established a basic infrastructure to train professional manpower in the RI-Biomics field by developing/operating optimum training program through expert interviews and survey. The developed program has four organized sections to understand overall procedure of RI-Biomics. To evaluate our training program, we performed test operations with eight students who have a major related to RI-Biomics for three weeks in KARA (Seoul) and KAERI (Jung-eup). In detail, radioisotope usage and safety management were conducted for one week as basic course, RI-Biomics application technology was conducted for two weeks as professional course. To verify performance results of training program, we conducted to journal research, daily reports, and survey on participants. The results show a high level of satisfaction with training programs and continuous intention of involvement in our program. We also need to develop an intensive course to train high-quality human resources and to operate training program continuously. This training program will be used as basic materials for the development of RI-Biomics curriculum for university. Hence, we will expect that our training program contributes in training a professional manpower and develop RI-Biomics technology.
