Effects of Dietary n-3 Highly Unsaturated Fatty Acids on Growth and Biochemical Changes in the Korean Rockfish Sebastes schlegeli II. Changes of Blood Chemistry and Properties of Liver Cells (사료의 n-3계 고도불포화지방산 함량에 따른 조피볼락 Sebastes schlegeli의 성장 및 생화학적 변화 II. 혈액성분 변화 및 간세포 성상)

  • To define the effects of various levels $(0\~1.5\%)$ of dietary n-3HUFA on the physiological changes in the Korean rockfish, variations in blood variables and hepatocytes were studied. Biochemical serum analyses, lactate dehydrogenase (LDH) activity of the liver cytosol and ATPase activity of the liver microsomal membrane were also studied. The haematological values (red blood cell, hemoglobin, hematocrit, MCHC, MCV and MCH) were not significantly different in the experimental groups $(P\geq0.05)$. The total protein and glucose levels in the serum were affected by dietary n-3HUFA levels. These levels in groups fed n-3HUFA insufficient diets were significantly lower than those of n-3HUFA sufficient groups (P<0.05). Serum levels of total cholesterol, free cholesterol, glutamic pyruvic transaminase (GPT) and glutamic oxaloacetatic transaminase (GOT) showed significantly higher values in the fish fed n-3HUFA deficient diets (P<0.05). The LDH in the serum was dropped with increasing dietary n-3HUFA levels, but the LDH activity of the liver cytosol was elevated. Histologically, the hepatic cell in the fish fed n-3HUFA free diet was abnormal and showed a necrotic condition. $Ca^{2+}-ATPase$ activities of the liver microsomal membrane were significantly lower in the fish fed n-3HUFA deficient diets than in those fed n-3HUFA sufficient diets (p<0.005). These results suggested that the liver cell membrane was affected by dietary fatty acid compositions and cell membrane of the fish fed n-3HUFA insufficient diets showed abnormalities.

Effect of Transport Time on the Blood Profile and Meat Quility of Slaughter Pigs (수송시간이 돼지의 혈액성상과 육질에 미치는 영향)

  • In a trial involving 120 pigs, the effects of transport time on blood profile and meat quality in pigs were investigated. One group of 60 animals was subjected to 20 min and the others to 2 h transport time, and held in lairage for 1 h 30 min. There was not significantly different in the carcass weight, backfat thickness and carcass grade between groups. Cortisol and lactic dehydrogenase(LDH) concentrations were significantly(P〈0.05) higher in the group transported for 2 h compared with the group transported for 20 min. There was not significantly different(P〉0.05) in meat quality(pH$_1$, pH$_{u}$, drip loss, cooking loss, hardness, CIE L$^{*}$, a$^{*}$, b$^{*}$ and NPPC) and skin damage of pork carcass between groups. These results imply that the stress could be affected by transport time in transit without meat quality.

Changes in water quality and hematological parameters according to the stocking density of olive flounder, Paralichthys olivaceus raised in bio-floc environment (바이오플락 양식기술 적용 넙치, Paralichthys olivaceus 수용밀도에 따른 수질변화 및 혈액학적 성상 변화)

  • The Paralichthys olivaceus (mean weight 34.3±3.5 g) was used in the conduction of density experiment in this study to establish the optimal density determination for a period of 13 weeks. The density consisted of four groups. These were group 1 (500 fish, 40 fish m-2), group 2 (750 fish, 60 fish m-2), group 3 (1,000 fish, 80 fish m-2) and group 4 (1,250 fish, 100 fish m-2), each in 12.56 ㎡ circular water tanks. In the high density groups, such as groups 1 and 2, the nitrite was increased to over 21 mg L-1 (group 3) and 25 mg L-1 (group 4). The experiment of the groups was terminated by skin ulcers and mortality after 49 days for group 3 and 43 days for group 4. The water quality environment, such as the water temperature, dissolved oxygen, salinity and pH, remained constant. The ammonia and nitrite in groups 1 and 2 remained stable after ammonia and nitrite peak. However, the experiments in groups 3 and 4 were terminated due to the mortality induced by high nitrite. Hematological parameters, such as hemoglobin and hematocrit, were significantly decreased in the high density groups. The plasma components were significantly changed in the high density groups, such as groups 3 and 4. The mean weight of groups 1 and 2 after 13 weeks were 91.3 g and 83.7 g, respectively. The survival rates were 99.4% and 98.2%, respectively. The final FCR was 0.6 in both groups. The results of this study show that the density of 80 fish m-2 or more induce mortality due to high nitrite with hematological changes. Additionally, they indicate that the 60 fish m-2 indicate proper density in bio-floc environment in olive flounder weighing less than 100 g.

Effects of Fasting and Handling Stress on Plasma Concentrations of Glucose, Stress-associated Enzymes and Carcass Quality of Market Pigs (절식과 출하취급이 출하돈의 스트레스 관련 혈액성상과 도체품질에 미치는 영향)

  • One hundred and forty-four cross-bred market pigs weighing approximately 110 kg were randomly divided into six groups in a 3 (duration of fasting prior to loading; 0, 12 and 24 h) ${\times}$ 2 (handling stress; minimal vs stimulated handling stress) factorial arrangement of treatments. The stimulated handling stress group received overally rough handling including electric prod stimulation during loading, transport and lairage at least once at each step. All the animals received 3-h lairage prior to slaughter. Blood and longissimus dorsi muscle (LM) samples were taken at slaughter and after overnight chilling of the carcass, respectively. Mean plasma glucose concentration, as expected, was less in the 12 h- or 24 h-fasting group than in the 0 h-fasting, whereas cortisol concentration was greater (P<0.05) in the 24 h- vs 0 h-fasting group. Plasma concentrations of stress indicators glucose, cortisol, creatine kinase and lactate dehydrogenase were greater in the stimulated vs minimal handling stress group. There were no interactions between the duration of fasting and handling stress in their effects on these blood variables. The incidence of pale, soft and exudative (PSE) carcass and drip loss of LM were reduced in the 12 h- or 24 h- vs 0 h-fasting group, whereas the 24-h postmortem LM pH and color including the lightness and redness were not affected by the duration of fasting. The incidence of PSE carcass and physicochemical characteristics of LM, however, were not changed by the stimulated vs minimal handling stress. In conclusion, results suggest that fasting the market pig overnight prior to transport is desirable in terms of reducing the incidence of PSE carcass. Rough handling of market pigs may not affect the carcass quality of the animals when an enough lairage time is provided. However, rough handling inflicts a stimulated stress on the animal, which is manifested by increased blood concentrations of stress indicators, and therefore should be avoided fer animal welfare.

Study on the Winter Mass Mortality of red sea bream, Pagrus major in South sea area (동절기 남해안 참돔(Pagrus major)의 대량폐사에 관한 연구)

  • Pathological symptoms and hematological parameters of red sea bream, Pagrus major and water temperature in the culture ground was investigated to clarlify the cause of winter moratility. Dead fish showed green liver and accumulation of ascites in the cavity. A few Bivagina tai were also found on the gill but either bacteria or virus were not. When hematological parameters from fish taken before/after the winter mortality were compared, blood glucose, serum AST and ALT, and total cholesterol, trigyceride and total protein were significantly decreased in the fish after the winter mortality. These results may explain that the nutritional level of fish was decreased because fish could not fed during winter season. According to the CORI monitoring system operating by KODC, NFRDI a long term water temperature from Dec. 25, 2005 to Feb. 24, 2006 (60 days) were exposed the low water temperature environments to the red sea bream. First of all, mass mortality began at Sarayngdo where low temperature below 8℃ continued for 42 days. The winter mortality did not occured in the depths of 9 m to 19.2 m where difference of water temperature in the surface and bottom was only within 0.1℃. But in the depth of 7.5 m to 11 m winter mortality occured where water temperature in the surface and bottom showed much variation ranged from 0.2℃ to 1.4℃. From these results, great difference of water temperature in the surface and bottom of the culturing area might results in winter mortality of red sea bream.

급격한 수온변화 스트레스에 의한 넙치(Paralichthys olivaceus)의 생리학적 변화

  • 본 연구에서는 넙치(Paralichthys olivaceus)를 이용한 고ㆍ저온에서의 혈액 생리학적인 변화뿐만 아니라 항산화 효소인 CAT 및 SOD등의 효소 변화를 조사하여 스트레스에 대한 생리적인 변화에 대한 지표를 설정하고자 실시하였다. 실험어는 평균 전장의 29.0$\pm$1.5 cm, 평균 중량 246.5$\pm$25 g인 것을 사용하였다. 실험구는 각각 15$^{\circ}C$, 2$0^{\circ}C$(대조군), $25^{\circ}C$ 및 3$0^{\circ}C$로 설정하였으며, 실험 개시 후 1, 2, 4, 6 및 8 hr후에 혈액성상을 분석하였다. 측정항목으로는 헤마토크리트(Ht)수치, 혈색소 농도(Hb), 적혈구 수(RBC), 코티졸, Catalase (CAT) 및 Superoxide dismutase (SOD)를 각각 측정하였다. 실험결과 아가미 운동은 대조구에 비해 3$0^{\circ}C$$25^{\circ}C$ 에서 유의한 증가를 나타내었고, 저온인 15$^{\circ}C$ 및 1$0^{\circ}C$에서는 감소하는 결과를 보였다. Ht수치는 3$0^{\circ}C$에서 실험 개시 후 6시간까지 높은 값을 나타내었으며, $25^{\circ}C$에서는 1 시간까지는 증가하였다가 이후 6 시간까지는 감소되었다. 그러나 저 수온인 15$^{\circ}C$ 및 1$0^{\circ}C$에 서는 전체적으로 감소된 값을 나타내었다. RBC는 3$0^{\circ}C$$25^{\circ}C$에서 전반적으로 증가하였으며, 저 수온인 15$^{\circ}C$는 4 시간까지 낮아지다가 이후 증가 되었다. Hb농도는 3$0^{\circ}C$$25^{\circ}C$에서 전반적으로 증가하는 경향을 보였으나 4 시간째 일시적으로 감소하는 경향을 보였다. 15$^{\circ}C$에서는 4시간째까지는 감소되었다가 6 시간째에는 높아지는 경향을 보였다. 이러한 증가된 경향은 온도가 높아질수록 용존 산소의 포화도가 낮아져 산소부족 요인에 의해 적혈구수의 증가와 함께 Ht수치도 높아지며 아가미운동을 통한 혈액으로의 산소운반이 능동적으로 이루어지고 있음을 나타내고 있다. 혈장 코티졸의 농도는 $25^{\circ}C$에서는 증가하는 경향을 보였으며, 1$0^{\circ}C$및 3$0^{\circ}C$에서는 1 시간까지 증가되었다가 회복되는 경향을 나타내었다. 신장에서의 SOD활성은 $25^{\circ}C$인 경우 대조구보다 높은 값을 유지하였다. 하지만, 30도에서는 4 시간째까지 증가하는 경향을 보였으나 이후 감소하는 경향을 보였으며, 저온인 1$0^{\circ}C$에서는 감소하였다. 간장에서의 CAT의 활성은 3$0^{\circ}C$인 경우 4시간까지 $25^{\circ}C$ 및 15$^{\circ}C$에서는 2 시간까지 증가하였다가 이후 낮아지는 경향을 보였다. 이상의 결과로 고ㆍ저 수온변화에 따른 스트레스가 장기화될 경우 어체의 건강상에 문제점의 발생하여 대량 폐사 및 질병발생의 야기될 것으로 사료된다.

Effect of Mahwang (Ephedrae herba) extract on serum Biological Values and Blood cell Composition (마황추출액이 혈액의 생물학적 수치 및 혈구 구성에 미치는 영향)

  • Effects of mahwang(Ephedrae herba) extract on plasma lipid composition, GOT, GPT activity and blood cell composition were investigated in rats. Concentrations of FFA was not significantly different in all treatment groups. Concentrations of triglyceride and total cholesterol in plasma were decreased in mahwang(Ephedrae herba) extract groups. However concentrations of HDL-cholesterol and LDL-cholesterol showed no significant difference in all treatment groups. GOT activities were increased in mahwang(Ephedrae herba) extract 300mg/kg group. However the values of GPT showed no significant difference in all treatment groups. Counts of RBC were increased in mahwang(Ephedrae herba) extract 300mg/kg group. Counts of WBC showed no significant difference in all treatment groups. In WBC composition, neutrophils were increased, but other cells were not affected by mahwang extract treatment.

Alterations in Hematological Parameters and Antioxidant Responses in the Biofloc-reared Flatfish Paralichthys olivaceus Following Ammonia Exposure (암모니아 급성노출에 의한 바이오플락 사육 넙치(Paralichthys olivaceus)의 혈액학적 성상 및 항산화반응의 변화)

  • Flatfish Paralichthys olivaceus raised in biofloc system (mean length $27.6{\pm}3.1cm$, mean weight $280.4{\pm}26.5g$) were exposed for to different concentrations of ammonia (0, 8, 16, 32, and $64mg\;{NH_4}^+/L$) for 7 days. Following ammonia exposure, hematological parameters such as hemoglobin and hematocrit were significantly reduced. Plasma components such as magnesium, glucose, aspartate aminotransferase (AST), and alanine aminotransaminase (ALT) were significantly altered by ammonia exposure, whereas there were no significant changes in calcium, cholesterol, or total protein. Antioxidant responses, such as superoxide dismutase (SOD) and catalase (CAT) levels were significantly elevated following ammonia exposure. The results of this study indicate that ammonia exposure induces significant changes in hematological parameters and antioxidant responses in biofloc-reared Paralichthys olivaceus as a toxic response.

Alterations of Hematological Parameters, Plasma Constituents and Antioxidant Responses in the Sablefish Anoplopoma fimbria Depending on Salinity (염분농도에 따른 치어기 은대구(Anoplopoma fimbria)의 혈액학적 성상, 혈장성분 및 항산화반응의 변화)

  • Juvenile Anoplopoma fimbria (mean length $15.6{\pm}1.4cm$, mean weight $68.7{\pm}4.3g$) were exposed to 4 months with the different levels of salinity [100 (35.0), 90 (31.5), 80 (28.0), 70 (24.5), 60 (21.0), 50 (17.5), and 40 (14.0) % (psu)] for 4 months. Hematological parameters such as red blood cell (RBC) counts, hematocrit (Ht), and hemoglobin (Hb) concentrations were substantially decreased under salinities of 50% psu or lower. Of the measured inorganic plasma constituents, magnesium was notably decreased, whereas there was no effect on calcium. Among organic plasma components, glucose and cholesterol were significantly increased, and total protein was decreased. Among enzyme plasma components, glutamic oxalate transaminase (GOT), glutamic pyruvate transaminase (GPT), and alkaline phosphatase (ALP) were significantly increased under salinities of 50% psu or lower. Antioxidant responses such as glutathione S-transferase (GST) and glutathione (GSH) were significantly decreased at salinities of 50% psu or lower. The results of this study indicate that salinity affects the blood parameters, plasma constituents, and antioxidant responses of A. fimbria.

Toxic Effects of Waterborne Copper Exposure on the Hematological Parameters and Plasma Components of Mirror Carp Cyprinus carpio nudus (수인성 구리 급성노출에 의한 향어(Cyprinus carpio nudus)의 혈액학적 성상 및 혈장성분의 독성영향)

  • Mirror carp Cyprinus carpio nudus (weight 42.0±3.8 g, length 14.3±0.4 cm) were exposed to different concentrations of waterborne copper (0, 100, 200, 400, 800, and 1,600 ㎍ Cu2+·L-1) at 20.3℃ for 96 h. The lethal concentration 50 of waterborne copper was 1,176.45 ㎍ Cu2+·L-1. Among hematological parameters, red blood cell count was significantly decreased, whereas there were no significant changes in the hemoglobin concentration and hematocrit value. Among the inorganic plasma components, calcium was significantly decreased following copper exposure. Conversely, organic plasma components such as glucose and total protein were significantly increased. Similarly, enzymatic components, such as aspartate transaminase, alanine transaminase, and alkaline phosphatase, were also significantly increased. These findings suggest that the copper exposure is detrimental to the survival rates and physiology of C. carpio nudus.
