Experimental Planting of the Seedlings for the Restoration of Abies koreana forest in Hallasan (한라산 구상나무림 복원을 위한 묘목식재시험)

  • To seek for restoration method of Abies koreana forest with planting seedlings, Abies koreana seedlings(2, 3 and 5 year old) were planted by date(spring and summer) and surface treatment (coir-mat, coir-net and control) at Janggumok area in Hallasan. Survival rate and shoot growth of the seedlings were measured and compared by planting date, seedling age and surface treatments. For summer planting, survival rates of the seedlings are increased with increasing seedling ages, and those are not differed between surface treatments. For spring planting, survival rates of the seedlings are increased with decreasing seedling ages, and those are not differed between surface treatments. For summer planting, shoot growth of the seedlings are significantly differed between surface treatments. Shoot growth of the seedlings was the highest value 2.11mm at coir-matted plot, and the lowest value 1.49mm at control B)lot. Shoot growth of the seedlings are not differed between the seedling ages, but the highest shoot growth was 2.26mm at coir-matted and 3-year old plot. For spring planting, survival rates and shoot growth of the seedlings are not differed between seedling ages and surface treatments.

Effects of Aluminum Solution Treatment on the Growth of Forsythia koreana and Platanus occidentalis Cuttings(2) (알루미늄용액 처리가 개나리와 플라타너스삽수의 생장에 미치는 영향(2))

  • To examine aluminum toxicity on woody plants, Forsythia koreans and Platanus occidentalis cuttings were grown in the pot(48$\times$33$\times$9cm) filled with sand, and treated aluminum solution and ground water (pH 6.75) 3times per week from April 28, 1993 to June 16. Aluminum solution were prepared 1.0, 2.5 and 5.0mM aluminum potassium sulfate, dilulted with ground water. Growth-related characters (Shoot growth, leaf number and leaf chlorophyll content ) and root growth were measured and compared among the treatments. In all growth-related characters(Shoot growth, leaf number and leaf chlorophyll content), differences among the treatments were highly significant. In root growth, differences among the treatments were highly significant for Forsythia koreana cuttings, but not for Platanus occidentalis cuttings.

Induction of Mutants by Irradiation of $\gamma$-Ray on In vitro Shoots of Persimmon (기내$\gamma$-선 처리에 의한 감(Diospyros kaki Thunb.) 돌연변이 유기)

  • This study was carried out to know the optimum dose of gamma-ray for the induction of mutation in vitro and the characteristics of the mutants induced by gamma-ray in persimmon (Diospyros kaki Thunb.). The LD50 (50% lethal dose) for in vitro shoots of the cultivar, Nishimurawase was between 1 krad and 2 krad and about 1 krad for the cultivar, Ichikikeijiro. As the dose of gamma-ray increased, the length of shoots decreased and necrosis of buds increased. For the cultivar, Nishmurawase, 37.5∼58.3% shoots rooted and the rooting rate and the number of roots per shoot was low in high gamma-ray. The irradiated young plants which were grown in the growth cabinet for 6 weeks were shorter in shoot length and had more branches than non-irradiated plants. The survival rate of irradiated plants grown in the green house for 3 months was 33%, while 77% for control plants.

Forsythia koreana cuttings were severely stressed in aluminum solution more than 1.0mM concentration.

In Vitro Shoot Tip Culture of Pear 'Niitaka' as Related to Tree Vigor Sampling Time and Plant Growth Regulators (배 '신고'의 경정배양에 있어서 모수의 수세, 경정의 채취시기 및 생장조절제의 영향)

  • In vitro shoot tip culture technique was established in pear (Pyrus pyrifolia 'Niitaka') as related to tree vigor, sampling time, and plant growth regulators and sucrose supplemented to medium. Shoot tips excised in June from the tree having medium-vigor developed good shoots. BA (1.0 and 2.0 mg/L) without NAA produced shoots suitable for proliferation, and NAA supplemented to medium resulted in poor shoot growth and excessive callus formation. BA of 2.0 mg/L combined with 0.01 mg/L NAA provided shoots suitable for rooting and sucrose of 30 g/L was recommended for proliferation medium. A fourth strength MS medium supplemented with 0.1 mg/L NAA produced plantlets in good quality of root number and root length.

Photosynthesis, Shoot Growth and Fruit Quality in 'Fuji'/M.9 Mature Apple Trees in Response to Prohexadione-calcium Treatments (Prohexadione-calcium 처리에 따른 성목기 '후지'/M.9 사과나무의 광합성, 신초생장 및 과실품질)

  • This study was conducted to elucidate the influence of prohexadione-calcium (Pro-Ca) application on shoot growth and fruit quality in 'Fuji'/M.9 mature apple trees. Pro-Ca was applied at concentrations ranging from 100 to $400mg{\cdot}L^{-1}$ one time at petal fall (PF; 5-10 cm terminal shoot growth) to the whole canopy of the tree, or at 100, 125, $150mg{\cdot}L^{-1}$ concentrations two times, at PF and then 4-8 weeks after PF. Pro-Ca treatment generally reduced mean shoot growth, with its effect being proportional to the application rate. Pro-Ca at $400mg{\cdot}L^{-1}$ significantly reduced the shoot growth by 15-22% compared to the control, while its effect was not significantly different from Pro-Ca $200mg{\cdot}L^{-1}$. Pro-Ca treatments induced higher occurrence of secondary growth compared to the control. Pro-Ca treatment increased the photosynthetic rate by 5-10% relative to the control, and also increased soluble solid concentration and fruit red color. However, fruit weight was significantly reduced by $400mg{\cdot}L^{-1}$ Pro-Ca, which was attributable to the greater secondary growth caused Pro-Ca $400mg{\cdot}L^{-1}$ treatment compared to the control and Pro-Ca $200mg{\cdot}L^{-1}$ treatment.

Effect of kinds of medium and plant growth regulators for plantlets regeneration by bud culture in Disporum sessile (윤판나물(Disporum sessile) 아배양에 의한 식물체 재분화에 영향하는 배지 종류 및 생장조절물질 효과)

  • This study was conducted to establish the optimal condition for in vitro propagation of Disporum sessile. In the experiments with different kinds of media (MS, B5 and WPM) and explant types for shoot induction and elongation, the highest number of shoot inductions (2.5/explant) was shown when the axillary bud explants were cultured on MS medium without plant growth regulators (PGRs). The best shoot elongation (7.2 cm) was also obtained when the apical buds were cultured on MS medium. The effect of BA pulse treatments with in vitro shoots was also examined. The highest in shoot induction (2.29/explant) and elongation (7.28 cm) was observed when the shoots from axillary buds were cultured on the media without PGRs. However, it decreased with increasing duration of BA pulse-treatment. The highest rooting rate (100%) and number of root inductions (21.3/explant) were achieved with 1.0 mg/L indole-3-butyric acid (IBA) treatment, whereas no differences were observed by different shoot types. The regenerated in vitro plantlets were hardened and successfully established in soil.

Effects of Soil Moisture Control and Dormancy Breaking Agents on Bud Burst and Fruiting for Double Cropping System in a Year in 'Kyoho' Grapes (포도 '거봉' 2기작재배를 위한 하계휴면타파에서 토양수분 조절과 휴면타파제 처리가 발아에 미치는 영향)

  • Double cropping system in a year in Kyoho grapes (Vitis labruscana L.) has currently been attempted in the plastic greenhouse. One of the problems in double cropping system is the promotion of bud break in summer season and shoot fertility. Effects of the control of soil moisture tension near the root zone and treatments of bud dormancy breaking agents on bud breaking in summer were examined to promote the bud break for the second fruiting. The lignification of shoots was induced in July or August by the control of soil moisture tension in root zone environment. The first shoot growth was almost the same as that in common plastic greenhouses. The highest bud break value appeared in the plot of cyanamide chemicals mixed with merit blue as over 75% bud break rate. The bud break rate in the discontinuing plot in irrigation showed significantly higher in bud break than that in the continuing plot in irrigation. Despite of the final high bud break rate, the time of bud break was irregular.

Differences in Regrowth and Terminal Flower Bud Formation of 'Fuji' and 'Jonagold' Apple Trees in Response to Summer Heading Back Pruning of Current Season's Shoots (신초의 하계 절단전정에 의한 사과품종 '후지' 및 '조나골드'의 2차생장과 정화아 형성의 차이)

  • Current season's shoots on 2-year-old branches of 'Fuji' and 'Jonagold' apple trees were heading back pruned to 5 leaves from early May to mid July at about 12-day intervals. The summer heading back pruning induced regrowth on the pruned shoots with different responses between the two cultivars. Generally, greater regrowth occurred on the pruned shoots of 'Fuji' trees than on those of 'Jonagold', irrespective of the time of the heading cut. The shoots of 'Fuji' trees pruned in late May or in June exhibited greater regrowth compared with those pruned in early May or in July, whereas the summer heading back pruning in June resulted in the greatest regrowth for 'Jonagold'. The heading cut induced terminal flower bud formation on the pruned shoots, the percentage of which was higher in 'Fuji' than in 'Jonagold'. The highest percentages of terminal flower bud formation for 'Fuji' and 'Jonagold' were obtained with the heading cut in late May and in mid June, respectively. Percent flowering of the buds was similar in both cultivars, but percent fruit set was slightly higher in 'Fuji' than in 'Jonagold'. The time of the heading cut did not affect percent fruit set in both cultivars. Our results demonstrate that summer heading back pruning of current season's shoots induces regrowth and terminal flower bud formation therefrom when done at appropriate time, but the specific responses to the heading cut are cultivardependent.

  • To investigate the effect of media and growth regulators in micropropagation of persimmon (Diospyros kaki Thunb.), dormant axillary buds taken from trees of persimmon cultivars such as Ichikikeijiro, Tonawase and Hiratenenashi were used. Shoot tips were successfully cultured in full or half of nitrogen strength of MS medium. The most effective cytokinins for shoot proliferation and elongation of persimmon cv. Ichikikeijiro were 5 mg/L and 2 mg/L zeatin, respectively. Shoots were successfully rooted in 1/2N-MS medium with 1 mg/L IBA.
