The aim of this study was to evaluate the total phenol, total flavonoids contents and antioxidant activity of 80% ethanolic extract from Ixeris dentata Nakai(IDE) as well as to assess the lipid accumulation during adipogenesis of 3T3-L1 cells. The results demonstrated that the total phenolic and flavonoids contents of the IDE were $4.01{\pm}0.63$ GAE mg/g and $0.05{\pm}0.01$ RE mg/g, respectively. The antioxidative activities of the IDE were significantly increased in a dose dependent manner on DPPH(1,1-Diphenyl-2-picrylhydrazyl) radical scavenging, ABTS(2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt) radical scavenging, reducing power value. During adipocyte differentiation, IDE significantly inhibited lipid accumulation compared with the control cells. These results indicated that the anti-adipogenesis effect of Ixeris dentata Nakai could be attributed to phenolic compounds that may potentially inhibit ROS(reactive oxygen species) production.

Culinary mushroom Pleurotus pulmonarius has been popular in Asian countries. In this study, the anti-oxidant, cholinesterase, and inflammation inhibitory activities of methanol extract (ME) of fruiting bodies of P. pulmonarius were evaluted. The 1,1-diphenyl-2-picryl-hydrazy free radical scavenging activity of ME at 2.0 mg/mL was comparable to that of butylated hydroxytoluene, the standard reference. The ME exhibited significantly higher hydroxyl radical scavenging activity than butylated hydroxytoluene. ME showed slightly lower but moderate inhibitory activity against acetylcholinesterase (AChE) and butyrylcholinesterase than galantamine, a standard AChE inhibitor. It also exhibited protective effect against cytotoxicity to PC-12 cells induced by glutamate ($10{\sim}100{\mu}g/mL$), inhibitory effect on nitric oxide (NO) production and inducible nitric oxide synthase protein expression in lipopolysaccharide-stimulated RAW 264.7 macrophages, and carrageenan-induced paw edema in a rat model. High-performance liquid chromatography analysis revealed the ME of P. pulmonarius contained at least 10 phenolic compounds and some of them were identified by the comparison with known standard phenolics. Taken together, our results demonstrate that fruiting bodies of P. pulmonarius possess antioxidant, anti-cholinesterase, and inflammation inhibitory activities.
Taurine is known to suppress oxidant-induced tissue injury by stabilizing biomembrane and scavanging free radicals. The purpose of this study was to determne the antioxidative and antimutabenic acitvities of taurine, ad to compare those acitivities with major antioxidants. For the measurement of antioxidative capacity, 0.05 , 0.1,0.5 and 1.0mg/ml of taurine, L-Ascorbic acid, alpha-tocopherol, and BHT (dibuty hydroxiy toluene)were prepared and tested for their ability to donate electrons to DPPH (1,1-diphenyl-2-picryl-hydrazyl). Antimutagenic acitivity was examined using the Ames salmonela test system at concentrations of 600, 900 and 1200ug/ml. Results indicated that taurine possesses electron-donating capacity, however, the degree of donation was very weak compared to the major antioxidants tested. However, taurine was evaluated as a potent mutation suppressor. Antimutagenic capacity was in increasing order BHT>taurine>L-ascorbic acid>alpha-tocopherol at concentrations of 600 and 900ug/ml. There was a dose-dependent increase in antimutabenicity of these compounds , however, antimutagenity of the 900ug taurie/plate was not significantly differently from that of 1200ug taurine/plate. These results indicate that taurine effectively suppresses the mutagenicity of AFB1 without noticeable elelctron donating ability.
The antioxidant, whitening, and anti-wrinkle activity of Spirodela polyrhiza extracts and fractions were evaluated to determine its efficacy as a functional cosmetic material. 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azinobis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activities were 44.2 and 74.3%, respectively, at 100 ㎍/mL of SE-E (the ethyl acetate fraction of 70% ethanol extract). To measure anti-wrinkle effects, procollagen biosynthesis and matrix metalloproteinase-1 (MMP-1) inhibition activity were determined. At 25 ㎍/mL of SE (70% ethanol extract), the biosynthesis activity was 48.5%, and SE-E showed the best activity (57.8%) at the same concentration. MMP-1 inhibition activity of SE and SE-E was 13.4 and 28.5%, respectively, at 25 ig/mL. Finally, the inhibition of cellular melanin synthesis and cellular tyrosinase were measured to determine the whitening effect; at 25 ㎍/mL, the inhibition activities of SE were 9.6 and 13.8%, respectively, and those for SE-E were 15.4 and 22.0%, respectively. Our results confirmed the possibility of SE and SE-E as effective functional materials. Further research investigating the antimicrobial, anti-inflammatory, and anticancer activities of S. polyrhiza is necessary to confirm its potential use in the food, cosmetics, and drug industries.
As part of our ongoing study focused on the discovery of antioxidants from natural products by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity, methanol extract of flowers of Carthamus tinctorius L. was found to show potent antioxidant activity. Activity-guided fractionation of the methanol extract lead to the isolation of twenty compounds including two flavonol glycosides, quercertin-3-O-${\beta}$-D-glucopyranoside (12) and kaempferol-3-O-${\alpha}$-L-rhamnopyranosyl-${\beta}$-D-glucopyranoside (18), two flavanone glycosides, (2S)-4',5,6,7-tetrahydroxyflavanone 6-O-${\beta}$-D-glucopyranoside (15) and (2R)-5,7,8',4-tetrahydroxyflavanone 8-O-${\beta}$-D-glucopyranoside (16), and two acetylenic glycosides, 8Z-decaene-4,6-diyne-1-O-${\beta}$-D-glucopyranoside (13) and 4,6-decadiyne-1-O-${\beta}$-D-glucopyranoside (14). Their chemical structures were identified by using spectroscopic analysis. Among them, compounds 12-18 were tested in DPPH assay. Compounds 13-16 were first reported to their antioxidant activity. Quercertin-3-O-${\beta}$-D-glucopyranoside (12) showed the most potent inhibitory effect on DPPH with $IC_{50}$ value of 56.7 ${\mu}M$.
This study was conducted to compare the antioxidant, anticytotoxic, and anti-inflammatory properties of Euphorbia maculata ethanol extract with those of E. supina ethanol extract. 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical and superoxide scavenging activities of E. maculata at $50{\mu}g/mL$ were $38.3{\pm}3.7$ and $21.5{\pm}1.2%$, respectively, whereas those of E. supina at the same concentration were $109.4{\pm}0.9$ and $59.5{\pm}4.8%$, respectively. Oxygen radical absorbance capacities of E. maculata and E. supina at $10{\mu}g/mL$ were $14.70{\pm}0.63$ and $26.17{\pm}1.36nmol/mL$ Trolox, respectively. Cupric reducing antioxidant capacities of E. maculata and E. supina at $10{\mu}g/mL$ were $10.22{\pm}0.97$ and $62.99{\pm}5.28nmol/mL$ Trolox, respectively. Total phenolic contents of E. maculata and E. supina at $50{\mu}g/mL$ were $29.03{\pm}0.14$ and $87.89{\pm}0.20nmol/mL$ gallic acid, respectively. E. maculata and E. supina were reported to prevent supercoiled DNA breakage induced by peroxyl and hydroxyl radicals in a concentration-dependent manner, where protection against the supercoiled DNA breakage provided by E. supina was greater than that provided by E. maculata. E. maculata and E. supina at $100{\mu}g/mL$ inhibited tert-butyl hydroperoxide-induced cytotoxicity in HepG2 cells by $49.4{\pm}4.3$ and $87.3{\pm}4.5%$, respectively. E. maculata and E. supina at $500{\mu}g/mL$ inhibited lipopolysaccharide-induced nitric oxide production in RAW 264.7 cells by $63.1{\pm}7.0$ and $85.2{\pm}1.6%$, respectively. The antioxidant capacities including DPPH radical scavenging, superoxide scavenging, oxygen radical absorbance, and cupric reducing antioxidant activity were found to be highly correlated with total phenolic content (0.896 < r < 0.983, p < 0.01) and anticytotoxic activities (0.915 < r < 0.960, p < 0.01). However, the superoxide scavenging activity was not significantly correlated (r = 0.604, p > 0.05) with the anti-inflammatory activity. Thus, these findings demonstrated that the radical scavenging, anticytotoxic, and anti-inflammatory capacities of E. supina were more potent than those of E. maculata. Further studies are needed to elucidate the properties of polyphenolic constituents in E. supina responsible for these effects and the underlying mechanisms.

Vacuum deposited N,N-di-1-naphthyl-N,N-diphenyl-1,1'-biphenyl-4,4'-diamine (NPD) as a hole transporting (HTL) materials in OLEDs was placed on PEDOT-PSS, a hole injection layer (HIL). PEDOT-PSS was spin-coated on to the ITO glass. $C_{60}$-doped NPD-$C_{60}$(10 wt%) film was formed via co-evaporation process and the morphology of NPD-$C_{60}$ films was investigated using XRD and AFM. The J - V, L - V and current efficiency of multi -layered devices were characterized. According to XRD results, the deposited $C_{60}$ thin film was partially crystalline, but NPD-$C_{60}$ film was observed not to be crystalline, which indicates that $C_{60}$ molecules are uniformly dispersed in the NPD film. By using $C_{60}$-doped NPD-$C_{60}$ film as a HTL, the current density and luminance of multi-layered ITO/PEDOT-PSS/NPD-$C_{60}/Alq_3$/LiF/Al device were significantly increased by about 80% and its efficiency was improved by about 25% in this study.
Allium senescens L. is perennial plant of the Liliaceae family that grows throughout Korea. In this study, we investigated the effect of Allium senescens L. methanol extracts on reactive oxygen species (ROS) production and lipid accumulation during adipogenesis. Our results indicated that 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity of Allium senescens L. methanol extracts increased in a dose-dependent manner. Allium senescens L. methanol extracts suppressed ROS production and lipid accumulation during adipogenesis. In addition, Allium senescens L. methanol extracts inhibited the mRNA expression of the pro-oxidant enzyme, such as G6PDH and lead to a reduction in the mRNA levels of the transcription factors, such as sterol regulatory element binding proteins 1c, peroxisome proliferator-activated receptor ${\gamma}$, and CCAAT/enhancer-binding proteins ${\alpha}$. These results indicate that Allium senescens L. methanol extracts inhibit adipogenesis by modulating ROS production associated with ROS-regulating genes and directly down-regulating adipogenic transcription factors.
Measurement of toxicologically relevant polychlorinated biphenyl (PCB) congeners such as non-ortho(IUPAC#) 77, 81, 126, 169 and mono-ortho 105, 114, 118, 123, 156, 157, 189 and di-ortho 170, 180 and polybrominated diphenyl ethers (PBDEs) such as 47, 66, 85, 99, 100, 138, 153, 154 in environmental samples become almost mandatory in several countries now. However, most of the available methods involve expensive instrumentations such as HRGC-HRMS or ECNI-LRMS, apart from expensive extraction and clean-up (with large volume of solvents) steps. A method has been devised combining the analytical separation power of PYE [2-(1-pyrenyl)ethyldimethysilylated silica] column HPLC and high-resolution gas chromatographic techniques including micro-electron capture detection (ECD) and two dimensional gas chromatograpy-ECD techniques to determine these eco-toxic substances at parts-per-trillion (ppt) levels. This combination resolves co-elution of congeners that occur in disproportionate ratios (e.g. CB-110 and -77) and allows accurate congener-specific determination of target compounds. This method is cost effective as it requires only hexane, that in small quantities (10 mL) and GC-ECD. The elution and analysis time are optimized to less man hours. This method is effectively utilized in the analysis of co-planar PCBs and PBDEs from archived solvent extracts of samples previously analyzed for pesticides and PCBs. Structure based separation of contaminant classes improves GCECD determination at ppt levels.
This study characterized the anti-Helicobactor pylori activity of Xanthium strumarium L. extract obtained by lactic acid fermentation. The growth of the Lactobacillus strains was typically robust upon lactic acid production in monocultures containing Xanthium strumarium L. extract. Lactic acid fermentation in mixed cultures of Lactobacillus brevis KCTC 3498 and Lactobacillus casei KCTC 3109 produced higher of anti- H. pylori activity than monocultures. Concerning antioxidant activity of fermented extracts, total polyphenol contents were the highest in co-cultures of Lactobacillus brevis KCTC 3498 and Lactobacillus helveticus KCTC 3545. Electron donating ability using diphenyl picryl hydrazyl (DPPH) showed 70% scavenging in Lactobacillus casei KCTC 3109.
