들깨 잎 추출물의 항산화 및 신경세포 보호작용 (Antioxidant and Neuroprotective Effects of Perilla frutescens var. japonica Leaves)

  • The leaves of Perilla frutescens Britt. var. japonica Hara (Labiatae) are often used in gourmet food in several Asian countries. Two kinds of perilla cultivars, Namcheon (NC) and Bora (BR), have been respectively developed in Korea by the pure line of 'deulkkae' from the local variety and by the cross of 'deulkkae' and 'chajogi'. The present study evaluated and compared antioxidant and neuroprotective effects of the fractions prepared from the leaves of the two cultivars using cell-free bioassay systems and primary cultured rat cortical cells. We found that the spirit, chloroform, hexane and butanol fractions from NC and BR leaves inhibited lipid peroxidation initiated in rat brain homogenates by $Fe^{2+}$ and L-ascorbic acid. In contrast, only the spirit and butanol fractions from both cultivars exhibited 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity. Among the fractions tested, the butanol fractions from NC and BR leaves exhibited the most potent antioxidant properties, and the butanol fraction from BR was more potent than the NC fraction. In consistence with these findings, the butanol fractions from both cultivars protected primary cultured cortical cells from the oxidative damage induced by $H_2O_2$ or xanthine and xanthine oxidase, with the BR butanol fraction being more active. The butanol fractions from NC and BR did not produce cytotoxicity in our cultures treated for 24 h at the concentrations of up to $100\;{\mu}g/ml$. Taken together, these results indicate that the leaves of the two cultivars of Perilla frutescens exert antioxidant and neuroprotective effects, and that the butanol fraction from BR leaves exhibits the most potent antioxidative neuroprotection among the fractions tested in this study.

n-Decane과 sec-Butanol 혼합물의 최소자연발화온도의 예측 (Prediction of Autoignition Temperature of n-Decane and sec-Butanol Mixture)

  • 최소자연발화온도는 가연성물질이 주위의 열에 의해 스스로 발화하는 최저온도이다. 최소자연발화온도는 유기혼합물 중 가연성 액체혼합물의 안전한 취급을 위해서 중요한 지표가 된다. 본 연구에서는 ASTM E659 장치를 이용하여 가연성 혼합물인 n-Decane+sec-Butanol 계의 최소자연발화온도를 측정하였다. 2성분계를 구성하는 순수물질인 n-Decane과 sec-Butanol의 최소자연발화온도는 각 각 $211^{\circ}C$, $447^{\circ}C$로 측정되었다. 그리고 측정된 n-Decane+sec-Butanol계의 최소자연발화온도는 제시된 식에 의한 예측값과 적은 평균절대오차에서 일치하였다.

  • Sr hexaaluminate($Sr_{1-x}La_xMnAl_{11}O_{19-\alpha}$)와 금속 alkoxide를 전구체로, 1-butanol과 ethylene glycol을 각각 용매로 사용한 sol-gel 법으로 합성하였다. 용매 변화에 따른 Sr hexaaluminate의 물리적 특성을 TG/DTA, XRD 및 $N_2$ adsorption을 이용하여 분석하였다. 합성 후 건조한 시료에 대한 열분해 거동을 분석한 결과는 1-butanol을 용매로 사용하여 합성한 hexaaluminate와 비교할 때, ethylene glycol을 용매로 이용한 경우 용매의 분해반응과 dehydroxylation 반응이 관찰되었고, 결정생성 온도도 상승하였다. Dehydroxylation 반응과 결정생성온도의 상승은 hexaaluminate의 소결현상을 가속시켜 낮은 비표면적의 원인이 되었다. 메탄에 대한 연소 반응으로 표면적 차이가 촉매 활성에 영향을 주었음을 확인하였다.

  • 인화점은 화재 및 액체의 폭발 위험의 가능성을 결정하는 데 사용되는 가장 중요한 물리적 특성이고, 산업공정에서 안정성 평가시 중요한 연소특성 가운데 하나이다. 따라서 본 연구는 4류 위험물 중 알코올계 혼합물인 {methanol + 1-butanol}, {ethanol + 1-butanol} 그리고 {2-propanol + 1-butanol} 이성분계 101.3 kPa에서 최소인화점을 SETA closed cup flash point tester를 이용하여 측정하였다. 각 이성분계에 대하여 Wilson, NRTL 및 UNIQUAC 파라미터를 이용하여 혼합물에 대한 인화점 예측하고 실험 결과와 비교하였다. 비교결과 모든 예측값과 실험값은 유사한 값을 보였고 편차가 1.14 K 이내의 결과를 보였다.

  • We examined butanol fermentation by Clostridium beijerinckii NCIMB 8052 using various hydrolyzates obtained from rice bran which is one of the most abundant agricultural by-products in Korea and Japan. In order to increase the amount of fermentable sugars in the hydrolyzates of rice bran, various hydrolysis procedures were applied. Total eight different hydrolyzates were prepared using rice bran (RB) and defatted rice bran (DRB) with enzyme or acid treatment and both. Each hydrolyzate was evaluated in terms of total sugar concentration and butanol production after fermentation by C. beijerinckii NCIMB 8052. Acid treatment yielded more sugar than enzyme treatment and combined treatment with enzyme and acid yielded even more sugars as compared to single treatment with enzyme or acid. As a result, the highest sugar concentration (33 g/L) was observed from the hydrolyzate from DRB (100 g/L) with combined treatment using enzyme and acid. Prior to perform fermentation of the hydrolyzates, we examined the effect of P2 solution containing yeast extract, buffer, minerals, and vitamins on production of butanol during the fermentation. Fermentation of the hydrolyzates with or without additionof P2 was performed using C. beijerinckii NCIMB 8052 in a 1 L anaerobic bioreactor. Although the hydrolyzates RB were able to support growth and butanol production, addition of P2 solution into the hydrolyzates significantly improved cell growth and butanol production. Highest butanol production (12.24 g/L) was observed from the hydrolyzate of DRB with acid and enzyme treatment after supplementation of P2 solution.

재래식고추장 숙성과정 중의 휘발성 향기성분의 특성 (Changes of Volatile Flavor Compounds in Traditional Kochujang during Fermentation)

  • 찹쌀로 담금한 재래식 고추장의 발효과정 중 휘발성 향기성분을 purge and trap 장치로 포집하여 GC-MSD로 분석, 동정하였다. 재래식 고추장에서 alcohol류 19종, ester류 13종, acid류 7종, aldehyde류 3종, alkane류 1종, ketone류 2종, amine 2종, benzene류 1종, alkene 1종, phenol류 1종, 기타 1종 등 51종의 향기성분이 동정되었고 동정된 향기성분은 담금 직후에 22종이었으나 30일에 41종으로 증가되었고 120일에는 51종으로 향기성분수가 가장 많았다. 이들 성분 중 숙성 전과정을 통하여 공통으로 검출된 성분은 acetic acid-ethyl ester, ethanol, butanoic acid-ethyl ester, 1-butanol, 2-methyl-1-propanol, 3-methyl-1-butanol, butanoic acid, ethenone등 22종이었다. 휘발성 향기성분의 면적비율은 담금 직후에 1-butanol이 30일 이후는 ethanol이 가장 높았다. 고추장 향기성분의 면적비율(peak area%)은 숙성기간에 따라 다소 차이가 있으나 acetic acid-ethyl ester, ethanol, butanoic acid-ethyl ester, 1-butanol, 3-methyl-1-butanol, 2-methyl-1-propanol 등이 높아 재래식고추장의 휘발성 향기성분의 주성분으로 추측된다.

  • This Study was carried out develop antitumor effect of the n-butanol soluble of fraction of Perilla frutescens on human skin melanoma cells. The antitumor activity of various fractions obtained form n-butanol soluble fraction of Perilla frutescens was evaluated in human skin melanoma cells. The antitumor activity of the n-butanol soluble fraction in human skin melanoma cells was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay, neutral red (NR) assay and sulforhordamine B protein (SRB) assay of colorimetic assay methods. The light microscopic study was carried out to observe morphological changes of cultured human skin melanoma cells. These results were obtained follows; The fractions 5 and 6 of the n-butanol soluble fraction of P frutescens were shown significant antitumor activities. The number of human skin melanoma cells were decreased and tend to form cell cluster by treatment with actions 5 and 7 of the n-butanol soluble fraction of P. frutescens. The fraction 6 of the the n-butanol soluble fraction showed the highest antitumor activity on P. frutescens. These results suggest that the fraction 6 of the n-butanol soluble fraction of P. frutescens may be a valuable choice for the studies on the treatment of human skin tumors.
