Total Phenol Contents and DPPH Radical Scavenging Activity of Entomopathogenic Fungi (곤충병원성진균의 총 페놀 함량 및 DPPH 라디칼 소거능)

  • This study was conducted to investigate the total phenol contents, antioxidative activities and antibacterial activities of twenty species of entomopathogenic fungi. The total phenol content was highest in Aspergillus flavus ($553.0{\pm}52.15{\mu}g/g$) and A. parasiticus ($529.9{\pm}60.10{\mu}g/g$). On the other hand those in other strains were within the range of $26.6{\sim}121.9{\mu}g/g$. The antioxdative activity was shown in the most of strains and the highest DPPH (1, 1-diphenyl-2-picrylhydrazyl) radical scavenging activity was observed in A. flavus ($90.9{\pm}2.90%$) and A. parasiticus ($77.9{\pm}4.13%$). This result indicated that the antioxidative activities were very correlated with the total phenol contents. The antibacterial activitiy was found in the every tested pathogenic bacteria. Especially, the antibacterial activity was strongest against Listeria monocytogenes and Escherchia coli.

Effects of Oxyfluorination on Surface Graft Polymerization of Low Density Polyethylene Film and Its Surface Characteristics (함산소불소화가 저밀도 폴리에틸렌 표면의 그라프트 중합 및 그 표면 특성에 미치는 영향)

  • The surface of low density polyethylene (LDPE) film was oxyfluorinated under different reaction conditions to introduce hydroperoxide groups and change surface characteristics. Hydroperoxide functional groups created by oxyfluorination were used as active sites for graft polymerization with hydrophobic monomer, acryl amide (AM), and hydrophilic monomer, methyl methacrylate (MMA) to carry out the second modification of the LDPE film surface. The surface properties of the OFPE films and grafted OFPE films were characterized by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging method, ATR-IR, contact angle measurement and DSC. From the results of DPPH method, the amount of hydroperoxide groups on the oxyfluorinated LDPE film continuously increased as the total pressure in the oxyfluorination and the partial pressure of fluorine gas increased. The water contact angle and surface free energy measurements showed that hydrophilic liquid (water) contact angle on LDPE film surface decreased with hydrophilic AM grafting and hydrophobic liquid (methylene diiodide) contact angle on LDPE film surface decreased with hydrophobic MMA grafting. These were attributed to AM or MMA monomer grafting and the wettability of LDPE filmsurface to hydrophilic and hydrophobic liquids were improved.

Inhibitory Activity of Blueberries on UVB-induced Oxidative Stress and Matrix Metalloproteinase Expression in Human Skin Fibroblasts (인간피부 섬유아세포에서 UVB 유도된 산화 스트레스와 기질금속단백질가수분해효소 발현에 블루베리의 저해능)

  • Intermediate-wavelength solar radiation, also known as ultraviolet B (UVB: 290-320 nm) radiation, may cause premature aging and oxidative damage-dependent skin cancer in humans. UVB-induced formation of reactive oxygen species (ROS)-often a consequence of excessive exposure to these rays-could activate matrix metalloproteinases (MMPs) such as MMP-1 and MMP-3. These enzymes break down type I collagen in human fibroblasts. In this study, we assessed the antioxidant and anti-aging effects of ethyl acetate extract of blueberry (EEB). An antioxidant test in blueberries evaluated ROS production using CCD-986sk cells and DPPH assay. In order to evaluate the anti-wrinkle efficacy of blueberries, the MMP-1 production and type 1 procollagen synthesis evaluated and the expression of MMP 1, 3 were tested through Western blot and RT- PCR. EEB exhibited 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and reduced the production of UVB-induced ROS. Also, EEB inhibited UVB-induced processes associated with photoaging and skin cancer, such as reduction in procollagen production and increase in MMP-1 production. More precisely, EEB (50 ㎍/ml) markedly suppressed mRNA and protein levels of MMP-1 and -3. The anti-aging effects are attributable to the antioxidant activity of EEB. These findings indicate that EEB has a protective effect against UVB-induced aging in human fibroblast cells by regulating the levels of type-1 procollagen, MMP-1, and MMP-3.

Evaluation of the Antioxidant Potential of Korean Indigenous Plant Extracts by Free Radical Scavenging Activity

  • Since reactive oxygen radicals play an important role in carcinogenesis and other human diseases including neurodegenerative states, antioxidants present in natural products have received considerable attention for alleviation of these disease states. Therefore, in order io identify antioxidants in plant extracts, fifty-seven methanolic extracts derided from indigenous Korean plants were primarily assessed for potential to scavenge stable 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals. As a result, nine plant extracts were found to exhibit the DPPH free radical scavenging activity in the criteria of $IC_{50}<40\;{\mu}g/ml$. In particular, the extracts of Melioma oldhami $(IC_{50}=0.1\;{\mu}g/ml)$, Myrica rubra $(IC_{50}=16.2\;{\mu}g/ml)$, Sympolocos paniculata $(IC_{50}=23.0\;{\mu}g/ml)$, Carpinus laxiflora $(IC_{50}=25.1\;{\mu}g/ml)$, and Cleyera japonica $(IC_{50}=26.2\;{\mu}g/ml)$ showed a potent radical scavenging activity. Further study for the identification of active compounds from these lead extracts might be warranted.

Anti-oxidant activities of ethanol extract and fractions from defatted Camellia japonica L. seeds (동백 유박 에탄올추출물 및 분획물의 항산화 활성)

  • The aim of this study was to investigate in vitro antioxidant activities of defatted Camellia japonica L. seeds (DCJS). The DCJS were extracted using ethanol and then fractionated with butanol (BuOH), ethyl acetate (EtOAc), chloroform, and hexane. To evaluate antioxidant activity of extract and fractions from DCJS, we investigated free radical scavenging activities such as 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+), hydroxyl radical (OH), and superoxide anion (O2-) radicals. The five extract and fractions of DCJS dose-dependently increased DPPH, ABTS+ and O2- radical scavenging activities. The BuOH fraction of DCJS showed the highest free radical scavenging activities among other extract and fractions. The contents of total polyphenol and flavonoid in BuOH fraction of DCJS were 23.26 mg GAE/g and 32.39 mg QE/g, respectively. The polyphenol and flavonoids contents of BuOH fraction has highest than other extract and fractions. In addition, BuOH and EtOAc fraction of DCJS contained 102.37 and 165.05 ㎍/g of camelliaside B, respectively. Therefore, DCJS has higher antioxidant activity and may be useful as a natural antioxidant material.

Effects of Mole Crickets (Gryllotalpa orientalis) Extracts on Anti-oxidant and Anti-inflammatory Activities. (땅강아지(Gryllotalpa orientalis) 추출물의 항산화 및 항염증 활성)

  • Tremendous natural product extracts were used as a herb medicine remedy or therapy for centuries. Because these extracts have various biological activities, we examined the effects of Gryllotalpa orientalis extract for anti-oxidant and anti-inflammation activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH), ferric reducing ability of plasma (FRAP), hydroxy radical scarvenging and cyclooxygenase-2 promoter assays. Gryllotalpa orientalis extracts were prepared from solvents such as distilled water (DW), dimethyl sulfoxide (DMSO), ethanol and methanol. The results showed that Gryllotalpa orientalis extracts have potent DPPH (methanol extract), FRAP (DW extract) and hydroxy radical scavenging (DW and methanol extracts) activity than any other extracts used. A significant inhibition of cyclooxygenase-2 (Cox-2) promoter activity was detected in the presence of DMSO extract or ethanol extract. Collectively, the present results suggested that Gryllotalpa orientalis extract could be used for anti-oxidant and/or anti-inflammation agent for human or agricultural purposes.

Antioxidant Potential of Enzymatic Extracts from Blueberry (Vaccinium corymbosum L.) (블루베리 (Vaccinium corymbosum L.) 유래 효소 추출물의 항산화성)

  • Enzymatic extracts were prepared from the blueberry (Vaccinium corymbosum L.) collected in Jeju, Korea. Five carbohydrases namely AMG, Celluclast, Termamyl, Ultraflo and Viscozyme, and five proteases namely Alcalase, Flavourzyme, Kojizyme, Neutrase and Protamex were used to prepare the enzymatic extracts. Antioxidant properties of each extracts were studied using stable 1,1-diphenyl 2-picrylhydrazyl (DPPH), reactive oxygen species (ROS), nitric oxide (NO) scavenging, metal chelating assays and lipid peroxidation inhibitory activity in hemoglobin-induced linoleic acid system. The phenolic content of all enzymatic extracts was in the range of 517.85-597.96 mg/100 g dried sample. DPPH and NO${\cdot}$scavenging, and metal chelating assays exhibited prominent activities. Viscozyme showed the highest DPPH activity $(0.046{\pm}0.002\;mg/mL)$ while AMG Showed the highest activity in NO${\cdot}$scavenging $(0.339{\pm}0.011\;mg/mL)$. All the extracts exhibited strong metal chelating activities. Blueberry enzymatic extracts also showed relatively good activity in hydrogen peroxide scavenging. AMG showed the highest lipid peroxidation inhibitory activity $(0.28{\pm}0.01\;mg/mL)$ in hemoglobin-induced linoleic acid system. In this results, the blueberry, which has potential antioxidant components, may be a good candidate as a natural antioxidant source.

Comparative Study on Antioxidant Activity and Multi-drug Resistance Reversing Activity in Korean Colored Soybean Cultivars

  • The purpose of this study is to analyze antioxidant activity and multidrug resistance reversing activity in several Korean colored soybean (Glycine max Merr.) cultivars. Antioxidant activity of methanol extracts from colored soybean cultivars was evaluated by TBARS (thiobarbituric acid reactive substances), DPPH (1, 1-diphenyl-2-picrylhydrazyl) and ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) methods. By means of TBARS, cultivar "Jeonnam #1" showed the highest activity until 7 days, and followed by "Black #1", "Jinyul" and "Black #3", showing lower activity than that of BHT (butylated hydroxytoluene). Methanol extracts of all cultivars proved that DPPH radical scavenging activity is dose-dependent. Methanol extract from cultivar "Jeonnam #1" showed highest DPPH radical scavenging activity, and followed by cultivars "Black #1". MDR (multi-drug resistance reversing) activity, however, showed the highest effect in "Black #3" and the lowest "Black #1" cultivar. These results suggest that seed colors of soybean may play an important role in antioxidant activity and MDR activity.

Isolation and identification of lignans as Antioxidant from loaves of Catalpa ovata G. $D_{ON}$ (개오동나무 잎으로부터 항산화 활성을 갖는 lignan 화합물의 분리 및 동정)

  • The methanol extract from leaves of Catalpa ovata 6. DoN showed DPPH (1,1-diphenyl-2-picrylhydrazyl) radical-scavenging activity, and its antioxidative compounds were studied. The ethyl acetate-soluble neutral fraction from the methanol extract was successively purified with silica gel adsorption column chromatography, Sephadex LH-20 column chromatography and HPLC. Three antioxidative compounds were isolated and identified as piperitol, pinoresinol and lariciresinol by HR-MS and NMR spectroscopic analyses. The DPPH radical-scavenging activity of the identified compounds decreased in the order of lariciresinol > pinoresinol > piperitol.

Antioxidant activity of Opuntia robusta and its total polyphenol and flavonoid contents

  • The aim of this study was to investigate the antioxidant effects of O. robusta stem extract (ORE) and to determine the total polyphenol and flavonoid contents. Free radical scavenging properties were evaluated using DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid)), and ·OH (hydroxyl radical) scavenging assays. Total polyphenol and flavonoid contents were measured using Folin-Ciocalteu reagent and aluminum chloride colorimetric methods, respectively. Active compounds of ORE were determined using high-performance liquid chromatography (HPLC). The results of the study showed that ORE exhibited DPPH, ABTS, and ·OH radical scavenging activities in a dose-dependent manner. Especially, 1,000 ㎍/mL of ORE showed the strongest radical scavenging properties against DPPH, ABTS, and ·OH. ORE contained total polyphenol content of 57.4 mg GAE/g and total flavonoid content of 5.4 mg QE/g, which may contribute to their antioxidant effects. As a result of HPLC, the contents of active compounds in ORE, dihydrokaempferol (0.65 ㎍/mL), and 3-O-methylquercetin (1.10 ㎍/mL) were confirmed. In conclusion, ORE may be useful as a functional material with antioxidant properties.
