Effects of gradual loaded exercise on antioxidative enzymes response in normal and obese men (비만인과 정상인간 일회성 점증부하 훈련이 항산화 효소에 미치는 효과)

  • The purpose of this study was to identify the effects of acute exercise on antioxidative enzymes in noramal and obese men. The written informed consent was obtained from 16 participants of eight normally healthy persons and eight obesity ones. The baseline level of obesity was determined at 25 % of the body fat. Any subjects had no experiences in participating in exercise and did not take vitamin or mineral supplement. Blood was sampled at five minutes before exercise, at exhaustion, and after 30 minutes of recovery. All statistical analyses and description methods were computed by SPSS window 14.0. Analysis of Covariance(ANCOVA) was performed to know the differences in values between pre- and post- exercise in each group. We found that after exhaustion, the normal group showed higher erythrocyte superoxide dismutase (SOD) activity compared to the obesity group. At exhaustion, the normal group had lower HSP activity compared to the obesity group. Based on the results, an acute exercise of normally healthy people induced an up-regulation of superoxide dismutase (SOD) activity. This plays an important role in the protection of extra-cellular fluid components against peroxide-mediated damage.

Radioprotective effect of naringin and naringenin against cellular damage and oxidative stress of γ-irradiated mice (감마선을 조사한 마우스의 세포 손상과 산화적 스트레스에 대한 나린진과 나린제닌의 방사선방호 효과)

  • The present study was designed to evaluate the antioxidant activity and radioprotective effects of Naringin and Naringenin in ${\gamma}$-irradiated mice. The antioxidant activity of Naringin and Naringenin was evaluated by 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and ferric reducing antioxidant power (FRAP) assays. Healthy female BALB/c mice were administered Naringin and Naringenin orally ($90{\mu}M/dose$ and $180{\mu}M/dose$) for 7 consecutive days prior to ${\gamma}$-irradiation (6 Gy). Naringenin displayed a much higher antioxidant activity in ABTS and FRAP than naringin. ${\gamma}$-irradiation resulted in cellular damage with decreased spleen and thymus indices and white blood cells (WBC) count. Additionally, ${\gamma}$-irradiation significantly increased lipid peroxidation and decreased the levels of antioxidant enzymes and glutathione (GSH) in the liver tissue. Strikingly, prior administration of Naringenin resulted in considerable prevention of these symptoms. Protection against ${\gamma}$-irradiation-induced cellular damage by Naringenin is likely due to its higher its antioxidant activity. Together, these results confirm that Naringenin is a potent antioxidant and radioprotector.

Antioxidant Activity and Anti-inflammatory effects of Sicyos angulatus L. extract (가시박 추출물의 항산화 및 항염증 효과)

  • This study was carried out to find out the physiological activity effect of Sicyos angulatus L. extract which is known as an ecosystem disturbance plant and confirm the availability as a functional cosmetic material. Total polyphenol and flavonoid contents were measured, and DPPH radical scavenging, intracellular ROS, and its inhibitory effect on the expression Nitric oxide of and COX-2 were evaluated. The content of polyphenol and flavonoid was found to be 3.079 mg(CA)/100g and 72 mg(Q)/100g of Sicyos angulatus L. extract and antioxidant activity through high radical scavenging activity was confirmed. ~Significant cytotoxicity was not observed up to a concentration of $100{\mu}g/mL$ in RAW 264.7 cells and HDF cells and concentration-dependent inhibition of ROS production in HDF cells, inhibition of high NO production and inhibition of COX-2 protein expression in RAW 264.7 cells were confirmed. Through these results, we found the possibility of use as a functional cosmetic material with excellent antioxidant effect and antioxidant and anti-inflammatory effects through inhibition of intracellular ROS generation, inhibition of NO generation, and inhibition of COX-2 protein expression.

Physicochemical Properties and Antioxidant Activities of Maillard Reaction Products from Hydrolyzed Wheat Gluten (소맥 gluten 산가수분해물을 이용한 마이야르 반응물질의 이화학적 특성 및 항산화성)

  • Maillard reaction products (MRPs) were produced from aqueous solution of various sugars with hydrolyzed wheat gluten (HWG) with different temperatures, pressures, pH values and solvents. The physicochemical properties of MRPs were investigated and DPPH and hydroxyl radical scavenging activity and sensory properties were also assessed. MRPs from ribose and HWG evidenced the highest preference for meaty flavor and antioxidant activity and also evidenced higher antioxidant activity with larger pH reductions and higher browning index increases than were observed in other MRPs. The antioxidant activities were increased with increased reaction temperature and pressure. The most preferred meaty flavor was obtained from MRPs with ribose at 140$^{\circ}C$ in an oil bath with the pH adjusted to 9 in water as a solvent, and heated for 30 mins.

Antioxidative Activities and Whitening Effects of Ethyl Acetate Fractions from The Immature Seeds of Abeliophyllum distichum (미선나무 미성숙 종자의 항산화 및 미백 활성)

  • Abeliophyllum distichum Nakai is deciduous shrubs of flowering plant in Oleaceae. It is important plant resources and consists of one species in the world. Also the endemic plant of A. distichum has been protected and designed endangered plant in Korea. For this reason, study on the immature seeds of A. distichum (ADS) hasn't progressed. In the present study, we evaluated the antioxidant activity and inhibitory effects on proteins and mRNA levels were related in the whitening effect in B16F10 cells. ADS was effective for reaction oxygen species (ROS). ROS causes various diseases such as aging, inflammation, cancer, and etc. Antioxidant properties were evaluated DPPH, ABTS radical scavenging activity and Reducing power. Plants were known that contained phenolic compounds were related in antioxidant activity. Phenolic compounds were phytochemicals commonly named natural polyphenols. These are secondary metabolites of plants involved in the defense against different types of stresses. In results, ADS suppressed the expression and transcription of Tyrosinase, TRP-1, TRP-2, and Microphthalmia-associated transcription factor (MITF). Tyrosinase, tyrosinase-related protein 1 (TRP-1), tyrosinase-related protein 1 (TRP-2) are known to play an important role in melanin biosynthesis. MITF regulated the expression and transcription of Tyrosinase, TRP-1, and TRP-2. In conclusion, ADS was effective in both antioxidant activities and whitening effects. Also, they were associated with the content of phenolic compounds. We suggested that ADS can be use antioxidants and skin-whitening functional cosmetics material derived from natural plant resources.

The Physiological Responses of Spotted Seahorse Hippocampus kuda to Low-pH Water (사육수의 pH변화가 복해마(Hippocampus kuda)에 미치는 생리적 영향)

  • The rising concentration of atmospheric carbon dioxide is causing ocean acidification and global warming. The seahorse is an important species in marine ecosystems and fishery markets, however, their populations have recently decreased due to ocean acidification. As a result, we examined changes in the physiological responses of the spotted seahorse Hippocampus kuda when it was exposed to acidic sea water (pH 6.0, 6.5, and 7.0) and normal seawater (pH 8.0 as the control) over a period of 15 days. As the pH decreased, the seahorses' body weight and length also decreased. Components in body of ash, the crude lipids and crude proteins also differed significantly with changes in pH, due to stress caused by the seahorses' exposure to the acidic water conditions. The superoxide dismutase levels were significantly lower in the pH 6.0 and 6.5 groups than they were in the pH 7.0 and pH 8.0 groups. However, the catalase and glutathione levels were significantly higher in the acidic sea water groups. We suggest that decreasing the pH level of rearing water induces a stress response in H. kuda, damaging their ability to maintain their homeostasis and energy metabolism. Antioxidant enzymes are generally sensitive to acidic stress; in this study, the antioxidant activity was significantly affected by the pH level of the rearing water. These results indicate that physiological stress, induced by exposure to acidification, induces an antioxidant reaction, which can reduce general components in the body and the growth of H. kuda.

The Effect of Powdered Herb of Aster scaber Thunb. on Antioxidant System in Ethanol-Treated Rats (참취 분말이 에탄올을 투여한 흰쥐의 항산화계에 미치는 효과)

  • The present study was conducted to investigate effect of powdered herb of Aster scaber Thunb. (chamchwi) on antioxidant system in ethanol-administrated rats. Four week-old Sprague Dawley male rats which had initial body weights of 97.10$\pm$4.50 g were randomly divided into three groups: control (ethanol treated, vitamin E-deficient group); 5% chamchwi (ethanol-treated, 5% chamchwi powder-supplemented group): 10% chamchwi (ethanol-treated, 10% chamchwi powder-suplemented group). Three groups of rats were suplemented with three experimental diets for 4 weeks and orally administrated 10% ethanol (v/v) daily via drinking water in the last experimental week. Contents of TBARS (thiobarbituric acid reactive substance). glutathione in liver and kidney and serum albumin were determined. The activities of superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH-px) in liver and kidney were also analyzed. Relative weight of liver and spleen to body in chamchwi groups was lower than that in control group (p<0.05). The most remarkable result was that liver TBARS contents in chamchwi groups (5% chamchwi group, 46 $\mu\textrm{g}$ in MDA value; 10% chamchwi group, 35 $\mu\textrm{g}$) were significantly lower (p<0.05) than that in control group (66 $\mu\textrm{g}$). The supplement of chamchwi powder lowered the activity of manganese- superoxide dismutase (Mn-SOD), catalase in liver and GSH-px in kidney. The levels of glutathione in liver and kidney and serum albumin were not significantly different in all experimental groups (p<0.05). These results indicate that powdered herb of Aster scaber decreases lipid peroxidation and acitvity of Mn-SOD increased by alcohol-induced oxidative stress in liver of rats.

Stabilization of Polyphenolic Antioxidants Using Inclusion Complexation with Cyclodextrin and Their Utilization as the Fresh-food Preservative (폴리페놀계 천연 항산화제의 cyclodextrin inclusion complexation을 통한 안정화와 식품 보존제로의 활용)

  • Insoluble polyphenol antioxidants, quercetin and catechin, were stabilized through the complexation with cyclodextrin to increase heat and pH stabilities. Comparison of inclusion complex formabilities of quercetin and catechin with ${\alpha}-,\;{\beta}-$, and ${\gamma}-CDs$ revealed ${\beta}-CD$ to be the most suitable result. Optimal molar mixing ratio of ${\beta}-CD$ and quercetin or catechin for inclusion complex formation was found to be 1 : 1. Inclusion complexation was confirmed using differential scanning calorimetry. Solubility of ${\beta}-CD-antioxidant$ inclusion complexes increased compared with native antioxidants, Stability against temperature and pH of ${\beta}-CD-antioxidant$ inclusion complex analyzed revealed antioxidant activities of ${\beta}-CD-quercetin$ and catechin inclusion complexes have higher stabilization compare to raw quercetin and catechin. Peroxide value of linoleic acid dissolved in water decreased substantionally after using ${\beta}-CD-quercetin$ inclusion complex. ${\beta}-CD-antioxidant$ inclusion complex can be used effectively as a fresh-food preservative.

Comparison of Antioxidant Activities of Rice Bran Extracts by Different Extraction Methods (추출방법에 따른 미강 추출물의 항산화 활성 비교)

  • The objective of this study was to determine the antioxidant activities of rice bran extracts by three different extraction methods. Rice bran was extracted by solvent extraction, saponification extraction, and supercritical fluid extraction. The antioxidant activities of the rice bran extracts were determined based on ABTS and DPPH radical scavenging activities, reducing power, and lipid peroxidation inhibitory activity. The unsaponifiable matter (USM) extracted by the saponification method showed higher ABTS (671.7 mg Trolox equivalent antioxidant capacity (TEAC)/g) and DPPH (330.7 mg TEAC/g) radical scavenging activities as well as reducing power ($A_{700}=1.14$) than those of the solvent extract (ME) and supercritical fluid extract (SFE). Inhibitory effect on lipid peroxidation was higher in USM (68.7%) and SFE (75.4%) compared to ME (47.8%). USM indicated relatively higher antioxidant activities compared with those of SFE and ME. These results show that the saponification method for extraction of USM from rice bran extracted was the most effective method for enhancement of antioxidant activity. In addition, these extracts from rice bran could be used as functional ingredients in the food industry.

Radioprotective Effect of Quercetin Post-Treatment against γ-Irradiation-Induced Hepatocellular and Hematopoiectic System Damage in Mice (감마선 조사로 유도된 간세포와 조혈계 손상 마우스에서 퀘르세틴 투여 후의 방사선방호 효과)

  • This study was designed to evaluate the therapeutic effect of quercetin against radiation-induced hepatocellular and hematopoiectic damage in BALB/c mice. Mice were exposed to 6 Gy of ${\gamma}$-radiation and orally administered quercetin (25, 50 mg/kg b.w.) for 7 consecutive days. ${\gamma}$-Irradiation caused marked elevation of serum aspartate aminotransferase and alanine aminotransferase, levels as well as reduction of spleen index, thymus index, and the number of white blood cells. In addition, ${\gamma}$-irradiation induced significant elevation of lipid peroxidation as well as reduction of antioxidant enzyme activities, including superoxide dismutase, catalase, and glutathione peroxidase. However, post-treatment with quercetin resulted in a significant recovery of all of these parameters. These results suggest that quercetin acts as a potent radioprotector against irradiation-induced cellular damage in mice.
