이 연구는 인삼 성분 약물이 도파민 수용체에 미치는 영향을 연구하기 위한 첫 단계로써 그 기본적인 assay system을 정착 시키기 위함이 목적이었다. 우리는 이 연구를 수행하기 위하여 생후 2-4 일의 쥐를 사용하여 흑질, 선조체, 해마구, nucleus accumbens등의 뇌 부위에서 신경세포 배양을 시도하였다. 흑질로 부터 배양한 신경세포들의 경우엔 면역 세포학적방법을 써서 살펴본 결과 대부분의 신경이 도파민성 신경이나 GABA성 신경들 이었다. 또한 이들 세포들의 전기 생리학적 상태를 알아보기 위하여 흑질에서 신호 전달체계가 잘 확립된 GABA 수용체의 작용을 살펴 본 결과 이 신경세포들은 GABA-A 및 GABA-B 수용체의 발현은 물론 이온 채널에 미치는 신호 전달체계를 완전히 갖추고 있었다. 도파민 수용체의 작용을 전기 생리학적으로 연구하기 위하여 배양한 신경세포에 도파민agonist를 가해서 이온 채널에 미치는 효과를 살펴 보았다. 선조체에서 배양한 신경세포들은 Dl 과 D2 agonist에 대해서 상반되는 반응을 나타냈다. 즉 Dl agonist는 선조체 신경세포를 활성화 시켰으나 D2 agonist는 선조체 신경세포들을 억제 하였다. 한편 해마구의 CAI 과 CA3 부위로 부터 배양한 신경세포에 대한 도파민 agonists의 작용은 선조체의 신경세포에 대한도파민 agonists의 작용과는 상반되는 반응 이었다. Dl agonist는 해마구로 부터 배양한 신경세포의 활성을 억제 하였으나 D2 agonist는 이들 신경세포들의 활성을 증가 시켰다. Nucleus accumbens 에서 배양한 신경세포들은 도파민에 의해서 그 활성이 억제 되었다. 이러한 결과로 미루어 봐서 같은 도파민 수용체라도 분포되어 있는 조직에 따라서 신호전달 에 관여 하고 있는 C-단백이나 이차 전령물질이 달라서 신경세포에 대한 작용이 다르든지. 약리학적으로는 구분되지 않으나 뇌의 조직에 따라서 분포가 다른 도파민 수용체의 아그룹이 존재 한다고 생각된다.

Curcumin is a natural phenolic yellow curry spice, derived from the tumeric, which has been used for the treatment of diseases associated with oxidative stress and inflammation. Curcumin is known to have both anti-oxidative and anti-inflammatory properties. These properties can be beneficial to protect the brain from the neurodegenerative diseases. We now report the neuroprotective effects of curcumin pretreatment in primary hippocampal neurons to glutamate-induced excitotoxicity. Pretreatment of embryonic mouse hippocampal cell cultures with low does of curcumin protected neurons against glutamate-induced death, however, this neuroprotection was not correlated with the modulation of oxidative stress. Interestingly, high dose of curcumin showed the cytotoxicity in primary cultured hippocampal neurons. Immunoblot analyses showed that levels of stress response. protein HSP70 were significantly elevated in neurons exposed to low dose of curcumin, whereas levels of cleaved PARP were increased in neurons exposed to high dose of curcumin. These findings show that curcumin can modulate neuronal responses to glutamate, and suggest possible use of curcumin and related compounds in the prevention and/or treatment of neurodegenerative disorders.
Reversible brain ischemia was produced by occluding both common carotid arteries for 5 min, and the effects of aminoguanidine (AG), $DL-{\alpha}-difluoromethylornithine$ (DFMO), MK-801, and nimodipine (NM) on the ischemia induced changes of the polyamine, glutamate and acetylcholine levels in the hippocampus CA1 subfield and the specific $[^3H]\;MK-801$ binding to the hippocampus synaptosomal membranes were studied with a histological reference of the cresyl violet stained hippocampus. The basal putrescine level $(PT:\;74.4{\pm}8.8\;nM)$ showed a rapid increase (up to 1.7 fold) for 5 min of ischemia, remained significantly increased for 6 h, and then resumed the further increase to amount gradually up to about 3 fold 96 h after recirculation. However, the level of spermidine was little changed, and the spermine level showed a transient increase during ischemia followed by a sustained decrease to about 40% of the preischemic level after recirculation. The increase of PT level induced by brain ischemia was enhanced with AG or MK-801, but it was reduced by DFMO or NM. The basal glutamate level $(GT:\;0.90{\pm}0.l4\;{\mu}M)$ rapidly increased to a peak level of $8.19{\pm}1.14\;{\mu}M$ within 5 min after onset of the ischemia and then decreased to the preischemic level in about 25 min after recirculation. And NM reduced the ischemia induced increase of GT level by about 25%, but AG, DFMO and MK-801 did not affect the GT increase. The basal acetylcholine level $(ACh:\;118.0{\pm}10.5\;{\mu}M)$ did little change during/after brain ischemia and was little affected by AG or NM. But DFMO and MK-801, respectively, produced the moderate decrease of ACh level. The specific $[^3H]\;MK-801$ binding to the hippocampus synaptosomal membrane was little affected by brain ischemia for 5 min. The control value (78.9 fmole/mg protein) was moderately decreased by AG and MK-801, respectively but was little changed by DFMO or NM. The microscopic findings of the brains extirpated on day 7 after ischemia showed severe neuronal damage of the hippocampus, particularly CA1 subfield. NM and AG moderately attenuated the delayed neuronal damage, and DFMO, on the contrary, aggravated the ischemia induced damage. However, MK-801 did not protect the hippocampus from ischemic damage. These results suggest that unlike to the mode of anti-ischemic action of NM, AG might protect the hippocampus from ischemic injury as being negatively regulatory on the N-methyl-D-aspartate (NMDA) receptor function in the hippocampus.

목적 : 뇌의 신경세포 증식은 해마 치상회와 뇌실하영역에서만 나타나는 현상이다. Kainic acid(KA)를 이용한 간질 동물모델을 연구하던 중 침이 해마 치상회의 신경세포증식을 촉진하는 현상을 발견하여 이를 보고하고자 한다. 방법 : 수컷 ICR계 생쥐를 Saline(n=8), KA(n=8), KA+Acu(n=8)의 세 군으로 나누고, 모든 생쥐들에게 KA 주입 3일 전부터 1일 1회씩 5'-bromodeoxyuridine(BrdU)을 3일간 주입하였다. Saline군에는 멸균된 생리식염수를 뇌실 내에 주입하였고, KA군 및 KA+Acu군에는 $0.1{\mu}g$의 KA를 뇌실 내에 주입하였으며, KA+Acu군에 속한 쥐들에게는 KA 주입 2일전, 1일전, 주입 직후에 양쪽 소부(少府)(HT8)에 자침하였다. KA 주입 3시간 후 쥐의 뇌를 적출하고 해마 치상회부위의 BrdU 및 neuropeptide Y (NPY)의 발현을 측정하였다. 결과 : 소부(少府) 자침이 KA의 독성으로 인한 신경세포의 파괴를 줄여주었으며, BrdU 양성 세포 및 NPY를 유의하게 증가시켰다. KA 주입시 세포증식이 일어나긴 하나, 3시간 안에는 거의 일어나지 않는다. 결론 : 소부(少府) 자침이 해마 치상회의 신경세포증식을 촉진하며, 이는 KA의 효과가 아닌 KA 투여 전 소부(少府) 자침으로 인한 것으로 사료된다.

The current study was conducted to evaluate beneficial effects of a new formula (CWC-9) using four traditional Oriental medicinal herbs, Cynanchum wilfordii, Rehmannia glutinosa, Polygala tenuifolia, and Acorus gramineus, on hippocampal cells and memory function. To examine the neuroprotective effects of a new four-herb extract, cell viability, cytotoxicity, and reactive oxygen species (ROS) assays were performed in HT22 cells and behavioral tests (Morris water maze and passive avoidance retention), Western blot, and immunohistochemistry were performed in a mouse model of focal cerebral ischemia. In HT22 hippocampal cells, pretreatment with CWC-9 resulted in significantly reduced glutamate-induced cell death with suppression of ROS accumulation caused by glutamate. In a mouse model of focal cerebral ischemia, we observed significant improvement of spatial and short-term memory function by treatment with CWC-9. Phosphorylated p38 mitogen-activated protein kinases (MAPK) in hippocampus of ischemic mice were decreased by treatment with CWC-9, but phosphorylated phosphatidylinositol-3 kinase (PI3K) and cAMP response element binding protein (CREB) were significantly enhanced. By immunohistochemical analysis, we confirmed higher expression of phosphorylation of CREB in the hippocampal neurons of CWC-9 treated mice. These results suggest that new multi-herb formula CWC-9 mainly exerted beneficial effects on cognitive function through regulation of neuro-protective signaling pathways associated with CREB.
목적:임신 중 소음 스트레스가 태아에 어떤 영향을 미치는 가를 관찰하고자, 임신 중인 흰쥐에 소음 스트레스를 가한 후 출생한 새끼 흰쥐에 인삼을 투여하여 신경적 발육에 대한, 특히 새끼 흰쥐 해마의 c-Fos 관점에서 연구하였다. 결과:CIA 영역의 c-Fos-positive 세포의수는, 출생 전 소음 스트레스를 받은 대조군에 비해, 10 mg/kg의 인삼을 투여한 군에서 유의성 있는 감소를 보였다. 해마의 CA2, CA3 영역에서의 c-Fos-positive 세포의 수는 10 mg/kg의 인삼을 투여한 그룹에서, 출생 전 소음 스트레스를 받은 대조군에 비해 유의성 있는 감소를 보였다. 해마의 dentate gyrus 영역에서 c-Fos-positive 세포의 수는, 10 mg/kg의 인삼을 투여한 그룹에서, 출생 전 소음 스트레스를 받은 대조군에 비해 유의성 있는 감소를 보였다. 결론: 인삼은 태아기 소음 스트레스로 야기되는 c-Fos의 변화로 인한 해마의 기능장애에 중화제로서 사용될 수 있다. 그리고 임신 중 소음 스트레스를 경험한 엄마의 아이들에 있어서 정신과적 문제를 치료하는데 유용할 것이다.
Deep seawater (DSW) generally refers to seawater at depths equal to or greater than 200 meters. DSW is rich in inorganic materials which have attracted attention for its various applications. In this study we investigated the effects of the DSW upwelled from the East Sea, offshore Yang Yang (KangWon-do, Korea), on the expression of ${\mu}$-opioid receptor (MOR) of cultured rat hippocampal neurons. Neurons were grown in a minimal essential medium containing 10% (v/v) fetal bovine serum and either 25% (v/v) distilled water, or hardness (H) 800, or H 1000 DSW. Cultures grown in the presence of DSW with H 800 and H 1000 exhibited robust MOR immunoreactive signals in both neurons and astrocytes. Interestingly, the increase in MOR immunoreactive signals was more dramatic in astrocytes than in neurons. Statistical analysis revealed that the relative intensities for MOR clusters increased approximately 4-fold in astrocytes cultured in H 800 and H 1000 media. These increases were statistically very significant (p<0.001). In contrast, the increase in intensities for MOR immunoreactive signals was relatively less dramatic in neurons, where only the increase in the H 1000 culture was statistically very significant (p<0.001). These results indicated that DSW promotes expression of MOR in both neurons and astrocytes, and more significantly in the latter.
Local protein synthesis in neuronal dendrites is important for site-specific regulation of synaptic plasticity. In this study, we investigated whether translation initiation factors (eIFs) are present at the postsynaptic sites. High resolution confocal microscopy showed that the eIF4E and eIF4G (which bind the 5'-terminal mRNA cap), eIF5 (which is important during the 3' direction scanning to find an initiation codon), eIF6 (which mediates upregulation of translation by external stimuli), and eIF5A (which mediate translation upregulation under adverse conditions) were localized to the post-synaptic sites. Immunoblot and detergent extraction experiments also indicated that these eIFs were present in the synapse in association with the postsynaptic density (PSD). Our data provide evidence for the strategic positioning of eIFs at the postsynaptic site for initiation of translation in diverse situations.

Deep seawater (DSW) refers to water extracted from the ocean, usually at depths of 200 meters or more, which is rich in inorganic materials and has attracted attention for various applications. We investigated the effects of the DSW on the synaptic maturation of cultured rat hippocampal neurons. Immunocytochemical examination of DIV21 showed that PSD-95, $\alpha$CaMKII, and synGAP$\alpha1$clusters were strengthened and coupling rates of SV2 and NR2B were significantly increased in neurons grown in the presence of H-800 and H-1000 DSW. Our results indicate that DSW promotes the formation of excitatory postsynaptic signal transduction complexes NRC/MASC and functional synapses.
This study investigated the effects of treadmill exercise on memory ability, cell proliferation, BDNF, and TrkB in the hippocampus and forebrain cholinergic cells in adolescent rats. Male Sprague-Dawley rats (4 weeks old) were randomly assigned to the following two groups: the sedentary group (n=10) and the exercise group (n=10). Rats in the exercise group were forced to run on a treadmill for 30 min, five times per week for 4 weeks. The latency of the step-through avoidance task was used in order to evaluate memory ability. Hippocampal brain-derived neurotrophic factor (BDNF) and tropomyosin-related kinase B (TrkB) expression were assessed by Western blotting. Hippocampal cell proliferation and forebrain cholinergic cells were assessed by immunohistochemistry. The present study showed that treadmill running during the adolescent period significantly improved memory capability, increased hippocampal cell proliferation, up-regulated hippocampal BDNF and TrkB expression, and enhanced the number of forebrain cholinergic cells. These results suggest that regular exercise during the adolescent period may enhance memory function.
