Sensory Characteristics of Pork Sausages with Added Citrus Peel and Dried Lentinus edodes Powders (감귤과피분말 및 건 표고버섯을 첨가한 돈육 소시지의 관능적 특성)

  • The effects of addition of citrus peel powders (C 0, 0.5, 1 & 2%), dried Lentinus edodes powders (L 0, 0.5, 1 & 2%), and their combination (C-L) on the chemical, sensory and textural properties of pork sausages were studied. Addition of 0.5, 1 or 2% C, L, and C-L all significantly decreased moisture content, pH, and color a-values of sausage samples, whereas ash content and color b-value were increased (p<0.05). C, L, and C-L did not affect protein, fat, carbohydrates contents or texture characteristics. Sensory evaluation was performed by multivariate data analysis, namely principal component analysis (PCA). Eighty-two percent total variation was observed in the main structured information among the test groups: the first (PC1) and second (PC2) components of variation were 59 and 23%, respectively. Eight parameters (sweet flavor, pork aroma, bitterness, rancidity, salty flavor, color, sour flavor and citrus aroma) were utilized to describe the main sensory characteristic of the sausages. Addition of 0.5, 1 & 2% citrus peel was obviously correlated with PC1 (salty flavor, sour flavor and citrus aroma, pork aroma, and sweet flavor and rancidity), whereas addition of 0.5 & 1% Lentinus edodes was related with PC2 (aroma and rancidity).

The Vertical Changes of Zooplankton Dynamics and Community Filtration Rate in the Artificial Lake (Okjeong Lake) (인공호수(옥정호)내 동물플랑크톤 동태 및 군집 여과율의 수직적 변화)

  • The vertical changes (upper layer: 0$\sim$0.5 m, middle layer: 4$\sim$5 m and lower layer: 8$\sim$10 m) of the limnological variables, zooplankton dynamics and community filtration rate on the phytoplankton were evaluated monthly intervals from November 2004 to June 2007 at the artificiallake (Okjeong Lake). During the study, the annual mean of water temperatures were 15.2$\pm$7.8$^{\circ}C$ (upper layer), 14.5$\pm$7.4$^{\circ}C$ (middle layer) and 13.0$\pm$6.2$^{\circ}C$ (lower layer), respectively (n=30). The vertical variations of most limnological parameters (DO, pH, conductivity and turbidity) were fairly similar, except for chl.-${\alpha}$ concentration. Annual mean of chl.-${\alpha}$ concentration was highest at the upper layer (21.4 ${\mu}g$ L$^{-1}$), while annual means of concentrations were low at the middle layer and lower layer (4.3 ${\mu}g$ L$^{-1}$ and 3.5 ${\mu}g$ L$^{-1}$, respectively). Annual mean of Secchi depth was 261 cm (n=30). A total 45 species of zooplankton were identified (28 rotifers, 12 cladocerans and 5 copepods) during the study. The mean abundance of rotifers was highest (124$\pm$232 ind. L$^{-1}$, n =90) and that of cladocerans and copepods were relatively low (22$\pm$56 ind. L$^{-1}$ and 13$\pm$30 ind. L$^{-1}$) in the whole layer. Total zooplankton abundance increased from late spring to summer, and then declined and remained low throughout the winter. The annual mean of total zooplankton filtration rate for phytoplankton at the lower layer was much higher than that of both layers (upper layer: 44.4$\pm$107.8, middle layer: 95.1$\pm$436.4 and lower layer: 158.2$\pm$436.4 mL L$^{-1}$ day$^{-1}$). Among the major zooplankton community, relative community filtration rate (RCFR, %) of copepods for phytoplankton (50.2$\sim$54.8%) was much higher than that of cladocerans (27.7$\sim$36.3%) and rotifers (8$\sim$17.6%). The seasonal variation of RCFRs of major zooplankton community was observed. The RCFRs of rotifers were high in winter (44.6%, n=9), while the RCFRs of cladocerans were high in summer (58.0%, n=7). In spring and fall, the RCFRs of copepods were high in the whole layer (spring: 67.4%, n=9; fall: 74.4%, n=5).

Effect of Surfactant Addition on Se Absorption and Growth of Pak-choi and Leaf Lettuce in Hydroponics (수경 재배 시 계면 활성제 첨가가 상추와 청경채의 생육과 Se 흡수에 미치는 영향)

  • Surfactant polyvinyl alcohol (PVA-95) 1, 2, 4mg. $L^{-1}$ and calcium lignosulfonate (CLS) 5, 25, 50, 100mg. $L^{-1}$ were treated to the nutrient solution containing 1.0mg. $L^{-1}$ selenium (Se) to evaluate Se absorption effect using small DFT apparatus. The growth of leaf lettuce and pak-choi did not show unique significance by surfactant kinds and concentration in the tested range and physiological disorder. Vitamin C in leaves of two leafy vegetables was not significantly affected by surfactants also. Among major cations K content in pak-choi was highest in Se I+PVA-95 4mg. $L^{-1}$ treatment, and high in Se I+CLS 5 mg. $L^{-1}$ treatment. Ca content in two leafy vegetables showed increasing pattern by the two kinds of surfactants. Mg content was high in Se I+CLS 25mg. $L^{-1}$ treatment compared to control. Pak-choi absorbed Se about 10 times higher than leafy lettuce. Se content in leaves of two leafy vegetables increased by the treatment of two kinds of surfactants with the exception of CLS 100mg. $L^{-1}$ treatment.

Toxicity Assessment of Phenanthrene using the Survival and Population Growth Rate of the Marine Rotifer, Brachionus plicatilis (해산로티퍼(Brachionus plicatilis)의 생존 및 개체군 성장률을 이용한 Phenanthrene의 독성평가)

  • The oxicity assesment of Phenanthrene (PHE) has been investigated by using the rate (r) of survival and population growth in rotifer Brachionus plicatilis. The survival rate was determined after 24 h of exposure to PHE. The survival rate of PHE had no effect at a maximum of $300mg\;L^{-1}$. The r was determined after 72 h of exposure to PHE. It was observed that r in the controls (absence PHE) was greater than 0.5, but that it suddenly decreased with an increased concentration of PHE. PHE reduced r in a dose-dependent manner and a significant reduction occurred at a concentration of greater than $37.5mg\;L^{-1}$. The $EC_{50}$ value of r in PHE exposure was $63.7mg\;L^{-1}$. The no-observed-effect-concentration (NOEC) of r in PHE exposure was $18.8mg\;L^{-1}$. The lowest-observed-effect-concentration (LOEC) of r in the PHE exposure was $37.5mg\;L^{-1}$. From the results, the concentration of PHE (greater than $37.5mg\;L^{-1}$) has a toxic effect on the r of B. plicatilis in natural ecosystems. These results(including NOEC, LOEC and $EC_{50}$) might be useful for the Polycyclic aromatic hydrocarbons(PAHs) toxicity assessment in marine ecosystems.

In vitro micropropagation of M.26 (Malus pumila Mill) apple rootstock and assessment of the genetic diversity of proliferated plantlets using simple sequence repeat markers (사과 대목 M.26 (Malus pumila Mill)의 기내 대량번식 및 simple sequence repeat 마커를 이용한 증식된 식물체의 유전적 다양성 평가)

  • The objective of this study was to determine the most effective medium condition of shoot proliferation and root formation for the efficient in vitro micropropagation of M.26 (Malus pumila Mill). Simple sequence repeat (SSR) markers were used to analyze the genetic diversity of micro-propagated and greenhouse grown M.26. Shoot proliferation was carried out in MS (Murashige and Skoog) containing benzyladenin (BA, $0.5{\sim}5.0mg{\cdot}L^{-1}$) and thidiazuron (TDZ, $0.01{\sim}0.1mg{\cdot}L^{-1}$). The highest number of shoots (10.67 shoots per explant) was induced by adding BA at a concentration $1.0mg{\cdot}L^{-1}$. TDZ treatments caused higher hyperhydricity rate in cultured explants than in BA treatments. There was no significant effect of both BA and auxin on shoot proliferation, and the optimum proliferation medium for M.26 was MS medium containing $1.0mg{\cdot}L^{-1}$ BA. To find a suitable medium composition for shoot rooting, we tested different concentrations indole-3-butyric acid (IBA) and ${\alpha}$-naphthaleneacetic acid ($0.5{\sim}5.0mg{\cdot}L^{-1}$), MS medium (1/4-1), sucrose ($0{\sim}30g{\cdot}L^{-1}$). The shoots showed good rooting on half-strength MS medium containing $1.0mg{\cdot}L^{-1}$ IBA and $15-20g{\cdot}L^{-1}$ sucrose. The rooting rate (100%), number of roots (10.45 ~ 13.60 roots per explant), root length (7.41 ~ 8.33 cm), and shoot length (4.93 ~ 5.38 cm) were good on this medium. Fifteen SSR primers were detected in a total of 30 alleles in 20 micro-propagated plantlets, all SSR profiles from micro-propagated plantlets were monomorphic and similar to greenhouse grown control plantlet M.26 plant. The results indicated that M.26 micro-propagated plantlets were genetically stable.

Culture characteristics of Leuconostoc mesenteroides WiKim32 in kimchi cabbage juice without supplements and sterilization (비살균 무첨가 배추즙에서 Leuconostoc mesenteroides WiKim32의 배양 특성)

  • This study was performed to develop an economical culture method of lactic acid bacteria (LAB) for kimchi fermentation. Leuconostoc mesenteroides WiKim32 was grown to $1{\times}10^9CFU/mL$ and maintained at 8.88 log CFU/mL for four days by culturing in kimchi cabbage juice (KCJ) without supplements and sterilization. Leuconostoc mesenteroides WiKim32 was cultured in 100 mL of KCJ by inoculating with 0.1% starter culture and adding 100 mL of KCJ everyday by adjusting pH to 5.5 using 1 M NaOH at $20^{\circ}C$ for four days. KCJ was prepared by extraction of kimchi cabbage leaves after washing them with citric acid and ethanol. Adjusting pH over 6.0 was favorable for the growth of LAB in the initial stage, but LAB growth was retarded in the later stage. In contrast, adjusting pH below 5.0 was not beneficial for the growth of LAB; therefore, pH was adjusted to 5.5.

Chromosomal Analysis of Hanwoo Embryos by In Vitro Culture Condition (한우 체외 수정란의 체외 배양 조건에 따른 염색체 분석)

  • Antioxidants were well known to be essential supplements in the complex media and serve as a reservoir of oxygen. In this study, Hanwoo COCs (cumulus oocytes complexes) were matured and developed in L-cysteine-TCM199 and analyzed metaphase chromosome. Maturation rate of Hanwoo COCs were 73.4%, 94.6% in 0.1% PVA, 0.1 mM L-cysteine, respectively and showed significantly different between the treatments (p<0.05). Blastocyst formation were revealed 20.3%, 10.0% in 5% FBS+TCM199, 0.1 mM L-cysteine+1% BSA, respectively. There were no significant difference among treatment groups. Metaphase chromosome were showed 18.3%, 12.0% in 5% FBS-TCM199, 0.1 mM L-cysteine, respectively and analyzable chromosome were 6.1%, 4.0% and had no differences between the treated groups. In the case of in vitro developmental stages, metaphase chromosome were showed 18.3%, 12.0% in $4{\sim}16$ cells stage, 43.1%, 13.0% in morulae stage and 94.8%, 100.0% in blastocyst stage. These results suggested L-cysteine has beneficial role for in virto maturation and development in Hanwoo COCs.

L-Arabinose Production from Diluted Sulfuric Acid Hydrolysis of Corn-fiber (Corn-fiber의 희석된 황산 가수분해에 의한 L-arabinose의 생산)

  • The demand of L-arabinose has been increased recently because of its advantages including clinical effect. L-arabinose can be produced from dilute acid hydrolysis of agricultural wastes. In this study, optimum conditions of L-arabinose production using dilute acid hydrolysis of agricultural wastes and nutshells were determined. Among the tested various agricultural wastes and nutshells, corn fiber was selected as the best raw material for the production of arabinose. The highest arabinose production was achieved an acid hydrolysis of corn fiber for 1 h at 130$^{\circ}C$ with 0.4% sulfuric acid. Above optimal conditions, it was obtained 20.1 g/L glucose, 10.1 g/L xylose, 7.8 g/L arabinose and 1.8 g/L galactose from 90 g/L of corn fiber. For the purification of arabinose, it was carried out to remove all of sugars except arabinose by the Candida tropicalis cultivation of acid hydrolyzate and an organic contaminants such as pigments by the active carbon treatment of fermentation broth. Moreover, experiments were carried out to eliminate an ions by exchange chromatography. Finally, we obtained 3.1 g of partially purified L-arabinose powder with about 40% yield by evaporation and vacuum drying.

Synthesis and Antitumor Activity of $N^1$-derivatives of 5-Fluorouracil (5-Fluorouracil 유도체 합성 및 항암작용)

  • In order to find out a proper connecting bridge between 5-fluorouracil(5-FU) and a macromolecule such as a polypeptide, potentially hydrolytic N$^{1}$-derivartives of 5-FU have been systhesized and evaluated for their biological activity. When tested with in vitro leukemic L$_{1210}$ cells all the obtained derivartives exhibited slightly higher antitumor activity than the parent 5FU. Among them the N$^{1}$ -carbamoyl analogue 2 and N$^{1}$-acetamido analogue 6b showed 50% inhibition of the L$_{1210}$ cell growth at the concentrations of 5.01$\times$10$^{-8}$M and 1.03$\times$10$^{-7}$M, respectively. When tested against sarcoma 180 tumor cells inoculated into mice, the compounds 2 and 6b exhibited, respectively, 62% and 54% inhibition of the solid tumor growth at the 5-time doses of 100 mg/kg/day. Both compounds, N$^{1}$-carbamoyl analogue 2 and N$^{1}$-acetamido analogue 6b, realeased the parent 5-FU when incubated in the L$_{1210}$ cell cultural media for 5 hrs.

p-Fluorophenylalanine Resistant Cell Line Selection and Enzyme Activity from Diploid and Hapliod calli of Nicotiana tabacum cv. BY4 (담배 (Nicotiana tabacum cv. BY4)의 캘러스로부터 p-Fluorophenylalanine 저항성 캘러스 선발 및 효소활성도 측정)

  • Calli were induced on MS medium supplemented with 0.5 mg/L 2,4-D by using the leaf explants of haploid which were derived from the diploid and haploid of Nicotiana tabacum cv BY4. These calli were subcultured on MS medium with the combination of 2.0 mg/L 2,4-D, 1.0 mg/L kinetin and 0.1 mg/L BAP. Cell propagation of diploid plants were good in a combination of 2.0 mg/L 2,4-D, 0.1mg/L BAP in vitro conditions, suspension cultures were conducted in equal condition. Homogenized suspension cultured cells were smeared 2.0 mL each on MS medium with 0~100 $\mu$M PFP, to select the resistant colony to PFP, and were examined after 10d, 20d and 30d. Measurment of fresh weight of cells after 30d of culture shows that with more concentration of PFP in medium the fresh weight of the cells decreased. In case of diploid, selected callus was the highest in vitro treated with 5 $\mu$M PFP. It was higher than control until 100 $\mu$M PFP. The active degree of catalase was the highest in vitro with 5 $\mu$M PFP but the lowest in vitro with 10 $\mu$M PFP on the other hand, in case of haploid plant, the active degree of peroxidase and catalase was the highest in vitro treated with 50 $\mu$M PFP. It's sure that enzyme active degree of between diploid and haploid had big differences.
