Antioxidant Effect and Component Analysis of Cardiospermum halicacabum Leaf Extracts (풍선덩굴 잎 추출물의 항산화 효과 및 성분 분석)

  • In this study, the antioxidant effect and component analysis for extract and fractions of Cardiospermum halicacabum leaf were investigated. All experiments were performed with 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from dried C. halicacabum leaf. The yields of extract and fractions were 16.4, 0.9 and 0.3% per dried powder, respectively. DPPH (1,1-phenyl-2-picrylhydrazyl) radical scavenging activity ($FSC_{50}$) of ethyl acetate fraction ($92.5{\mu}g/mL$) was the greatest radical scavenging activity, but lower than (+)-${\alpha}$-tocopherol ($8.9{\mu}g/mL$). In reactive oxygen species (ROS) scavenging activity (total antioxidant capacity, $OSC_{50}$) on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system, aglycone fraction ($4.2{\mu}g/mL$) was the highest total antioxidant capacity and similar to L-ascorbic acid ($1.5{\mu}g/mL$). The cellular protective effects of C. halicacabum leaf extract and fractions on the $^1O_2$-induced cellular damage of human erythrocytes were exhibited at all concentration-dependent ($5.0-25.0{\mu}g/mL$). Especially, aglycone fraction (${\tau}_{50}$, 76.4 min) in $25.0{\mu}g/mL$ showed the most protective effect among extracts. Components of the ethyl acetate fraction obtained from C. halicacabum extracts were analyzed by TLC, HPLC chromatogram and LC/ESI-MS. Results showed that the ethyl acetate fraction contained some flavonoids, such as apigenin-7-O-glucuronide, apigenin-7-glucosdie and quercitrin hydrate. These results suggest that the extracts and fractions of C. halicacabum leaf may be applied as antioxidant functional cosmetic raw materials.

  • It is important to discriminate between tuberculosis and tuberculosis-like disease by Mycobacteria other than tuberculosis in the serodiagnosis of tuberculosis. But because common antigens share among Mycobacteria, their antigenicities to human are similar. Therefore degree of cross-reactivity of antibody in the sera of patients with tuberculosis between M. tuberculosis and Mycobacteria other than tuberculosis should be checked to increase the specificity in the serodiagnosis of tuberculosis. The activity levels of IgG antibody in the sera of 106 patients confirmed as active pulmonary tuberculosis and 30 normal healthy control person to the pressate extract antigen (TE, BE, AE, and FE antigen) from M. tuberculosis, M. bovis, M. avium, and M. fortuitum were measured by enzyme-linked immunosorbent assay and the crossreactivity of IgG antibody with mycobacterial species was analysed. The results were as follows; 1. The activity level(O.D. at 492nm) of IgG to TE antigen in sera of patients with pulmonary tuberculosis was $0.228{\pm}0.167$ in minimal tuberculosis; moderately advanced, $0.556{\pm}0.616$; far advanced, $1.116{\pm}0.651$ and $0.315{\pm}0.245$ in miliary tuberculosis. 2. The activity level (O.D. at 492nm) of IgG to BE antigen in sera of patients with pulmonary tuberculosis was $0.190{\pm}0.162$ in minimal tuberculosis; moderately advanced, $0.337{\pm}0.361$; far advanced, $0.713[\pm}0.460$ and $0.204{\pm}0.162$ in miliary tuberculosis. 3. The activity level (O.D. at 492nm) of IgG to AE antigen in sera of patients with pulmonary tuberculosis was $0.165{\pm}0.114$ in minimal tuberculosis; moderately advanced, $0.392{\pm}0.494$; far advenced, $0.751{\pm}0.512$ and $0.233{\pm}0.191$ in miliary tuberculosis. 4. The activity level (O.D. at 492nm) of IgG to FE antigen in sera of patients with pulmonary tuberculosis was $0.280{\pm}0.227$ in minimal tuberculosis; moderately advanced, $0.460{\pm}0.564$ ; far advanced, $0.845{\pm}0.573$ and $0.257{\pm}0.103$ in miliary tuberculosis. 5. The activity level (O.D. at 492nm) of IgG in sera of healthy control person was $0.126{\pm}0.084$ to TE antigen. $0.105{\pm}0.041$ to BE antigen, $0.103{\pm}0.052$ to AE antigen, and $0.095{\pm}0.061$ to FE antigen. 6. Degree of correlation(r) in activity level of IgG between TE antigen and BE antigen was 0.905 ; between TE antigen and AE antigen, 0.760; between TE antigen and FE antigen, 0.790, and between AE antigen and FE antigen, 0.945. 7. As O.D. above 0.200 was determined positive for the serodiagnosis of pulmonary tuberculosis, the sensitivity and specificity in ELISA using TE antigen were 80% and 87% respectively, whereas in the case of using BE antigen, 66% and 100%; in the case of using AE antigen, 62% and 100%, and in the case of using FE antigen, 72% and 93%, respecitively.

  • The following results were obtained, microstructures and tensile properties in arc brazed joints of DP(dual phase) steel using Cu-5.3wt%Sn insert metal was investigated as function of brazing current. 1) The Fusion Zone was composed of ${\alpha}Fe+{\gamma}Cu$ and Cu23Sn2. The reason for the formation of these solid solutions. Despite, Fe & Cu were impossible to solid solution at room temperature. It's melting & reaction to something of insert metal & Base Metal (DP Steel) by Arc. Brazing Process has faster cooling rate then Cast Process, Supersaturated solid solution at room temperature. 2) The increase Hardness of Fusion Zone was directly proportional to the rise of welding current. Because, ${\alpha}Fe+{\gamma}Cu$ phase (higher hardness than the Cu23Sn2.(104.1Hv < 271.9Hv)) Volume fraction was Growth, due to increasing the amount of base metal melting by High current. 3) The results of tensile shear test by Brazing, All specimens happen to fracture in Fusion Zone. On the other hand, when Brazing Current increasing tend to rise tensile load. but it was very small, about 26-30% of the base metal. 4) The result of fracture analysis, The crack initiate at Triple Point for meet to Upper B.M/Under B.M/Fusion Zone. This Crack propagated to Fusion zone. So ruptured by tensile strength. The Reason to in the fusion zone fracture, Fusion zone by Brazing of hardness (strength) was very lower then the base metal (DP steel). In addition the Fusion Zone's thickness in triple point was thin than the base metal's thickness in triple point.

Optimum Feeding Rate and Frequency in Juvenile Korean Rockfish Sebastes schlegeli Fed a Commercial Diet (조피볼락(Sebastes schlegeli) 치어의 배합사료 적정 공급률과 공급횟수)

  • Two feeding trials were conducted to investigate the effects of feeding rate and frequency on growth performance and body composition of juvenile Korean rockfish Sebastes schlegeli reared at $15.0{\pm}0.3^{\circ}C$. In the first trial, three replicate fish groups averaging $2.07{\pm}0.03g$ were fed a commercial diet with one of seven different daily feeding rates: 1.00%, 2.00%, 2.50%, 2.75%, 3.00%, 3.25%, and satiation (3.34%) based on body weight (BW). After four weeks of feeding, fish fed the 3% diet showed significantly higher weight gain (WG) and feed efficiency (FE) than fish that received the other feeding rates. In the second trial, the optimum feeding frequency was evaluated with three replicate fish groups averaging $2.04{\pm}0.03g$. Fish were fed a commercial diet at 2.87% BW with six different daily feeding frequencies: 2, 3, 4, 5, 7, or 9 meals. After four weeks of feeding, WG, specific growth rate, FE, and protein efficiency rate in fish fed 3 meals $d^{-1}$ were significantly higher than those of fish fed 7 or 9 meals $d^{-1}$. In both feeding trials, proximate composition of the entire body changed depending on the feeding rate or frequency. A broken-line regression analysis based on weight gain suggested that the optimum daily feeding rate and frequency for juvenile Korean rockfish at $15^{\circ}C$ was 2.9% BW $d^{-1}$ and 2-3 meals $d^{-1}$, respectively.

Analytical Prediction for Electromagnetic Characteristics of Tubular Linear Actuator with Halbach Array Using Transfer Relations

  • This paper deals with analytical prediction for electromagnetic characteristics of a tubular linear actuator with Halbach array using transfer relations, namely, Melcher's methodology. Using transfer relations derived in terms of magnetic vector potential and a two-dimensional (2-d) cylindrical coordinate system, this paper derives analytical solutions for magnetic vector potential due to permanent magnets (PMs) and stator winding currents. On the basis of these analytical solutions, this paper also achieves analytical solutions for the magnetic fields distribution produced by PMs, stator windings current and axial thrust. The analytical results are validated extensively by finite element (FE) analyses. In particular, test results such as thrust measurements are given to confirm the analysis. Finally, this paper estimates control parameters using analytical solutions and test results such as thrust, back-emf, inductance and resistance measurements.

Antioxidative and Cellular Protective Effects of Dolwoe (Gynostemma pentaphyllum Makino) Extracts against Oxidative Stress (돌외(Gynostemma pentaphyllum Makino) 추출물의 산화적 스트레스에 대한 항산화 및 세포보호효과)

  • In this study, we investigated the total phenolic and flavonoid contents, component analysis, antioxidative activity and cellular protective effects against oxidative stress on human skin cells in 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from Gynostemma pentaphyllum (G. pentaphyllum) Makino. The DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging activites ($FSC_{50}$) of the 50% ethanol extracts, ethyl acetate fraction and aglycone fraction were 246.8, 147.2, $128.9{\mu}g/mL$, respectively. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of the 50% ethanol extract, ethyl acetate fraction and aglycone fraction on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay were 37.15, 10.74, $7.19{\mu}g/mL$, respectively. We investigated the cellular protective activity and the results showed that treatment of aglycone fraction ($0.05-0.39{\mu}g/mL$) protect human skin cells in a concentration-dependent manner when the skin cell damages were induced by treating them with $H_2O_2$. These results suggest that extract/fractions of G. pentaphyllum Makino may be applicable as natural antioxidants in cosmetics.

Shaft Group Efficiency of Friction Pile Groups in Deep Soft Clay (대심도 마찰무리말뚝의 주면 무리효율 분석)

  • In this study, the behaviors of friction pile groups are investigated using 3D finite element (FE) analysis. The emphasis was quantifying on the shear load transfer (f-w) characteristics of pile groups and the shaft group effects. A framework for determining the f-w curve is proposed based on both theoretical analysis and field load test database. Through comparisons with case histories and FE results, it is shown that the proposed f-w curve is capable of predicting the behavior of a friction pile in deep soft clay. Additionally, a numerical analysis that takes into account the group efficiency factors were performed for major parameter on group pile-soil interaction, such as the pile spacing, pile arrangement, soil condition, and location of pile cap. Based on these results, the shaft group efficiency factors were also proposed.

Study for Manufacturing of Zinc Sulfate from Electric Arc Furnace Dust by Hydrometallrugical Process (제강분진으로부터 습식제련공정에 의한 황산아연의 제조 연구)

  • Herein, we selectively recovered Zn and produced ZnSO4 from electric arc furnace dust using a hydrometallurgical process. The analysis of the properties of the electric arc furnace dust revealed that the Fe content (9.9%) was relatively low while the Mn content (19%) was high as compared to the composition of general dust. Therefore, an appropriate hydrometallurgical process was designed based on the properties of the raw materials. In the leaching process involving the use of 1.6 M sulfuric acid and 20% solid-liquid ratio at 60℃ for 1 h, 85% of the Zn and Mn got dissolved while the Fe was not leached. To selectively recover Zn, a solvent extraction process using D2EHPA as the extractant was chosen, and 99% of the Zn was extracted using 0.8 M D2EHPA with 32% saponification and an O/A ratio of 2 using counter-current 3-stage extraction. Mn was entirely scrubbed with an aqueous sulfuric acid solution of pH 1.5. Finally, Zn was concentrated and stripped using 1.5 M sulfuric acid at an O/A ratio of 4 using counter-current 4-stage stripping. The stripping solution contained 40 g/L of Zn, and 99.9% of ZnSO4∙H2O was obtained by vacuum distillation.

Failure Analysis of Ferroelectric $(Bi,La)_4Ti_3O_{12}$ Capacitor in Fabricating High Density FeRAM Device (고밀도 강유전체 메모리 소자 제작 시 발생하는 $(Bi,La)_4Ti_3O_{12}$ 커패시터의 불량 분석)

  • 고밀도 FeRAM (Ferroe!ectric Random Access Memory) 소자를 개발하기 위해서는 강유전체 물질을 이용한 안정적인 스텍형의 커패시터 개발이 필수적이다. 특히 $(Bi,La)_4Ti_3O_{12}$ (BLT) 강유전체 물질을 이용하는 경우에는 낮은 열처리 온도에서도 균질하고 높은 값의 잔류 분극 값을 확보하는 것이 가장 중요한 과제 중의 하나이다. 불행히도, BLT 물질은 a-축으로는 약 $50\;{\mu}C/cm^2$ 정도의 높은 잔류 분극 값을 갖지만, c-축 방향으로는 $4\;{\mu}C/cm^2$ 정도의 낮은 잔류 분극 값을 나타내는 동의 강한 비등방성 특성을 보인다. 따라서 BLT 박막에서 각각 입자들의 크기 및 결정 방향성을 세밀하게 제어하는 것은 무엇보다 중요하다. 본 연구에서는 16 Mb의 1T/1C (1-transistor/1-capacitor) 형의 FeRAM 소자를 BLT 박막을 적용하여 제작하였다. 솔-젤 (sol-gel) 용액을 이용하여 스핀코팅법으로 BLT 박막을 증착하고, 후속 열처리 공정을 RTP (rapid thermal process) 공정을 이용하여 수행하였다. 커패시터의 하부 전극 및 상부 전극은 각각 Pt/IrOx/lr 및 Pt을 적용하였다. 반응성 이온 에칭 (RIE: reactive ion etching) 공정을 이용하여 커패시터를 형성시킨 후, 32k-array (unit capacitor: $0.68\;{\mu}m$) 패턴에서 측정한 스위칭 분극 (dP=P*-P^) 값은 약 $16\;{\mu}C/cm^2$ 정도이고, 웨이퍼 내에서의 균일도도 2.8% 정도로 매우 우수한 특성을 보였다. 그러나 단위 셀들의 특성을 평가하기 위하여 bit-line의 전압을 측정한 결과, 약 10% 정도의 커패시터에서 불량이 발생하였다. 그리고 이러한 불량 젤들은 매우 불규칙적으로 분포함을 확인할 수 있었다. 이러한 불량 원인을 파악하기 위하여 양호한 젤과 불량이 발생한 셀에서의 BLT 박막의 미세구조를 분석하였다. 양호한 셀의 BLT 박막 입자들은 불량한 셀에 비하여 작고 비교적 균일한 크기를 갖고 있었다. 이에 비하여 불량한 셀에서의 BLT 박막에는 과대 성장한 입자들이 존재하고 이에 따라서 입자 크기가 매우 불균질한 것으로 확인되었다. 또 이러한 과대 성장한 입자들은 거의 모두 c-축 배향성을 나타내었다. 이상의 실험 결과들로부터, BLT 박막을 이용하여 제작한 FeRAM 소자에서 발생하는 불규칙한 셀 불량의 주된 원인은 c-축 배향성을 갖는 과대 성장한 입자의 생성임을 알 수 있었다. 즉 BLT 박막을 이용하여 FeRAM 소자를 제작하는 경우, 균일한 크기의 입자 및 c-축 배향성의 입자 억제가 매우 중요한 기술적 요소임을 알 수 있었다.

Phase evaluation of Fe/Co pigments coated porcelain by rietveld refinement (리트벨트 정밀화법에 의한 Fe/Co 안료가 코팅된 도자기의 상분석)

  • Porcelain (white ware, celadon ware) coated with a ferrous sulfate and ferrous/cobalt sulfate was sintered at 1250℃. The specimens were investigated by HR-XRD, FE-SEM, HR-EDS, and UV-vis spectrophotometer. Through X-ray rietveld quantitative analysis, quartz and mullite were found to be the main phases for white ware, and mullite and plagioclase were found to be the main phases for celadon ware. When the pigment of ferrous/cobalt sulfate was applied, were identified as an andradite phase for celadon ware and a spinel phase for the white ware, and the amorphous phase, respectively. The L* value, which was the brightness of the specimen, was 72.01, 60.92 for white ware and celadon ware, respectively. The ferrous and ferrous/cobalt pigment coated porcelain had L* values of 44.89, 52.27 for white ware and celadon ware, respectively; with a* values of 2.12, 1.40, an d at b* values of 1.45 and 13.79. As for the color of the specimens, it was found that the L* value was greatly affected by the white ware, and the b* value differed greatly depending on the clay. It was thought to be closely related to the production of the secondary phase such as Fe2O3 and andradite phase produced in the surface layer.
