Plant Regeneration from Mesophyll Protoplasts Culture of Solanum sisymbriifolium

  • The optimal culture conditions were studied for plant regeneration from mesophyll protoplasts of Solanum sisymbriifolium. Axenic seedlings of S. sisymbriifolium were used as a explant for protoplast culture. Many viable protoplasts were isolated by incubating leaf slices in an enzyme solution containing 0.25% Meicerase and 0.05% Macerozyme for 16 hr at $25^{\circ}C$ without shaking. Protoplast density of $5.0{\times}10^4\;ml^{-1}$ in Kao medium containing 5.0 mg/L NAA, 1.0 mg/L 2,4-D and 1.0 mg/L BA was optimal for colony formation. Most colonies were formed when protoplasts were cultured at $25^{\circ}C$ after initial culture at $30^{\circ}C$ for one week. On the MS agar medium with 1.0 mg/L zeatin, 38.4% of protoplast-derived calli differentiated shoots. These shoots rooted on 1/2MS medium with 5.0 g/L sucrose and 2.5 g/L gellan gum, and developed into whole plants.

Microspore-derived Embryo Formation in Response to Cold Pretreatment, Washing Medium, and Medium Composition of Radish (Raphanus sativus L.)

  • Cold pretreatment, washing medium and composition of nutrient media may have marked effects on microspore embryogenesis. When microspores isolated from radish (Raphanus sativus L. cv. Gwanhun) flower buds were washed with Nitsch & Nitsch (NLN) medium liquid medium containing $130g{\cdot}L^{-1}$ sucrose (NLN-13), yields of microspore-derived embryos were greater than when using B5 liquid medium containing $130g{\cdot}L^{-1}$ sucrose. Microspore viability is known to decrease rapidly with storage; however, in this experiment, microspore viability was maintained for 24 h at $4^{\circ}C$ without media. Among the various medium concentrations used ($0.25{\times}$, $0.5{\times}$, $1.0{\times}$, $2.0{\times}$, and $4.0{\times}$ NLN liquid medium), $0.5{\times}$ NLN liquid medium induced the most efficient formation of microspore-derived embryos. In addition, microspore-derived embryos yields were greater when microspores were cultured in $0.5{\times}$ NLN liquid medium supplemented with $0.25{\times}$, $0.5{\times}$, and $1.0{\times}$ NLN microelements, compared to medium not supplemented with microelements. In this study, the highest yield of microspore-derived embryos was observed when the microspores derived from flower buds were washed using NLN-13 liquid medium and then cultured on $0.5{\times}$ NLN liquid medium supplemented with $0.25{\times}$ NLN microelements, followed by incubation at $25^{\circ}C$ for 30 days.

Sucrose, 칼슘, 붕소 첨가가 복숭아 화분 발아에 미치는 영향 (Effect of Sucrose, Calcium and Boron Added in the Medium on Pollen Germination of Peach(Prunus persica SIEB))

  • 우리나라 과수 중 개화기가 빠른 복숭아는 저온에 의하여 화분발아 장해로 결실율이 저하되고 있다. 본 연구는 저온에서 화분 발아율을 증진시킬 수 있는 방법을 강구하기 위한 기초연구로서 화분 발아조건과 배지내에 sucrose, $CaCl_2$$H_3BO_3$ 첨가농도를 구명하고자 일련의 실험을 실시하였다. 복숭아 화분은 배지에 치상한 후 30분부터 발아가 시작되어 3시간 30분까지 지속되었으나, 그 이후는 더 이상 발아되지 않았다. 따라서 복숭아의 화분발아 판정을 위한 적정 시간은 치상 후 4시간이 좋았다. 복숭아 화분의 적정발아 온도는 $20^{\circ}C$였고, 이보다 저온이거나 고온에서는 화분발아율이 감소하였다. Sucrose는 화분발아를 촉진하였고 최적농도는 10%였다. 무기염류 중 $H_3BO_3$ 첨가는 '장택백봉'과 '수봉' 품종에서 화분 발아율을 무처리에 비해 $20{\sim}30%$ 증진시켰으며, 그 효과는 저온인 $10^{\circ}C$에서 현저하였다. 최적처리 농도는 $100\;mg\;{\cdot}\;L^{-1}$ 처리였다. 반면 $CaCl_2$ 첨가하면 화분발아율이 감소하였다. $10^{\circ}C$의 저온에서 화분발아율은 품종간에 뚜렷한 차이가 있었으며, '장택백봉', '백봉', '월미', '백도', '대구보', '유명', '수봉', '백미조생'의 순으로 화분발아율이 낮았다.

소리쟁이 (Rumex crispus)의 부정근과 모상근 배양에 의한 Anthraquinone 생산성 (Anthraquinone Productivity by the Cultures of Adventitious Roots and Hairy Roots from Curled Dock (Rumex crispus))

  • 형질전환된 소리쟁이 모상근의 기내 배양시 그 생장 특성과 anthraquinone생산성을 조사하기 위하여 소리쟁이의 모상근을 Agrobacterium rhirogenes strain $A_4$ 접종에 의하여 잎 절편으로부터 유기하여 부정근과 비교하였다. 잎 절편에서의 부정근 형성은 5 $\mu$M NAA를 첨가한 MS배지에서 가장 양호하였다. Paper electrophoresis를 통한 opine분석 결과 모상근에서만 mannopine이 검출되고 부정근에서는 검출되지 않았다. 부정근과 모상근 모두에서 2차근인 측근 근단은 호르몬을 첨가하지 않은 배지에서 신장만 할 뿐 분지하지 않았으나, 1차근인 주근의 근단은 많은 측지를 내며 빠르게 생장하였다. Anthraquinone 함량 증대에 따른 부정근과 모상근의 생장에는 MS기본 배지가 가장 양호하였다. 부정근과 모상근 배양시 생장량과 anthraquinone함량은 5% sucrose를 포함한 0.05 $\mu$M NAA와 0.1 $\mu$M kinetin으로 조성된 MS 액체 배지에서 가장 높은 것으로 나타났으며, 모상근의 anthraquinone함량은 배양 일수에 따라 증가하다가 배양 25일 이후에는 감소하였다.

발아중인 대두콩에서의 Lipoxygenase의 국재(局在) (Localization of Lipoxygenase in Germinating Soybeans)

  • 발아중인 대두콩에서의 lipoxygenase의 세포내 위치를 차별 원심 분리법과 밀도 구배법을 이용하여 규명하였다. 차별 원심 분리법을 사용하였을때, lipoxygenase활성은 거의 supernatant fraction에서 나타났으며, 1.5%의 활성만이 particulate fractions에서 나타났다. 밀도 구배법에서는 lipoxygenase와 acid phosphatase의 활성이 1.19, 1.23, $1.25g/cm^3$의 높은 밀도들에서 일치하였으며, 이것은 발아중 가수분해되는 단백질체내에 이 효소가 국재되어지는 것으로 사료되어진다. 다른 세포내 소기관인미토콘드리아, ER, 그리고 glyoxysomes에 lipoxygenase가 존재한다는 증거는 보이지 않았다.

  • 흥국으로부터 분리된 Monascus 균주로부터 항균활성이 우수한 2균주, 분리주 No.116및 No.481을 선정하여 영양원 및 배양조건의 변화에 따른 항균활성에 대한 검토한 결과 분리주에 따라 차이를 보였으며, 분리주 No.116의 경우 탄소원으로서는 sucrose 8%, 질소원으로 $(NH_4)_2SO_40.8%,$ 인산염으로 $KH_2PO_40.5%$ 그리고 금속성분으로 $MgSO_40.5%$ 첨가시 뚜렷이 향상된 결과를 보였으며, 분리주 No.481의 경우에는 각각 sucrose 8%, $(NH_4)_2SO_41.6~2%,$ $K_2HPO_40.1%,$ $MgSO_40.5%$ 그리고 $FeSO_40.5%$ 첨가시 항균활이 크게 향상된 결과를 보였다. 또한 두 균주 모두 배양 초기의 pH는 5.3, 배양온도는 $32.5^{\circ}C에서$ 가장 우수한 항균물질을 생성할 수 있었다. 상기 배양 최적조건에서 분리주 No.116과 No.481 모두Bacillus subtilis, Staphyloccus aureus, Listeria monocytogenes 및 Enterococcus faecium에 대해 51시간 이상 뚜렷한 생육저해를 나타냈다.

Dynamics of Early Fermentation of Italian Ryegrass (Lolium multiflorum Lam.)Silage

  • The dynamics of fermentation were studied with Italian ryegrass ensiled in the laboratory silos. The silos were kept in the room set at 25$^{\circ}C$, and then were opened on 0.5, 1, 2, 3, 5, 7 and 14 days after ensiling, respectively. The samples were taken from three silos at each sampling time for chemical analyses. Mono-and disaccharides composition was determined for glucose, fructose and sucrose by high performance liquid chromatography. The Italian ryegrass silage succeeded to achieve lactate type fermentation; high values of lactic acid (85.83 g/kg) and lactic acid/acetic acid at the end of ensiling (14 day), low values of pH (3.74), acetic acid (5.38 g/kg), ethanol (19.20 g/kg) and $NH_3-N/Total\;N$ (75.84 g/kg), no or only small amounts of butyric acid, valeric acid and propionic acid. The fermentation dynamics showed a fast and large pH decrease caused by a fast and large production of lactic acid during the first 5 days. Mono-and disaccharides composition largely decreased within initial 0.5 day (12 h) of ensiling. Sucrose disappeared rapidly within initial 0.5 day of ensiling, and fructose and glucose contents showed an initial rise during ensiling, and then decreased gradually. These indicated that the enzymes of plant tissue were active within 2 days of ensiling, which caused the initial rise in fructose and glucose from the hydrolysis of sucrose and fructans. After 5 days of ensilage, glucose was consumed completely, suggesting that glucose was the first fermentation substrate. After 2 days of ensiling, sum amounts of lactic acid and remaining mono-and disaccharides proved to be larger than the quantity of mono-and disaccharides in the initial grass. From the facts mentioned above, it was suggested that considerable amounts of lactic acid were produced from some other substrate such as fructans than initial mono-and disaccharides.

Influence of Cooling Rate, Developmental Stage and Addition of Sugar on Cryopreservation of Pearl Oyster (Pinctada Fucata Martensii) Larvae

  • This study was conducted to investigate cryopreservation of pearl oyster, Pinctada fucata martensii larvae. Four cooling rates (-0.25, -0.5, -0.75 and -1.0$^{\circ}C$/min.) were used to examine a proper cooling rate during cryopreservation of trochophores before seeding temperature (-12$^{\circ}C$). Seven developmental stages (early and late trochophores, early and late D-shaped larvae and early, middle and late umbo stage larvae) and different sugars (fructose, glucose and sucrose) were used to investigate optimal larval stage and effective sugar in cryopreservation of larvae. The survival rates of frozen-thawed trochophores increased at cooling rate of -1.0$^{\circ}C$/min. As larval developing, survival rate of frozen-thawed larvae increased, except umbo stage larvae, and especially late D-shaped larvae highly survived as 91%. Addition of sugar revealed positive effect on cryopreservation in this experiment and 0.2 M glucose and sucrose mixed with 2.0 M dimethyl sulfoxide significantly enhanced survival rate of larvae (P<0.05). The results of our study indicate that desirable cooling rate, developmental stages of larvae and effective sugar far cryopreservation of pearl oyster, P. fucata martensii larvae are -1$^{\circ}C$/min, late D-shaped larvae and 0.2 M glucose and sucrose, respectively.

  • Shoot tips of in vitro propagated plantlets were cryopreserved using encapsulation/dehydration procedures. Shoot tips were excised under filter sterilized antioxidants solution (0.2M phosphate buffer, pH 5.7 supplemented with 5g/1 ascorbic acid and 15g/1 sodium borate). They were drawn up into a sterile 10 $\textrm{cm}^3$disposable pipette and were dropped into the culture medium with 2.5w/v Na-alginate, then into 100mM CaCl$_2$.2$H_2O$. Encapsulated shoot tips were transferred into 10㎤ of liquid culture medium with a range of sucrose concentrations (0.25-1.0M) and were incubated in dark for 24 hours in 18C at 40rpm. Beads were then dehydrated in silica gel for different time intervals (1-24 hours). Then they were freeze dried either rapidly (plunge directly into liquid N2 or in two stages (samples were kept at 20C for 10 minutes, then reduced to 35C at 1C per minute. Variations in the Chemical Compositions of Peanut Mutants Induced by Gamma Radiation

  • This study was carried out to investigate the chemical composition of peanut mutants induced by gamma radiation (Co-60) at 300 Gy in seeds of the cultivar Shinnamkwang. The protein contents of twenty-eight peanut mutants ranged from 23.3% to 31.7% and were increased by from 0.5% to 8.4% in fifteen mutants lines from the 26.8% of the original variety, cv. Shinnamkwang. Lipid contents of in mutants ranged from 43.2% to 53.5%, an increase of 0.2% to 5.7% from the 47.8% of the original variety. The range of unsaturated oleic acid in 28 mutants was from 38.9% to 56.9% an increase of from 1.3% to 14.0% from the 50.6% in the original variety. Linoleic acid, the highest unsaturated fatty acid, constituted 32.6% meanly of mutants, it was 17.4% lesser than oleic acid, ranging from 25.9% to 42.0%. Palmitic acid (16:0) contents ranged from 8.6% to 11.1%, and the mutant line-9 had the highest content. The ratio of oleic to linoleic acid was 0.9 to 2.2. A negative coefficient (r=-0.98**) was obtained between oleic and linoleic acid, but for other fatty acids, no significant relation was observed. Similarly, a negative coefficient of r=-0.68**was observed between saturated and unsaturated acids. The sucrose compositions of mutants ranged from 2.6% to 6.2%.
