Cyclofarnesane sesquiterpene glucoside from the whole plant of Loranthus tanakae and its cytotoxicity ('꼬리겨우살이'(Loranthus tanakae) 전초로부터 cyclofarnesane sesquiterpene glucoside의 분리 동정 및 세포독성)

  • The whole plants of Loranthus tanakae were repeatedly extracted with 80% aqueous MeOH and the concentrate was partitioned into ethyl acetate (EtOAc), n-butyl alcohol (n-BuOH) and $H_2O$ fractions. The repeated silica gel ($SiO_2$), octadecyl $SiO_2$ (ODS), and Sephadex LH-20 column chromatographies for the n-BuOH fraction led to isolation of a cyclofarnesane sesquiterpene glucoside. The chemical structure including stereo structure was determined to be a (1'R,3'S,5'R,8'S,2E,4E)-dihydrophaseic acid $3^{\prime}-O-{\beta}-{\text\tiny{D}}$-glucopyranoside on the basis of spectroscopic analyses. This compound was isolated for the first time from L. tanakae in this study. Compound 1 showed a moderate dose-dependent cytotoxicity against human Caucasian gastric adenocarcinoma cells and human hepatocyte cari-noma cells cell lines at higher than $50{\mu}g/mL$.

  • Six compounds were isolated from the EtOAc and $Et_2O$ fractions of the bark of Rhus chinensis by repeated column chromatography with $SiO_2$ and Sephadex LH-20. The structures were determined by instrumental analysis using MS and NMR spectrophotometer as: gallic acid (1), methyl gallate (2), 6, 7-dimethoxycoumarin (3), orcinol-${\beta}$-D-glucoside (4), scopoletin (5), semialactone (6). Among these compounds, 6,7-dimethoxycoumarin (3) was isolated from this plant for the first time. To measure the antioxidant activity, the DPPH radical scavenging activity test was performed. Gallic acid (1) showed the strongest activity, while orcinol-${\beta}$-D-glucoside (4), semialactone (5) and scopoletin (6) had the low activities.

  • A furanofuranoid lignan glycoside, with radical scavenging on peroxynitrite, total reactive oxygen species (ROS) and 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical, was isolated from the rhizome of Salvia miltiorrhiza and characterized as (+)-1-hydroxypinoresinol-1-Ο-$\beta$-D-glucoside based on spectroscopic evidence. The compound exhibited peroxynitrite, total ROS and DPPH radical scavenging activities with $IC_{50}$ values of 3.23$\pm$0.04, 2.26$\pm$0.07 and 32.3$\pm$0.13 $\mu$M, respectively. Penicillamine, Trolox (6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid) and L-ascorbic acid, acting as positive controls, showed radical scavenging activities with $IC_{50}$ values of 6.72$\pm$0.25, 1.43$\pm$0.04 and 11.4$\pm$0.07 $\mu$M, respectively.

  • n-Alkanes (n-nonacosane and n-hentriacontane), phytosterols (campesterol and ${\beta}$-sitosterol) and phytosteryl glucosides (${\beta}$-sitosterol 3-O-${\beta}$-D-glucopyranoside and campesterol 3-O-${\beta}$-D-glucopyranoside) were isolated from the underground parts of Epimedium koreanum (Berberidaceae) and characterized by spectral data.

  • Daucosterol and naphthalene glycoside were isolated from the seeds of Cassia mimosoides var. nomame Makino. The isolated compounds were identified by spectral means including 1D, 2D-NMR and FAB-MS experiments. Daucosterol was characterized as ${\beta}$-sitosterol-3-O-${\beta}$-D-glucoside and naphthalene glucoside was done as 2-acetyl-3-methyl-6-methoxynaphthalene-1,8-diol 8-O-glucosyl-(1${\rightarrow}$6)-glucoside. These compounds were isolated for the first time from Cassia mimosoides var. nomame Makino.

  • From the polar fractions of 70% EtOH extract of Paeonia lactiflora roots (Paeoniaceae), ten monoterpene glucosides were isolated and identified as lactiflorin (1), benzoylpaeoniflorin (2), mudanpioside C (3), $1-O-{\beta}-D-glucosylpaeonisuffrone$ (4), paeonidanin (5), $1-O-{\beta}-D-glucosyl-8-O-benzoylpaeonisuffrone$ (6), paeoniflorin (7), albiflorin (8), oxypaeoniflorin (9) and mudanpioside E (10) by spectroscopic methods. Among these glucosides, 3-6 and 10 were isolated for the first time from this plant.

  • From the legumes of Albizzia julibrissin Durazzini (Leguminosae), a mixture of long-chain alcohols, ${\alpha}-spinasterol$, a mixture of acylated sterol glycosides and ${\alpha}-spinasteryl$ $3-O-{\beta}-D-glucoside$ were isolated. Two mixtures of long-chain alcohols and acylated sterol glycosides were characterized as 1-hexacosanol (major) and 1-tetracosanol, and $3-O-[6'-O-palmitoyl-{\beta}-D-glucosyl]-{\alpha}-spinasterol$ (major) and its 6'-O-stearoyl ester. All compounds were identified on the basis of spectral data and chemical reactions.

  • Eight compounds were isolated from the BuOH extract of the stem of Clematis trichotoma (Ranunculaceae). On the basis of spectroscopic evidences, the structures of these compounds were established as rutin, kaempferol 3-O-neohesperidoside, adenosine, adenin, hirustrin, caffeic acid $4-{\beta}-glucoside$, 3-methoxyarbutin and uridine.

  • The methanol extract of the rootbark of Fraxinus sieboldiana Blume(Oleaceae) gave two coumarin glycosides, fraxin (fraxetin-8-glucoside), $C_{16}H_{18}O_{10},\;mp\;204^{\circ}\;and\;esculin\;(esculetin-6-glucoside),\;C_{15}H_{16}O_{9},\;mp\;204{\sim}205^{\circ}$. Beside, the methanol extract of the root bark was found to contain mannitol. ${\beta}-sitosterol$ is confirmed by massspectrometry.

  • Two flavonoid glycosides and three secoiridoide glycosides were isolated from the leaves of Lonicera maackii. On the basis of spectral and physico-chemical data, the structures of isolated compounds were identified as hesperetin-7-O-glucoside (1), luteolin-7-O-${\beta}$-D-glucopyranoside (2), secologanin dimethylacetal (3), epi-vogeloside (4), and vogeloside (5), respectively. Hesperetin-7-O-glucoside (1) was isolated for the first time from this plant.
