  • The regeneration and differentiation of the cutaneous pigment system in the goldfish, Carassius auratus during the wound healing process were studied with high magnification electron microscope. The cutaneous pigment cells of the normal tissues were composed of three kinds of dermal chromatophores-xanthophores, leucoiphores and melanophores. While xanthophores contain two kinds of pigment granules-pterinosomes and carotenoid vesicles, leucophores and melanophores contain amorphous pigment granules (leucosomes) and oval shaped electron dense melanin pigment granules (melanosomes) respectively. After injury, primary wound healing responses being carried out by migration of epidermal cells and hemocytes spreading over the wound surface at the day of wounding. And at the time of primary wound closure, 5 to 7 days after wounding, rER rich cells-presumably common precursors of dermal chromatophores-immigrated into the wound area. First redifferentiated chromatophores appeared 3 weeks after wounding. Pigment granules of the chromatophores were emerged from the cytoplasmic Golgi complex via rough endoplasmic reticulum. Pinocytotic vesicles which associated with accumulation of pigment material, appeared only at the inner surface of the chromatophores adhering to the rER rich cells, characteristically. The differentiation of each chromatophore in addition to integumental wound repair were accomplished within 4 weeks after wounding at most cases, however the total numbers and densities of these repaired chromatophores still primitive state. Moreover, It has been revealed that complete repair of chromatophores at wounded tissues from burns requirs more than 3 months in normal environment.

  • Safflower is natural red dye largely used for dyeing on protein and cellulose fabric. It contains safflower yellow and carthamin red. Safflower yellow is water-soluble dye, while carthamin red is soluble in alkaline condition. Therefore the former was extracted by cold water. Cartamon obtained by adding acidic solution to carthamin red shows the original hue of safflower. In this study, the condition of extraction with bean stem ash solution and dyeing behavior of carthamon in safflower were examined by using the traditional dyeing method. The relationship between the dye-uptake(K/S) of silk and ramie fabric and the various extractions and dyeing conditions was investigated.

  • To improve poor dye uptake of safflower yellow colorants, cellulose fibers were pretreated with chitosan. The effect of chitosan pretreatment on the dyeability of safflower yellow colorants to cotton, ramie, and rayon was investigated in terms of dye uptake, color, and colorfastness. Irrespective of fiber types, dye uptake increased continuously with increase in chitosan concentration. Chitosan pretreatment improved dye uptake up to 5.6 times for cotton, 7.2 times for ramie, and 3.7 times for rayon. For cotton and ramie, the shade of dyed fabric changed YR color to Y color with increase in chitosan concentration. Dyed rayon fabrics showed Y color irrespective of chitosan concentration. Shades got darker and deeper with increasing chitosan concentration. Shades of chitosan pretreated fabrics were shifted differently depending on dyeing temperature within same fabrics. In common, the color of all dyed fabrics changed to YR at 50 and $70^{\circ}C$ while Y color at 30 and $90^{\circ}C$. V and C value decreased with increase in dyeing temperature and resulted in darker and duller color, in general. Light fastness was fair while washing fastness was poor. It was confirmed that ultrasonic dyeing method enhanced dye uptake more than 30% for cotton and ramie fabrics compared to the conventional automatic machine dyeing method. However, no difference in dye uptake between two dyeing methods was found for rayon.

  • The dorsal skin of Rana temporaria dybowskii Guenther was examined under electron microscope. The results of the fine structures in the xanthophores, iridophores and melanophores were as follows: Xanthophores: Xanthophores were filled with pterinosomes and carotenoid vesicles. Type I pterinosomes had a clear limiting membrane. Type II pterinosomes had the inner fibrous structures. Tyep III pterinosomes were characterized by a few superficial lamellae and type IV pterinosomes by multiple concentric lamellae. Especially typical type II and type III pterinosomes were evenly distributed in the cytoplasm. Iridophores: Iridophores were situated between a xanthophore and a melanophore in the outer part of the dermis just below the basement membrane. Iridophores were filled with reflective platelets, each of which is rectangular and convex lens-like in shape. These platelets were closely contiguous and leave no interspace between them. Endoplasmic reticulum and a few mitochondria were observed in the supranuclear cytoplasm. Melanophores: Dermal melanophores contained numerous melanosomes. The dendritic precesses of the melanophore containing the melanin granules extented up the lateral sides of the iridophore. Epidermal melanophores were filled with melanin granules which appered as the same electron density. A few melanin granules were observed in a cornified surface cell.

  • The antibacterial activities of silver nanoparticles (Ag-NPs) were studied with respect to Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli by observing the bacterial cells treated or not with Ag-NPs by FE-SEM as well as measuring the growth curves, formation of bactericidal ROS, protein leakage, and lactate dehydrogenase activity involved in the respiratory chain. Bacterial cells were treated with Ag-NPs powder, and the growth rates were investigated under varying concentrations of Ag-NPs, incubation times, incubation temperatures, and pHs. As a result, S. aureus and E. coli were shown to be substantially inhibited by Ag-NPs, and the antibacterial activity of Ag-NPs did not fluctuate with temperature or pH. These results suggest that Ag-NPs could be used as an effective antibacterial material.

  • We investigated the molecular epidemiological characteristic of methicillin resistant Staphylococcus aureus (MRSA) strains isolated from stool samples in Busan from 2004 to 2006. Among 142 isolates of S. aureus, 49 isolates (34.5%) were confirmed as MRSA. With the antimicrobial susceptibility tests, 37 isolates (75.5%) showed multiple resistance to more than 10 antibiotics, but all isolates were sensitive to vancomycin. All of MRSA had enterotoxin A in 30.6%, B 4.1%, C 8.3%, D, C/G, A 2.0% and None 51%. PFGE of SmaI-digested chromosomal DNA was performed on 49 sporadic MRSA isolates. Restriction fragment patterns consisted of 8 to 14 fragments ranged in size from 48.5 to 630.5 kbp. We could divided the isolates into 7 groups ($I{\sim}VII$) by analyzing PFGE patterns. Group I subdivided as 2 subgroups and 17 (34.7%) strains belong to the group I. Dendrogram of PFGE patterns showed that MRSA strains in Busan were heterogeneous but we could find out minor homogeneity in hospital.

  • The propose of this study was to evaluate the microbiological hazards of the customers' hands in university foodservice operation. A total of 190 customers' hands were sampled to test about aerobie plate counts, coliforms, E. coli and Staphylococcus aureus. The average number of aerobic plate counts was 3.11 log CFU/hand Ifrom 0.70 to 6.47 log CFU/hand) and that of coliform counts was 1.06 log CFU/hand (from not detected to 5.04 log CFU/hand). The mean level of aerobic plate counts (t=9.87, p<0.001) and coliforms (t=7.91, p<0.05) was significantly different by gender. E. coli was not detected, but Staphylococcus aureus was detected in 39 (20.5%) of 190 samples. The average of hand-washing frequency was 6.46 time per day. By the hand-washing frequency, the mean level of aerobic plate counts was significantly different (F=527, p<0.001), but coliforms was not significantly different. This study shows that it is needs to be given attention to hygiene handlers but also for customers in foodservice operations.

  • Genipin was obtained from hydrolysis of geniposide isolated from gardenia fruits with ${\beta}-glucosidase$. Reaction of genipin with glycine, alanine, histidine, lysine, phenylalanine and glutamate in aqueous buffer solution converted colorless starting materials to blue pigments. Effect of pH for the formation of blue pigments was tested using UV/Vis spectrophotometer. The optimum pH for the formation of blue pigments was 7.0. No pigment and trace amounts were formed at acidic (pH 3.0) and alkaline (pH 12.0) conditions, respectively. The amount and tincture of blue color were distinct with different amino acids. In contrast with lysine $({\lambda}_{max}=573\;nm)$, glycine $({\lambda}_{max}=595\;nm)$, phenylalanine $({\lambda}_{max}=602\;nm)$ and alanine $({\lambda}_{max}=595\;nm)$, the reaction of genipin with histidine $({\lambda}_{max}=601\;nm)$ and glutamate $({\lambda}_{max}=601\;nm)$ produced relatively small amounts of blue pigments. Rate constants for the formation of blue pigments from genipin with amino acids at various temperatures $(60,\;70,\;80,\;90^{\circ}C,\;pH\;7.0\;phosphate\;buffer)$ were obtained. Rate constants of genipin with basic amino acids were larger than neutral or acidic amino acids. Arrhenius activation energies of the formation of blue pigments indicated that activation energy of glycine $(E_A=9.8\;kcal/mol)$ was especially lower than those of other amino acids $(E_A=13.3{\sim}15.4\;kcal/mol)$.

  • Plankton samples used for the present study were collected by the NORPAC net during the CSK cruises in the Korean waters in March and August, 1967. Regional and seasonal variations in the zooplankton biomass (wet weight, mg/㎥) were noticed in the Korean waters. In March the highest biomass, 130mg/㎥ on the average, occurred in the southern part of Japan Sea, but the lowest biomass of less than 50mg/㎥ occurred in the Yellow Sea and the western sea of Cheju Island Contrally, in August, the average biomass of 120mg/㎥ was measured in the Yellow Sea, the western sea of Cheju Island and the coastal waters of southern Korea, while the biomass of Japan Sea was the lowest of the regions surveyed. In comparison with the zooplankton biomass, total number of zooplankton per cubic meter of water strained also showed regional and seasonal fluctuations. In general, variations in the number of zooplankton specimens follows the same trend as in the biomass. The largest number, up to 800mg/㎥ on the average, occurred in the southern part of Japan Sea in March and the lowest number, less than 200mg/㎥ occurred in the Yellow Sea and the western sea of Cheju Island. In August, as shown by the biomass fluctuations, the largest number of zooplankton 850mg/㎥ on the average occurred in the Yellow Sea, the western sea of Cheju Island and the coastal region of southern Korea. But the lowest number of less than 500mg/㎥ was found in the Japan Sea. Among the various groups of zooplankton examined, the following were dominant components of the zooplankton population: Copepoda, Chaetognatha, Siphonophora, Euphausiacea, Cladocera, Appendicularia, and Amphipoda. The zooplankton conposition was significantly differed between the Japan Sea and Yellow Sea. Copepods which usually occupied over 66% in the Japan Sea and thd Korean Strait samples occupied only 42% of the catches in August, while cladocerans and chaetognaths were relatively abundant, i. e., 15 and 18% of the total organisms. The most dominant species of copepods and chaetognaths were Paracalanus parvus, Oithona similis, Acartia clausi, Calanus helgolandicus, Sagitta enflata, S. bedoti, S. elegans and S. crassa.
