녹차와 목단피의 항산화 성분 (Antioidants of Theae Folium and Moutan Cortex)

  • 산화성 프리 라디칼과 반응함으로써 항산화제로 작용할 수 있는 물질을 얻을 목적으로 1,1-diphenyl-2-picryl hydrazyl(DPPH)을 프리 라디칼 모델로 사용하여 여러 식물 생약의 에탄올 추출물을 검색하고, 그 결과로 선별된 녹차와 목단피의 추출물에 대해서는 용매에 의한 분획과 컬럼 크로마토그라피를 실시하여 각각의 유효 성분을 분리, 규명하고 그 항산화 효과를 평가하였다. 녹차의 경우 그 유효 성분이(-)-epigallocatechin$(100\;{\mu}M\;DPPH$의 50%를 환원 시키는데 필요한 농도 즉, $SC_{50}=3.2\;{\mu}g/ml)$과 (-)-epigallocatechin gallate$(SC_{50}=2.6\;{\mu}g/ml)$로 나타났으며, 목단피의 경우는 gallic acid$(SC_{50}=2.0\;{\mu}g/ml)$가 유효 성분으로 동정되었다. 이들 화합물들은 $Fe^{3+}-ADP/ascorbate$계에 의한 microsome 분획에서의 지질 과산화 반응을 잘 억제하였으며, 그 항산화력은 (-)-epigallocatechin(1mg protein/ml의 microsome 분획에 대하여 지질 과산화를 50% 억제하는데 필요한 농도 즉, $(IC_{50}=0.2\;{\mu}g/ml)>(-)-epigallocatechin\;gallate(IC_{50}=2.5\;{\mu}g/ml)$ 순으로 나타났다. 본 연구의 결과로 부터 강력한 항산화 물질들을 함유한 녹차와 목단피등의 식물 생약이 각종 프리 라디칼 반응에 의한 생체 손상을 막아줄 수 있을 것으로 기대되었다.

까마중내 (Solanum nigrum L.) 항산화방어계의 항산화력 및 물질의 동정 (Evaluation of the Antioxidant Potential and ldentification of Active Principles of Solanum nigrum L. on Antioxidant Defense Systems)

  • 까마중의 부위별 DPPH free radical 소거활성은 뿌리에서 가장 높았고, 줄기, 전부위, 열매 잎 그리고 꽃 순으로 높았다. 추출방법간의 DPPH free radical 소거활성은 80%methanol에 침지하여 추출한것보다는 $80^{\circ}C$에서 80% meth-anol에 흰류추출한것이 높았다. Sephades LH-20 column chromatography에 의하여 분획된 L6의 DPPH free radical 소거활성은 ethy acetate 분획 추출물보다 6.7배 높았다. 분획물내의주요물질은 2, 6-methano-3-benzazocin-11-ol, 2[1h]-pyridinethione 및 2-hydroxy-5 methylbenzaldehyde로 phenolic 화 물이었다. 줄기와 뿌리의 POD및 SOD 활성은 타 부위에 비하여 높은 활성을 나타내었고, 생육기간에는 파종 후 60일후에서 활성이 가장 높았다. PODdmlehddnl gth 패턴은 뿌리에서 10개의 band로 가장 많았고 그중 8개의 band가 식물체의 부위별로 차이가 있어Tdmaul, SOD의 동위효소 패턴은 잎에 5개의 band로 가장 많았고 그중 2개의 band가 식물체의 부위별로 차이가 있었다.이가 있었다.

  • Effects of Taeumjowetang on Lipid Peroxidation by Free Radicals and Oxidative Damage of Hepatocytes by tert-Butyl Hydroperoxide. 1. Purpose The present study was carried out to evaluate the antioxidant effects of Taeumjowetang in vitro. 2. Methods In this study, antioxidant effects of TJT on lipid peroxidation were determined according to the method of TBA. (Abbreviation) TJT : Taeumjowetang, TBA : 2-thiobarbituric acid. 3. Results : 1) TJT inhibited markedly peroxidation of linoleic acid during the autoxidation. 2) TJT inhibited lipid peroxidation induced by hydroxyl radical derived from H2O2-Fe2+ in rat liver homogenate. 3) TJT showed 66% scavenging effect on DPPH radical. 4) TJT exhibited a 25% inhibitory effect on superoxide generation from xanthine-xan thine oxidase system. 5) To investigate the antioxidative effects of TJT on the hepatocytes, cultured normal rat liver cells(Ac2F) were prepared and incubated with or without TJT. After 16~18hr, cells placed in DMEM medium without serum, and then incubated with 1mM t-BHP for 2hr. Viable cells were detected by MTT assay. In this test, TJT protected the cell death induced by t-BHP and significantly increased cell viability in the normal rat liver cell. Effects of mushroom waster medium and stalk residues on the growth performance and oxidative status in broilers

  • Objective: The study developed mushroom stalk residues as feed additives in the broiler diet for improving the growth performance and immunity of broilers as well as to increase the value of mushroom stalk residues. Methods: In total, 300 ROSS 308 broilers were randomly allocated into fifteen pens with five dietary treatments: i) control, basal diet; ii) CMWM, supplemented with 1% Cordyceps militaris waster medium (CM); iii) CMPE, supplemented with 0.5% CM+0.5% Pleurotus eryngii stalk residue (PE); iv) CMPS, supplemented with 0.5% CM+0.5% Pleurotus sajorcaju stalk residue (PS); v) CMFV, supplemented with 0.5% CM+0.5% Fammulina velutipes stalk residue (FV). Results: The chemical analysis results showed that CM extracts, PE extracts, PS extracts, and FV extracts contain functional components such as polysaccharides and phenols and have both 2, 2-diphenyl-1-picryl-hydrazyl-hydrate scavenging and Ferrous scavenging capacities. The group CMWM saw increased body weight gain and feed conversion rate and the promotion of jejunum villus growth, but there is no significant difference in the intestinal bacteria phase. Antioxidant genes in the nuclear factor (erythroid-derived 2)-like 2 (Nrf2)- antioxidant responsive element pathway among the groups are significantly higher than that of the control group, especially in group CMWM. Conclusion: The mushroom stalk residues have antioxidant functional components, can improve the intestinal health and body weight gain of chickens, and can activate the antioxidant pathway of Nrf2 to increase the heme oxygenase-1 expression. The treatment with 1% CM was the most promising as a feed additive.

인진청간탕(茵蔯淸肝湯)의 알코올성 산화스트레스에 대한 보호효과 연구 (Protective Effects of Yinjinchunggan-tang (YJCGT) on Alcohol-induced Oxidative Stress)

  • Objectives : Oxidative stress seems to play a major role in mechanisms by which ethanol causes liver injury. Previous studies have shown that treatment with Yinjinchunggan-tang (Yinchenqinggan-tang, YJCGT) has protective effects on alcoholic liver disease. The aim of this study was to investigate the protective effects of YJCGT on alcohol-induced oxidative stress. Materials and Methods : In vitro, we evaluated the inhibitory activities of YJCHT on DPPH(1,1-diphenyl-2-picryl-hydrazyl), xanthine oxidase, trypsin, and hyaluronidase. In a cell culture model, we measured cell viability and proliferation, and the activities of superoxide dismutase (SOD), and catalase (CAT) after YJCGT treatment in C34 and E47 cell lines, and HepG2 cells transfected with/ without cytochrome P450IIE1 (CYP2E1) gene. In vivo, we estimated serum level of hepatic biochemical markers, and alcohol concentration in the blood. Results : YJCGT showed significant free radical scavenging activity against DPPH and xanthine oxidase and decreased hyaluronidase activity effectively in vitro. YJCGT also increased cell viability, and proliferation in C34 and in E47 cell lines, and increased activities of superoxide dismutase, and catalase in C34 and in E47 cell lines. YJCGT reduced serum AST, LDH, and total cholesterol level in some of the results, and reduced blood alcohol concentration in vivo, as well. Conclusions : This study suggests that YJCGT has protective effects on oxidative stress by inhibiting alcohol-induced suppression of antioxidant enzyme activities.

Comparative Study of White and Steamed Black Panax ginseng, P. quinquefolium, and P. notoginseng on Cholinesterase Inhibitory and Antioxidative Activity

  • This study evaluated the anti-cholinesterases (ChEs) and antioxidant activities of white ginseng (WG) and black ginseng (BG) roots of Panax ginseng (PG), P. quinquefolium (PQ), and P. notoginseng (PN). Ginsenosides $Rg_1$, Re, Rf, $Rb_1$, Rc, $Rb_2$, and Rd were found in white PG, whereas Rf was not found in white PQ and Rf, Rc, and $Rb_2$ were not detected in white PN. The major ginsenoside content in steamed BG including $RK_3$, $Rh_4$, and 20(S)/(R)-$Rg_3$ was equivalent to approximately 70% of the total ginsenoside content. The WG and BG inhibited acetylcholinesteras (AChE) and butyrylcholinesterase (BChE) in a dose dependent manner. The efficacy of BG roots of PG, PQ, and PN on AChE and BChE inhibition was greater than that of the respective WG roots. The total phenolic contents and 2, 2-diphenyl-1-picryl-hydrazyl (DPPH) scavenging activity were increased by heat treatment. Among the three WG and BG, white PG and steamed black PQ have significantly higher contents of phenolic compounds. The best results for the DPPH scavenging activity were obtained with the WG and BG from PG. These results demonstrate that the steamed BG roots of the three studied ginseng species have both high ChEs inhibition capacity and antioxidant activity.

Cotoneaster horizontalis Decne 추출물의 항산화 및 항염증 활성 (Anti-Oxidative and Anti-Inflammatory Activities of Cotoneaster horizontalis Decne Extract)

  • Cotoneaster horizontalis Decne 에탄올 추출물(CHEE)의 항산화능과 항염증 생리활성을 분석하였다. CHEE의 항산화능을 DPPH radical scavenging activity로 분석한 결과 radical 소거능의 정도가 양성 대조군으로 사용한 ascorbic acid와 유사한 정도의 높은 활성을 보여 매우 강한 항산화능을 보유함을 확인하였다. 또한 RAW 264.7 세포주를 이용하여 H2O2 유도에 의해 생성된 ROS에 대한 소거능을 분석한 결과에서도 강한 소거능을 보였다. 뿐만 아니라 항산화효소 HO-1 및 그 전사 인자인 Nrf2의 단백질 발현이 CHEE의 처리에 의해 증가되었다. 한편 CHEE가 LPS에 의해 유도된 NO 생성에 미치는 영향을 분석한 결과 농도의존적인 NO 생성 저해능을 보였으며 이는 NO 생성 단백질인 iNOS의 발현 저해에서 기인함을 확인하였다. 이러한 결과를 통해 C. horizontalis의 항산화능과 항염증 활성을 세포 수준에서 처음으로 확인하였으며 향후 기능성 소재로서 유용하게 활용될 수 있을 것으로 판단된다.

Natural Antioxidants to Improve Stability of Refined Anchovy Oil against Oxidation

  • The oxidation stability of fish oil containing omega-3 polyunsaturated fatty acids (PUFAs), which is very susceptible to oxidative deterioration, needs to be improved before it can be successfully applied to functional foods. The antioxidant activities of 17 species of materials in anchovy oil (AO) were compared and a potent antioxidant was determined to improve the shelf-life of refined AO. Antioxidant activities of the 0.05% (w/w) materials in AO were compared against control during storage at $30^{\circ}C$ for 10 days. While no antioxidant effect was shown in alpha tocopherol against control, 3 species of grapefruit seed extracts (GSEs), astaxanthin (AX), soybean lecithin, and green tea extract showed good antioxidant activities. Especially, GSE B, GSE C, and AX showed significantly high peroxide inhibitory activities (PIAs) of $16.2{\pm}2.1$, $20.{\pm}3.5$, and $17.7{\pm}3.5%$, respectively, after the 4th day (p<0.01). Radical scavenging activities (RSAs) of GSE B, GSE C, and AX were $85.1{\pm}0.8$, $95.3{\pm}0.3$, and $85.9{\pm}0.8%$, respectively. Correlation between PIAs and RSAs was high ($R^2=0.926$) in GSE B, GSE C, and AX. Therefore, we concluded that one of the main antioxidative mechanisms of GSEs and AX must operate through an RSA pathway. The $RC_{50}$ (concentration required for 50% reduction of 1,1-diphenyl-2-picryl-hydrazyl, DPPH) of GSE C was $258\;{\mu}g/mL$.

복분자 에탄올 추출물의 항산화 및 항용혈 효능 (Antioxidant and Antihemolytic Activity of Ethanol Extracts of Rubus coreanus Miquel)

  • 천연에서 유래한 항산화 및 항용혈 소재개발을 위해 복분자 에탄올 추출물의 생리활성 효능 평가를 실시하였다. 복분자를 에탄올 추출한 후, 획득한 추출물을 섬유아세포주인 HS68에 처리하여 DPPH 라디칼 소거능과 SOD 유사 항산화 효능을 평가하였다. 또한 Sprague Dawley 렛트 적혈구 세포에 과산화수소로 산화를 유도한 후 추출물의 항용혈 효능을 살펴보았다. 그 결과 추출물은 농도 의존적으로 DPPH 라디칼 소거능과 SOD 유사 활성이 증가하는 것으로 나타났다. 렛트 적혈구를 이용한 항용혈 평가에서는 과산화수소를 단독으로 처리한 양성대조군보다 추출물을 처리한 군에서 항용혈율이 농도 의존적으로 유의하게 증가하였다. 이들 결과는 복분자 추출물이 항산화효능과 항용혈 약용 식물 소재로서의 가능성을 내는 것으로 사료된다.

사매 에탄올 추출물의 항산화 및 항염증 효과 (Antioxidative and Anti-inflammatory Effect of Ethanol Extract from Duchesnea chrysantha)

  • Oxidative stress has been implicated in cutaneous damage in various inflammatory skin diseases, including atopic dermatitis. The present study was undertaken to investigate the antioxidative and anti-inflammatory activities of the extract of Duchesnea chrysantha (DCE). DEC was prepared by extracting with 80% ethanol. Total flavonoids and polyphenols were measured by a colorimetric assay. The free radical scavenging activity of the extract was analyzed by the DPPH (1,1-diphenyl-2-picryl hydrazyl), ABTS (2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) and Griess reagent assay. An oxidative product of nitric oxide (NO), was measured in the culture medium by the Griess reaction. The level of prostaglandin $E_2$ ($PGE_2$) was measured by enzyme-linked immunosorbent assay. The expressions of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) were measured by Western blot analysis. Total flavonoid and polyphenol contents of DCE were included $24.73{\pm}0.45$ and $178.77{\pm}2.65$, respectively. DCE significantly increased electron donating ability (DPPH), nitrite scavenging (NO) and ABTS reducing activity in dose dependant. We investigated the anti-inflammatory effects of DCE on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. DCE significantly suppressed NO and prdstaglandin $E_2$ ($PGE_2$) in dose dependant. Furthermore, the levels of iNOS and COX-2 protein expressions were markedly suppressed by the treatment with DCE in a dose dependent manner. These results suggest that DEC may has value as natural product with its high quality functional components, antioxidative and anti-inflammatory activities.
