Pseudomonas carboxydohydrogena, Cardiobacten'm hominis, lvIicrococcus lylae, XanfomonCls campestris p" juglandis, Micrococcus diversus, and Comamonas terrigencl as aerobic dominants, and Streptococcus bovis and Prevotella buccae as anaerobIc dominants were identified fi'om the supernatent of the primary settling tank. It seemed that microflora in the treatment process would consist of many kinds of microorganisms, whose dominant would change easily according to environmental conditions, They all grew well at $37^{\circ}C$ and at different initial medium pH's. Especially, some of them required sulfate ion for their growth, which came from a chemical coagulant of aluminium sulfate in the primary settling tank. Interestingly. many anaerobes grew well even in the aerobic wastewater treatment process and seemed to have some functions. Population of anaerobes increased three times in the supematant of primary settling tank and ten times in Lhe bottom sludge of primary settling tank than in the prime wastewater. Therefore, these anaerobes contributed to the producH tion of offensive gases, which would make some microorganisms not precLpitate and be buoyant.

Enrichment of anaerobic ammonium oxidation (ANAMMOX) bacteria is the essential step for operating full-scale ANAMMOX bioreactor because adding a significant amount of seeding sludge is required to stabilize the ANAMMOX reactor. In this study, the enrichment of ANAMMOX bacteria from an activated sludge using sequencing batch reactor was conducted and verified by analyzing changes in the microbial community structure. ANAMMOX bacteria were successfully enriched for 70 days and the substrate removal efficiencies showed 98.5% and 90.7% for $NH_4\;^+$ and $NO_2\;^-$ in the activity test, respectively. The phylogenetic trees of Planctomycetes phylum showed that the diverse microbial community structure of an activated sludge was remarkably simplified after the enrichment. All 36 clones, obtained after the enrichment, were affiliated with ANAMMOX bacteria of Candidatus Brocadia (36%) and Candidatus Anammoxoglobus (64%) genera. The quantification using real-time quantitative PCR (RTQ-PCR) revea ed that the 16S rDNA concentration of ANAMMOX bacteria was 74.8% compared to the granular ANAMMOX sludge obtained from an upflow ANAMMOX sludge bed reactor which had been operated for more than one year. The results of molecular analysis supported that the enriched sludge could be used as a seeding sludge for a full-scale ANAMMOX bioreactor.

혐기성 미생물의 특징은 성장 속도가 느리고 침강성이 좋지 않다는 점이다. 이 문제의 한 해결책인 막결합형 혐기성 소화는 고액분리를 완전하게 수행함으로써 미생물의 유출을 방지하여 반응조 내부에 미생물을 고농도로 유지할 수 있을 뿐만 아니라 에너지의 회수와 설비 면적의 축소 등 많은 장점이 있다. 이 막결합형 혐기성 소화의 경제성은 사용된 분리막의 투과 속도에 의해 크게 좌우된다. 분리막의 투과 속도에 영향을 미치는 인자로는 미생물 및 유입수를 비롯한 반응조 내부의 상태, 막모듈 압력 온도 막면유속 등의 운전 조건이 있다. 또한 사용되는 분리막 자체의 재질도 투과 유속에 큰 영향을 미친다. 본 연구의 목적은 관형의 지르코니아 스킨층과 탄소 소재 지지층으로 이루어진 복합 재료 분리막과 폴리프로필렌 분리막을 이용하여 막재질에 따른 막오염 특성을 비교 분석하고 효과적인 투과율 회복 방법을 확립하는 것이다.

Starting at the beginning q the 20th century, increasing amounts of tetrach1oroethene (PCE) and trichloroethene (TCE)were manufactured due to the extensive use of these compounds in industry, in the military, and in private households, mainly as nonflammable solvents. This widespread use, along with careless handling and storage, are among the most serious contaminants of soil, sediment and groundwater. Highly chlorinated ethenes are typically not degraded through oxygenation by aerobic bacteria Since complete reductive dechlorination of PCE and TCE to ethene (ETH) has been observed in anaerobic enrichment culture, anaerobic dehalorespiring bacteria have received increased attention in the last decade. Under anaerobic conditions, these compounds con be reductively dehalogenated to less-chlorinated ethenes or innocuous ethene by microorganism through dehalorespiration. We have been studying anaerobic enrichment culture which used lactate as the electron donor for reductive dechlorination of PCE to ETH the anaerobic mixed microbial culture was enriched from the sediment sample taken from site contaminated with PCE. PCE was consistently and completely converted to ethene. In addition, the accumulation of intermediate products such as 1,2-ds-dichloroethene (cis-DCE) and vinyl chloride (VC) was observed in the anaerobic mixed microbial culture. the established dechlorinating enrichment culture was analyzed by DGGE using primers specific to DefrJ1ococcoides 16S rRNA gene sequences. In conclusion, we established the PCE dechlorinating enrichment culture and confirmed the existence of Dehalococcoides in an enrichment culture.

다양한 혐기성 조건에서 다환방향족탄화수소(PAHs)로 오염된 토양의 미생물 분해 연구를 수행하였다. 대표적인 다환방향족탄화수소인 phenanthrene과 fluorene을 토양과 물에 오염시켜서 약 100일 동안 저감정도를 관찰하였고, 실제 다환방향족탄화수소로 오염된 현장 토양을 이용 혐기성하에서 다환방향족탄화수소의 생분해 가능성을 확인하였다. 미생물 접종원은 혐기성 조건에서 다환방향족탄화수소에 노출시킨 슬러리가 사용되었다. 황산염 환원조건, 질산염 환원조건, 메탄생성조건 등의 다양한 혐기성 조건에서 실험을 수행한 결과, 메탄생성조건 > 질산염 환원조건 > 황산염 환원조건의 순서로 분해가 잘 일어났다. 또한 현장오염토양의 경우 34일간 처리 후 메탄생성조건에서 최대 72%의 분해율을 보였다.

Extremely slow growing anammox(anaerobic ammonium oxidation) bacteria were cultivated using a combination of UASB(Upflow Anaerobic Sludge Blanket) reactor seeded with anaerobic granular sludge and carbon-fiber cultivating reactor. After 180 days of continuous cultivation, average nitrogen removal rate showed 0.54 kg $N/m^3-day$ when 0.6 kg $N/m^3-day$ of nitrogen loading was applied. The black granule was changed to brown and red granule as continuous operation, and the red granule was highly dependant on the high anammox activity. Microbial community structure of red granule in the UASB reactor was analyzed by molecular methods such as gene cloning, phylogenetic tree analysis, and FISH(Fluorescence In Situ Hybridization) method. As a result of gene cloning and phylogenetic tree analysis, 5 kinds of phylum were found to be Planctomycetes, Proteobacteria, Acidobacteria, Chlorobi and Chloroflexi. 13 clones were matched to anammox bacteria among 51 clones in the red anammox granule. In-silico test which used cloning information and FISH probe of the AMX368 was conducted to detect the presence of anammox bacteria in the red anammox granule. As a result of in-silico test only one clone was exactly matched to AMX368 but 11 clones was mutated one base among 18 bases representing all 12 clones are anammox bacteria. A filamentous Chloroflexi might be related to the granulation of anammox bacteria. As a result of FISH analysis, anammox bacteria was abundant in the red anammox granule.
