Extended Central Composite Designs with the Axial Points Indicated by Two Numbers

  • The central composite design is widely used for estimating second order response surfaces. This type of design is composed of $2^k$ factorial points, axial points and center points. In this paper, we suggest a version of central composite design where the positions of the axial points are indicated by two numbers, and study properties of this design. We obtain the variances and covariances of the estimators of the regression coefficients. Conditions are obtained for this design to be orthogonal and rotatable. This design is compared with other designs on the basis of efficiency.

Korean Nurses' Judgments of Child Sexual Abuse Situations: The Use of Vignettes (아동 성폭력 상황에 대한 한국 간호사의 판단)

  • 본 연구의 목적은 아동 성폭력 상황 자체와 성폭력 가해자에 대한 간호사의 태도가 상황 판단에 미치는 영향을 규명하는데 초점을 둔다. 503명의 병원 간호사와 526명의 보건교사를 대상으로 vignette 디자인 설문조사를 실시하였다. 아동성폭력 상황 vignettes은 7가지의 특성 (성폭력 행위, 성폭력 빈도, 피해자의 연령, 피해자의 저항, 가해자의 연령, 가해자와 피해자의 교차성별, 가해자와 피해자와의 관계)을 조합하여 구성하였다. 1/4 factorial design의 결과로서 64개의 vignettes 이 도출되었으며 이 중 16개 vignettes을 무작위로 추출하여 그 vignettes에 나타난 각 성폭력 상황들의 심각정도를 표시하도록 하였다. 성폭력 가해자에 대한 문항들은 1) 가해자가 아동 성폭력을 하는 이유 2) 전형적인 성폭력 가해자3) 아동성폭력 가해자의 처벌방법에 관한 간호사의 태도를 묻는 것이었다. 자료분석은 Logistic Regression을 실시하였다. 연구결과에서 간호사들은 아동 성폭력 상황의 심각도를 판단할 때 주로 성폭력 상황에 의해 영향을 받고 아동 성폭력 가해자에 대한 개인적 태도는 거의 영향을 미치지 않는 것으로 나타났으며, 아동 성폭력 상황 특성 중 피해자의 저항, 성폭력 행위, 성폭력 빈도, 가해자의 연령, 가해자와 피해자의 교차성별 순으로 아동 성폭력상황의 심각도에 영향을 미치는 것으로 나타났다. 결론적으로, 간호사들은 아동 성폭력 상황에 직면하여 사례를 판단할 때 아동 성폭력 문제에 대한 개인의 태도에 영향을 받기보다는 사례 그 자체에 근거를 두고 판단하는 경향을 나타낸다. 본 연구 결과는 법적, 전문가적 의미에서 아동성폭력 신고자로서의 의무와 사례중재에 개입된 의료인으로서의 역할 수행에 긍정적인 면을 시사한다.

Statistical Optimization for Monacolin K and Yellow Pigment Production and Citrinin Reduction by Monascus purpureus in Solid-State Fermentation

  • Monacolin K and yellow pigment, produced by Monascus sp., have each been proven to be beneficial compounds as antihypercholesterolemic and anti-inflammation agents, respectively. However, citrinin, a human toxic substance, was also synthesized in this fungus. In this research, solidstate fermentation of M. purpureus TISTR 3541 was optimized by statistical methodology to obtain a high production of monacolin K and yellow pigment along with a low level of citrinin. Fractional factorial design was applied in this study to identify the significant factors. Among the 13 variables, five parameters (i.e., glycerol, methionine, sodium nitrate, cultivation time, and temperature) influencing monacolin K, yellow pigment, and citrinin production were identified. A central composite design was further employed to investigate the optimum level of these five factors. The maximum production of monacolin K and yellow pigment of 5,900 mg/kg and 1,700 units/g, respectively, and the minimum citrinin concentration of 0.26 mg/kg were achieved in the medium containing 2% glycerol, 0.14% methionine, and 0.01% sodium nitrate at $25^{\circ}C$ for 16 days of cultivation. The yields of monacolin K and yellow pigment were about 3 and 1.5 times higher than the basal medium, respectively, whereas citrinin was dramatically reduced by 36 times.

  • The finite element method (FEM) is typically used in the process of motor design. However, the FEM requires computation time, Therefore, decreasing the number of FEM simulations may also decrease the simulation cost. Several optimal design methods overcoming this problem have been recently studied. This paper investigates the optimal design of the magnet pole of a BLDC motor through reducing simulation cost. The optimization minimizes the magnet volume and limits the average and cogging torques to certain values. In this paper, the response surface methodology and Taguchi's table for reducing the number of FEM simulations are used to approximate two constraints. The optimization result shows that the presented strategy is satisfactorily performed.

  • An oral adhesive tablet of acyclorir [9-(2-hydroxyethoxymethyl) guanine] for herpetic stomatitis was prepared and its physical properties were evaluated. 300 mg weighed tablets containing 30 mg of acyclovir were prepared with six kinds of polymers from direct compression, and the stickiness, fracture resistance and dissolution in pH 6.8 buffer solution were tested. HPMC and MC showed good stickiness and fracture resistance, and their dissolution rates were significantly different from each other. Three factors-HPMC:MC ratio, acyclovir content, compression force-were chosen as an important factor of manufacture and factorial analyses for these three factors were carried out. Eight kinds of formulations from different combination of three factors were prepared and tested in stickiness, fracture resistance and dissolution in pH 6.8 buffer solution. Dissolution rate was significantly affected by polymer ratio, fracture resistance was affected by compression force, and stickiness was not significantly affected by acyclovir content and polymer ratio.

Optimization of Culture Medium for Novel Cell-Associated Tannase Production from Bacillus massiliensis Using Response Surface Methodology

  • Naturally immobilized tannase (tannin acyl hydrolase, E.C. 3.1.1.20) has many advantages, as it avoids the expensive and laborious operation of isolation, purification, and immobilization, plus it is highly stable in adverse pH and temperature. However, in the case of cell-associated enzymes, since the enzyme is associated with the biomass, separation of the pure biomass is necessary. However, tannic acid, a known inducer of tannase, forms insoluble complexes with media proteins, making it difficult to separate pure biomass. Therefore, this study optimizes the production of cell-associated tannase using a "protein-tannin complex" free media. An exploratory study was first conducted in shake-flasks to select the inducer, carbon source, and nitrogen sources. As a result it was found that gallic acid induces tannase synthesis, a tryptose broth gives higher biomass, and lactose supplementation is beneficial. The medium was then optimized using response surface methodology based on the full factorial central composite design in a 3 l bioreactor. A $2^3$ factorial design augmented by 7 axial points (${\alpha}$ = 1.682) and 2 replicates at the center point was implemented in 17 experiments. A mathematical model was also developed to show the effect of each medium component and their interactions on the production of cell-associated tannase. The validity of the proposed model was verified, and the optimized medium was shown to produce maximum cell-associated tannase activity of 9.65 U/l, which is 93.8% higher than the activity in the basal medium, after 12 h at pH 5.0, $30^{\circ}C$. The optimum medium consists of 38 g/l lactose, 50 g/l tryptose, and 2.8 g/l gallic acid.

  • The components of the media used to elicit the biosynthesis of $poly-{\gamma}-glutamic$ acid $({\gamma}-PGA)$ by Bacillus subtilis ZJU-7 were investigated, particularly the carbon and nitrogen sources Of the 7 carbon sources investigated, sucrose induced the highest rate of ${\gamma}-PGA$ productivity; among the nitrogen sources, tryptone had the best effect for ${\gamma}-PGA$ production. A $2^{6-2}$ fractional factorial design was used to screen factors that influence ${\gamma}-PGA$ production significantly, and a central composite design was finally adopted to formulate the optimal medium. ${\gamma}-PGA$ productivity improved approximately 2-fold when the optimal medium was used compared with the original nonoptimized medium, and volumetric productivity reached a maximum of 58.2 g/L after a 24-h cultivation period.

  • A design of experiments was applied in order to investigate the effect of processing variables in UV irradiation on the electrical/optical properties in indium-zinc oxide thin films, The processing variables, equivalently input variables are listed as UV irradiation time, oxygen flow rate, and chamber pressure. The statistical significance of Ultra Violet (UV) treatment was confirmed using a paired-t test. The full factorial $2^3$ design was employed to determine significant main and interaction effects in UV irradiation process. The chamber pressure and the interaction between UV irradiation time and $O_2$ flow rate were found to be statistically significant at the significance level of 0.1. Furthermore, the optimized approach was proposed in achieving the improved conductivity after UV irradiation.

  • The leaching of Cu from waste PCBs was investigated with electro-generated chlorine as an oxidant. The leaching experiments were carried out according to the design of experiments to analyze quantitatively the effect of parameters on copper leaching. From the analysis of variance (ANOVA) it was suggested that the effective parameters were current density, temperature, concentration of HCl, and the interaction between the concentration of HCl and temperature. Especially, the effect of current density was analyzed to contribute to the interpretation of result for copper leaching up to 95.7%. A multiple regression model obtained from the analysis of effective parameters explained 99% of leaching results. From the model equation, it was found that the effect of HCl concentration on copper leaching increased with temperature.

  • In this study, the composition of optical glass for GMP(glass molding process) was designed with 'Design of Experiments' method. All the composition batch was performed by 'Create Factorial Design' method. Particularly, $SiO_2$, BaO and $Al_2O_3$ were chosen major parameters for investigating the effects of components on optical and thermal properties. BaO and $Al_2O_3$ strongly influenced on optical and thermal properties, respectively. Finally, the approximate values of desired optical and thermal values were obtained by microtuning of compositions. At the composition of $BaO:Al_2O_3:SiO_2$=10:4:48 (molar ratio), refractive index($n_d$) was 1.5833, coefficient of thermal expansion(CTE) was $104{\times}10^{-7}/^{\circ}C$.
