Antioxidative Activities of Diarylheptanoids from Alnus japonica and Their Structural Relationship (오리나무에서 분리된 Diarylheptanoid의 항산화작용 및 구조상관활성)

  • Anitioxidative activities againt 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical were evaluated for ten diarylheptanoids, $1,7-bis-(3,4-dihydroxyphenyl)-heptane-3-O-{\beta}-D-glucopyranosyl(1{\rightarrow}3)-{\beta}-D-xylopyranoside(1)$, $1,7-bis-(3,4-dihydroxyphenyl)-heptane-3-O-{\beta}-D-apiofuranosyl(1{\rightarrow}6)-{\beta}-D-glucopyranoside(2)$, $1,7-bis-(3,4-dihydroxyphenyl)-heptane-5-O-{\beta}-D-glucopyranoside(3)$, 1,7-bis-(3,4-dihydroxyphenyl)-5-hydroxyheptane(4), $1,7-bis-(3,4-dihydroxyphenyl)-heptane-3-one-5-O-{\beta}-D-glucopyranoside(5)$, oregonin(6), hirsutanonol(7), hirsutenone(8), $1,7-bis-(3,4-dihydroxyphenyl)-5-hydroxyheptane-3-O-{\beta}-D-xylopyranoside(9)$ and platyphylloside(10), which were isolated from Alnus japonica. Aglycones (4,7 and 8) and mono glycosides (3,6 and 9) showed more strong antioxidative activities than diglycosides (1 and 2) against the DPPH radical. Especially, hirsutenone(8) showed superior activity among ten diarylheptanoid.

Screening of Natural Resources with Inhibitory Activity on Free Radicals and Advanced Glycation end Products (AGEs) Formation (천연자원의 라디칼 소거능과 최종당화산물의 생성저해활성 검색)

  • Reactive oxygen species (ROS) and formation of advanced glycation end products (AGEs) play an important role in the pathogenesis of diabetic nephropathy by hyperglycemia. To find natural agents improving diabetic nephropathy, 63 natural resources which used to the treatment of diabetes mellitus in a folk remedy were investigated with an in vitro system employing radical scavenging activity and inhibitory activity of AGEs formation. In results, the extracts of Aspalathus linearis, Rubus coreanus, Rosa rugosa, and Epimedium koreanum significantly inhibited the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical with $IC_{50}$ values less than $10{\mu}g/ml$. The extracts of Zea mays, Cucurbita moschata, Cudrania tricuspidata, and Aspalathus linearis effectively reduced the formation of AGEs compared with the positive control $N-acetyl-_L-cystenine$ (NAC) and aminoguanidine (AG). In addition, the extracts of Aspalathus linearis, Commelina communis, Cornus officinalis, and Lespodeza cuneata showed the all inhibitory activity against DPPH radical and AGEs formation. Also, these resources definitely showed the radical scavenging activity against peroxynitrite $(ONOO^-)$ and hydroxyl radical $({\cdot}OH)$ relating to high glucose-induced ROS production. Thus, these results suggest that some natural resources may regulate the initiation and progression of diabetic nephropathy through inhibition of ROS production and AGEs formation.

Antioxidant Activity of Black Panax ginseng (흑삼의 항산화 활성)

  • This study was performed to investigate the antioxidant activity of extracts of black Panax ginseng (BGE) and its crude saponin (BGECS). The antioxidant activities of BGE and BGECS were evaluated for free radical scavenging activity against stable free radical (1,1-diphenyl-2-picrylhydrazyl) DPPH, nitrite, hydrogen peroxide and superoxide. In addition, the antioxidant activity of BGE and BGECS against peroxyl radicals, hydroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay. As a result, BGE and BGECS were found to have a strong inhibitory activity with >90% against the DPPH radical at $1000{\mu}g/m{\ell}$ concentrations. Also, BGE and BGECS exhibited strong inhibitory activity with >80% against hydrogen peroxide at lower concentration ($125{\mu}g/m{\ell}$). Moreover, specific TOSC values (405 and 473 TOSC/mM) of BGE and BGECS against peroxynitrites were higher than GSH (347 TOSC/mM) used a positive control. These results suggest that BGE and BGECS could be useful to develop functional foods against disease related oxidative stress.

Evaluation of Antioxidant Potential of Extract/Fractions of Equisetum arvense (I) (쇠뜨기 추출물의 항산화 작용에 관한 연구(I))

  • In this study, the antioxidative effects of Equisetum arvense extracts were investigated. The free radical (1,1 diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$) of extract/fractions of Equisetum arvense was in the order: 50 % ethanol extract ($182.04{\mu}g/mL$) < ethylacetate fraction ($54.50{\mu}g/mL$) < deglycosylated flavonoid aglycone fraction ($14.13{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of some Equisetum arvense extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated using the luminol- dependent chemiluminescence assay. The order of ROS scavenging activity was deglycosylated flavonoid aglycone fraction ($OSC_{50}$, $3.54{\mu}g/mL$) < 50 % ethanol extract ($0.80{\mu}g/mL$) < ethylacetate fraction ($0.006{\mu}g/mL$). Ethylacetate fraction showed the most prominent scavenging activity. The protective effects of extract/fractions of Equisetum arvense on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethanol extract (50%) suppressed photohemolysis in a concentration dependent manner, particularly deglycosylated aglycone extract exhibited the most prominent celluar protective effect ($\tau_{50}$, 161.10 min at $10{\mu}g/mL$). These results indicate that extract/fractions of Equisetum arvense can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS.

Antioxidant Activities of Essential Oils from Chamaecyparis obtusa (편백정유의 항산화활성)

  • This study was carried out to investigate the potential promise of Chamaecyparis obtusa oil as a natural antioxidant. C. obtusa oil and its fractions were subjected to screening for their antioxidant activities by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and ammonium thiocyanate method. In the first case, $IC_{50}$ value of the C. obtusa oil was determined as $40{\mu}{\ell}/m{\ell}$. At $0.5{\mu}{\ell}/m{\ell}$ concentration level, free radical scavenging effect of fraction G determined as 66.94% was the highest among the fractions of C. obtusa oil. In the ammonium thiocyanate method, essential oil of C. obtusa and its fraction C, D, and E showed activities of 72.0%, 71.2%, 71.9% and 71.1%, respectively. Fraction G, most active fraction, was mainly consisted of $\alpha$-terpineol, elemol, widdrol and $\alpha$-cadinol.

  • Crude extract of young dangyuja (Citrus grandis Osbeck) fruit was investigated for its antioxidant activity as measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity. Among the samples, including 4 Citrus species and various solvent-extracted-fractions of young dangyuja fruit, the water-extracted fraction (WF) and butanol-extracted fraction (BF) showed the greatest DPPH free radical scavenging activity. WF and BF were further examined for their antioxidant activities by three different in vitro assays. The cell viability tests using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) reduction assay showed that both fractions significantly reduced $H_2O_2$-induced cytotoxicity in HepG2 cells dose-dependently. Generation of the reactive oxygen species (ROS) was also reduced in cells pretreated with both fractions. In addition, BF showed a higher level of lipid peroxidation inhibitory capacity than WF in $H_2O_2$-treated HepG2 cells. Taken together, these results indicate that young dangyuja fruit can be used as an easily accessible source of natural antioxidants.

Antioxidant and Protective Effects of Palmul-tang on Ultraviolet B (UVB)-induced Damage in Human Keratinocytes (팔물탕의 항산화 효과와 자외선으로 유도된 각질형성세포 손상에 대한 보호효과)

  • Objective : In this paper, we investigated the anti-oxidative capacities and protective effects of water extract of palmul-tang (PMT) against Ultraviolet B(UVB)-induced oxidative damage in human keratinocytes(HaCaT). Method : To evaluate the anti-oxidative activities of PMT, we measured scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation and reducing power of PMT. To give an oxidative stress to HaCaT cells, UVB was irradiated with $40mJ/cm^2$ to HaCaT cells. To detect the protective effects of PMT against UVB, we measured cell viability, apoptotic bodies and reactive oxygen species(ROS) generation. Results : PMT showed the anti-oxidative activities by scavenging DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation. Also PMT showed high reducing values. The UVB-induced oxidative conditions led to the cell apoptosis. However, treatment with PMT reduced oxidative stress conditions, including inhibition of cell apoptosis and expression of ROS. Conclusion : PMT had anti-oxidative activities and exhibited protective effects against UVB on HaCaT cells. PMT would be useful for the development of cosmetics treating UVB-induced skin aging.

Antioxidant Activity on Ethanol Extract from Boiled-water of Hizikia fusiformis (톳 자숙액 에탄올 추출물의 항산화 활성)

  • Antioxidant activity of the ethanol extract from boiled-water of Hizikia fusiformis (EBH) were compared with those of BHA, L-ascorbic acid, gallic acid, caffeic acid and (-)-catechin. The free radical scavenging ability against DPPH (1,1-diphenyl-2-picrylhydrazyl), authentic peroxynitrite and reducing power were measured as indices of antioxidant activity. EBH showed the potent DPPH radical and peroxynitrite scavenging activities, showing 85.23 and 96.97% at final concentration of $1000{\mu}g/ml$, respectively. The reducing power increased with the increasing amount of EBH (final concentration of 1, 10, 100 and $1000{\mu}g/ml$). Total phenolic content of EBH was 588 mg (-)-catechin/g at the final concentration $1000{\mu}g/ml$. Total phenolic contents correlated with DPPH radical scavenging activity $(R^2=0.766)$ and reducing power $(R^2=0.944)$. These results suggested that EBH could be a natural antioxidative source containing antioxidative components.

Antioxidant Activities of Aroma Extracts in Commercially Available Red Wines in Korea

  • The antioxidant activities of aroma extracts from commercially available red wines in Korea were evaluated. The aroma extracts of the red wines were extracted by simultaneous steam distillation. Antioxidant activity was measured by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity and ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid) radical cation scavenging activity. DPPH radical scavenging activity of the aroma extracts in the red wines increased with increases in the amount of wine used for aroma extraction. Antioxidant activities of domestic wine 1, imported wine 7, and imported wine 12 were 97.16, 96.72 and 94.52%/20 mL wine by DPPH assay and 7.09, 8.07 and 7.28 mg ascorbic acid equivalents per mL wine by ABTS assay, respectively. This study demonstrates potent antioxidant activities of the aroma extracts of commercially available red wines in Korea.

Antioxidant Activity and Protection from DNA Damage by Water Extract from Pine (Pinus densiflora) Bark

  • Water extract from Pinus densiflora (WPD) was investigated for its antioxidant activity and its ability to provide protection from DNA damage. A series of antioxidant assays, including a 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging assay, a reducing power assay, a metal-chelating assay, a superoxide radical scavenging assay, and a nitrite scavenging ability, as well as a DNA damage protection assay were performed. Total phenolic content was found to be 211.32 mg Tan/g WPD. The extract scavenged 50% DPPH free radical at a concentration of 21.35 ${\mu}g/mL$. At that same concentration, the reducing power ability of WPD was higher than that of ${\alpha}$-tocopherol. The extract chelated 68.9% ferrous ion at the concentration of 4 mg/mL. WPD showed better nitrite scavenging effect at the lower pH. Meanwhile, WPD exhibited a strong capability for DNA damage protection at 1 mg/mL concentration. Taken together, these data suggest water extract from Pinus densiflora could be used as a suitable natural antioxidant.
