Anti-inflammatory Effect of MeOH Extract of Cibotium barometz in IFN-$\gamma$ and LPS-stimulated Mouse Peritoneal Macrophage (IFN-$\gamma$와 LPS로 자극된 쥐의 복강 대식세포에서 구척 메탄올 추출물의 항염증 효과)

  • The rhizome of Cibotium barometz has been used for variety of bone disease as a traditional medicine. In the present study, we examined the antioxidant and anti-inflammatory activities of 85% methanol extract of C. barometz. C. barometz exhibited potent scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, superoxide radical and nitric oxide radical. In IFN-$\gamma$/LPS-stimulated mouse peritoneal macrophage model, C. barometz suppressed nitric oxide production and IL-6 secretion dose-dependently. Moreover, C. barometz showed decreased iNOS and COX-2 expression without notable cytotoxicity. These results suggest that C. barometz may be an useful agent as an antioxidant and anti-inflammatory agent.

Antioxidant Activities and Total Phenolics of Ethanol Extracts from Several Edible Mushrooms Produced in Korea

  • Eight edible mushrooms grown in Korea were extracted with ethanol at room temperature for 24 hr. The extracts were investigated for their antioxidant activities as measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical cation scavenging activities. Among the mushroom extracts evaluated in this study, the ethanolic extracts from Ganoderma lucidum and Pleurotus eryngii showed the greatest potential antioxidant activity, by producing 85 and 88% inhibition in DPPH radical scavenging method and 219 and 165 mg ascorbic acid equivalent antioxidant capacity (AEAC), respectively. Total phenolics and total flavonoids in the ethanolic extracts were determined by spectrophotometric method. Positive correlations were found between total phenolic contents in the extracts and their antioxidant activities, suggesting that phenolic contents in the mushrooms extracts are mainly responsible for their antioxidant activities.

Antioxidative Activity in Jeolpyun Containing Cnidium officinale M Extract (천궁 추출물 첨가 절편의 항산화활성)

  • The purpose of this study was to evaluate the antioxidative activities of jeolpyun containing Cnidium officinale M extract (2%, 4%, 6%, 8%) by total polyphenol contents, electron donating ability on 2,2-diphenyl-1-picrylhydrazyl (DPPH), scavenging ability of superoxide anion radical and decomposing ability of hydrogen peroxide. In chromaticity analysis, the brightness significantly decreased with increasing Cnidium officinale M extract content. Jeolpyun containing 8% Cnidium officinale M extract revealing the highest value for the redness and the yellowness, 1.07, 12.70, respectively. The total polyphenol contents of jeolpyun containing 8% Cnidium officinale M extract were the highest content of 4,213 ㎍ gallic acid equivalent (GAE)/mL. The total polyphenol contents revealed significant difference (p<0.05). Jeolpyun containing 8% Cnidium officinale M extract revealing the highest electron donating ability (83.55%). The electron donating abilities were significantly related at p<0.05. The scavenging abilities of superoxide anion radical for jeolpyun containing 4% Cnidium officinale M extract revealed the highest ability (0.01676). There was no significant difference. The hydrogen peroxide decomposing ability for jeolpyun containing 8% Cnidium officinale M extract revealed the most hydrogen peroxide decomposing ability (-0.193) and the hydrogen peroxide decomposing ability revealed a significant difference (p<0.05).

Antioxidant Constituents of the Aerial Parts of Curcuma longa (강황 지상부의 항산화 성분)

  • As part of ongoing study focused on the discovery of natural antioxidants from Korean plants by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging effect and superoxide quenching activity, ethanol extract of the aerial parts of Curcuma longa (Zingiberaceae) was found to show potent antioxidant activities. Subsequent activity-guided fractionation of the ethanol extract led to the isolation of two phenolic compounds, 1,2,3,4,6-penta-O-galloyl-${\beta}$-D-glucopyranoside (1) and gallic acid (2), as antioxidant compounds. Their structures were elucidated by spectroscopic studies. Compounds 1 and 2 were isolated for the first time from this plant. These compounds showed the significant antioxidative effects during the DPPH free radical scavenging test, and the riboflavin- and xanthine-originated superoxide quenching activity tests.

  • A new natural product, tetrastigmol A (1), and several known compounds as flavonoids (2 - 8), steroids (9 - 10), bergenin and its derivative (11 - 12), stilbens (13 - 15), lignan (16), benzenecarboxylic derivative (17) and two norisoprenoid (18 - 19) were isolated from the stem of Tetrastigma erubescens Planch. (Vitateae). Their structures were determined on the basis of NMR spectroscopic data. This is the first report on chemical constituents of this plant. Compounds 1, 6 - 8 and 12 - 15 showed strong antioxidant activity using two methods including DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical scavenging and lipid peroxidation inhibitory assays.

  • Phytochemical examination of Barks of Ulmus macrocarpa has led to the isolation and characterization of two fla-vanonol, taxifolin $7-O-{\beta}-D-glucopyranoside$ (1), taxifolin $3'-0-7-{\beta}-glucopyranoside$ (2), two flavanone eriodictyol $7-O-{\beta}-D-glucopyranoside$ (3), nalingenin $7-O-{\beta}-D-glucopyranoside$ (4), three flavan 3-ol, (+)-catechin (5), (-)-epicatechin (6), (+)-catechin 7-O-{\beta}-D-glucopyranoside (7) and one proanthocyanidin, procyanidin B-1 (8). Antioxidative activity of these compounds was determined by measuring the radical scavenging effect on 1,1-diphenyl-2- picrylhydrazyl (DPPH) radicals . (+)-Catechin (5) , (-)-epicatechin (6), (+)-catechin $7-O-{\beta}-D-glucopyranoside$ (7) and procyanidin B-1 (8) showed significant antioxidative activity.

A Study on the Biological Activities of Wild Mushroom Extracts from Jeju Island (제주 야생 버섯 추출물의 생리활성 연구)

  • In this study, we investigated the antioxidant, whitening and anti-inflammatory effects of 12 species of wild mushrooms in Jeju Island. Their anti-oxidative effects were measured by the free radical scavenging activity using 1,1-diphenyl-2-picrylhydrazyl (DPPH), and tyrosinase inhibitory activities were determined for the whitening effect. In addition, inhibitory activities of NO production for anti-inflammation were investigated. As a result, Elfvingia applanata extract and Fomitopsidaceae sp. extracts showed higher free radical scavenging activities ($FSC_{50}$; $74.8{\mu}g/mL$, $182.6{\mu}g/mL$, respectively) than other extracts. Elfvingia applanata extract ($IC_{50}$; $346.8{\mu}g/mL$) showed higher activity than the Arbutin ($IC_{50}$; $421.6{\mu}g/mL$) on tyrosinase inhibitory activity. Elfvingia applanata extract and Daedaleopsis styracina extract showed anti-inflammatory activity of 74.1% and 62.9% respectively, at the concentration of $100{\mu}g/mL$. Furthermore, the extracts inhibited NO production in a dose-dependent manner. In conclusion, we evaluated the biological activities of 12 species of wild mushrooms in Jeju Island, Elfvingia applanata, Fomitopsidaceae sp. and Daedaleopsis styracina could have the functional effects as a cosmetic raw material.

Studies on Antioxidant and Whitening Activities of Salix gracilistyla Extracts (갯버들 추출물의 항산화 및 미백활성 연구)

  • This study was carried out to evaluate the antioxidant and whitening activities of Salix gracilistyla extracts. The total polyphenol contents of the extracts were 142.60-151.95 mg GAE/g and total flavonoid contents were 83.43-92.60 mg CE/g. In 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, the 30% methanol extract showed the highest antioxidant activity ($IC_{50}$; $19.68{\mu}g/mL$). Tyrosinase inhibitory activity was similar in all four solvent extracts, and the highest inhibitory activity (35.18%) was obtained at a concentration of $200{\mu}g/mL$ of 30% ethanol extract. In addition, methanol extracts did not affect cell viability at all treatment concentrations and were found to significantly reduce the melanin content of B16F10 cells. As a result, it is considered that Salix gracilistyla extracts can be used as an effective cosmetic ingredient having antioxidant and whitening activity.

Assessment of Antioxidant Activity of Garlic (Allium sativum L.) Peels by Various Extraction Solvents

  • This study was carried out to investigate the antioxidant activities, total phenol, and flavonoid contents of 70% methanol, 70% ethanol, and chloroform-methanol (CM, 2:1, v/v) extracts from garlic (Allium sativum L.) peels. Antioxidant activity was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) radical and ABTS [2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, ${\beta}$-carotene bleaching assay, and ferric reducing antioxidant power. Total phenol and flavonoid were in a range of 32.59-63.05 mg CAE/g, 5.26-9.22 mg QE/g, respectively. Total phenol and flavonoid were predominant with the values of $63.05{\pm}0.20mgCAE/g$ and $9.22{\pm}0.17mgQE/g$ in 70% ethanol extract. Additionally, 70% ethanol extract also displayed the strongest antioxidant activity followed by 70% methanol extract and CM extract in all assays. These results showed that garlic peel can be used as natural antioxidant in nutraceutical preparations to prevent human diseases.

Investigation of Antioxidative Activity from Apple (Malus pumila Miller) Peel by Various Extraction Solvents

  • Consumption of fruits and vegetables has been conducted to be effective in the prevention of chronic diseases. In this study, 70% methanol, 70% ethanol and chloroform-methanol mixture (CM, 2:1, v/v) were used as solvents in the extraction of apple peels. The total phenol content, total flavonoid content and antioxidant activity of various extracts were investigated using in vitro assays. The extract obtained by 70% methanol showed the highest total phenol content ($20.87{\pm}0.17mgCAE/g$), ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity and ferric reducing antioxidant power. However, 70% ethanol extract possessed the strongest antioxidant activity assayed by DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity and ${\beta}$-carotene bleaching method. And CM extract was found to show the highest total flavonoid content with the value of $9.26{\pm}0.06mgQE/g$. These results indicated that apple peels can be used in dietary applications with a potential to reduce oxidative stress.
