Chitosan, a deacetylated derivative of chitin, is one of the abundant resources, and its biological properties such as antibactierial activity, hypocholesterolemic activity, and hypertensive action are remarked. However, increasing attention has recently been yen to converting chitosan to its oligosaccharides because they possess various functional properties like antitumor activity, immune-enhancing effects, enhancing protective effects against infection with some pathogens in mice, antifungal activity, and antimicrobial activity (Jeon et al., 2001). To investigate antitumor constituents of Hydnum repandum, which belongs to edible basidiomycetes, the mycelia separated from the carpophore were shake-cultured and subjected to hot water extraction. The extract was concentrated under vacuum and mixed with a three-fold volume of 95% ethanol to yield precipitates. The water soluble fraction of the precipitates was dialyzed and then lyophilized to yield a water soluble protein-polysaccharide fraction (= WPPF). This exerted antitumor activity against sarcoma 180 implanted in ICR mice. When administered i.p. at the dose level of 20mg/kg once daily for ten consecutive days, it showed an inhibition ratio of 54.3%. WPPF was found to be composed of a polysaccharide moiety (42% of WPPF) and a protein moiety (28% of WPPF) when determined by colorimetric method using anthrone reagent and Folin's phenol reagent. The polysaccharide moiety of WPPF was found to contain glucose (57.4%), mannose(19.3%), galactose (10.8%), xylose (6.8%), and fucose (5.7%), when the methanolysate of WPPF was analysed by GLC method.

To find antitumor components in Korean basidiomycetes, the carpophores of Armillariella mellea which were collected in Gyeong Gi Province were extracted with distilled water at $90{\sim}100^{\circ}C$ for eight hours. The hot water extract was concentrated under reduced pressure, mixed with three-fold volumes of ethanol and allowed to stand at $4^{\circ}C$ overnight. The precipitate was centrifugated and lyophilized to yield a protein-polysaccharide fraction. It was examined for antitumor activity against sarcoma 180 implanted in ICR mice. The fraction showed 75.7%, 83.9%, and 94.1% of tumor inhibition ratios at the doses of 10, 20 and 50 mg/kg/day, respectively. The chemical analysis of the fraction showed that it contained a polysaccharide(41.3%) and a protein (35.0%). The hydrolyzates of the polysaccharide moiety contained fucose (4.5%), xylose (1.1%), galactose (17.4%), glucose (55.4%), mannose(19.4%), and one unknown monosaccharide. The protein moiety contained seventeen amino acids. The protein-polysaccharide from A. mellea was administered, i.p., to mice and caused an influx of polymorphonuclear leukocytes (PMN) at $5{\sim}24$ hours which was followed by an accumulation of macrophages and disappearance of the PMN at $48{\sim}72$ hours.
In order to investigate the effects of Gamihagochosan Extract(加味夏枯草散抽出液) on antitumor effects after human cell lines (A549, hep3B, Caki-1, Ehrlich) transplantation into the peritoneal cavity or right groin in mice induced by RPMI1640 and GIBCO etc., the extracts of its herbal medicines were orally administered for 10 or 12 days. Experimental studies were performed for measurement of antitumor effect of Mitomycin C(MMC) and lysosomal enzyme's activities using colony forming efficiency, SRB assay which were regarded as a valuable method for the measurement of antitumor effects of unknown compound on tumor cell lines. The results obtained in this studies were as follows : 1. The change of colony-forming efficiency and SRB assay of Caki-1 cells, hep3B and A549 Cells after exposure to the extract of Gamihagochosan extract depressed the growth of tumor cells by concentration of Garnihagochosan. 2. Antitumor activity of the ethanol extract from Gamihggochosan extract and MMC on ascites form of Ehrlich carcinoma in mice is slightly improved. Especially the mean of survival times in the group of 200mg/kg and MMC 0.1mg/kg is improved over 34.9%. 3. When Gamihggochosan extract and MMC are administered together, the weight of tumor is more decreased than MMC alone. 4. The lysosomal enzyme's activities of the Gamihagochosan extract and MMC are more significantly improved than MMC alone. According to the above result, it could be suggested that Gamihagochosan extract has indirect antitumor effect by the increase of MMC uptake.

Conventional chemotherapeutic regimens often accompany severe side effects and fail to induce complete regression of chemoresistant or relapsing metastatic cancers. The need for establishing more efficacious anticancer strategies led to the development of a combined modality treatment of chemotherapy in conjunction with immunotherapy or radiotherapy. It has been reported that poly-gamma-glutamate (${\gamma}$-PGA), a natural polymer composed of glutamic acids, increases antitumor activity by activating antigen-presenting cells and natural killer (NK) cells. Here, we investigated the antitumor effect of ${\gamma}$-PGA in combination with cyclophosphamide in a murine melanoma model. Whereas cyclophosphamide alone directly triggered apoptosis of tumor cells in vitro, ${\gamma}$-PGA did not show cytotoxicity in tumor cells. Instead, it activated macrophages, as reflected by the upregulation of surface activation markers and the secretion of proinflammatory factors, such as nitric oxide and tumor necrosis factor ${\alpha}$. When the antitumor effects were examined in a mouse model, combined treatment with cyclophosphamide and ${\gamma}$-PGA markedly suppressed tumor growth and metastasis. Notably, ${\gamma}$-PGA treatment dramatically increased the NK cell population in lung tissues, coinciding with decreased metastasis and increased survival. These data collectively suggest that ${\gamma}$-PGA can act as an immunotherapeutic agent that exhibits a synergistic antitumor effect in combination with conventional chemotherapy.

According to the above results, it could be suggested that Soyosangamibang extract has indirect antitumor effect by strengthen the effect of MMC.
To investigate antitumor components of Korean higher fungi, the carpophores of Schizophyllum commune and Auricularia auricula-judae collected in Kyeong Buk Province were extracted with hot water or 0.1N-NaOH solution. The concentrated extracts were precipitated by addition of ethanol, and the precipitates were purified by dialyzing through visking tube and polysaccharide fractions were obtained. They were found to show antitumor activity against sarcoma 180 implanted in mice. Especially, the inhibition ratio of the extract of Auricularia adicula-judae was 90.8% in the doses of 100mg/kg/day for the period of ten days. The tumor in five of the eight mice was completely regressed. The components of these aqueous extracts were found to be polysaccharide and protein. The hydrolysis of the respective polysaccharide yielded four monosaccharides. After hydrolysis of the protein fraction, 15 amino acids were identified in the respective fraction of S. commune and A. auricula-judae.
Several nucleoside 5'-monophosphate analogues and lipophilic nucleoside 5'-(3-pyridinylcarbonyl)monophosphate analogues were synthesized. Antitumor activities of the synthesized nucleoside malogues against P338 mouse leukemia, FM3A murine mammary carcinoma, and U937 human histiocytic lymphoma cells were determined by MTT assay. Antitumor activities of the lipophilic uridine 5'-(3-pyridinylcarbonyl) monophosphate(7) and 2',3'-didehydro-3'-deoxy-thymidine-5'-(3-pyridinylcarbonyl) monophosphate(8) were stronger than those of uridine 5'-monophosphate(1) and 2',3'-didehydro-3'-deoxythymidine-5'-monophosphate(4). This preliminary experimental result suggests that nucleoside 5'-(3-pyridinylcar-bonyl)monophosphate analogues may be new prodrugs to overcome the clinical limit.
The antitumor effects of mice fed with cell lysate of Lactobacillus plantarum were studied. The abdominal cancer induced by Sarcoma-180 was markedly inhibited and the expected life span was extended by 60% for the Balb/c mice fed with L. plantarum cell lysate for two weeks. A similar result was obtained for the rat inoculated with Spontaneous Osteosarcoma(SOS). The primary tumor volume of SOS was reduced by 70% for the rats fed with L. plantarum cell lysate (100mg/kg/day) for one week before the inoculation of SOS, while only 42% for the rats fed with the same amount of cell lysate for one week after the inoculation of tumor cell line, SOS. As lung was the metastasis site of SOS, the weight of lung was measured to determine the degree of metastasis inhibition by the L. plantarum cell lysate feeding. The rats fed with cell lysate for one week showed a remarkable inhibition of lung metastasis by 63%(before) and 46%(after), respectively. These results indicate that the feeding of L. plantarum cell lysate to mouse or rat can induce a strong stimulation of mucosal or systemic immune system and these effects results in an efficient antitumor activity.
The mistletoe lectins are major active components in the extract of European mistletoe (Viscum album L) that have been widely used in adjuvant chemotherapy of cancer. This study was performed to investigate the antitumor activity of extract of Korean mistletoe (Viscum album var. coloratum) modified with Korean mistletoe lectin (Viscum album var. coloratum agglutinin, VCA). Compared with the results of VCA, survival rate was increased and experimental lung metastasis was reduced by treatment of modified extract (VCM). In addition, the treatment of VCM reduced angiogenesis and VCA-induced toxicity measured by a CAM assay. And VCM inhibited proliferation and induced apoptosis in vitro in tumor cells originated from tissues which are possible to apply topically without surgery. Taken together, the antitumor activities of VCM-treated group outperformed the activities of the VCA-treated group.
