Effect of Hexane Extract of Galla Rhois on Inflammatory Alveolar Bone Loss

  • One of functions of Galla Rhois (GR) is reportedly an anti-inflammatory effect on the several inflammatory diseases. However, an effect of GR related to periodontitis has not been investigated. In the present study, we examined the effect of the hexane extract of Galla Rhois (GR-H) on periodontitis. Cytotoxicity was assessed by MTS analysis using human gingival fibroblast (hGF) cells. Experimental periodontitis was induced by injecting E.coli LPS into the palatal gingiva maxillary molar thrice weekly for 3 weeks (LPS group). GR-H diluted in 1xPBS was orally administrated using a syringe at 30 mg/kg body weight and 100 mg/kg body weight once a day (GR-H group). GR-H effect on the alveolar bone loss (ABL) was digitized with a micro-CT. GR-H treatment at concentrations exceeding 0.5 mg/ml showed cytotoxic effect in hGF cells. The micro-CT among groups were presented for the different distances from cemento-enamel junction (CEJ) to alveolar bone crest (ABC). The results indicated an inhibitory effect on alveolar bone loss for orally administered GR-H in a model of LPS-induced periodontitis.

A Study on the Airborne Pollen Calender in Mt. Kwanak (冠岳山에 있어서 空中에 飛散하는 花紛層에 關한 硏究)

  • The airborne pollens were investigated by Durhan's pollen trap in Mt. Kwanak from March 1, 1984 to February 28, 1985. The observed airborn pollens were identified 22 families and 49 genera, which were divided into 27 tree pollens. Most airborne pollens were found from late-March to late-September and a few from early-November to late-February, 1985. The first observed pollen of the year was Alnus on March 5 and the last was Artemisia on Decomber 19, 1984. The maximum amount of daily airborne pollens was 823 grains/cm2 on May 16. Peak day and the maximum amount of major airborne pollens were as follows; 166 grains/cm2/day of Alnus pollen on April 4, 38 gr. of Populus on April. 13, 22 gr. of Quercus on May 10, 791 gr. of Pinus on May 26, 139 gr. on May 26 and 104 gr. of Graminae on September 6, 24 gr. of Ambrosia on Sep. 10 and 236 gr. on May 26 and 104 gr. of Graminae on September 6, 24 gr. of Ambrosia airborne pollen for one year was 12,733 grains/cm2 which were composed of 61% tree pollens and 39% herb ones. Decreasing order of pollen counts was Pinus, Alnus, Quercus and Populus in trees, and Graiminae, Artemisia and Ambrosia in herbs. Most of the airborn pollens were deposited in the day-time and a few at night, the maximum at 2∼4p.m.and the mininum at 8∼10 p.m.

Thermal aging of Gr. 91 steel in supercritical thermal plant and its effect on structural integrity at elevated temperature

  • In this study, the influence of thermal aging on structural integrity is investigated for Gr. 91 steel. A commercial grade Gr. 91 steel is used for the virgin material, and service-exposed Gr. 91 steel is sampled from a steam pipe of a super critical plant. Time versus creep strain curves are obtained through creep tests with various stress levels at 600 ℃ for the virgin and service-exposed Gr. 91 steels, respectively. Based on the creep test results, the improved Omega model is characterized for describing the total creep strain curve for both Gr. 91 steels. The proposed parameters for creep deformation model are used for predicting the steady-state creep strain rate, creep rupture curve, and stress relaxation. Creep-fatigue damage is evaluated for the intermediate heat exchanger (IHX) in a large-scale sodium test facility of STELLA-2 by using creep deformation model with proposed creep parameters and creep rupture curve for both Gr. 91 steels. Based on the comparison results of creep fatigue damage for the virgin and service-exposed Gr. 91 steels, the thermal aging effect has been shown to be significant.

  • 마오즈가 참고문헌 [4]에서 언급했듯이, GR(1) 합성이 소프트웨어 공학에서 활발히 사용되기 위해서는 명세 작성의 어려움이 해결되어야 한다. 명세 작성을 돕기 위해서 패턴 활용, 자연어 사용 및 그래픽 명세 언어 등이 제안되었지만 기대한 만큼 실효를 거두지는 못하고 있다. 일반 개발자들이 명세 작성을 배울 때 선호하는 방법이 예제에 의한 학습(learning by example) 이다. 본 논문에서는 GR(1) 합성을 이용하여 명세로부터 게임 예제의 제어기를 생성한다.

  • The type of graphite(Gr)/ppy, fullerene$(full^-)$ electrode, ppy one modified with $(full^-)$, was prepared with the cell type of Gr/5mM ppy, 1mM $(full^-)$, 0.1M $TBABF_4$, CH3CN/Pt. The values of the ionic formation rate of the it at electrode materials such as Pt/ppy, Pt, Gr and Au were $93.6,\;7.0\times10^2,\;42.6\;and\;1.3\times10^2cms^{-1}$ respectively. The admittance values of the Grippy electrode and the modified Grippy, $(full^-)$ one were five times enhanced $1.7\times10^{-3}S\;to\;8.3\times10^{-3}\;S$ and capacitance values of electrical double layer of them were 174 times increased $2.4\times10^{-6}\;F\;to\;4.2\times10^{-5}\;F$ respectively.

  • Cotyledon explants of lettuce were cocultured with Agrobacterium tumefaciens LBA4404::pBKS-GR1 harboring glutathione reductase(GR) gene in MS medium supplemented with 0.1 mg/L NAA and 1.0 mg/L 2ip for 48 hr. These explants were transferred to MS medium supplemented with 0.1 mg/L NAA and 1.0 mg/L 2ip, 50 mg/L kanamycin, and 500 mg/L carbenicillin. After 4 weeks of subculture, kanamycin-resistant shoots were obtained on selection medium. Leaves of putative transformants survived on selection medium containing 100 mg/L kanamycin. Incoporation of the GR gene into lettuce was confirmed by PCR analysis of genomic DNA. Southern blot analysis showed that ECL-labeled GR gene was hybridized to the expected amplified genomic DNA fragment of about 1.8 kb from transgenic lettuce. The presence of mRNA in transgenic lettuce was confirmed by RT-PCR with total RNA of transgenic lettuce. In progeny test of transformants, R$_1$ seeds were resistant to kanamycin (200mg/L) on MS medium.

An NMR study on the intrinsically disordered core transactivation domain of human glucocorticoid receptor

  • A large number of transcriptional activation domains (TADs) are intrinsically unstructured, meaning they are devoid of a three-dimensional structure. The fact that these TADs are transcriptionally active without forming a 3-D structure raises the question of what features in these domains enable them to function. One of two TADs in human glucocorticoid receptor (hGR) is located at its N-terminus and is responsible for ~70% of the transcriptional activity of hGR. This 58-residue intrinsically-disordered TAD, named tau1c in an earlier study, was shown to form three helices under trifluoroethanol, which might be important for its activity. We carried out heteronuclear multi-dimensional NMR experiments on hGR tau1c in a more physiological aqueous buffer solution and found that it forms three helices that are ~30% pre-populated. Since pre-populated helices in several TADs were shown to be key elements for transcriptional activity, the three pre-formed helices in hGR tau1c delineated in this study should be critical determinants of the transcriptional activity of hGR. The presence of pre-structured helices in hGR tau1c strongly suggests that the existence of pre-structured motifs in target-unbound TADs is a very broad phenomenon.

Protective effects of quercetin-3-glucosyl-(1-2)-rhamnoside from Schizophragma hydrangeoides leaves on ultraviolet A-induced photoaging in human dermal fibroblasts

  • Schizophragma hydrangeoides (S. hydrangeoides) is a vine endogenous to Jeju Island and Ulleungdo, where it grows attached to the foothills and rock surfaces. Previous research has mostly focused on the whitening effect of S. hydrangeoides leaf extract. In this study, we investigated S. hydrangeoides leaf extract further, and detected four phytochemicals in the extract: chlorogenic acid, quercetin-3-O-glucosyl-(1-2)-rhamnoside, quercetin-3-O-xylosyl-(1-2)-rhamnoside, and quercitrin. We pretreated human dermal fibroblast (HDFn) cells with previously established concentrations of the four compounds for 1 h before ultraviolet A (UVA) irradiation. Among the four compounds, quercetin-3-O-glucosyl-(1-2)-rhamnoside (Q-3-GR) best inhibited matrix metalloproteinase-1 (MMP-1) levels. Thus, we investigated the protective effects of Q-3-GR on photoaging and its underlying mechanisms. Q-3-GR significantly reduced MMP-1 production and inhibited MMP-1 protein expression in UVA-irradiated HDFn cells. Furthermore, Q-3-GR increased procollagen type I production and protein expression. Q-3-GR exerted its anti-photoaging effects by downregulating the mitogen-activated protein kinase/ activator protein-1 signaling pathway, and upregulating the transforming growth factor-β/Smad signaling pathway. Thus, S. hydrangeoides leaf-derived Q-3-GR is a potential potent cosmetic ingredient for UV-induced skin aging.

A Study on the Variation of Color and its Components of the Flue-Cured Tobacco Leaf During the Process of Curing (Flue -cured 잎담배의 건조과정중 색상과 색소성분 변화에 관한 연구)

  • Boring the process of curing of the tobacco ( Nicotiana tabacum L. clicks) leaf, the and other fundamental components of the tobacco leaf was observed The amount of total chlorophyll was reduced from 2902$\mu\textrm{g}$/gr to 1, 131$\mu\textrm{g}$/gr as a function of curing time. In the case of total carotene, the concentration was increased from 373-712$\mu\textrm{g}$/gr. to 3, 003~2, 215$\mu\textrm{g}$/gr. up to 70 hours of curing period. However, after 70 hours of curing, the concentration of total carotene stared to be decreased up to 1, 050~1, 039$\mu\textrm{g}$/gr. Simultaneously, the greenish yellow color becomes to be close an intermediate color between yellow and orange. The amount of total nicotine and nitrogen (2~2%) in the green harvested leaf was essentially identical after the curing process. However, the amount of total sugar was enhanced from 17%, in the green harvested leaf, to 35% at the time of 70 hours curing. At the end of the curing, the amount of total sugar was again reduced up to 16%. The correlation coefficient (r) between the axis of the color coordinate and the ratio of chlorophyll/carotene was estimated to be r=-0.96. This result indicates a linear relationship between those two parameters and the following linear equation is obtained as y=28.87-68.38 x. From the above equation, x=0.442 could be estimated when chlorophyll was completely decomposed (ylongrightarrowO). As a consequence of this result, we can suggest the tobacco leaf of Hicks belongs to the high quality level.

A new function of glucocorticoid receptor: regulation of mRNA stability

  • It has long been thought that glucocorticoid receptor (GR) functions as a DNA-binding transcription factor in response to its ligand (a glucocorticoid) and thus regulates various cellular and physiological processes. It is also known that GR can bind not only to DNA but also to mRNA; this observation points to the possible role of GR in mRNA metabolism. Recent data revealed a molecular mechanism by which binding of GR to target mRNA elicits rapid mRNA degradation. GR binds to specific RNA sequences regardless of the presence of a ligand. In the presence of a ligand, however, the mRNA-associated GR can recruit PNRC2 and UPF1, both of which are specific factors involved in nonsense-mediated mRNA decay (NMD). PNRC2 then recruits the decapping complex, consequently promoting mRNA degradation. This mode of mRNA decay is termed "GR-mediated mRNA decay" (GMD). Further research demonstrated that GMD plays a critical role in chemotaxis of immune cells by targeting CCL2 mRNA. All these observations provide molecular insights into a previously unappreciated function of GR in posttranscriptional regulation of gene expression. [BMB Reports 2015; 48(7): 367-368]
