Background: Previously, studies have observed that the leaf extract of Cotoneaster wilsonii Nakai has potent antioxidant and anti-inflammatory activites. Therefore, further research was conducted to separate the active antioxidant and anti-inflammatory compounds in the leaf of the C. wilsonii. Methods and Results: The anti-oxidant effects were evaluated by analyzing the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, estimating the totla phenolic content (TPC) and total flavonoid content (TFC), and using reducing power assay method. The ethyl acetate (EtOAc) fraction demonstrated the highest DPPH radical scavenging activity (IC50; 9.69 ㎍/㎖), and the highest TPC and TFC (345.98 mg·GAE/g, and 74.23 mg·QE/g). Moreover, it indicated the highest effects on nitric oxide production. Therefore, the active compound was separated using the EtOAc fraction, determined on the basis of spectral data, and identified as (+)-catechin. Conclusions: The first compound separated was made from the leaf of C. wilsonii was (+)-catechin, which produced potent anti-oxidant and anti-inflammatory effects.
To use mountain ginseng adventitious roots (MGAR), which is superior in functionality compared to ginseng, and to mass produce raw materials as a functional cosmetic material, the antioxidant and cosmeceutical activities of the extracts of MGAR at different ethanol concentrations (0, 20, 40, 60 and 80%) were compared and analyzed. At first, different ethanol concentrations were compared to determine the best solvent for the extraction of phenolic compounds from MGAR. An 80% ethanol extract with 10.07 mg of gallic acid equivalents/g sample and 0.94 mg of quercetin equivalents/g sample was the best solvent in the extraction of phenolic compounds. However, the 60% ethanol extract had the highest antioxidant activity, evident by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, Trolox equivalent antioxidant capacity (35.01 mM Trolox/g sample) and ferric reducing antioxidant power (460.94 mM $FeSO_4/g$ sample). However, the inhibitory activities of the 80% ethanol extract from the MGAR against tyrosinase and elastase were higher than those of the other extracts. The results of this study show that the extract of MGAR can be used as an easily accessible source of natural antioxidant and as a possible cosmetic ingredient in the cosmeceutical industry. However, the components responsible for the antioxidant and cosmeceutical activities of the MGAR extracts are currently unclear. Therefore, it is suggested that further studies should be carried out to isolate and identify the antioxidant and cosmeceutical components in MGAR.

The physiological activities of cultivated Korean blue mussel (Mytilus edulis) and New Zealand green-lipped mussel (Perna canaliculus) were analyzed and compared. Both hot water extracts of blue mussel flesh (BMF) and green-lipped mussel flesh (GMF) showed increased activities of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). BMF showed increased ADH and slightly decreased ALDH activities compared to GMF. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activity of BMF was higher than that of GMF at the same concentration. BMF and GMF showed similar inhibitory activity against angiotensin converting enzyme at a concentration of 30 mg/mL. These results suggest that cultivated Korean blue mussel has similar physiological activity with New Zealand green-lipped mussel.
The objective of this study was to evaluate the pre-heating treatment effects on the antioxidant properties of ethanolic garlic and onion extracts. Garlic and onion with or without heating ($100^{\circ}C$, 30 min) were extracted with ethanol, and the total phenolic content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability, iron chelating ability, reducing power, and antioxidant activity in a linoleic acid emulsion system were evaluated. Garlic (41%) had a higher drying yield than onion (11%). Regardless of pre-heating, ethanol extracts of onion resulted in an approximately 25-fold higher yield than those of garlic. Thermal treatment before extraction decreased the levels of ethanol-soluble phenolics for both garlic and onion. Regardless of pre-heating, the radical scavenging abilities of ethanol extracts from garlic were greater than the ethanol extracts from onion. The iron chelating abilities of ethanol extracts from fresh and heated garlic were 85 and 81% at 10 mg/mL, respectively, whereas those of onion extracts were 10 and 9% at the same concentration, respectively. However, no differences in reducing power between garlic and onion extracts were observed. Both garlic and onion inhibited the formation of hydroperoxide in linoleic acid emulsion systems when ethanol was used as a solvent. Overall, garlic extracts had greater antioxidant activity than onion extracts, and the antioxidant activity of garlic and onion extracts were not significantly affected by thermal treatment.
Objectives: The objective of this study was to investigate the antioxidant effects of manufactured Wen-pi-tang-Hab-Wu-ling-san (WHW$^{(R)}$) in vitro. Methods: WHW$^{(R)}$ was prepared by the pilot manufacture of WHW water extract from a GMP system appointed company. Antioxidative activities were determined by in vitro tests as follows: the scavenging activities of oxygen free radicals including 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, superoxide anion, hydrogen peroxide and nitric oxide radicals, as well as ferrous ion chelating capacity and Trolox equivalent antioxidant capacity (TEAC). Results: WHW$^{(R)}$ significantly scavenged oxygen free radicals such as DPPH (IC$_{50}$=115.28 $\pm$ 0.25 $\mu$g/$m\ell$), superoxide anion (IC$_{50}$=8.56 $\pm$ 0.08 $\mu$g/$m\ell$), hydrogen peroxide (IC$_{50}$=240.36 $\pm$ 3.41 $\mu$g/$m\ell$) and nitric oxide (IC$_{50}$=162.28 $\pm$ 0.21 $\mu$g/$m\ell$) radicals. WHW$^{(R)}$ also showed ferrous ion chelating activity (IC$_{50}$=543.19 $\pm$ 4.85 $\mu$g/$m\ell$) and Trolox equivalent effects (IC$_{50}$=45.311 $\mu$g/$m\ell$) in TEAC and ORAC assay, respectively. Conclusion: This study demonstrates that WHW$^{(R)}$ has strong antioxidative properties through free radical scavenging activity. These data suggest that WHW$^{(R)}$ be used as an antioxidant agent.

Antioxidative activities of catechin from green tea extracts were examined by the methods of 1,1-Diphenyl-2-picrylhydrazyl (DPPH) electron donating ability, hydroxy scavenging activity and the inhibitory effect on xanthine oxidase activity. The resulted demonstrated the fact that Catechin one (containing Green tea extracts plus catechin 51%) of green tea extracts product showed at 65.5% in electron donating activity on the DPPH. The electron donating ability on the DPPH of Catechin one was increased to 10% than purified catechin. Catechin one showed the activity at 64.5% in scavenging activity using hydroxy radical method. To the Catechin one provided to increase the hydroxy scavenging activity up to 3 fold. Inhibitory effects of the catechin one measured with xanthine oxidase method was 6.5%. Although the antioxidative activity of catechin (98% purified) was lower than that of Catechin one (containing Green tea extracts plus catechin 51%) in same catechin concentrations ($5{\mu}g$, respectively). Therefore, we may suggest that Catechin one can be used as a functional food additive possessing the potent antioxidative activity.

The seed of Vigna angularis (Ohwi) Ohwi & Ohashi (= Phaseolus angularis W. F. Wight, Leguminosae) is one of well-known folk foodstuffs in Korea, China and Japan. In the course of screening for antioxidants from natural plants in Korea by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) a methanol extract of the seeds of V. angularis were found to show a potent antioxidant activity. And the ethyl acetate soluble fraction of V. angularis (VAEA) showed the most potent DPPH radical scavenging and superoxide quenching activities. To reveal the effect of antioxidant activities of VAEA, we tested the thermal and oxidative stress tolerances, activities of SOD and catalase, and intracellular ROS level in Caenorhabditis elegans. Consequently, VAEA-fed worms lived longer than control worms under the thermal and oxidative stress conditions. And VAEA elevated SOD and catalase activities of worms, and reduced intracellular ROS accumulation in a dose-dependent manner.

Background: The Ministry of Environment, Korea recognizes Cotoneaster wilsonii Nakai as a second-class endangered plant. It is a native species that grows in Ulleung-do, Korea. To our knowledge, the bioactivity of this plant has not yet been reported. Therefore, in this study, we have reported the bioactivity of C. wilsonii Nakai. Methods and Results: The anti-oxidant activities of C. wilsonii methanolic extract were investigated in vitro. The anti-oxidant activity was evaluated using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical assay, and the total phenolic and flavonoid content were measured. The leaf methanolic extract had the highest DPPH radical scavenging activity ($IC_{50}$; $15.74{\mu}g/m{\ell}$), and it also had the highest total phenolic and flavonoid content ($220.95mg{\cdot}GAE/g$, and $36.46mg{\cdot}QE/g$ extract respectively). Through high performance liquid chromatography (HPLC) analysis, chlorogenic acid was found to be the predominant among all phenolic compounds, showing a concentration of 84.24 mg/g extract. More than 60% decrease in nitric oxide (NO) production was found in the leaf methanolic extract. Conclusions: To the best of our knowldege, this is the first report of the bioactivities of C. wilsonii. The results demonstrate that the leaf methanolic extract of C. wilsonii shows potent anti-oxidant, and anti-inflammatory activities.

This work was carried out to investigate the antioxidative effects of Acanthopnax sessiliflorus from Jeongseon County for the purpose of development of a novel antioxidant from natural products. The antioxidant activity was determined by using electron paramagnetic resonance (EPR), not measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) which have bun used for antioxidant activity of natural sources. Although DPPH radical scavenging activity assay have been generally used for antioxidant activity, this assay is nut appropriated for determinating which radical is scavenged by extracts from natural products. Using EPR, we determinated whether A. sessiliflorus extracts from Jeongseon County scavenge specific radicals or not. On experiment of scavenging superoxide anion radical, hydroxyl radical, nitrogen dioxide and peroxinitrite. Extracts from A. sessiliflorus showed high antioxidant activities to reactive oxygen and nitrogen species. These result suggest that extracts from A. sessiliflorus act as an antioxidant by scavenging reactive oxygen and nitrogen species and used as new cosmetic ingredients for anti-oxidative stress in skin.

In this study, the antioxidative effects, inhibitory effects on elastase, and components of Cayratia japonica extracts were investigated. The free radical(1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities($FSC_{50}$) of extract/fractions of Cayratia japonica were in the order: 50% ethanol extract(114.3 ${\mu}g/mL$)${\mu}g/mL$)${\mu}g/mL$). Reactive oxygen species(ROS) scavenging activities($OSC_{50}$) of some Cayratia japonica extracts in $Fe^{3+}-EDTA/H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The order of ROS scavenging activities were deglycosylated flavonoid aglycone fraction($OSC_{50},\;3.30{\mu}g/mL$)<50% ethanol extract(1.21 ${\mu}g/mL$)${\mu}g/mL$). Ethyl acetate fraction showed the most prominent scavenging activity. The protective effects of extract/fractions of Cayratia japonica on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The Cayratia japonica extracts suppressed photohemolysis in a concentration dependent manner($1{\sim}25{\mu}g/mL$), particularly deglycosylated flavonoid aglycone fraction exhibited the most prominent celluar protective effect(${\tau}_{50}$, 175.05min at 25 ${\mu}g/mL$). Aglycone fractions obtained from the deglycosylation reaction of ethyl acetate fraction among the Cayratia japonica extracts, showed 2 bands in TLC and 2 peaks in HPLC experiments(360 nm). Two components were identified as luteolin(composition ratio, 47.50%), apigenin(52.50). TLC chromatogram of ethyl acetate fraction of Cayratia japonica extract revealed 3 bands and HPLC chromatogram showed 4 peaks, which were identified as luteolin-7-O-${\beta}$-D-glucopyranoside(composition ratio, 11.14%), apigenin-7-O-${\beta}$-D-glucuronopyranoside(15.38%), luteolin(23.55%) and apigenin(49.92%) in the order of elution time. The inhibitory effect of aglycone fraction on elastase($IC_{50},\;70.5{\mu}g/mL$) was very high. These results indicate that extract/fractions of Cayratia japonica can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. And component analysis of Cayratia japonica extract and antioxidative effects could be applicable to new cosmetics.
