Anti-oxidative and Anti-inflammatory Activities of Polysaccharide isolated from Korean-Style Soy Sauce

  • Soy sauce is one of the representatives of traditional fermented foods in Korea. However, studies on soy sauce are relatively insufficient in Korea compared to Japan. In this study, antioxidant and anti-inflammatory activities of polysaccharides were measured by polysaccharides isolated from two different soy sauces, Korean and Japanese (KSS-0 and JSS-0). KSS-0 was purified into two fractions using gel chromatography and named them as KSS-I and KSS-II. To investigate the antioxidant activity of the polysaccharides, we measured the polyphenol content and radical scavenging activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS). To investigate the anti-inflammatory activity of polysaccharides, we used RAW 264.7 macrophage cells and induced inflammation using lipopolysaccharide (LPS). Then, we measured levels of inflammatory mediators such as nitric oxide (NO) and tumor necrosis factor (TNF)-α. Among the four polysaccharides, KSS-II showed the highest antioxidant activity and had good anti-inflammatory activity; KSS-II decreased inflammatory mediators in a dose-dependent manner. In conclusion, the polysaccharide isolated from Korean soy sauce (KSS-II) showed better anti-oxidant and anti-inflammatory activities than polysaccharides isolated from Japanese soy sauce, and may be useful as substances for functional foods.

청각(Codium fragile)을 첨가하여 제조한 막걸리의 발효 중 품질특성 및 항산화 효과 (Quality and Antioxidant Effects of the Korean Traditional Rice Wine Makgeolli Supplemented with Codium fragile during Fermentation)

  • This study investigated the physicochemical, microbiological, antioxidant, and sensory characteristics of Makgeolli supplemented with 10, 20, and 30% Codium fragile (Cf) during an 8-day fermentation period. The pH and acidity on day 0 were lower in Makgeolli supplemented with Cf (pH 3.55-3.65, acidity 0.26-0.29%) than in the unsupplemented Makgeolli (control) (pH 3.72, acidity 0.23%). The pH and acidity of Cf Makgeolli on day 8 were 3.08-3.12 and 0.67-0.75%, respectively. The sugar content increased steadily until day 4 (8.1-8.5 Brix) and then decreased steadily (6.5-6.8 Brix). The alcohol content increased steadily until day 8 (13.2-14.3%). The pH decreased, acidity increased, and alcohol content increased significantly (all P<0.05), with stepwise increases in fermentation time and Cf concentrations. The lactic acid bacteria counts were 9.28-9.37 log versus 8.18 log in the control. The DPPH (1,1-diphenyl-2-picrylhydrazyl) and ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)] radical scavenging activities were higher in Cf Makgeolli (DPPH 74.63-84.64%, ABTS 51.01-64.79%) than in the control (DPPH 30.97%, ABTS 21.21%). Makgeolli supplemented with 10 and 20% Cf had the highest taste, smell, and overall acceptance scores. This study suggests that Makgeolli supplemented with 10 and 20% Cf could be developed due to its good physicochemical and microbiological characteristics, including high antioxidant properties and good consumer acceptance.

식용 해조류 톳(Hizikia fusiformis) 유래 항산화 활성분획물의 산화적 손상이 유도된 인간피부섬유아세포 보호 효과 (Protective Effects of Antioxidant Active Fractions Derived from the Edible Seaweed Hizikia fusiformis in Oxidatively Stressed Human Dermal Fibroblasts)

  • We investigated the protective effects of antioxidant fractions from a 70% ethanolic extract of Hizikia fusiformis in human dermal fibroblasts (HDFs). Powdered H. fusiformis was extracted with 70% ethanol and then partitioned into three fractions according to polarity using n-hexane (HFH), chloroform (HFC), and ethyl acetate (HFEA). Antioxidant activity was observed in HFEA at 0.66 mg/mL based on the half maximal inhibitory concentration ($IC_{50}$) of 1,1-diphenyl-2-picrylhydrazyl (DPPH), and at 0.24 mg/mL based on alkyl radical scavenging. The protective effects of the HFEA antioxidant fraction against 2,2-azobis-(2-amidinopropane) dihydrochloride (AAPH)-damaged HDFs and the expression of Type I procollagen in HDFs were examined. HFEA caused the proliferation of HDFs with and without AAPH treatment and protected against AAPH damage to HDFs in a dose-dependent manner ($50-200{\mu}g/mL$). This implies that the antioxidant properties of the fractions depended on their proliferative and protective effects. The HFEA antioxidant fraction had significant effects and caused the dose-dependent expression of Type I procollagen, an important anti-wrinkle protein, in HDFs. In conclusion, antioxidant substances in H. fusiformis were found in the ethyl acetate fraction, and the resulting HFEA may have cosmetic applications.

반하사심탕 전탕액의 보관 온도 및 기간에 따른 안정성 및 유효성 연구 (Comparative study on stability and efficacy of Banhasasim-tang decoction depending on the preservation temperature and periods)

  • This study aimed to investigate the stability and biological activities of BHSST decoction depending on the preservation temperature and periods. Methods: BHSST decoction was preserved at room temperatures (R/T, $23{\pm}1^{\circ}C$) or refrigeration ($4^{\circ}C$) for 0, 30, 60 and 90 days. To evaluate the stability of BHSST decoction, pH and sugar content were estimated. In addition, high-performance liquid chromatography (HPLC) analysis was performed to determine marker compounds of BHSST decoction. To evaluate anti-inflammatory effect, nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) productions were measured in LPS-stimulated RAW 264.7 macrophages. Antioxidant activity was examined using the assays for 3-ethyl-benzothiazoline-6-sulfonic acid (ABTS) and 1-1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities. Results: There was no change in pH and sugar content depending on the preservation temperature and periods of BHSST decoction. Among the major components of BHSST, contents of liquiritin, baicalein and wogonin was reduced time-dependently both at R/T and $4^{\circ}C$. Inhibitory effects of BHSST decoction on NO and PGE2 productions were slightly decreased in a time-dependent manner by 90 days of preservation. In addition, BHSST decoction maintained ABTS and DPPH radical scavenging activities by 60 days while significantly reducing the activities in 90 days of preservation at R/T. By contrast, BHSST decoction had no significant change of ABTS and DPPH radical scavenging activities by 90 days at $4^{\circ}C$. Conclusions: Our results suggest that the stability and efficacy of BHSST decoction are maintained for 60 days at $4^{\circ}C$ rather than R/T.

굴피나무 추출물의 항산화, 항노화 효과 및 인체 시험 (Antioxidative and Antiaging Effects of Platycarya strobilacea Extract and Clinical Trial)

  • 본 연구에서는 굴피나무 수피 추출물의 항산화, 미백 및 주름생성억제에 관한 효과를 측정하였다. 추출물의 free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거활성($FSC_{50}$)은 50% 에탄올 추출물($6.75{\mu}g/mL$) < 당을 제거시킨 aglycone 분획(6.62) < ethyl acetate 분획(4.15) 순으로 증가하였다. Luminol-의존성 화학발광법을 이용한 $Fe^{3+}$-EDTA/$H_2O_2$ 계에서 생성된 활성산소종(reactive oxygen species, ROS)에 대한 총항산화능은 ethyl acetate 분획($OSC_{50}$, $0.56{\mu}g/mL$) < 50% 에탄올 추출물(0.02) < 당을 제거시킨 aglycone 분획(0.01) 순으로 당을 제거시킨 aglycone 분획에서 가장 큰 활성을 나타내었다. 굴피나무 추출물에 대하여 rose-bengal로 증감된 사람 적혈구의 광용혈에 대한 보호효과를 조사하였다. 50% 에탄올 추출물의 경우 농도 의존적($1{\sim}50{\mu}g/mL$)으로 광용혈을 억제하였다. 특히 ethyl acetate 분획은 $10{\mu}g/mL$ 농도에서 ${\tau}_{50}$이 717.27 min으로 매우 큰 세포보호 활성을 나타내었다. 미백 및 주름억제 효과측정으로는 각각 tyrosinase와 elastase의 활성 저해 효과를 측정하였다. Tyrosinase의 활성 저해 효과($IC_{50}$)는 50% 에탄올 추출물 ($243.98{\mu}g/mL$) < ethyl acetate 분획(153.87) < 당을 제거시킨 aglycone 분획(137.53) 순으로 나타났고, elastase의 활성 저해 효과($IC_{50}$)도 50% 에탄올 추출물($31.01{\mu}g/mL$) < ethyl acetate 분획(14.42) < 당을 제거시킨 aglycone 분획(1.48) 순으로, aglycone 분획이 매우 큰 활성 저해 활성을 나타내었다. 또한 굴피나무 추출물 중 ethyl acetate 분획을 함유하는 크림을 제조하고, 이를 피부에 도포한 후, 피부 수분 보유량, 경표피 수분 손실량 그리고 미백효과를 측정하였다. 추출물 함유 크림은 대조군(placebo)보다 수분 보유량을 $2{\sim}8%$ 증가시켰고, 경표피 수분 손실량은 감소시켰다. 또한 추출물 함유 크림은 대조군에 비해 멜라닌 생성을 9.55% 감소시켰다. 이상의 결과들은 굴피나무 추출물이 항산화 활성과 더불어 ROS에 대항하여 세포막을 보호함으로써 생체계, 특히 태양 자외선에 노출된 피부에서 항산화제로서 작용할 수 있음을 가리키며, tyrosinase 및 elastase의 활성 저해 효과와 이를 통한 인체 시험의 결과로부터 미백 기능성 화장품 원료로서 응용 가능성이 있음을 시사한다.

초음파 추출법에 의한 감태 추출물의 페놀성 화합물 함량, DPPH 라디칼 소거 활성 및 tyrosinase 저해 활성 분석 (Phenolic Content, DPPH Radical Scavenging, and Tyrosinase Inhibitory Activities of Ecklonia cava Extracted with the Ultrasonic Wave Method)

  • 해조류 유래 유용성분 추출을 위한 초음파 추출법의 적용성을 검토하였다. 동결건조한 감태 분말로부터 열수($65^{\circ}C$), 물($24^{\circ}C$), 50% 에탄올 및 100% 에탄올 뿐만 아니라 초음파 추출을 수행하여, 추출물을 제조하였다. 추출물의 페놀성 화합물 함량, DPPH 라디칼 소거 활성 및 tyrosinase 저해 활성을 분석하고, 초음파 추출과 전통적인 추출법의 차이를 비교하였다. 초음파 추출에 의한 감태 추출물의 페놀성 화합물의 함량(142.80 mg/g)은 열수 추출물의 페놀성 화합물 함량(10.03 mg/g)의 약 14배 이상 높았다. 열수 추출물에 비해 초음파 추출물의 DPPH 라디칼 소거 활성은 약 4배, tyrosinase 저해 활성은 약 14배 이상 높은 것으로 나타났다. 특히 초음파 추출법에 의한 추출물에서의 페놀성 화합물 함량과 DPPH 라디칼 소거활성 및 페놀성 화합물 함량과 tyrosinase 저해 활성간의 상관관계가 매우 높은 것(각각 $R^2$=99.47 및 99.99)으로 나타났다. 이상의 결과는 전통적인 용매 추출법보다 초음파 추출법이 감태의 유용성분 추출에 더 적합함을 의미한다.

택사탕의 항산화와 혈중지질에 대한 효과 (The Effects of Taeksa-tang on Blood Lipid Profile and Anti Oxidation)

  • Objectives We evaluated the improving effects of Taeksa-tang (TST) using 3T3-L1 cells and C57BL/6 mice were fed on a high-fat diet. Methods The anti-radical activities of TST were studied using 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid). The content of total polyphenol was measured using Folin-Ciocalteu reagent, whereas aluminum chloride colorimetric method was used for the content of total flavonoid. Moreover, the factors related to lipid profile and the protein expressions such as 𝛽-oxidation and anti-oxidant enzyme were analyzed using serum and western blotting of 3T3-L1 cells. Additionally, we examined lipolysis through glycerol appearance in mouse adipose tissue. Results TST treatment showed strong free radical scavenging activities with half maximal inhibitory concentration and the presence of a amount of total polyphenol and total flavonoid. TST treatment significantly increased factors related to 𝛽-oxidation such as carnitine palmitoyl transferase-1 and uncoupling protein 2 via the phosphorlyation of liver kinase B1 (LKB1) and AMP-activated protein kinase (AMPK). Moreover, the protein expressions of anti-oxidant enzyme and lipolysis were significantly elevated by TST administration. In addition, TST supplementation lowered serum malondialdehyde, triglyceride, and total cholesterol levels compared with the control group. Taken together, these data suggest that TST treatment regulated lipid parameters via the increase of 𝛽-oxidation by LKB1-AMPK signaling pathway. Conclusions TST may have a potential remedy in the prevention and treatment of obesity. Therefore, this study may provide the scientific basis for TST use.

당나귀 사골과 껍질의 항산화기능 및 SK-N-SH세포 보호효과 (Anti-oxidative Activity and the Protective Effect of Donkey's Bone and Skin Extracts on SK-N-SH Cells)

  • 본 실험에서는 당나귀 사골과 껍질을 추출하여 항산화 활성 및 산화적 스트레스로부터 유도되는 신경세포 보호효과를 구명하였다. DBSE의 항산화 활성은 처리 농도가 증가할수록 유의적으로 증가함을 나타내었는데(p<0.05), 40 mg/ml 일 때, 95.43%의 DPPH 라디칼 소거능을 나타내었고, 20 mg/ml일 때, 88.73%의 ABTS 라디칼 소거능을 나타내었으며, 10 mg/ml일 때, $132.53{\mu}M$ TE의 ORAC을 나타내었다(p<0.05). 신경모세포종 SK-N-SH 세포에 DBSE을 처리한 결과 세포 독성은 나타내지 않았으며, 신경세포에 활성산소종인 $H_2O_2$를 처리하여 DBSE의 산화적 스트레스 보호효과를 확인 한 결과, 처리농도 의존적으로 신경세포 보호효과를 나타내었다. 따라서 본 연구결과 당나귀 사골과 껍질 추출물은 산화를 억제하고 SK-N-SH 신경세포 보호효과를 보여 생리활성 기능소재로서 이용가능성이 있는 것으로 판단되며 추후 동물실험을 통한 안전성 및 생체대사조절기능과 효과증진기술에 대한 연구가 추가되어야 할 것으로 판단된다.

Quercetin의 급여가 우육의 항산화 효과 및 품질에 미치는 영향 (Effect of dietary supplementation of quercetin on antioxidant activity and meat quality of beef cattle)

  • This study was carried out to investigate the effects of dietary supplementation of quercetin ($Kocetin^{TM}$, QR) on antioxidative activity and meat quality of beef cattle (Holstein-Friesian). Beef cattle were divided into 3 groups; dietary supplementation of QR at 21 (n=4) and 42 ppm (n=3), and non-supplemented control (n=4). The QR comprised of 10% of quercetin. After slaughtering the beef cattle, loins were obtained and analyzed. Dietary supplementation of QR at 42 ppm showed significantly higher final pH of loin but did not affect the water holding capacity, drip loss, cooking loss, surface color, total phenolics content, 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity. Dietary QR showed no difference in both 2-thiobarbituric acid reactive substances and volatile basic nitrogen values. Textural characteristic results also showed no difference, except for cohesiveness. Cohesiveness was significantly higher in loin from beef cattle treated by dietary QR at 42 ppm when compared to control. Results suggest that dietary QR, which has only 10% of quercetin is not sufficient to have positive biochemical effects on beef meat quality.

Antioxidant Activity of Royal Jelly Hydrolysates Obtained by Enzymatic Treatment

  • Recently, research on the processing of raw functional materials with the aim of improving various physiological activities has been conducted. In this study, we investigated the antioxidant activity of royal jelly (RJ) hydrolysates obtained from three commercial proteases. Enzyme-treated royal jelly (ERJ), in which the RJ hydrolysates were converted into easy-to-absorb shorter chain monomers through the removal of two known allergen proteins, showed no difference in the content of (E)-10-hydroxydec-2-enoicacid (10-HDA) or the freshness parameter and showed a significant increase in total free amino acid content. The antioxidant activity of ERJ was determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and chemical assays. The ERJ showed about 80% DPPH-radical scavenging activity at same concentration of ascorbic acid. The antioxidant effect of ERJ was confirmed to be due to reduction of intracellular reactive oxidative species (ROS) and nitric oxide (NO) production in LPS-treated macrophages. Moreover, ERJ significantly increased the activity of the antioxidant enzyme superoxide dismutase (SOD) and the level of the antioxidant glutathione (GSH) in a dose-dependent manner. Interestingly, these antioxidant activities of ERJ were stronger than those of non-treated RJ. These findings indicate that ERJ has high potential as an antioxidant agent for use in human and animal diets.
