Luminance distribution of Illuminated Bridges by View Point (교량의 조망점에 따른 휘도분포 특성분석)

  • 본 연구에서는 서울시 한강에 위치한 교량을 대상으로 다양한 조망점에 따른 경관조명의 물리량을 측정하고 그 특성을 분석함으로써 동일한 연구대상에 대하여 조망점이 달라졌을 때 나타나는 물리적 변화를 통해 경관조명분야의 물리량평가방법론의 새로운 접근방법론을 제시하는 것이 목적이다. 이를 위하여 연구대상의 선정은 교량구조형식에 따라 분류하고. 경관조명이 설치된 교량 17개소 중 다양한 조망점과 현장측정의 편리성 등을 고려하여 광진교(거더교), 올림픽대교(사장교), 동작대교(트러스교), 동작대교(아치교)를 연구대상으로 선정하였다. 조망점은 한강둔치를 이용하는 방문객들이 바라보는 투시뷰, 교량주변 건축물에서 바라보는 건물뷰 그리고 교량 위를 이동하면서 바라보는 주행뷰로 구분하였다. 이러한 연구방법으로 도출된 결과는 동일한 경관조명연출을 토대로 교량별 조망점에 따른 휘도측정 및 분석에서 측정되는 각도, 거리, 방향에 따라 정형화된 비율은 아니지만 휘도값에 있어서 차이가 있다는 것을 확인 할 수 있었으며, 투시뷰와 건물뷰는 동일한 교량 구성요소를 갖고 있어 상호간 비교 분석이 가능하고 경관조명평가 요소로서 그 기능을 담당 할 수 있다고 판단되며, 주행뷰는 교량형식과 가로등의 영향을 많이 받으므로 평가요소로써 조망점을 활용하기에는 조금 더 깊은 연구가 선행되어야 할 것으로 보인다. 따라서 경관조명을 평가함에 있어 다양하게 바라보는 조망점은 조명연출로 인한 시각적 인식에 차이를 유발시킬 수 있어 경관조명을 평가할 때 다양한 조망점의 설정이 보다 객관적이고 정확한 분석이 가능할 것으로 판단된다.ridinium의 분포는 chlorophyll-a농도와 밀접한 관련성이 있었다. 담수적조의 현존량은 상류지역에서 $8.5\;{\times}\;10^3\;cell\;mL^L{-1}$로서 많았고, 댐부근의 하류지역($4.4\;{\times}\;10^2\;cell\;mL^L{-1}$)으로 갈수록 감소하는 양상을 보였다. 또한, 담수적조의 원인종으로 규조류 Synedra acus와 남조류 Microcystis aeruginosa도 소량관찰되었다. 저수지의 수질 부영양화 현상은 기존 경사사면형 농경지를 기반으로 조성되었으므로 저층으로부터 풍부한 영양환경을 내재하고 있었고, 담수 초기에 발생할 수 있는 가능성을 포함하고 있었다. 또한 유입수량의 부족과 현재 수질개선을 위해 적용하고 있는 수중폭기시설등의 물리적인 영향도 직 ${\cdot}$ 간접적으로 작용하였을 것으로 추정되었다 따라서 향후 이에 대한 중장기적인 모니터링이 필요하였고 유역과 저수지의 통합적인 수질관리계획 이 요구되었다.group으로, 용존산소량, 전기전도도, pH, 인산염이 정점 3, 4, 5에서 같은 group으로 묶였다.유의적인 CAT 활성 증가효과가 나타났고, 간조직의 시토졸 1획분에서는 대조군 대비 15${\sim}$27%의 CAT 활성 증가효과가 인정되었다. 이상의 결과에서 목초액의 장기간 투여는 간조직 중의 활성산소의 억제효과뿐만 아니라 방어시스템으로서 활성산소 제거효소의 역할도 충실히 수행하여 노화를 효과적으로 예방하고 억제할 수 있을 것으로 기

Antifungal Activity against Trichoderma spp. of Water Soluble Essential Oil extracted from Pinus densiflora and Chamaecyparis obtusa (소나무 및 편백나무 수용성 정유를 이용한 Trichoderma spp.의 생장억제 활성)

  • This study was carried out to investigate the antifungal activity of the water soluble essential oil against Trichoderma spp. Water soluble essential oils from Pinus densiflora and Chamaecyparis obtusa were obtained from GAP (Gas assisted process) extraction apparatus. When the water soluble essential oil was treated, the growth of the conidial germination was maximum under the cultivation condition at $25^{\circ}C$ and in the culture medium adjusted to pH 5.0. The yield of water soluble essential oil was 3.9% and 3.7% in P. densiflora and C. obtusa, respectively. 24 and 15 kinds of compounds were identified in water soluble essential oils of P. densiflora and C. obtusa, respectively. The major components in the essential oil of P. densiflora were $\alpha$-Terpineol acetate and Terpinen-4-ol and those of C. obtusa were Terpinen-4-ol and $\alpha$-Terpineol. Antifungal activity was the best in the 5000 ppm of P. densiflora against Trichoderma harzianum and in 5000 ppm of C. obtusa against Trichoderma atroviride.

Structural elucidation of immuno-stimulating polysaccharide, galactomannan isolated from Colocasia esculenta (토란으로부터 분리한 면역활성 다당 galactomannan의 구조적 특성 규명)

  • To elucidate the structure-function relationship of polysaccharides obtained from Colocasia esculenta, the immuno-stimulating polysaccharide, CE-4a was purified to homogeneity from the crude polysaccharide (CE) extracted from the corms of C. esculenta by two subsequent column chromatographies using DEAE-Sepharose FF and Sephadex G-100, and analysis of their immuno-stimulatory activities and structure were conducted. CE-4a showed an increase in anti-complementary activity in a dose-dependent fashion. The molecular mass was estimated to be 182.4 kDa, which mainly consisted of galactose (43.5%) and mannose (18.2%). Methylation analysis indicated that CE-4a comprised at least 10 different glycosyl linkages, such as terminal Galp, 3-linked Galp, and 4-linked Manp, as well as a characteristic linkage, 2,4,6-branched Manp residue. To analyze the fine structure of CE-4a, it was sequentially digested using endo-α-(1→4)-polygalacturonase, exo-α-galactosidase and endo-β-1,4-D-mannanase. These analyses suggested that CE-4a is to be a highly branched galactomannan with a (1→4)-mannan backbone and galactopyranosyl oligosaccharide side chains.

Properties of Acid Tolerance of Acid-Resistant Mutant Leuconostoc mesenteroides Which Was Improved as Kimchi Starter. (김치 starter용으로 개량된 내산성 변이주 Leuconostoc mesenteroides의 내산성 특성)

  • To investigate the increased acid tolerance of a acid-resistant mutant Leuconostoc mesenteroides M-100 improved as a kimchi starter, proton permeability, ATPase acitivity, glycolysis activity, $Mg^{2+}$ releasement, and membrane fatty acid composition were studied and comprised with its wild type Leuconostoc mesenteroides Mw. In the proton permeability experiment, the mininum values of the average half time ($t_{1/2}$) of pH equilibration through the cell membrane of the Mw and the M-100 were about 8.6 min and 9.2 min in 150 mM KCI solution, respectivily. In the 3% NaCl solution, the $t_{1/2}$ values of the Mw and the M-100 were 6min and 8.6 min, respectivily. The values and pHs of maximal specific activities of ATPase originated from the Mw and the M-100 were 0.6U at pH 5.5 and 0.8U at pH 5.5, respectivily. The result of pH dependence of glycolysis showed that the M-100 had higher activities than that of Mw except at pH 5.0. The releases of magnesium from the Mw and the M-100 were observed about 36.5% and 13% at pH 4.0 after 2 hours, respectivily. The results of comparison of membrane fatty acid composition of the Mw with the M-100 showed that $C_{12}$, $C_{14}$, $C_{18:1}$, and $C_{19:0cyclo}$ were major different fatty acids between two strains and the content of $C_{18:1}$, and $C_{19:0cyclo}$ were 23.4%, 10.2% in the Mw and 15.1%, 12.2% in the M-100. These results indicated that acid tolerance of the M-100 was significantly improved in comparison with its wild type Mw.

Physico-Chemical Analysis and Antioxidant Activities of Korea Aronia melanocarpa (국내산 아로니아의 이화학 분석 및 항산화 활성)

  • In this study, we examined the effects of cultivation adaptability and product quality of aronia (fruit of Aronia melanocarpa) cultivated in various domestic regions. Extracts of aronia cultivated in various domestic regions and Poland were measured for their total sugar contents, acidities, total polyphenol contents, anthocyanin contents, and antioxidant activities using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) assays. Our results showed that aronia extracts from the two countries had similar sugar contents, acidities, and anthocyanin contents. Anthocyanin is an important functional component of Aronia melanocarpa. Extracts of aronia from the two countries contained cyanidin-3-galactoside (65.5~69.1%) as the major anthocyanin compound. Aronia cultivated in C region showed higher polyphenol content (121.5%) than Poland aronia and we measured of antioxidant activities by DPPH ($IC_{50}$) and FRAP assay. Aronia cultivated in C region showed the highest antioxidant activity and polyphenol contents. Cultivation conditions of C region had sufficient sunshine and soil with pH of 6.5. From the above results, Korean aronia had similar activities with Poland aronia, which suggests that it can be a new potential development source and high technological foods.

In vitro Effect of Ecklonia stolonifera Okamura Extract on the Cell Growth in CCD-986sk Human Fibroblast and Melanin Formation Inhibition in Clone M-3 Mouse Melanocyte Cell Line (청정해역 곰피추출물의 세포생리활성 연구)

  • In order to investigate whether or not CCD-986sk cell line can be affected by Korean Ecklonia stolonifera Okamura, we examined the MTT assay when we treated Korean Ecklonia stolonifera Okamura extract in CCD-986sk human fibroblast cell line. The sample were tested for cell proliferation activity by means of a modification of the MTT assay. Ecklonia stolonifera Okamura extract showed significantly strong cell proliferation activity at the range of from 6.25 mg $mL^{-1}$ to 1.56 mg $mL^{-1}$ compared with control group. And in order to search for inhibition agents of skin melanin formation, we tested for inhibition effect of melanin pigmentation of Korean Ecklonia stolonifera Okamura using Clone M-3 mouse melanocyte cell lines. when we treated the extracts of Ecklonia stolonifera Okamura to the mouse melanocyte cell lines, the sample showed a significantly little formation of melanin pigments compared with control group at the only range of 200 mg $mL^{-1}$. These results suggest that extract of Korean Ecklonia stolonifera Okamura may represents an excellent candidate for inhibition of melanin pigmentation and for protection of human skin aging at in vitro level.

Comparison of Physicochemical Properties and Antioxidant Activities between Lentinula edodes and New Cultivar Lentinula edodes GNA01 (표고버섯(Lentinula edodes) 및 표고버섯 신품종 이슬송이버섯(Lentinula edodes GNA01)의 이화학적 특성 및 항산화 활성 비교)

  • The new cultivar Lentinula edodes, which is named 'Lentinula edodes GNA01', was bred by mating strains isolated from 'L26' and 'Kyoungwon9015' obtained from Sammyungjin Research Institute, Fujian, China. L. edodes GNA01 does not have stipes like L. edodes, although it generally has a similar spherical shape. Moisture and crude protein contents of L. edodes GNA01 were lower than those of L. edodes. Meanwhile, L. edodes GNA01 contained higher levels of crude ash, crude lipid, crude fiber, and carbohydrates than L. edodes. The ${\beta}$-carotene content ($19.05{\mu}g/100g$) of L. edodes GNA01 was about three times higher than that of L. edodes. In addition, vitamin D content ($118.53{\mu}g/100g$) of L. edodes GNA01 was more than twice that of L. edodes. L. edodes GNA01 was a good source of mineral elements, with K and Mg contents of 2,277.50 mg/100 g and 203.15 mg/100 g, respectively. The major fatty acids of L. edodes GNA01 were C16:0 and C18:2, and L. edodes GNA01 had the highest linoleic acid (C18:2) content of 1,087.66 mg/100 g. Total phenol content of L. edodes GNA01 was 12.52 mg GAE/g, and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities and ferric reducing antioxidant power (FRAP) values of L. edodes GNA01 were lower than those of L. edodes at all concentrations. However, DPPH radical scavenging activities and FRAP values of L. edodes GNA01 were above 80% and 0.9 at a concentration of 10 mg/mL, respectively.

Structural Analysis of Anti-metastatic Polysaccharides Isolated from Opuntia humifusa (천년초에서 분리한 항전이 다당의 구조 분석)

  • To examine the new practical utilization of mucilages in Opuntia humifusa, polysaccharides were isolated from O. humifusa and their anti-metastatic activity and structural analysis were carried out. In experimental lung metastasis of B16BL6 melanoma cells, prophylactically intravenous (i.v.) administration of the crude polysaccharide (CNC-0) from O. humifusa significantly inhibited lung metastasis in a dose-dependant manner. The main polysaccharide, CNC-Ia was purified to homogeneity from CNC-0 by two successive column chromatographies using DEAE-Sepharose FF and Sephadex G-100 and its structure was characterized. Molecular mass of CNC-Ia was estimated to be 700 kDa and it mainly consisted of arabinose, galactose and xylose in addition to two minor sugars such as rhamnose and fucose. Methylation analysis indicated that CNC-Ia comprised at least 18 different glycosyl linkages such as terminal Araf, 5-linked Araf, 4-linked Galp and terminal Xylp in addition to three characteristic linkages such as full branched Araf, 3,4,6-branched Galp and full branched Galp. To analyze the fine structure of CNC-Ia, it was sequentially digested by exo-${\alpha}$-L-arabinofuranosidase and endo-${\beta}$-1,4-D-galactanase. These analyses suggested that CNC-Ia belongs to be a highly branched Type I arabinogalactan which has a ($1{\rightarrow}4$)-${\beta}$-galactan backbone with arabinosyl oligosaccharide side chains.

Studies on Functional Salt Fortified with Seaweed Components (해조성분 강화 기능성소금에 대한 연구)

  • In an attempt to develop functional salts having beneficial health effects, we experimentally prepared three functional salts by fortification with soluble seaweed minerals (Hizikia mineral salt, HMS), fucoidan (fucoidan salt, FS) and laver extracts (laver salt, LS). To characterize the functional salts, their physicochemical properties and in vitro functionalities, such as pH, color, mineral composition, solubility, oxidation-reduction potential, sensory properties, angiotesin converting enzyme (ACE) inhibitory activity, and bile acid binding capacity were investigated. The functional salts revealed slightly lower NaCl concentrations, but showed a variety of pH values compared with conventional table salt. The pH values of HMS, FS, and LS were 11.3, 6.8, and 6.5, respectively. The oxidation-reduction potentials (ORP) of the functional salts varied from -229 mV to 38 mV, significantly lower than refined salt. The functional salts were significantly darker in color than refined salt, and the mineral composition of HMS was considerably enriched compared to refined salt, particularly in potassium ion. As a result of the sensory evaluation, FS and LS were comparatively palatable in saltiness, pungency, bitterness, and overall acceptance compared with refined salt. It was also found that one functional salt had ACE inhibitory activity (54.8% in LS) and another had bile acid binding capacity (80.7% in FS).

Antibacterial Action against Food-Borne Pathogens by the Volatile Flavor of Essential Oil from Chrysanthemum morifolium Flower (국화 꽃 휘발성 향기성분의 식중독균에 대한 항균 작용)

  • The aim of this study was to investigate antibacterial activities of essential oil from C. morifolium against four Grampositive bacteria and six Gram-negative bacteria. The antibacterial activity of the oils was determined by agar-well diffusion assay, minimum inhibitory concentration(MIC), and minimum bactericidal concentration(MBC). Essential oil of C. morifolium had a large inhibition zones especially against Salmonella enterica(21 mm) and Bacillus cereus(19 mm). Essential oil of C. morifolium generally showed higher antibacterial activity against Gram-positive bacteria than Gram-negative bacteria. MIC of essential oil from C. morifolium was 5 ${\mu}g/m{\ell}$ against ten food-borne pathogens. MBC values were determined to be from 5 to 20 ${\mu}g/m{\ell}$ against eight bacteria except Salmonella choleraesuis and Listeria monocytogenes. Therefore, the essential oil of C. morifolium and its components have a potent antibacterial activity against food-borne pathogens, and is expected to be used as a novel food preservative.
