Evaluation of Hydration Effect on Human Skin by $^1H$ MRS at 14.1T

  • Purpose: We achieved high resolution MR imaging and spectra of human skin in vitro with using a 14.1 T MRI/MRS system, and evaluated the hydration effect of various cosmetic products by measuring the skin's. moisture concentration. Materials and Methods: We used the Bruker 14.1 T MRI/MRS system with a vertical standard bore that was equipped with a DMX spectrometer gradient system (200 G/cm at a maximum 40 A), RF resonators (2, 5 and 10 mm) and Para Vision software. Spin echo and fast spin echo pulse sequences were employed for obtaining the high resolution MR images. The 3D-localized point resolved spectroscopy (PRESS) method was used to acquire the MR spectra. Results: The high resolution MR images and spectra of human skin in vitro were successfully obtained on a 14.1 T system. The water concentration of human skin after applying a moisturizer was higher than that before applying a moisturizer. Conclusions: The present study demonstrated that the high-resolution MR images and spectra of human skin from a high field NMR instrument could be applicable to evaluating the hydration state of the stratum corneum.

  • Bioassay-directed fractionation of the lipophilic extract of a marine sponge Sarcotragus sp. led to the isolation of a known trisoxazole containing macrolide, mycalolide B (1). Its structure was identified by NMR and MS analyses. This is the first report on the isolation of macrolide from a sponge of the genus Sarcotragus (Order: Dictyoceratida).

  • Polymeric nanoparticles are recognized as promising drug carriers [1]. Here, novel tri-block copolymers based on poly PPDO, PCL and PEG were synthesized and employed for the formulation of reproducible polymeric nanoparticles [2]. To estimate the feasibility of the polymer to form polymeric nanoparticles, nanoparticles were prepared by co-solvent evaporation technique. Polymerization and structural features of the polymer were analyzed by different physico-chemical techniques. Existence of hydrophobic domains as a core of nanoparticles was characterized by $^{1}H-NMR$ spectroscopy, and further confirmed by fluorescence technique using pyrene as probe.

  • Michael Reaction을 이용하여 상업적으로 이용 가능한 APS(3-aminopropyltrime thoxysilane)과 AEAPS(N-[3-(trimethoxysilyl)propy1] ethylenediamine)을 다수의 Michael acceptor(ethyl acrylate, acrylonitrile, acrylamide, 2-cyanoethyl acrylate, 2-hydroxyethyl acrylate 그리고 3-(trimethoxysilyl)propylmethacrylate)와 반응시켜 10종류의 aminosilane ([3-{N-2-carboethoxyethyl)aminopropyl]triethoxysilane, [3-(N-2-cyanoethyl)aminopropyl] triethoxysilane, [3-(N-di-2-carboethoxyethyl) aminopropyl]triethoxysilane, [3-(N-di-2-cyanoethyl)aminopropyl]triethoxysilane, [3-(N-2-cyanoethoxypropionyl)aminopropyl] triethoxysilane, [3-(N-di-2-cyanoethoxypropionyl)aminopropyl] triethoxysilane, [3-(N-di-2-hydroxyethoxypropionyl) aminopropyl]-triethoxysilane, [3-(N-2-amidoethyl aminopropyl]triethoxysilane, {3-[N-(N-di-2-cyanoethyl)ethyl]aminopropyl)triethoxysilane, {3-[N-(3-trimethoxy-silylpropyl)-2-methylpropionyl]aminopropyl)triethoxysilane 등을 35-70% 수율로 제조하였으며, 이들의 구조는 $^1$H-NMR과 FT-IR spectroscopy를 이용하여 확인하였다.

  • A chemical investigation of the polar extract of the sea cucumber Stichopus japonicus, collected from Jeju Island, Korea, has led to isolation of five new fatty acid derivatives (1, 4 - 7) along with known compounds (2 - 3, 8 - 14). Their structures were elucidated by a combination of MS and NMR spectroscopy.

  • The flavonoids from plants is very widly used as natural dye for food and medicine etc. In this study, red pine which is widespread in Korea was studied to find new chemicals which may use as raw material for the special purpose. The fIavonoids of red pine bark were separated with Sephadex LH-20 and Toyo pearl HW-40F as packed materials and the structure of separated f1avonoids was determined by $^1H$-and $^{13}C$-NMR spectroscopy. The (+) catechin which is widespread in nature and dihydroquercetin-3'-0-${\beta}$-galactoside were found in red pine bark. The dihydroquercetin-3'-0-${\beta}$-galactoside is newly found in this species.

유기발광디바이스용 정공수송재료의 합성 (Synthesis of Hole Transport Materials for Organic Light Emitting Device)

  • This study was based on organic electroluminescence display. Especially, TPD and $\alpha-NPD$ for the hole transport materials were synthesized by Ullmann reaction. This reaction was conducted between 3­methylphenylamine, 1-naphthylamine and 4,4'-diiodobiphenyl in toluene containing CuCl catalyst and KOH base. The structural property of reaction products were analyzed by FT-IR, $^1H-NMR$ spectroscopy, and thermal stability, reactivity and PL property were analyzed by melting point, yield and emission spectrum, respectively. The photoluminescence spectra of a pure TPD and $\alpha-NPD$ were observed at approximately 416nm and 438nm respectively. In this study, it was known that the melting point, yield, PL properties of TPD and $\alpha-NPD$ were changed by substituent group of amines.

Synthesis of (Diamine)platinum(II) and (Diamine)platinum(IV) Complexes of Isopropylidenmalonate Ligand and Their Interaction with Guanosine-$5^{\prime}$-Monophosphate

  • A series of (diamine)isopropylidenmalonatoplatinum(Ⅱ) complexes and the oxidation products, (diamine)Pt (OOC)2C=C(CH3)2(X)2, (diamine=ethylenediamine(en), 1,2-diaminopropan(dap), N-methylethylenediamine(men); X=OH, OCOCH3, OCOCF3), have been prepared, and their interaction with guanosine-5'-monophosphate (5'-GMP) have been examined by means of 1H NMR spectroscopy. The present platinum(Ⅱ) complexes have shown to interact with 5'-GMP through N7 coordination in two concecutive steps in a similar way as with cisplatin, but no interaction between the present platinum(Ⅳ) complexes and 5'-GMP was observed. However, in the presence of ascorbic acid, the platinum(Ⅳ) complexes have been found to interact with 5'-GMP with the reaction rate depending on their reduction rate.

화장품중 생약성분의 분석(II) 화장비누중의 쑥추출액의 함량분석 (Analysis of Medicinal Plants in Cosmetics(II) The HPLC Separation and Quantitation of Artemisia Extract in Cosmetic SOAP)

  • A method was described for the analysis of Artemisia extract in cosmetic soap. Esculetin-6-methylether was used as indicator ingredient for analysis of Artemisia extract. It was isolated from the plant and purified with silicagel column. The structure was conformed with IR, NMR and MASS spectroscopy. The Artemisia extract in sample was isolated with chloroform and interfering com- pounds were eliminated with silicagel column. The Artemisia extract was determined by reverse phase high- performance liquid chromatography with a fluorescence detector and a solvent system of H2O/THF/MeOH. The recorveries of Artemisia extract were 93.9-106.1% in the cosmetic soap.

Phytopathogenic fungus Alternaria brassicicola SW-3가 생산하는 항암활성 물질의 분리 정제 (Isolation and Purification of an Antitumor Metabolite from Alternaria brassicicola SW-3, the Cause of Brassica Black Leaf Spot Disease.)

  • 국내 토양에서 분리한 식물성 병원균인 Alternaria brassicicola SW-3리 항암활성 물질 생산능을 조사하고, 활성물질을 분리 정제하여 구조를 확인하였다. A. brassicicola SW-3는potato dextrose broth를 이용하여 15$^{\circ}C$에서 2주간 진탕 배양한 다음, MTT assay를 실시하여 항암활성을 확인하였으며, 배양여액 중의 항암물질은 ethyl acetate로 추출하고, silica gel column chromatography로 정제하여 무색의 oily product를 얻었다(수율 22mg/m1). 분리된 물질은 물이나 hexane에는 녹지 않고, chloroform, ethyl acetate, ethanol 등에는 잘 녹는 특징을 보였으며, , $IR^{1}$H-NMR, $^{13}$C-NMR 등을 통해 구조를 분석한 결과, 최근에 일본에서 분리되어 항암효과가 있는 것으로 알려진 depudecin과 동일한 물질로 추정되었다. 본 실험에서 분리된 depudecin은 인체간암세포와 mouse 피부암세포에 대한 세포독성을 나타내었으며, 각각의$ IC_50$$57\mu$g/ml, $69\mu$g/ml로 나타났다. Alternaria brassicicola SW-3에 의해 생산된 물질이 기지의 물질이지만, depudecin은 아직 작용 기작이나 적용범위 등이 밝혀지지 않은 초기 연구 단계에 있는 물질로서, 새로운 항암제로서의 가능성이 매우 높아 이의 유도체를 합성하거나, 다른 항암제와의 혼용에 의해 부작용이 적은 강력한 항암제를 개발하기 위한 선도물질로 활용될 수 있을 것이다.
