In this study, the antibacterial, antioxidative and inhibitory effects of Allium cepa peel extracts on tyrosinase and elastase were investigated. MIC values of the ethyl acetate fraction of Allium cepa peel on especially, S. aureus among the skin resident flora (Staphylococcus aureus, S. aureus; Propionibacterium acnes, P. acnes; Pityrosporum ovale, P. ovale; Escherichia coli, E. coli) were 0.06%. The aglycone fraction showed more excellent free radical (1,1-diphenyl-2-picrylhydrazyl radical, DPPH) scavenging activity ($FSC_{50}=5.05{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of the ethyl acetate fraction and aglycone fraction in the luminol-dependent $Fe^{3+}-EDTA/H_2O_2$ system were 0.05 and $0.03{\mu}g/mL$, respectively. The cellular protective effect of the aglycone fraction on the rose-bengal sensitized photohemolysis of human erythrocytes exhibited more prominent (${\tau}_{50}$, 480 min at $25{\mu}g/mL$). The inhibitory effects ($IC_{50}$) of the ethyl acetate fraction and aglycone fraction on tyrosinase were 9.16 and $8.68{\mu}g/mL$, the inhibitory effect ($IC_{50}$) of the aglycone fraction on elastase was $14.12{\mu}g/mL$ The transepidermal water loss of the cream containing 0.1% ethyl acetate fraction was decreased from $8.3g/m^2h$ in control to $6.8g/m^2h$ in the subjects applied with cream containing the ethyl acetate fraction. These results indicate that extract/fractions of Allium cepa peel can function as antioxidant in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS, and possibly as antiaging agents. Allium cepa peel extract could be used as a new cosmeceutical for whitening and anti-wrinkle products.

This study was carried out to investigate the anti-oxidative and anti-inflammatory actions of genistein in BALB/c mice injected with lopopolysaccharide (LPS), called endotoxin. Mice (10 weeks of age) weighing approximately 20 g were divided into 4 groups. Endotoxin shock was induced by intraperitoneal injection of LPS (100 mg/kg BW). LPS and genistein+LPS groups were injected with LPS 30 min after phosphate buffered saline (PBS) solution and genistein (200 mg/kg BW) injections, respectively. Genistein group was injected with genistein, followed by PBS, while PBS group received two injections of PBS. Superoxide anion generation of peritoneal macrophage cells was significantly (p<0.05) lower in the genistein+LPS group than in the LPS injection group at 8 h after intraperitoneal injection, while SOD activity was significantly higher in genistien+LPS group than LPS group. Tumor necrosis factor-$\alpha$ levels of plasma were significant lower (p<0.05) in the genistein+LPS injection group than LPS group at 8 h after intraperitoneal injection. Plasma TBARS was lower in genistein+LPS group than LPS group, while hepatic TBARS were not different among groups. Hepatic glutathione concentrations and antioxidant enzyme activities were ignificantly higher in the genistein+LPS group than in the LPS group at 1 h and 8 h after intraperitoneal injection. Nuclear factor-kappa B (NF-${\kappa}B$) transactivation was significantly (p<0.05) inhibited in LPS group. These results demonstrate genistein may ameliorate inflammatory diseases through inhibition of NF-${\kappa}B$ transactivation and oxidative stress, which may be mediated partially by anti-oxidative effect of genistein.
This study was aimed to investigate that training of horses is related with the activity of superoxide dismutase(SOD) in erythrocyte of racing horses. The SOD activity was assayed from erythrocyte of six Thoroubred horses having final stage of training, about 21 month-old, 474~509 kg body weight for race trainig. During 7 weeks of training period from 24th Sep. to 6th Nov, horses were bled very carefully 4 times at 1st Oct, 16th Oct, 30th Oct. and 6th Nov. As the training period passed, erythrocyte of the horses have gradually increased the MnSOD activity(p<0.05) and lowered the CuZnSOD activity. The plasma ceruloplasmin and peroxidase activities, and lactate levels were reduced gradually while peroxide and glucose levels gradually increased. The calculated oxygen consumption(Eaton, 1995) for training of horses were linearly related with the MnSOD activity(r=0.650, n=32) but negatively with CuZnSOD activity in erythrocyte and lactate levels(r=-349, n=32) in plasma. Also, peroxide levels in plasma of horses had positive relation with the MnSOD activity in erythrocyte(r=0.616, n=48). In conclusions, as the training is progressed, the raised MnSOD activity in erythrocytes and peroxide levels in plasma indicated balances between oxidant and antioxidants for the protection from ROS during race of horses. The results showed that the MnSOD activity in erythrocyte and peroxide levels in plasma may be used as marker for the intensity of training racing horses.

Cucumber fermentation has been used as a means of preservation. This study was performed to investigate the effects of fermented cucumber beverage (CF) containing beneficial materials for an ethanol hangover based on Hovenia dulcis (SKM) on ethanol-induced hepatotoxicity. Male Sprague-Dawley rats were randomly divided into three groups: ethanol control, ethanol plus SKM, and ethanol plus CF+SKM. SKM or CF+SKM was orally administered at a dose of 7 mL/kg body weight once per day for 5 weeks. Control rats were given an equal amount of water. CF+SKM significantly lowered plasma ethanol levels, whereas SKM tended to decrease the levels compared to the control. Both SKM and CF+SKM significantly lowered the plasma acetaldehyde levels and serum transaminase activities compared to those in the control. SKM and CF+SKM did not affect hepatic alcohol dehydrogenase activity; however, it significantly inhibited cytochrome P450 2E1 (CYP2E1) activity. Hepatic aldehyde dehydrogenase (ALDH) activity was significantly higher in the SKM and CF+SKM groups than that in the control group. Plasma acetaldehyde concentration was significantly correlated with hepatic CYP2E1 (r=0.566, p<0.01) activity and ALDH (r=-0.564, p<0.01) activity. Hepatic superoxide dismutase and catalase activities as well as glutathione content increased with the SKM and CF+SKM administration, whereas lipid peroxide content decreased significantly. Furthermore, SKM and CF+SKM lowered plasma and hepatic lipid content and lipid droplets compared to those in the control group. These results indicate that SKM and CF+SKM exhibit hepatoprotective properties partly by inhibiting CYP2E1 activity, enhancing ALDH activity and stimulating the antioxidant defense systems in ethanol-treated rats.

This study was designed to examine the effect of sea buckthorn (SBT) leaves on hepatic antioxidative enzyme levels in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats by an injection of streptozotocin (STZ). Sprague-Dawley rats were then fed for four weeks, with experimental groups receiving a modified diet containing 10% or 20% powder derived from SBT leaves. The experimental groups were divided into six groups: a normal (N)-control group, N-SBT 10% and N-SBT 20% treated groups, STZ-control, STZ-SBT 10% and STZ-SBT 20% treated groups. Malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX), glutathione reductase (GR), glutathione-S-transferase (GST) and xanthine oxidase (XOD) levels were measured in liver cytosol. The results showed that the level of SOD was significantly increased in the N-SBT 20% group but not statistically different in the diabetic group. The level of CAT was significantly higher in the N-SBT 20% group compared to the control group. The level of GPX was significantly increased in the N-SBT 20% group and the diabetic supplementary group. In contrast, the level of XOD was significantly decreased in the diabetic group supplemented with SBT leaves.
This review showed a discussion on the biofunctional activities of citrus flavonoids. The major flavonoids of citrus species, hesperidin, hesperetin, naringin, and naringenin, were selected to evaluate their biological effects on the lipid metabolism in rats and hamsters, the proliferation of human hepatocyte HepG2 cells, and the antioxidative effect in lipid peroxidation models. These flavonoids showed hypotriglyceridemic effect in hamsters and hypochloesterolemic effect in rats. They also significantly inhibited the activities of phosphatidate phophohydrolase and acyl-CoA: cholesterol acyltransferase, which are key enzymes for biosynthesis of triglyceride and cholesterol, repectively, in vivo and in vitro experiments. These biofunctional activities by citrus flavonoids were shown more potent in the aglycone flavonoids, hespreretin and naringenin, than their corresponding glycoside flavonoids, hesperidin and naringin. These aglycone flavonoids also have inhibitory effects on proliferation of human hepatocyte cancer HepG2 cells. Hesperidin showed lowering activities of cellular triglyceride and cholesterol concentrations in HepG2 cells. Citrus flavonoids have significant importance in functional food industry as biofunctional active ingredients.
In this study, we produced bioethanol from the original hydrolysate obtained during oxalic acid pretreatment of lignocellulosic biomass. The bioethanol was separated and concentrated by pervaporation and the residue after pervaporation was evaluated for its antioxidant activity. Xylose ($37.28g/{\ell}$) was the major product in the original hydrolysate. The original hydrolysate contained acetic acid, furfural and total phenolic compounds (TPC) as fermentation inhibitors. Acetic acid was removed by electrodialysis (ED), and $12.21g/{\ell}$ of bioethanol was produced from ED-treated hydrolysate. The TPC of ethyl acetate extracts from the residue obtained (OA-E) during pervaporation was 86.81 mg/100 g (extract). The $IC_{50}$ values of DPPH and ABTS radical scavenging activities, and reducing power of OA-E were $0.87mg/m{\ell}$, $0.85mg/m{\ell}$, and $0.59mg/m{\ell}$, respectively. Sugar degradation products and the phenolic compounds in OA-E were determined by GC-MS.

In order to investigate the cellular peroxidative damage due to heated oil intake and the preventive effect of vitamin E on it rats were fed heated corn oil with acid value of 4.02 at the level of 10 Cal% and three different levels of vitamin E that were 0, 40 and 200 mg/kg diet. Control group was fed fresh corn oil and 40mg/kg diet of vitamin E. After ech feeding period of 0, 3 and 6 weeks, liver superoxide dismutase (SOD), glutathione peroxidase (GPX) activities and microsomal content of vitamin E and lipid peroxide (LPO) were measured as well as cellular morphology was examined. SOD activities and LPO contents were higher, while GPX activities and vitamin e contents were lower in heated oil groups than control group. Electromicroscopic observation revealed the loss of inner mitochondrial membrane and cristae and irregular arrangement of nuclear membrane and chromatin in heated oil groups. As dietary vitamin e level was increased, SOD activity and LPO content were decreased, but GPX activity and vitamin E content in the liver increased and cellular peroxidative damage reduced progressively. This phenomena was more remarkable in 6 weeks of feeding than 3 weeks.

This study was performed to determine the effects of pretreatment and drying methods on the quality and physiological activities of garlic powders. Fresh garlic was boiled, baked, steamed or left untreated before hot-air drying or freeze-drying. Electron-donating abilities were higher in the untreated group for each drying method. Relative EDA of baked group was 32.8%, whereas those of the boiling and steaming groups were 12.4% and 20.1%, respectively. Nitrite-scavenging effects were slightly affected by processing methods. Polyphenol content of the processing group was higher in the hot-air drying than freeze-drying group, and it was significantly highest in the untreated freeze-dried group. The value of polyphenol in the baked group was significantly higher than those of the boiling and steaming groups. Tyrosine contents of garlic powder were higher in the hot-air drying than freeze-drying group, and it was significantly highest in the untreated group of each drying method. Antimicrobial activities of the garlic powders were higher in freeze-drying group compared to the hot-air drying group. Therefore, antimicrobial activities of garlic powder were affected by heating treatments.

The effects of methanol extract of barely (Hordeum vulgare) on alcohol-induced damages of liver were investigated in male ICR mice. Mice were divided into three groups, control, ethanol, and ethanol plus 0.15% of barley extract. After four weeks of ethanol feeding, ethanol group significantly increased the P450 content, CYP2E1 and CYP1A2 enzyme activities, whereas ethanol plus barely group markedly decreased to levels similar to control group. Catalase activity in ethanol group was significantly lower than that in control group; however, ethanol plus barely group stimulated catalase activity as well as SOD activity significantly. These results indicated that barely extract modulated P450 enzymes for ethanol-induced liver damage and might be useful in developing functional food for alcoholic liver damage.
