In order to find new functional materials for the cosmetics application, we investigated the anti-oxidant and anti-melanogenic properties of Gentiana scabra extracts (GSE), which were prepared by the various extraction methods. Results showed that GSE had high DPPH radical scavenging activity in a dose-dependent manner. Also, GSE inhibited the prodution of melanin in B16F10 melanoma cell as well as tyrosinase activity. We also found that GSE inhibited mRNA expression of tyrosinase-related protein (TRP)-1 and 2. In conclusion we suggest that GSE is applicable to cosmetics as a potential ingredient for their anti-oxidant and whitening effects.
18 flowers of Compositae family were collected and extracted in aqueous methanol (MeOH). The concentrated extract was partitioned into n-hexane, ethyl acetate (EtOAc), n-BuOH, and water fractions. The extract and fractions were evaluated for total phenolics, total flavonoids, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and tyrosinase inhibition activity. n-Hexane and EtOAc fractions of Aster yomena, n-hexane fraction of Cosmos bipinnatus White, n-hexane and EtOAc fractions of C. bipinnatus Pink showed high total phenolics. And EtOAc fractions of A. yomena, C. bipinnatus White, C. bipinnatus Red, C. morifolium Froggy, and C. morifolium Himaya exhibited high total flavonoids. EtOAc fractions of A. yomena, C. bipinnatus White, C. bipinnatus Pink, C. morifolium Yellowmable, and MeOH extract of C. morifolium Rosa significantly scavenged DPPH radical. EtOAc fractions of C. chinensis, C. bipinnatus White, C. bipinnatus Red, C. morifolium Himaya, and C. morifolium Hongsim highly inhibited the tyrosinase activity. A. yomena, C. bipinnatus White, C. bipinnatus Pink, C. bipinnatus Red and C. morifolium Himaya are evaluated as good source for whitening cosmetics materials.

Seaweeds have a number of secondary metabolites, such as polyphenols, polysaccharides, and carotenoids, and have received much attention as a source of natural antioxidants. Thus, this study was carried out to examine the antioxidant activities from ethanol (EE) and water (WE) extracts of Zostera marina. Their antioxidant effects were investigated using total polyphenol contents (TPC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, reducing power, and chelating effect. TPC of EE and WE was 2.12 mg/g and 3.88 mg/g, respectively. DPPH radical scavenging activities of EE and WE were increased in a dose-dependent manner. In particular, EE had DPPH radical scavenging activity of 93% at a concentration of 0.5 mg/ml, and was higher than that of WE (71%). EE and WE increased reducing power in a concentration-dependent manner, but their effects were lower than that of the control (ascorbic acid). In case of chelating effect, WE was 66% at a concentration of 1 mg/ml, and was stronger than EE (6%). These results suggest that extracts of Zostera marina can be potentially used as proper natural antioxidants in the food industry.
In the present work, novel orange peel was extracted with 100%EtOH (ethanol) and fractionated into four fractions namely F1, F2, F3, F4 which were eluted from paper chromatographs using 100%EtOH, 80%EtOH, 50%EtOH and pure water respectively. The crude extract and its four fractions were evaluated for their total polyphenol content (TPC), total flavonoid content (TFC) and radical scavenging activity using DPPH (1,1-diphenyl-2-picrylhydrazyl) assay. Their cytotoxic activity using WST assay and DNA damage by agarose gel electrophoresis were also evaluated in a human leukemia HL-60 cell line. The findings revealed that F4 had the highest TPC followed by crude extract, F2, F3 and F1. However, the crude extract had the highest TFC followed by F4, F3, F2, and F1. Depending on the values of $EC_{50}$ and trolox equivalent antioxidant capacity, F4 possessed the strongest antioxidant activity while F1 and F2 displayed weak antioxidant activity. Further, incubation HL-60 cells with extract/fractions for 24h caused an inhibition of cell viability in a concentration-dependent manner. F3 and F4 exhibited a high antiproliferative activity with a narrow range of $IC_{50}$ values ($45.9-48.9{\mu}g/ml$). Crude extract exhibited the weakest antiproliferative activity with an $IC_{50}$ value of $314.89{\mu}g/ml$. Analysis of DNA fragmentation displayed DNA degradation in the form of a smear-type pattern upon agarose gel after incubation of HL-60 cells with F3 and F4 for 6 h. Overall, F3 and F4 appear to be good sources of phytochemicals with antioxidant and potential anticancer activities.

The attempts to develop natural antioxidants have been made with aerial part of 16 Compositae species by analyzing their phenolic compound contents, scavenging activities on 1,l-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) radicals, ferrous ion chelating effects and inhibition effects on peroxidation of linoleic acids. Total polyphenol and flavonoid contents were highest in Matricaria recutica. Scavenging activity on DPPH and ABTS radicals were highest with extracts of Echinacea angustifolia and Serratula coronata var. insularis f. insularis, respectively. Aerial palt extracts of all species showed lower DPPH scavenging activity than ascorbic acid and 2,6-Di-tert-butyl-4-methylphenol (BHT). But Serratula coronata var. insularis f. insularis demonstrated higher ABTS scavenging activity than ascorbic acid and BHT. In Hieracium pilosella, Echinacea angustifolia, Matricaria reculica extracts showed higher ABTS scavenging activity than BHT. Ferrous ion chelating effects was highest with Matricaria recutica extract, but the effects were much lower than ethylenediaminetetraacetic acid (EDTA). The inhibition activity in lipid peroxidation of linoleic acids was highest in Eupatorium japonicum with 90.06% inhibition 4 days after reaction and 40.52% after 24 days. This demonstrated higher inhibition activity and longer lasting than BHT. Aerial part of Matricaria recutica for extraction source, rather than flower, has higher potential for antiox.idant material. In conclusion, development of natural antioxidants in Compositae is possible by studying antioxidant activity of each species.
This study was conducted to investigate the effects of the hot water extract from buckwheat (WEB) in RAW-264.7 macrophage cells against lipopolysaccharide (LPS). In these experiments, we evaluated the anti-inflammatory effects of WEB by measuring MTT assay, nitric oxide (NO), inducible NOS (iNOS) production, and cyclooxygenase-2 (COX-2) expression by Western blotting. The extracts showed a protective effect by increasing cell viability on LPS in RAW264.7 cells. WEB (0.25, 0.5, and 1.0 mg/mL) significantly suppressed LPS-stimulated production of NO. Also, WEB reduced the expression of iNOS and COX-2 proteins. The present results show that WEB has potent anti-inflammatory effects on RAW264.7 cells. In addition, WEB has various antioxidant effects as a result of 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 2,2-Diphenyl-1-picrylhydrazyl (DPPH) which possess a radical scavenging activity. The total polyphenol and flavonoid contents of the WEB were $13.22{\pm}3.69mg\;GAE/g$ extract and $38.53{\pm}5.20mg\;CE/g$ extract respectively. The present results give the understanding of biological activities of buckwheat and encourage their application for supplements.

Drought is one of the key limiting factors that adversely affects the growth and productivity of crop plants. For the enhancement of drought tolerance in crop plants, the identification of basic mechanisms of a plant to drought stress is necessary. In this study, we compared physiological and biochemical responses of five local Arundenilla hirta ecotypes to drought stress. These ecotypes were previously collected from various parts of Korean peninsula, including Youngduk, Gunsan, Jangsoo, Jinju-1 and Yecheon. A. hirta plants were exposed to drought stress for 14 and 17 days respectively, followed by re-watering for 3 days. The results showed that the lipid peroxidation (MDA), hydrogen peroxide ($H_2O_2$), DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity, and proline level were significantly increased while the chlorophyll content was decreased by drought stress in A. hirta leaves. The highest proline content and DPPH scavenging activity were shown in Ecotype of Youngduk with least MDA and $H_2O_2$ levels while the highest MDA and $H_2O_2$ contents, and least proline and DPPH levels were shown in Gunsan, respectvely. These results indicate that the Youngduk is the most tolerant and Gunsan is the most sensitive ecotype among the five different collections. Together, these results provide a new insight of overall physiological responses of A. hirta to drought stress.

The purpose of this study was to investigate anti-inflammatory and antioxidant effects of essential oil from seed of Zanthoxylum schinifolium on cultured RAW 264.7 cell line. Antioxidant activity of essential oil was evaluated by two different assays as 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and super oxide dismutase (SOD) like activities. This essential oil was tested for cell viability on RAW 264.7 cell line by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay. The effects of anti-inflammatory on LPS-induced RAW 264.7 cell line was studied by the content of nitric oxide (NO) and prostagladin $E_2$ ($PGE_2$) in cells. The essential oil of seed from Zanthoxylum schinifolium obtained dose-dependently increased the scavenging activity on DPPH radical scavenging activity and SOD like activity. The essential oil showed low cytotoxicity as more than 98% cell viability in $40{\mu}g/mL^{-1}$ concentration. The essential oil of seed extracted from Zanthoxylum schinifolium presented obvious effect on inflammation. These results suggest that essential oil of seed from Zanthoxylum schinifolium may have value as the potential anti-inflammatory effects by decreasing the action of NO and $PGE_2$ and preventing the activation of oxidative.

In this study, the antioxidative effects, inhibitory effects on elastase, and components of Quercus glauca extracts were investigated. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity $(FSC_{50})$ of extract I fractions of Quercus glauca leaf was in the order: 50% ethanol extract $(12.45{\mu}g/mL)$ < ethyl acetate fraction $(10.47{\mu}g/mL)$ < deglycosylated flavonoid aglycone fraction $(8.57{\mu}g/mL)$. Reactive oxygen species (ROS) scavenging activities $(OSC_{50})$ of some Quercus glauca leaf extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The order of ROS scavenging activity was 50% ethanol extract $(OSC_{50},\;4.2{\mu}g/mL)$ < deglycosylated flavonoid aglycone fraction $(1.58{\mu}ug/mL)$ < ethyl acetate fraction $(0.66{\mu}g/mL)$. Ethyl acetate fraction showed the most prominent scavenging activity. The protective effects of extract / fractions of Quercus glauca leaf on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The Quercus glauca leaf extracts suppressed photohemolysis in a dose dependent manner, particularly deglycosylated flavonoid aglycone fraction exhibited the most prominent celluar protective effect $({\tau}_{50}$, 398.67 min at $50{\mu}g/mL$). Aglycone fractions obtained from the deglycosylation reaction of ethyl acetate fraction among the Quercus glauca leaf extracts, showed 2 bands in TLC and 2 peaks in HPLC experiments (360 nm) as well. Two components were identified as quercetin (55.77%), and kaempferol (44.23 %). TLC chromatogram of ethyl acetate fraction of Quercus glauca leaf extracts revealed 6 bands $(QG1{\sim}QG6)$, Among them, isoquercitrin (QG3), hyperin (QG4), and rutin (QG6) were identified. The inhibitory effect of aglycone fraction on tyrosinase $(IC_{50},\;73.5{\mu}g/mL)$ and elastase $(IC_{50},\;16.2{\mu}g/mL)$ was high. These results indicate that extract / fractions of Quercus glauca can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. And component analysis of Quercus glauca leaf extract and inhibitory activity on tyeisinase and elastase of the aglycone fraction could be applicable to new functional cosmetics.

Nano-emulsion with phase inversion temperature (PIT) emulsifying system was prepared to use rapeseed oil from originating Jeju in order to apply various cosmetic applications. Natural rape seed oil (NRSO) extraction was extracted using n-hexane as a solvent. NRSO extract showed a light yellowish color of viscous liquid as well as yield was $43{\pm}2.5%$. Acid value was $2.76{\pm}0.5$ and gravity was $0.89{\pm}0.05$. Droplet size of PIT-Yuche-NE with 20wt% of rapeseed oil was 50-120nm (average: $82{\pm}5.8nm$) and zeta potential was -29.5mV. It was thermodynamically good stable emulsion due to $(PEG)_{5-30}$fattyacidether. Some conclusions from the result of characteristic experiment were obtained as follows. First, the anti-oxidative activity was measured by free radical scavenging activity using DPPH (1,1-diphenyl-2-picrylhydrazyl radical). Anti-oxidative activity of PIT-Yuche-NE was $37.2{\pm}6.7%$ on 10mg/mL compared with PIT-Toco-NE (Natural tocopherol nano-emulsion, $28.8{\pm}6.5%$ on 10 mg/mL) and PIT-Nokcha-NE (Green tea extract nano-emulsion, $29.6{\pm}7.2%$ on 10mg/mL). Second, the collagen synthesis activity of PIT-Yuche-NE was $148{\pm}15.2%$ compared with PIT-Toco-NE (Natural tocopherol nano-emulsion, $121{\pm}13.5%$ on 10mg/mL) and PIT-Nokcha-NE (Green tea extract nano-emulsion, $95{\pm}12.7%$ on 10mg/mL). Third, the effectiveness of moisturizing activity of Yuche-CRM with Aramo-TS after 6 hours increase $47{\pm}3.9%$ (*p-value£0.05, n=7) whereas Both Toco-CRM was $30{\pm}5.2%$ (*p-value£0.05, n=7) and Nokcha-CRM was $35{\pm}4.5%$. Therefore, Yuche-CRM has higher moisturizing effect than other two creams. Finally, Nano-emulsion stabilizing rapeseed oil using PIT emulsifying system of this study can be used to apply cosmetics industry and pharmaceutical industry.
